1Therapeutic Effects of DNA Vaccine on Allergen-Induced Allergic Airway Inflammation in Mouse Model显示文摘Vaccination with DNA encoding Dermatophagoides pteronyssinus group 2 (Der p 2) allergen previously showed its effects of immunologic protection on Der p 2 allergen-induced allergic airway inflammation in mice. In present study, we investigated whether DNA vaccine encoding Der p 2 could exert therapeutic role on allergen-induced allergic airway inflammation in mouse model and explored the mechanism of DNA vaccination in asthma specific-allergen immunotherapy. After sensitized and challenged by Der p 2, the BALB/c mice were immunized with DNA vaccine. The degrees of cellular infiltration were scored. IgE levels in serum and IL-4/IL-13 levels in BALF were determined by ELISA. The lung tissues were assessed by histological examinations. Expressions of STAT6 and NF-κB in lung were determined by immunohistochemistry staining. Vaccination of mice with DNA vaccine inhibited the development of airway inflammation and the production of mucin induced by allergen, and reduced the level of Der p 2-specific IgE level. Significant reductions of eosinophil infiltration and levels of IL-4 and IL-13 in BALF were observed after vaccination. Further more, DNA vaccination inhibited STAT6 and NF-κB expression in lung tissue in Der p 2-immunized mice. 2Effects of total soy saponins on free radicals in the quadriceps femoris,serum testosterone,LDH,and BUN of exhausted rats显示文摘Purpose:The aim of this study was to investigate the impact of total soy saponins(TS) on the free radical metabolism from the quadriceps femoris muscle,serum testosterone,lactate dehydrogenase(LDH),and blood urea nitrogen(BUN) in rats exercised to exhaustion.Methods:A one-time exhausted treadmill exercise session was used.Sprague-Dawley rats were divided into 4 groups:a control group—animals receiving no TS and no exercise(NTSNE),animals receiving TS but no exercise group(TSNE),animals receiving no TS but exercised to exhaustion group(NTSE),and animals receiving TS and exercised to exhaustion group(TSE).The TSNE and TSE groups were fed TS at a dosage of 20 mg/kg body weight once per day for 2 weeks.The NTSE group was given a placebo,and the NTSNE group was not given any treatment.The NTSE and TSE groups were exercised at speed of 30 m/min on treadmill until exhausted.The exercise time and exercise distance were recorded when the rats became exhausted and the rats were then decapitated and anatomized immediately.A 10% homogenate of the quadriceps femoris tissue was prepared.The levels of superoxide dismutase(SOD),catalase(CAT),malondialdehyde(MDA),glutathione peroxidase(GSH-Px),glutathione reductase(GR),reduced glutathione(GSH),total antioxidant capacity(T-AOC),LDH,BUN,and serum testosterone were tested.Results:TS significantly increased the exercise time to exhaustion by 20.62%(p < 0.05).The MDA levels were decreased significantly in the TSNE group than in NTSNE group(p < 0.05);the T-AOC levels increased significantly in the TSNE group than in the other 3 groups(p < 0.01,p < 0.05,p < 0.05).The LDH activity significantly increased in the NTSE group than in TSNE group(p < 0.05).The BUN levels significantly increased in the NTSE group than in the other 3 groups(p < 0.01,p < 0.01,p < 0.05),and significantly increased in the TSE group than in NTSNE and TSNE groups(both p < 0.01).The serum testosterone levels increased significantly in the TSNE group than in the other 3 groups(all p < 0.01).SOD,CAT,GSH-Px,GR,and GSH were not statistically different among the groups.Conclusion:TS can significantly improve the exercised rats' serum testosterone level and antioxidant activity in their quadriceps femoris to varying degrees,decrease MDA and serum LDH and BUN levels,increase the exercise time,and delay the occurrence of the fatigue.Zhigang Liu Ruixin Nie Yun Liu Zhouhong Li Chenxi Yang Zhengying Xiong 2017Journal of Sport and Health Science2017,6,3:8

4In vivo transfection of enhanced green fluorescent protein in rat retinal ganglion cells mediated by ultrasound-induced microbubbles显示文摘BACKGROUND:Studies have demonstrated that ultrasound-mediated microbubble destruction significantly improves transfection efficiency of enhanced green fluorescent protein(EGFP) in in vitro cultured retinal ganglial cells(RGCs). OBJECTIVE:To investigate the feasibility of ultrasound-mediated microbubble destruction for EGFP transfection in rat RGCs,and to compare efficiency and cell damage with traditional transfection methods. DESIGN,TIME AND SETTING:In vivo,gene engineering experiment.The study was performed at the Central Laboratory,Institute of Ultrasonic Imaging,Chongqing Medical University from March to July 2008. MATERIALS:Eukaryotic expression vector plasmid EGFP and microbubbles were prepared by the Institute of Ultrasonic Imaging,Chongqing Medical University.The microbubbles were produced at a concentration of 8.7×10^(11)/L,with a 2-4μm diameter,and 10-hour half-life in vitro. METHODS:A total of 50 Sprague Dawley rats were randomly assigned to four groups.Normal controls(n=5) were infused with 5μL normal saline to the vitreous cavity;the naked plasmid group (n=15) was infused with 5μL EGFP plasmid to the vitreous cavity;in the plasmid with ultrasound group(n=15),the eyes were irradiated with low-energy ultrasound wave(0.5 W/cm^2) for a total of 60 seconds(irradiated for 5 seconds,at 10-second intervals) immediately following infusion of EGFP plasmids to the vitreous cavities.In the microbubble-ultrasound group(n=15),the eyes were irradiated with the same power of ultrasonic wave immediately following infusion of microbubbles containing EGFP plasmids to the vitreous cavities. MAIN OUTCOME MEASURES:After 7 days,retinal preparations and EGFP expression in RGCs were observed by fluorescence microscopy.RGC quantification in the retinal ganglion cell layer was performed.In addition,EGFP mRNA expression was semi-quantitatively determined by RT-PCR. RESULTS:The transfection efficiency of EGFP to RGCs by microbubbles with ultrasound was significantly greater than the other groups,and no obvious damage was detected in the RGCs. 5Microstructural Evolutions of AA7055 Aluminum Alloy Under Dynamic and Quasi-static Compressions显示文摘The microstructural evolution of AA7055 aluminum alloy under dynamic impact loading with the strain rate of1.3*104s-1controlled by a split Hopkinson pressure bar was investigated,and compared with that under quasi-static mechanical loading in compression with strain rate of 1.0*10-3s-1.The quasi-static-compressed sample exhibited equiaxed dislocation cells,which were different from the elongated and incomplete dislocation cells for the alloy undergoing dynamic compression.The high strain-rate compression also induced the formation of localized shear bands in which the recrystallizations characterized as fine equiaxed grains were observed.The microstructural evolutions under both quasi-static and dynamic compressions are rationalized in terms of the dislocation cell model combined with the dislocation kinetics,in addition to the adiabatic temperature rise in shear bands at high strain rate.Yuanyuan Xiong Ning Li Huawen Jiang Zhigang Li Zhu Xu Lin Liu 2014Acta Metallurgica Sinica(English Letters)2014,27,2:2
6Case study on stratospheric and mesospheric concentric gravity waves generated by deep convection显示文摘Concentric gravity waves(CGWs)in the middle and upper atmosphere show wave-coupling processes between the lower atmosphere and the middle and upper atmosphere.In this research,we analyzed a case of CGWs detected simultaneously by the AIRS(Atmospheric Infrared Sounder)and the VIIRS/DNB(Day/Night Band of the Visible Infrared Imager Radiometer Suite)in the stratosphere and mesosphere.Results showed that gravity waves(GWs)were generated by the collocated Hurricane Bejisa on the island of Mauritius.The AIRS data showed arc-like phase fronts of GWs with horizontal wavelengths of 190 and 150 km at 21:08 coordinated universal time(UTC)on 1 January 2014 and at 10:00 UTC on 2 January 2014,whereas the DNB observed arced GWs with horizontal wavelengths of 60 and 150 km in the same geographic regions at 22:24 UTC.The characteristics of CGW parameters in the stratosphere(~40 km)and the mesosphere(~87 km),such as the vertical wavelength,intrinsic frequency,and intrinsic horizontal phase speed,were first derived together with the background winds from ERA5 reanalysis data and Horizontal Wind Model data through the dispersion relationship of GWs and the wind-filtering theory.GuoChun Shi Xiong Hu ZhiGang Yao WenJie Guo MingChen Sun XiaoYan Gong 2021Earth and Planetary Physics2021,5,1:2
7Accelerating Packet Classification with Counting Bloom Filters for Virtual Open Flow Switching显示文摘The growing trend of network virtualization results in a widespread adoption of virtual switches in virtualized environments. However, virtual switching is confronted with great performance challenges regarding packet classification especially in Open Flow-based software defined networks. This paper first takes an insight into packet classification in virtual Open Flow switching, and points out that its performance bottleneck is dominated by flow table traversals of multiple failed mask probing for each arrived packet. Then we are motivated to propose an efficient packet classification algorithm based on counting bloom filters. In particular, counting bloom filters are applied to predict the failures of flow table lookups with great possibilities, and bypass flow table traversals for failed mask probing. Finally, our proposed packet classification algorithm is evaluated with real network traffic traces by experiments. 9Experimental Analysis of Microscale Laser Shock Processing on Metallic Material Using Excimer Laser显示文摘Microscale laser shock processing (μLSP), also known as laser shock processing in microscale, is a technique that uses microscale focused laser beam to induce high pressure plasma and generates plastic deformation and compressive residual stress in target materials, thus improves fatigue or stress corrosion cracking resistance of MEMS (Micro Electromechanical Systems) devices made of such a material. Many works have been reported about the research and experiment for μLSP. But the diameters of 50–200 μm were used at the first time for this field, which was useful for treating micro-device components with larger area and curved surface. The excimer laser was used firstly on μLSP for shorter wavelength than that of used in previous researches. The determination method of laser spot size at micro-level spatial resolution was presented. Under these conditions, plastic deformation, the stress analysis and microhardness with different pulse number, pulse energy and pulse spacing were investigated. Especially the residual stress distribution with depth treated by μLSP, was first investigated. 11大豆总皂苷改善力竭大鼠心肌抗氧化能力与运动能力(英文)显示文摘目的:本实验旨在研究大鼠运动至力竭过程中,大豆总皂苷(TS)对心肌抗氧化能力的影响。方法:用跑台一次运动力竭模型,将受试大鼠分为4组:对照组、TS组、力竭组、TS力竭组。按每千克体重20毫克、每天一次的剂量持续两周喂服TS组和TS力竭组大鼠。力竭组喂服安慰剂,对照组不给予任何处理。将跑步机速度预设为30 m/min,让力竭组和TS力竭组大鼠进行跑台运动直至力竭,立即处死并制备10%的心肌组织匀浆。结果:与力竭组相比,TS力竭组运动时间显著延长20.62%(p<0.05)。与对照组相比,TS组CAT、GSH-Px和GR酶活性显著增强(p<0.01),TS力竭组GSH-Px与GR酶活性显著增强(p<0.01)而MDA水平显著降低(p<0.05)。与力竭组相比,TS力竭组GSH-Px活性显著增强(p<0.01),TS组CAT、GSH-Px和GR酶活性均显著提高(p<0.01)。与TS组相比,TS力竭组CAT和GR活性显著增强(p<0.01)。结论:TS可不同程度地改善运动大鼠的心肌抗氧化活性,降低大鼠心肌MDA水平、血清AST和LDH水平,延长运动时间并延缓运动疲劳的发生。Zhigang Liu Yun Liu Zhengying Xiong Yue Feng Wenkun Tang 2016Journal of Sport and Health Science2016,5,4:1

14Compressional behavior of MgCr_2O_4 spinel from first-principles simulation显示文摘The compressional behavior of the MgCr_2O_4 spinel has been investigated with the CASTEP code using density functional theory and planewave pseudopotential technique. We treated the exchange-correlation interaction by both the local density approximation(LDA) and generalized gradient approximation(GGA) with the Perdew-Burker-Ernzerhof functional. Our simulation was conducted for the pressure range of 0–19 GPa. We obtained the isothermal bulk modulus(K_T) of the MgCr_2O_4 spinel as 181.46(48) GPa(GGA; low boundary) or 216.1(11) GPa(LDA; high boundary), with its first derivative(K'_T) as 4.41(6) or 4.5(1), respectively. The oxygen parameter u is not constant but negatively correlated with P, and decreases by about 0.5–0.6% for the investigated P range. 15Sinoatrial node pacemaker cells share dominant biological properties with glutamatergic neurons显示文摘Activation of the heart normally begins in the sinoatrial node(SAN).Electrical impulses spontaneously released by SAN pacemaker cells(SANPCs)trigger the contraction of the heart.However,the cellular nature of SANPCs remains controversial.Here,we report that SANPCs exhibit glutamatergic neuron-like properties.By comparing the single-cell transcriptome of SANPCs with that of cells from primary visual cortex in mouse,we found that SANPCs co-clustered with cortical neurons.Tissue and cellular imaging confirmed that SANPCs contained key elements of glutamatergic neurotransmitter system,expressing genes encoding glutamate synthesis pathway(G/s),ionotropic and metabotropic glutamate receptors(Grina,Gria3,Grm1 and Grm5)t and glutamate transporters(Slc17a7).SANPCs highly expressed cell markers of glutamatergic neurons(Snap25 and S/-c17a7)t whereas Gad1,a marker of GABAergic neurons,was negative.Functional studies revealed that inhibition of glutamate receptors or transporters reduced spontaneous pacing frequency of isolated SAN tissues and spontaneous Ca2+transients frequency in single SANPC.Collectively,our work suggests that SANPCs share dominant biological properties with glutamatergic neurons,and the glutamatergic neurotransmitter system may act as an intrinsic regulation module of heart rhythm,which provides a potential intervention target for pacemaker cell-associated arrhythmias.Dandan Liang Zhigang Xue Jinfeng Xue Duanyang Xie Ke Xiong Huixing Zhou Fulei Zhang Xuling Su Guanghua Wang Qicheng Zou Yi Liu Jian Yang Honghui Ma Luying Peng Chunyu Zeng Gang Li Li Wang Yi-Han Chen 2021Protein & Cell2021,12,7:1

18Bio-inspired untethered fully soft robots in liquid actuated by induced energy gradients显示文摘Soft robotics with new designs,fabrication technologies and control strategies inspired by nature have been totally changing our view on robotics.To fully exploit their potential in practical applications,untethered designs are preferred in implementation.However,hindered by the limited thermal/mechanical performance of soft materials,it has been always challenging for researchers to implement untethered solutions,which generally involve rigid forms of high energy-density power sources or high energy-density processes.A number of insects in nature,such as rove beetles,can gain a burst of kinetic energy from the induced surface-energy gradient on water to return to their familiar habitats,which is generally known as Marangoni propulsion.Inspired by such a behavior,we report the agile untethered mobility of a fully soft robot in liquid based on induced energy gradients and also develop corresponding fabrication and maneuvering strategies.The robot can reach a speed of 5.5 body lengths per second,which is 7-fold more than the best reported,0.69(body length per second),in the previous work on untethered soft robots in liquid by far.Further controlling the robots,we demonstrate a soft-robot swarm that can approach a target simultaneously to assure a hit with high accuracy.Without employing any high energy-density power sources or processes,our robot exhibits many attractive merits,such as quietness,no mechanical wear,no thermal fatigue,invisibility and ease of robot fabrication,which may potentially impact many fields in the future.Liang Xiong Lyu Fen Li Kang Wu Pan Deng Seung Hee Jeong Zhigang Wu Han Ding 2019National Science Review2019,6,5:1
