| 1 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 2 | The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. | 3 | Effect of a starch-based filter aid on the dewatering of fine clean coal显示文摘The dewatering of fine, flotation cleaned coals from Huaibei and Xuzhou (bituminous) and Yongcheng (anthracite) were studied. The supernatant and filter cake were examined to determine the rate and extent of flocculation and dewatering. A starch-based filter aid was used to increase flocculation and dewatering rates. The filtration constant, K, and compression index, s, of the Yongcheng slurry were measured under various conditions. A designed experiment was performed to determine optimum conditions for dewatering. The results showed that the filter aid enhanced flocculation and coagulation of the fine cleaned coal slurry, enhanced the structure of the filter cake and promoted dewatering of the cake. | 4 | Methylesterification of cell-wall pectin controls the diurnal flower-opening times in rice显示文摘Flowers are the core reproductive organ of plants, and flowering is essential for cross-pollination. Diurnal flower-opening time is thus a key trait influencing reproductive isolation, hybrid breeding, and thermostability in plants. However, the molecular mechanisms controlling this trait remain unknown. Here, we report that rice Diurnal Flower Opening Time 1 (DFOT1) modulates pectin methylesterase (PME) activity to regulate pectin methylesterification levels of the lodicule cell walls, which affect lodicule swelling to control diurnal flower-opening time. DFOT1 is specifically expressed in the lodicules, and its expression gradually increases with the approach to flowering but decreases with flowering. Importantly, a knockout of DFOT1 showed earlier diurnal flower opening. We demonstrate that DFOT1 interacts directly with multiple PMEs to promote their activity. Knockout of PME40 also resulted in early diurnal flower opening, whereas overexpression of PME42 delayed diurnal flower opening. Lower PME activity was observed to be associated with higher levels of pectin methylesterification and the softening of cell walls in lodicules, which contribute to the absorption of water by lodicules and cause them to swell, thus promoting early diurnal flower opening. Higher PME activity had the opposite effect. | 5 | Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. | 6 | Identifying genomic regions controlling ratoon stunting disease resistance in sugarcane(Saccharum spp.) clonal F_(1) population显示文摘The ratoon stunting disease (RSD) of sugarcane,caused by the bacterium Leifsonia xyli subsp.xyli,is one of the major concerns to sugarcane production and breeding programs worldwide.Due to no obvious external symptoms,RSD cannot be easily detected by the growers,hence has reduced the world’s sugarcane production significantly.This study aimed to identify quantitative trait loci (QTL) associated with RSD resistance and to assist in the development of linked molecular markers for marker-assisted breeding to minimize the reduction in sugarcane yield by the RSD infection.A set of 146 individuals derived from a self-crossing of CP80-1827 were evaluated for RSD resistance in a mechanically duplicated inoculated field trial from 2014 to 2017 using tissue blot immunoassay.Leveraging the genetic data and the four years phenotyping data of CP80-1827 selfing population,linkage map construction and QTL analysis were conducted based on clonal F_1 and F_2 mapping population types with GACD V.1.2 and Ici Mapping V.3.3,respectively.A total of 23 QTL associated with RSD resistance were identified,which explained 6%to13%of the phenotypic variation with the two types of software.A total of 82 disease resistance genes were identified by searching these 23 QTL regions on their corresponding regions on the Sorghum bicolor genome (44 genes),sugarcane R570 genome (20 genes),and S.spontaneum genome (18 genes),respectively.Compared with Ici Mapping V.3.3,GACD V.1.2 identified more major (6 vs.3) and stable QTL (2vs.0),and more disease resistance genes (51 vs.31),indicating GACD V.1.2 (clonal F_1 mapping type) is most likely to be more efficient than Ici Mapping (F_2 mapping type) for QTL analysis of a sefling population or clonal F_1 population in clonal species.The identified QTL controlling RSD resistance along with the associated SNP markers will assist sugarcane molecular breeding programs in combating this disease. | Qian You Sushma Sood Ziliang Luo Hongbo Liu Md.Sariful Islam Muqing Zhang Jianping Wang | 2021 | The Crop Journal2021,9,5: | 1 |
| 7 | circCAPRIN1 interacts with STAT2 to promote tumor progression and lipid synthesis via upregulating ACC1 expression in colorectal cancer显示文摘Background:Circular RNAs(circRNAs)generated by back-splicing of precursor mRNAs(pre-mRNAs)are often aberrantly expressed in cancer cells.Accumulating evidence has revealed that circRNAs play a critical role in the progression of several cancers,including colorectal cancer(CRC).However,the current understandings of the emerging functions of circRNAs in CRC lipid metabolism and the underlying molecular mechanisms are still limited.Here,we aimed to explore the role of circCAPRIN1 in regulating CRC lipid metabolism and tumorigenesis.Methods:circRNA microarray was performed with three pairs of tumor and non-tumor tissues from CRC patients.The expression of circRNAs were determined by quantitative PCR(qPCR)and in situ hybridization(ISH).The endogenous levels of circRNAs in CRC cells were manipulated by transfection with lentiviruses overexpressing or silencing circRNAs.The regulatory roles of circRNAs in the occurrence of CRC were investigated both in vitro and in vivo using gene expression array,RNA pull-down/mass spectrometry,RNA immunoprecipitation assay,luciferase reporter assay,chromatin immunoprecipitation analysis,and fluorescence in situ hybridization(FISH).Results:Among circRNAs,circCAPRIN1 was most significantly upregulated in CRC tissue specimens.circCAPRIN1 expression was positively correlated with the clinical stage and unfavorable prognosis of CRC patients.Downregulation of circCAPRIN1 suppressed proliferation,migration,and epithelial-mesenchymal transition of CRC cells,while circCAPRIN1 overexpression had opposite effects.RNA sequencing and gene ontology analysis indicated that circCAPRIN1 upregulated the expressions of genes involved in CRC lipid metabolism.Moreover,circCAPRIN1 promoted lipid synthesis by enhancing Acetyl-CoA carboxylase 1(ACC1)expression.Further mechanistic assays demonstrated that circCAPRIN1 directly bound signal transducer and activator of transcription 2(STAT2)to activate ACC1 transcription,thus regulating lipid metabolism and facilitating CRC tumorigenesis.Conclusions:These findings revealed the oncogenic role and mechanism of circCAPRIN1 in CRC.circCAPRIN1 interacted with STAT2 to promote CRC tumor progression and lipid synthesis by enhancing the expression of ACC1.circCAPRIN1 may be considered as a novel potential diagnostic and therapeutic target for CRC patients. | Yufei Yang Dakui Luo Yang Shao Zezhi Shan Qi Liu Junyong Weng Weijing He Ruoxin Zhang Qingguo Li Ziliang Wang Xinxiang Li | 2023 | Cancer Communications2023,43,1: | 0 |

| 10 | The E3 ubiquitin ligase CSIT1 regulates critical sterility-inducing temperature by ribosome-associated quality control to safeguard two-line hybrid breeding in rice显示文摘Two-line hybrid breeding can fully utilize heterosis in crops.In thermo-sensitive genic male sterile(TGMS)lines,low critical sterility-inducing temperature(CSIT)is vital to safeguard the production of two-line hybrid seeds in rice(Oryza sativa),but the molecular mechanism determining CSIT is unclear.Here,we report the cloning of CSIT1,which encodes an E3 ubiquitin ligase,and show that CSIT1 modulates the CSIT of thermo-sensitive genic male sterility 5(tms5)-based TGMS lines through ribosome-associated quality control(RQC).Biochemical assays demonstrated that CSIT1 binds to the 80S ribosomes and ubiquitinates abnormal nascent polypeptides for degradation in the RQC process.Loss of CSIT1 function inhibits the possible damage of tms5 to the ubiquitination system and protein translation,resulting in enhanced accumulation of anther-related proteins such as catalase to suppress abnormal accumulation of reactive oxygen species and premature programmed cell death in the tapetum,thereby leading to a much higher CSIT in the tms5-based TGMS lines.Taken together,our findings reveal a regulatory mechanism of CSIT,providing new insights into RQC and potential targets for future two-line hybrid breeding. | Guoqing Peng Minglong Liu Liya Zhu Wenlong Luo Qinghua Wang Mumei Wang Huiqiong Chen Ziliang Luo Yueping Xiao Yongjie Zhang Haona Hong Zhenlan Liu Lingyan Zhou Guoqiang Guo Yingxiang Wang Chuxiong Zhuang Hai Zhou | 2023 | Molecular Plant2023,16,10: | 0 |
