The altered expression of inflammation-related microRNAs with microRNA-155 expression correlates with Th17 differentiation in patients with acute coronary syndrome显示文摘MicroRNAs(miRNAs)are a novel class of small,non-coding RNAs that play a significant role in both inflammatory and cardiovascular diseases.Immune cells, especially T helper(Th)cells, are critical in the development of atherosclerosis and the onset of acute coronary syndrome(ACS).To assess whether inflammation-related miRNAs(such as miR-155,146a,21,125a-5p,125b,31)are involved in the imbalance of Th cell subsets in patients with ACS,we measured the expression of related miRNAs in patients with acute myocardial infarction(AMI),unstable angina(UA),stable angina(SA)and chest pain syndrome(CPS);analyzed the relationship between miRNA expression and the frequency of Th cell subsets;and observed the co-expression of miR-155 and IL-17A in peripheral blood mononuclear cells(PBMCs)of patients with ACS.The results showed that the expression of miR-155 in the PBMCs of patients with ACS was decreased by approximately 60%,while the expression of both miR-21 and miR-146a was increased by approximately twofold. The expression patterns of miRNAs in plasma correlated with those in PBMCs,except for miR-21,which was increased by approximately sixfold in the AMI group and showed no significant difference between the UA group and the CPS group. We also found that the expression of miR-155 inversely correlated with the frequency of Th17 cells(r520.896,P,0.01)and that miR-155 was co-expressed with IL-17A in patients with ACS.In conclusion, our study revealed the expression patterns of inflammation-related miRNAs in patients with ACS and found that miR-155 may be associated with Th17 cell differentiation.Rui Yao Yulan Ma Youyou Du Mengyang Liao Huanhuan Li Wei Liang Jing Yuan Zhijun Ma Xian Yu Hong Xiao Yuhua Liao 2011Cellular & Molecular Immunology2011,8,6:32
Report on Childhood Obesity in China (9): Sugar-sweetened Beverages Consumption and Obesity显示文摘Objective To explore the associations between sugar-sweetened beverage (SSB) consumption and obesity as well as obesity-related cardiometabolic disorders among children in China. Methods A total of 6974 (boys 3558, girls 3412) children aged 6-13 years participated in the study. Each participant's height, weight, waist circumference, fasting glucose, triglycerides, total cholesterol, high- density lipoprotein cholesterol, and low-density lipoprotein cholesterol were measured. The type of beverage consumption was determined using a self-administered questionnaire. Results SSBs were consumed regularly by 46.1% of the children. The prevalence [adjusted odds ratio (95% confidence internal (CI)] of obesity was 7.6% [as the reference group (ref.)], 10.1% [1.36(1.07, 1.74)], and 11.6% [1.46(1.21, 1.75)], among children who regularly drank milk, other beverages and SSBs, respectively. Regularly drinking SSBs elevated the likelihood of abdominal obesity [adjusted odds ratio (95% CI): 1.36 (1.17, 1.59)]. The prevalence [adjusted odds ratio (95% CI)] of obesity among children who regularly drank sports/caloric beverages, carbonated beverages, sweet tea, and plant protein beverages was 16.8% [2.00(1.31, 3.07)], 12.7% [1.52(1.23, 1.88)], 11.5% [1.52(1.18, 1.95)], and 10.4% [1.41(1.03, 1.94)], respectively, which was higher than that of regular milk drinkers [7.6 % (ref.)]. The prevalence [adjusted odds ratio (95% CI)] of abdominal obesity among children who regularly drank sweet tea, fruit/vegetable juices, and carbonated beverages was 17.7% [1.55(1.26, 1.90)], 16.2% [1.36(1.09, 1.70)], and 15.3% [1.24(1.03, 1.50)], respectively, which was much higher than that of regular milk drinkers [12.8% (ref.)]. Conclusions Regular SSB consumption was positively related to obesity and abdominal obesity. This relationship should be investigated further using a longitudinal study design.SHANG Xian Wen LIU Ai Ling ZHANG Qian HU Xiao Qi DU Song Ming MA Jun XU Gui Fa LI Ying GUO Hong Wei DU Lin LI Ting Yu MA Guan Sheng 2012Biomedical and Environmental Sciences2012,25,2:23
Electroacupuncture activates enteric glial cells and protects the gut barrier in hemorrhaged rats显示文摘AIM:To investigate whether electroacupuncture ST36 activates enteric glial cells, and alleviates gut inflammation and barrier dysfunction following hemorrhagic shock.METHODS:Sprague-Dawley rats were subjected to approximately 45% total blood loss and randomly divided into seven groups:(1) sham:cannulation, but no hemorrhage;(2) subjected to hemorrhagic shock(HS);(3) electroacupuncture(EA) ST36 after hemorrhage;(4) vagotomy(VGX)/EA:VGX before hemorrhage, then EA ST36;(5) VGX:VGX before hemorrhage;(6) a-bungarotoxin(BGT)/EA:intraperitoneal injection of a-BGT before hemorrhage, then EA ST36; and(7) a-BGT group:a-BGT injection before hemorrhage.Morphological changes in enteric glial cells(EGCs) were observed by immunofluorescence, and glial fibrillary acidic protein(GFAP; a protein marker of enteric glial activation) was evaluated using reverse transcriptase polymerase chain reaction and western blot analysis.Intestinal cytokine levels, gut permeability to 4-k Da fluorescein isothiocyanate(FITC)-dextran,and the expression and distribution of tight junction protein zona occludens(ZO)-1 were also determined.RESULTS:EGCs were distorted following hemorrhage and showed morphological abnormalities.EA ST36 attenuated the morphological changes in EGCs at 6 h,as compared with the VGX,a-BGT and HS groups.EA ST36 increased GFAP expression to a greater degree than in the other groups.EA ST36 decreased intestinal permeability to FITC-dextran(760.5 ± 96.43 ng/m L vs 2466.7 ± 131.60 ng/m L,P < 0.05) and preserved ZO-1 protein expression and localization at 6 h afterhemorrhage compared with the HS group. However, abdominal VGX and a-BGT treatment weakened or eliminated the effects of EA ST36.EA ST36 reduced tumor necrosis factor-a levels in intestinal homogenates after blood loss, while vagotomy or intraperitoneal injection of a-BGT before EA ST36 abolished its antiinflammatory effects.CONCLUSION:EA ST36 attenuates hemorrhageinduced intestinal inflammatory insult, and protects the intestinal barrier integrity, partly via activation of EGCs.Sen Hu Zeng-Kai Zhao Rui Liu Hai-Bin Wang Chun-Yu Gu Hong-Min Luo Huan Wang Ming-Hua Du Yi Lv Xian Shi 2015World Journal of Gastroenterology2015,21,5:15

Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. Therefore the observation of Insight-HXMT constitutes an important chapter in the full context of multi-wavelength and multi-messenger observation of this historical GW event.TiPei Li ShaoLin Xiong ShuangNan Zhang FangJun Lu LiMing Song XueLei Cao Zhi Chang Gang Chen Li Chen TianXiang Chen Yong Chen YiBao Chen YuPeng Chen Wei Cui WeiWei Cui JingKang Deng YongWei Dong YuanYuan Du MinXue Fu GuanHua Gao He Gao Min Gao MingYu Ge YuDong Gu Ju Guan ChengCheng Guo DaWei Han Wei Hu Yue Huang Jia Huo ShuMei Jia LuHua Jiang WeiChun Jiang Jing Jin YongJie Jin Bing Li ChengKui Li Gang Li MaoShun Li Wei Li Xian Li XiaoBo Li XuFang Li YanGuo Li ZiJian Li ZhengWei Li XiaoHua Liang JinYuan Liao CongZhan Liu GuoQing Liu HongWei Liu ShaoZhen Liu XiaoJing Liu Yuan Liu YiNong Liu Bo Lu XueFeng Lu Tao Luo Xiang Ma Bin Meng Yi Nang JianYin Nie Ge OU JinLu Qu Na Sai Liang Sun Yin Tan Lian Tao WenHui Tao YouLi Tuo GuoFeng Wang HuanYu Wang Juan Wang WenShuai Wang YuSa Wang XiangYang Wen BoBing WU Mei Wu GuangCheng Xiao He Xu YuPeng Xu LinLi Yan JiaWei Yang Sheng Yang YanJi Yang AiMei Zhang ChunLei Zhang ChengMo Zhang Fan Zhang HongMei Zhang Juan Zhang Qiang Zhang Shu Zhang Tong Zhang Wei Zhang WanChang Zhang WenZhao Zhang Yi Zhang Yue Zhang YiFei Zhang YongJie Zhang Zhao Zhang ZiLiang Zhang HaiSheng Zhao JianLing Zhao XiaoFan Zhao ShiJie Zheng Yue Zhu YuXuan Zhu ChangLin Zou 2018Science China(Physics,Mechanics & Astronomy)2018,61,3:12

Estimation of the quantity of aeolian saltation sediments blown into the Yellow River from the Ulanbuh Desert,China显示文摘The Ulanbuh Desert borders the upper reach of the Yellow River.Every year,a mass of aeolian sand is blown into the Yellow River by the prevailing wind and the coarse aeolian sand results in serious silting in the Yellow River. To estimate the quantity of aeolian sediments from the Ulanbuh Desert blown into the Yellow River,we simulated the saltation processes of aeolian sediments in the Ulanbuh Desert. Then we used a saltation submodel of the IWEMS(Integrated Wind-Erosion Modeling System)and its accompanying RS(Remote Sensing)and GIS(Geographic Information System)modules to estimate the quantity of saltation sediments blown into the Yellow River from the Ulanbuh Desert. We calibrated the saltation submodel by the synchronous observation to wind velocity and saltation sediments on several points with different vegetation cover. The vegetation cover, frontal area of vegetation, roughness length, and threshold friction velocity in various regions of the Ulanbuh Desert were obtained using NDVI(Normalized Difference Vegetation Index)data, measured sand-particle sizes, and empirical relationships among vegetation cover,sand-particle diameters, and wind velocity.Using these variables along with the observed wind velocities and saltation sediments for the observed points, the saltation model was validated. The model results were shown to be satisfactory(RMSE less than 0.05 and|Re|less than 17%).In this study,a subdaily wind-velocity program,WINDGEN,was developed using this model to simulate hourly wind velocities around the Ulanbuh Desert.By incorporating simulated hourly wind-velocity and wind-direction data, the quantity of saltation sediments blown into the Yellow River was calculated with the saltation submodel. The annual quantity of aeolian sediments blown into the Yellow River from the Ulanbuh Desert was 5.56×106t from 2001 to 2010,most of which occurred in spring(from March to May);for example,6.54×105tons of aeolian sand were blown into the Yellow River on 25 April,2010.However,in summer and winter, the saltation process occasionally occurred. This research has supplied some references to prevent blown sand hazards and silting in the Yellow River.HeQiang DU Xian XUE Tao WANG 2014Journal of Arid Land2014,6,2:11

Neuroprotective effects of INT-777 against Aβ1-42-induced cognitive impairment, neuroinflammation, apoptosis, and synaptic dysfunction in mice显示文摘OBJECTIVE To investigate the neuroprotective effects of TGR5 agonist INT-777 in the Aβ_(1-42)-treated mouse model of acute neurotoxicity.METHODS Cognitive impairment was induced by single intracerebroventricular injection of aggregated Aβ_(1-42)(410 pmol per mouse;5 μL)into the mouse brain in normal ICR mice.After3 d,INT-777(0.75,1.5 or 3.0 μg per mouse) was infused into the same site.Another 3 d later, the cognition function was evaluated by Morris water maze(MWM),novel objective recognition(NOR),and Y maze tests.Furthermore, the levels of TGR5,TNF-α,IL-1β,IL-6,NF-κB p65,Iba1,caspase 3,Bcl-2,Bax,PSD95,and synaptophysin in the hippocampus and frontal cortex were detected using Western blotting.Microglia activation in the hippocampal CA1,CA3 and DG regions and frontal cortex were detected by immunohistochemistry.TUNEL staining was used to detect the apoptotic cells. The number of spines in the hippocampal CA1,CA3 and DG regions and frontal cortex was detected by GolgiCox staining.RESULTS INT-777(1.5 or 3.0 μg per mouse) significantly ameliorated memory impairment in the Aβ_(1-42)-treated mice, evidenced by increased the time spent in the target quadrant(P<0.05),and the number of target crossings(P<0.05) in the MWM test, increased the discrimination index in the NOR test(P<0.05),and increased the number of correct choices(P<0.05,P<0.01) and decreased the latency(P<0.05) to enter the shock-free compartment in the Y-maze test.Importantly, this treatment reversed Aβ_(1-42)-induced TGR5 down-regulation(P<0.05,P<0.01),suppressed the increase of nuclear NF-κB p65(P<0.05,P<0.01) and mitigated neuroinflammation, evidenced by lower proinflammatory cytokines such as TNF-α(P<0.05),IL-1β(P<0.05),IL-6(P<0.05,P<0.01),and less Iba1-positive cells in the hippocampal CA1,CA3 and DG regions and frontal cortex(P<0.05,P<0.01) as well as the protein level of Iba1.INT-777(1.5 or3.0 μg per mouse) also pronouncedly suppressed apoptosis through the reduction of TUNEL-positive cells(P<0.05,P<0.01),decreased activation of caspase 3(P<0.05),increased ratio of Bcl-2/Bax(P<0.05,P<0.01),and ameliorated synaptic dysfunction via promoting dendritic spines generation(P<0.05,P<0.01) with the upregulation of PSD95(P<0.05) and synaptophysin proteins(P<0.05).CONCLUSION INT-777 has potent neuroprotective effects against Aβ_(1-42)-induced neurotoxicity, which suggests that the activation of TGR5 could be a novel and promising strategy for the treatment of AD.WU Xian LYU Yang-ge DU Yi-feng CHEN Fang Miranda N REED HU Mei Vishnu SUPPIRAMANIAM TANG Su-su HONG Hao 2018中国药理学与毒理学杂志2018,32,9:6

Effects of Environmental Temperature on the Regeneration Frequency of the Immature Embryos of Wheat(Triticum aestivum L.)显示文摘The immature embryos(IEs) of wheat are the most widely used tissues for in vitro culture and genetic transformation due to its high regeneration competency. However, this explant can only be maintained in 4°C daily cooler for a short period time for its use in plant tissue culture or transformation experiments. This study aimed to investigate the effects of environmental temperature, cryopreservation storage temperature, and heat shock culture(HSC) temperature on the regeneration frequency of wheat IEs. Results indicated that environmental temperature significantly affected the induction of embryonic calli. The optimum total accumulated temperature(TAT) during the time of anthesis and sampling for regeneration of these tissues was around 280°C for spring wheat type cv. CB037 and approximately 300°C for winter wheat type cv. Kenong 199. Regeneration ability obviously declined when the highest environmental temperature was over 35°C for 1 d or a high temperature between 30 and 33°C lasted for 5 d during anthesis and sampling. This finding was verified by culturing the freshly isolated IEs under different temperatures from 29 to 37°C in different controlled growth incubators for 5 d; the IEs almost completely lost regeneration ability when the temperature rose to 37°C. Cryopreservation of-20°C caused the wheat samples lost ability of producing callus or embryonic callus in a few days, and cryopreservation of-10°C more than 10 d made the regeneration potential of the tissues dramatically declined. Comparatively, the temperature that best maintained high regeneration ability was-5°C, at which the materials can be maintained for around 1 mon. In addition, the preservation of the immature samples at-5 or-10°C inhibited the direct germination of the IEs, avoiding the embryo axis removing process. Our results are useful for ensuring that fi eld collection and cryopreservation of the wheat IEs are done correctly to enable tissue culture and genetic transformation.WANG Xin-min REN Xian YIN Gui-xiang WANG Ke LI Jia-rui DU Li-pu XU Hui-jun YE Xing-guo 2014Journal of Integrative Agriculture2014,13,4:6
The Medium Energy X-ray telescope (ME) onboard the Insight-HXMT astronomy satellite显示文摘The Medium Energy X-ray telescope(ME) is one of the three main telescopes on board the Insight hard X-ray modulation telescope(Insight-HXMT) astronomy satellite. ME contains 1728 pixels of Si-PIN detectors sensitive in 5-30 ke V with a total geometrical area of 952 cm^2. The application specific integrated circuit(ASIC) chip, VA32TA6, is used to achieve low power consumption and low readout noise. The collimators define three kinds of field of views(FOVs) for the telescope, 1°×4°, 4°×4°,and blocked ones. Combination of such FOVs can be used to estimate the in-orbit X-ray and particle background components. The energy resolution of ME is ~3 ke V at 17.8 ke V(FWHM) and the time resolution is 255 μs. In this paper, we introduce the design and performance of ME.XueLei Cao WeiChun Jiang Bin Meng WanChang Zhang Tao Luo Sheng Yang ChunLei Zhang YuDong Gu Liang Sun XiaoJing Liu JiaWei Yang Xian Li Ying Tan ShaoZhen Liu YuanYuan Du FangJun Lu YuPeng Xu Ju Guan ShuangNan Zhang HuanYu Wang TiPei Li ChengMo Zhang XiangYang Wen JinLu Qu LiMing Song XiaoBo Li MingYu Ge YuPeng Zhou ShaoLin Xiong Shu Zhang YongJie Zhang ZeHao Cheng Fei Zhang MaoShun Li XiaoHua Liang Min Gao EnBo Yang XiaoHang Liu HongWei Liu YiJung Yang Fan Zhang 2020Science China(Physics,Mechanics & Astronomy)2020,63,4:5

ADAMTS-7 promotes vascular smooth muscle cells proliferation in vitro and in vivo显示文摘Vascular smooth muscle cell(VSMC) proliferation and migration are pivotal for the pathogenesis of atherosclerosis and post-angioplasty restenosis. We have recently reported that a disintegrin and metalloproteinase with thrombospondin motifs-7(ADAMTS-7), a novel metalloproteinase, contributes directly to neointima formation by mediating VSMC migration. However, whether ADAMTS-7 affects VSMC proliferation remains unclear. In this study, we found that luminal adenoviral delivery of ADAMTS-7 aggravated intimal hyperplasia 7 d after injury, paralleled by an increased percentage of PCNA-positive cells in both intima and media. In contrast, perivascular administration of ADAMTS-7 si RNA, but not scrambled si RNA to injured arteries attenuated intimal thickening at day 7, paralleled with reduced intimal VSMC replication, without alteration of VSMC proliferation in the media. In accordance, [3H]-thymidine incorporation assay in primary cultured rat VSMCs revealed an enhanced replication rate(by 61%) upon ADAMTS-7 overexpression and retarded proliferation(by 23%) upon ADAMTS-7 si RNA administration. Our data demonstrates that ADAMTS-7 promotes VSMC proliferation both in vitro and in vivo. ADAMTS-7 may therefore serve as a novel therapeutic target for atherosclerosis and post-angioplasty restenosis.ZHANG Lu YU Fang WANG Li ZHENG JinGang DU YaoYao HUANG YaQian LIU Bo WANG Xian KONG Wei 2015Science China(Life Sciences)2015,58,7:4
Solvent extraction and separation of light rare earths from chloride media using HDEHP-P350 system显示文摘Solvent extraction is the most important method for rare earth extraction and separation.Currently,di(2-ethylhexyl)phosphoric acid(HDEHP)and 2-ethylhexyl phosphonic acid mono-2-ethylhexyl ester(HEH/EHP)are widely used in industrial production, but there are still obvious deficiencies that require further research to resolve. In this paper, the unsaponification extraction of light rare earth ions in a hydrochloric acid medium by di(2-ethylhexyl)phosphoric acid-di(1-methyl-heptyl)methyl phosphonate(HDEHPP350)system was studied. The results show that the addition of P350 reduces the extraction capacity of HDEHP,and also greatly reduces the concentration of acidity required for the back-extraction.It still has a good separation factor for light rare earths without saponification, and the extractant is not easy to emulsify. With an aqueous phase of pH=2.85,and HDEHP mole fraction XHDEHP=0.9(compared with O/A=2),the separation effect of light rare earth is the best, resulting in the separation coefficientβCe/La=3.39,βPr/Ce=1.67 andβNd/Pr=1.45,respectively.The loaded light rare earth ions extracted by HDEHP-P350 can be easily stripped when 2 mol/L HCl is used as the stripping agent. Finally, the extraction mechanism is discussed using a slope method, and the final structure of the extracted complex is determined to be RECl[(DEHP)_(2)]_(2)P350_((o)),based on a combination of infrared spectra and 1 H NMR and 31P NMR analyses.Wenjie Zhang Dongxia Feng Xian Xie Xiong Tong Yunpeng Du Yang Cao 2022Journal of Rare Earths2022,40,2:3
Obtaining Ultra-High Strength and Ductility in a Mg–Gd–Er–Zn–Zr Alloy via Extrusion,Pre-deformation and Two-Stage Aging显示文摘The Mg–12Gd–1Er–1Zn–0.9 Zr(wt%)alloy with ultra-high strength and ductility was developed via hot extrusion combined with pre-deformation and two-stage aging treatment. The age-hardening behavior and microstructure evolution were investigated.Pre-deformation introduced a large number of dislocations, resulting in strain hardening and higher precipitation strengthening in the subsequent two-stage aging. As a result, the alloy showed a superior strength–ductility balance with a yield strength of 506 MPa,an ultimate tensile strength of 549 MPa and an elongation of 8.2%at room temperature. The finer and denserβ’precipitates significantly enhanced the strength, and the bimodal structure,smallβ-Mg5RE phase as well as denseγ’precipitates ensured the good ductility of the alloy. It is suggested that the combination of pre-deformation and two-stage aging treatment is an eff ective method to further improve the mechanical properties of wrought Mg alloys.Lin-Yue Jia Wen-Bo Du Jin-Long Fu Zhao-Hui Wang Ke Liu Shu-Bo Li Xian Du 2021Acta Metallurgica Sinica(English Letters)2021,34,1:3
