| 3 | Frequent Mutation of rs13281615 and Its Association with PVT1 Expression and Cell Proliferation in Breast Cancer显示文摘The q24 band of chromosome 8(8q24)is frequently amplified in human cancers including breast cancer,and several SNPs(single nucleotide polymorphisms)at 8q24,including rs13281615,have been identified for their association with cancer risks.These SNPs are in a'gene desert'region,and their functions in cancer development remain to be illustrated,although several of the SNPs appear to influence the genes in the'desert'in a long-range manner,including the v-myc avian myelocytomatosis viral oncogene homolog(MYC)and the nonprotein coding plasmacytoma variant translocation 1(PVT1),both of which have been implicated in human cancers.In the current study,we examined rs13281615 for its potential role in breast cancer using normal and cancer tissues from 121 Chinese women with breast cancer.In addition to confirming the association of the GG genotype of rs13281615 with breast cancer risk,we found that germline GG genotype was significantly associated with estrogen receptor(ER)positivity,higher tumor grade and higher proliferation index.We also found frequent somatic mutations(22/121 or 18.2%)of this SNP in breast cancer.Interestingly,the majority of the mutations(17/22 or 77%)involved a G/A change,resulting in a decrease in the number of cancers with the GG risk genotype and subsequent loss of GG association with higher tumor grade and proliferation index in cancers.Furthermore,PVT1 expression was increased in cancers,and the increase was associated with the GG genotype of rs13281615.These results suggest that the GG genotype of SNP rs13281615 plays a role in breast cancer likely by influencing PVT1 expression,and that during oncogenesis,'protective'mutations could occur. | Zhiqian Zhang Zhengmao Zhu Baotong Zhang Weidong Li Xin Li Xiao Wu Lijuan Wang Liya Fu Li Fu Jin-Tang Dong | 2014 | Journal of Genetics and Genomics2014,41,4: | 12 |
| 4 | Counteraction of ABA-Mediated Inhibition of Seed Germination and Seedling Establishment by ABA Signaling Terminator in Arabidopsis显示文摘Seed germination and seedling establishment are important for the reproductive success of plants,but seeds and seedlings typically encounter constantly changing environmental conditions.By inhibiting seed germination and post-germinative growth through the PYR1/PYL/RCAR ABA receptors and PP2C co-receptors,the phytohormone abscisic acid(ABA)prevents premature germination and seedling growth under unfavorable conditions.However,little is known about how the ABA-mediated inhibition of seed germination and seedling establishment is thwarted.Here,we report that ABA Signaling Terminator(ABT),a WD40 protein,efficiently switches off ABA signaling and is critical for seed germination and seedling establishment.ABT is induced by ABA in a PYR1/PYL/RCAR-PP2C-dependent manner.Overexpression of ABT promotes seed germination and seedling greening in the presence of ABA,whereas knockout of ABT has the opposite effect.We found that ABT interacts with the PYR1/PYL/RCAR and PP2C proteins,interferes with the interaction between PYR1/PYL4 and ABI1/ABI2,and hampers the inhibition of ABI1/ABI2 by ABA-bound PYR1/PYL4,thereby terminating ABA signaling.Taken together,our results reveal a core mechanism of ABA signaling termination that is critical for seed germination and seedling establishment in Arabidopsis. | Zhijuan Wang Ziyin Ren Chunhong Cheng Tao Wang Hongtao Ji Yang Zhao Zhiping Deng Liya Zhi Jingjing Lu Xinying Wu Shimin Xu Mengmeng Cao Hongtao Zhao Liu Liu Jiankang Zhu Xia Li | 2020 | Molecular Plant2020,13,9: | 10 |
| 5 | Autologous hematopoietic stem cell transplantation in chemotherapy-sensitive lymphoblastic lymphoma: treatment outcome and prognostic factor analysis显示文摘Objective: The study evaluated the effectiveness of autologous hematopoietic stem cell transplantation(AHSCT) in the treatment of lymphoblastic lymphoma(LL).Methods: We retrospectively analyzed the data from 41 patients with chemotherapy-sensitive LL who underwent hematopoietic stem cell transplantation(HSCT) from December 1989 to December 2009 in a single institution.Results: HSCT was conducted as first-line consolidation therapy and salvage therapy in 36 and 5 patients, respectively. The median follow-up was 97.1 months(range, 24.6-173.1 months). The 5-year overall survival(OS) and event-free survival(EFS) rate were 64% and 47% for the initially treated patients, respectively, and were both 20% for the relapsed ones. | 7 | Methylesterification of cell-wall pectin controls the diurnal flower-opening times in rice显示文摘Flowers are the core reproductive organ of plants, and flowering is essential for cross-pollination. Diurnal flower-opening time is thus a key trait influencing reproductive isolation, hybrid breeding, and thermostability in plants. However, the molecular mechanisms controlling this trait remain unknown. Here, we report that rice Diurnal Flower Opening Time 1 (DFOT1) modulates pectin methylesterase (PME) activity to regulate pectin methylesterification levels of the lodicule cell walls, which affect lodicule swelling to control diurnal flower-opening time. DFOT1 is specifically expressed in the lodicules, and its expression gradually increases with the approach to flowering but decreases with flowering. Importantly, a knockout of DFOT1 showed earlier diurnal flower opening. We demonstrate that DFOT1 interacts directly with multiple PMEs to promote their activity. Knockout of PME40 also resulted in early diurnal flower opening, whereas overexpression of PME42 delayed diurnal flower opening. Lower PME activity was observed to be associated with higher levels of pectin methylesterification and the softening of cell walls in lodicules, which contribute to the absorption of water by lodicules and cause them to swell, thus promoting early diurnal flower opening. Higher PME activity had the opposite effect. | 16 | A tailored and rapid approach for ozonation catalyst design显示文摘Catalytic ozonation is widely employed in advanced wastewater treatment owing to its high mineralization of refractory organics.The key to high mineralization is the compatibility between catalyst formulation and wastewater quality.Machine learning can greatly improve experimental efficiency,while fluorescence data can provide additional wastewater quality information on the composition and concentration of organics,which is conducive to optimizing catalyst formulation.In this study,machine learning combined with fluorescence spectroscopy was applied to develop ozonation catalysts(Mn/g-Al_(2)O_(3)catalyst was used as an example).Based on the data collected from 52 different catalysts,a machine-learning model was established to predict catalyst performance.The correlation coefficient between the experimental and model-predicted values was 0.9659,demonstrating the robustness and good generalization ability of the model.The range of the catalyst formulations was preliminarily screened by fluorescence spectroscopy.When the wastewater was dominated by tryptophan-like and soluble microbial products,the impregnation concentration and time of Mn(NO_(3))_(2) were less than 0.3 mol L^(-1)and 10 h,respectively.Furthermore,the optimized Mn/g-Al_(2)O_(3)formulation obtained by the model was impregnation with 0.155 mol L^(-1)Mn(NO_(3))_(2)solution for 8.5 h and calcination at 600℃ for 3.5 h.The model-predicted and experimental values for total organic carbon removal were 54.48% and 53.96%,respectively.Finally,the improved catalytic performance was attributed to the synergistic effect of oxidation(·OH and ^(1)O_(2))and the Mn/g-Al_(2)O_(3) catalyst.This study provides a rapid approach to catalyst design based on the characteristics of wastewater quality using machine learning combined with fluorescence spectroscopy. | Min Li Liya Fu Liyan Deng Yingming Hu Yue Yuan Changyong Wu | 2023 | Environmental Science and Ecotechnology2023,,3: | 0 |
