| 2 | OsWRKY03, a rice transcriptional activator that functions in defense signaling pathway upstream of OsNPR1显示文摘WRKY family proteins are a class of plant specific transcription factors that involve in many stress response pathways. It has been shown that one Arabidopsis WRKY protein, AtWRKY29/22, is activated by MAP kinase signaling cascade and confers resistance to both bacterial and fungal pathogens. However, little is known about the biological roles of WRKY proteins in rice. In this study, we investigated the expression patterns of rice AtWRKY29/22 homolog, OsWRKY03, under different conditions, and also its possible role involved in plant defense. Our results showed that OsWRKY03 was up-regulated by several defense signaling molecules or different treatments. Further analysis revealed that the expres- sion of OsWRKY03 was light dependent. Transcriptional activation activity of OsWRKY03 was also demonstrated by yeast functional assay. Transient expression of OsWRKY03-GFP fusion protein in onion epidermis cells showed that OsWRKY03 was a nuclear localized protein. OsNPR1 as well as several other pathogenesis-related genes, such as OsPR1b, phenylalanine ammonia-lyase (ZB8) and peroxidase (POX22.3), were induced in OsWRKY03-overexpressing transgenic plants. | 3 | Pre-collision Granites and Post-collision Intrusive Assemblage of the Kelameili-Harlik Orogenic Belt显示文摘The main types of intrusive rocks in the Kelameili-Harlik Hercynian erogenic belt include calc-alkaline granites, diabase dykes, kaligranites and alkaline granites. Investigation in field geology, petrology, mineralogy and geochemistry shows that the calc-alkaline granites belong to the syntexis-type (or I-type) and were formed in a pre-collisional magmatic arc environment. In consideration of the fact that kaligranites have many features of alkaline granites with higher consolidation temperatures than the calc-alkaline granites and show a discontinuity of minor element and REE evolution in respect to the calc-alkaline granites, they could not have been derived by differentiation of magmas for the calc-alkaline granites, but are likely to have been generated in an environment analogous to that for alkaline granites. The triplet of basic dyke swarms, kaligranites and alkaline granites could be regarded as a prominent indication of the initial stage of post-collisional delamination and extension. In vivo experiments, exogenous gene was transferred into tumor vascular endothelial cells and tumor cells of subcutaneously transplanted human colon cancer LOVO, human malignant melanoma A375 and human hepatoma graft in nude mice. This system could also target gene to intrahepatically transplanted human hepatoma injected via portal vein in nude mice. | 6 | Investigation of quality of life in patients with lung cancer by the EORTC QLQ-C30(V3.0) Chinese version显示文摘Objective To explore quality of life(QOL) and its influencing factors in patients with lung cancer.Methods A QOL questionnaire(European Organization for Research and Treatment of Cancer [EORTC] Quality of Life Questionnaire-Core 30 [QLQ-C30] Chinese version) was used with 138 patients with lung cancer participating in the study. A statistical description of the general characteristics of the patients with lung cancer was performed. The patients' QLQ-C30 scores were compared with the reference value for each dimension. To analyze the influence of QOL in different genders, age groups, and cancer stages, ttests and rank sum tests were used to compare the differences in QOL using a 5% significance level. Results The QLQ-C30 function scores in PF(physical functioning), EF(emotional functioning), SF(social functioning), and GH(global functioning), and symptom scales in NV(nausea and vomiting), DY(dyspnea), SL(insomnia), and FI(financial difficulties) were significantly different(P < 0.05) in comparison to the reference values. Female patients were worse than males(P < 0.05) in EF, NV, and DI(diarrhea). The later the stage of lung cancer, the worse the quality of life became; the functional scales in RF(role functioning), EF, CF, SF, and GH, and symptom scales in PA(pain), AP(appetite), and SL differences were statistically significant(P < 0.05). | 9 | Inhibition of angiogenesis and HCT-116 xenograft tumor growth in mice by kallistatin显示文摘AIM: To investigate the inhibitory effect of kallistatin (KAL) on angiogenesis and HCT-116 xenograft tumor growth. METHODS: Heterotopic tumors were induced by subcutaneous injection of 2 × 106 HCT-11 cells in mice. Seven days later, 2 × 1011 rAAV-GFP or rAAV-KAL was injected intratumorally (n = 5 for each group). The mice were sacrificed at d 28, by which time the tumors in the rAAV-GFP group had grown to beyond 5% of the total body weight. Tumor growth was measured by calipers in two dimensions. Tumor angiogenesis was determined with tumor microvessel density (MVD) by immunohistology. Tumor cell proliferation was assessed by Ki-67 staining. RESULTS: Intratumor injection of rAAV-KAL inhibited tumor growth in the treatment group by 78% (171 ± 52 mm3) at d 21 after virus infection compared to the control group (776 ± 241 mm3). | 10 | The Genesis Mechanism of the Mantle Fluid Action and Evolution in the Ore-Forming Process: A Case Study of the Laowangzhai Gold Deposit in Yunnan, China显示文摘Based on petrological studies of the wall rocks, mineralizing rocks, ores and veins from the Laowangzhai gold deposit, it is discovered that along with the development of silication, carbonation and sulfidation, a kind of black opaque ultra-microlite material runs through the spaces between grains, fissures and cleavages. Under observations of the electron microprobe, scanning electronic microscopy and energy spectrum, this kind of ultra-microlite material is confirmed to consist of ultra microcrystalline quartz, silicate, sulfides and carbonates, as well as rutile, scheelite and specularite (magnetite), showing characters of liquation by the analyses of SEM and energy spectrum. The coexistence of immiscibility and precipitating co-crystallization strongly suggests that the mineralizing fluid changed from the melt to the hydrothermal fluid. Combined with the element geochemical researches, it is realized that the ultra-microlite aggregate is the direct relics of the mantle fluid behaving like a melt and supercritical fluid, which goes along with the mantle-derived magma and will escape from the magma body at a proper time. | 11 | Genome-wide characterization and analysis of b HLH transcription factors in Panax ginseng显示文摘Ginseng(Panax ginseng C.A. Meyer) is one of the best-selling herbal medicines, with ginsenosides as its main pharmacologically active constituents. Although extensive chemical and pharmaceutical studies of these compounds have been performed, genome-wide studies of the basic helix-loop-helix(b HLH) transcription factors of ginseng are still limited. The b HLH transcription factor family is one of the largest transcription factor families found in eukaryotic organisms, and these proteins are involved in a myriad of regulatory processes. In our study, 169 bHLH transcription factor genes were identified in the genome of P. ginseng, and phylogenetic analysis indicated that these PGb HLHs could be classified into 24 subfamilies. A total of 21 RNA-seq data sets, including two sequencing libraries for jasmonate(JA)-responsive and 19 reported libraries for organ-specific expression analyses were constructed. Through a combination of gene-specific expression patterns and chemical contents,6 PGbHLH genes from 4 subfamilies were revealed to be potentially involved in the regulation of ginsenoside biosynthesis. These 6 PGbHLHs, which had distinct target genes, were further divided into two groups depending on the absence of MYC-N structure. | 12 | Global Pharmacopoeia Genome Database is an integrated and mineable genomic database for traditional medicines derived from eight international pharmacopoeias显示文摘Genomic data have demonstrated considerable traction in accelerating contemporary studies in traditional medicine. However,the lack of a uniform format and dispersed storage limits the full potential of herb genomic data. In this study, we developed a Global Pharmacopoeia Genome Database(GPGD). The database contains 34,346 records for 903 herb species from eight global pharmacopoeias(Brazilian, Egyptian, European, Indian, Japanese, Korean, the Pharmacopoeia of the People’s Republic of China, and U.S. Pharmacopoeia’s Herbal Medicines Compendium). In particular, the GPGD contains 21,872 DNA barcodes from 867 species, 2,203 organelle genomes from 674 species, 55 whole genomes from 49 species, 534 genomic sequencing datasets from 366 species, and 9,682 transcriptome datasets from 350 species. Among the organelle genomes, 534 genomes from 366 species were newly generated in this study. Whole genomes, organelle genomes, genomic fragments, transcriptomes, and DNA barcodes were uniformly formatted and arranged by species. The GPGD is publicly accessible at http://gffzzf938e9d610ce4c4ahx6pkuww5o5fn60on.ffgz.tsg.suse.edu.cn and serves as an essential resource for species identification, decomposition of biosynthetic pathways, and molecular-assisted breeding analysis. | 14 | Reversal of multi-drug resistance by pSUPER-shRNA-mdr1 in vivo and in vitro显示文摘AIM:To explore the possibility of reversing multi-drug resistance(MDR) to HepG2/mdr1 in vitro and in vivo with RNA interference(RNAi) . METHODS:HepG2/mdr1 was obtained by cloning the whole gene mdr1 into HepG2 cells.shRNA targeting sequence was designed to be homologous to the P-gp encoding MDR1 mRNA consensus sequence.pSUPERshRNA/mdr1 was constructed using the enzymedigested technique.HepG2/mdr1 cells were transfected with vectors of pSUPER-shRNA/mdr1 to measure their efficacy by real-time PCR for mdr1 mRNA,flow cytometry(FCM) for P-gp expression,and Rhodamine efflux,MTT method for HepG2/mdr1 function,respectively.In vivo,mice tumors were treated by injecting pSUPER-shRNA/mdr1 in situ and into intraabdominal cavity.Tumors were collected to create cell suspension and cryosections after chemothearpy with adiramycin and mytomycin.The cell suspension was incubated in RPMI-1640 supplemented with G418 to screen stable cells for appreciating the reversal of MDR. Cryosections were treated with immunohistochemistry technique to show the effectiveness of transfection and the expression of P-gp. RESULTS:pSUPER-shRNA/mdr1 was successfullyconstructed,which was confirmed by sequencing. The MDR phenotype of HepG2/mdr1 was decreased significantly in vitro transfection.HepG2/mdr1 showing its MDR was reversed notably in P-gp expression(11.0%vs 98.2%,P<0.01) .Real-time PCR showed that mRNA/mdr1 was lower in test groups than in control groups(18.73±1.33 vs 68.03±2.21,P<0.001) . Compared with HepG2,the sensitivity of HepG2/mdr1 and HepG2/mdr1-dsRNA cells to ADM was decreased by 1.64 times and 15.6 times,respectively. The accumulation of DNR in positive groups was decreased evidently.In vivo,the p-gp expression in positive groups was significantly lower than that in control groups(65.1%vs 94.1%,P<0.05) .The tumor suppressing rate in test groups was 57.8%. After chemotherapy,the growth rate in test groups was lower than that in control groups(700.14±35.61 vs 1659.70±152.54,P<0.05) .Similar results were also observed under fluorescence microscope,and confirmed by Image-Pro Plus 4.5 analysis. | 15 | A study on the Dushiling tungsten-copper deposit in the Miao'ershan-Yuechengling area, Northern Guangxi, China:Implications for variations in the mineralization of multi-aged composite granite plutons显示文摘The polymetallic Dushiling W-Cu deposit is a large, altered, skarn-type deposit, located in the northeastern part of the Miao'ershan-Yuechengling pluton, China. Two types of granite have been identified in the deposit: a medium-grained porphyritic biotite granite, and a medium- to fine-grained biotite granite. Both are spatially and temporally related to ore bodies, suggesting they may be the source of mineralization in the deposit. A medium- to fine-grained porphyritic biotite granite is exposed at the surface in the region of mineralization. U-Pb dating of zircons yielded magmatic ages of 423 Ma for the medium-grained porphyritic biotite granite and 421 Ma for the medium- to fine-grained porphyritic biotite granite, while a younger age(217 Ma) obtained for surface samples indicates later diagenesis. Thus, magmatism occurred during the Caledonian and Indosinian, respectively. The petrological and geochemical characteristics of the two Caledonian granites show that both are calc-alkaline and peraluminous.They are moderately enriched in Cs, Rb, U, and REE, and strongly depleted in Sr,Ba,P,and Ti; they show similar REE behavior,including negative Eu anomalies. These geochemical similarities suggest that the two granites were derived from the same source,although they were emplaced during different stages of the evolution of the magma. Furthermore, the granites are associated with mineralization, suggesting they were the source of mineralization in the Dushiling W-Cu deposit. Sm-Nd ages of scheelite from the Dushiling W-Cu deposit indicate that metallogenesis occurred at 417±35 Ma, while the two types of titanite, intergrown with scheelite, yield U-Pb ages of 423–425 Ma(in altered granite sample) and 218 Ma(in skarn sample). These ages place the main mineralization event in the late Caledonian, and later magmatic-hydrothermal activity occurred in the Indosinian. | 16 | A telomere-to-telomere gap-free reference genome of watermelon and its mutation library provide important resources for gene discovery and breeding显示文摘Watermelon,Citrullus lanatus,is the world's third largest fruit crop.Reference genomes with gaps and a narrow genetic base hinder functional genomics and genetic improvement of watermelon.Here,we report the assembly of a telomere-to-telomere gap-free genome of the elite watermelon inbred line G42 by incorporating high-coverage and accurate long-read sequencing data with multiple assembly strategies.All 11 chromosomes have been assembled into single-contig pseudomolecules without gaps,representing the highest completeness and assembly quality to date.The G42 reference genome is 369321829 bp in length and contains 24205 predicted protein-coding genes,with all 22 telomeres and 11 centromeres characterized.Furthermore,we established a pollen-EMS mutagenesis protocol and obtained over 200000M1 seeds from G42.In a sampling pool,48 monogenic phenotypic mutations,selected from 223M1and 78 M2 mutants with morphological changes,were confirmed.The average mutation density was 1 SNP/1.69Mband1 indel/4.55 Mb per M1 plant and 1SNP/1.08Mb and 1 indel/6.25 Mb per M2 plant.Taking advantage of the gap-free G42 genome,8039 mutations from 32 plants sampled from M1 and M2 families were identified with 100%accuracy,whereas only 25% of the randomly selected mutations identified using the 97103v2 reference genome could be confirmed.Using this library and the gap-free genome,two genes responsible for elongated fruit shape and male sterility(CiMs1)were identified,both caused by a single basechange from G to A.The validated gap-free genome and its EMS mutation library provide invaluable resources for functional genomics and genetic improvement of watermelon. | Yun Deng Shoucheng Liu Yilin Zhang Jingsheng Tan Xiaopeng Li Xiao Chu Binghua Xu Yao Tian Yudong Sun Bosheng Li YunbiXu Xing Wang Deng Hang He Xingping Zhang | 2022 | Molecular Plant2022,15,8: | 9 |
| 17 | Repeatability and Reproducibility of Quantitative Assessment of the Retinal Microvasculature Using Optical Coherence Tomography Angiography Based on Optical Microangiography显示文摘Objective The aim of this study was to determine the repeatability and reproducibility of optical coherence tomography angiography(OCTA) based on optical microangiography(OMAG) measurements of macular vessels in normal eyes. Methods In this prospective cohort study, 40 eyes of 40 healthy volunteers underwent repeated OCTA(Cirrus HD-OCT 5000 angiography system, Carl Zeiss Meditec, Inc.) scans on two separate visit days. On each visit day, the eyes were scanned three times. The following parameters were used to quantitatively describe the OCTA images of the superficial vascular network: vessel area density(VAD), vessel skeleton density(VSD), vessel diameter index(VDI), vessel perimeter index(VPI), vessel complexity index(VCI), flux, and foveal avascular zone(FAZ). Coefficient of variation(CV) and intraclass correlation coefficient(ICC) were calculated for evaluating intravisit and intervisit repeatability, as well as interobserver reproducibility. Results The measurements showed high repeatability [CVs ≤ 4.2%(intravisit) and ≤ 4.6%(intervisit)] and interobserver reproducibility(ICCs ≥ 0.923) for all parameters. | 18 | Cholesterol transport through the peroxisome-ER membrane contacts tethered by PI(4,5)P2 and extended synaptotagmins显示文摘Most mammalian cells take up cholesterol from low-density lipoproteins(LDLs) via receptor-mediated endocytosis.After reaching lysosomes,LDL-derived cholesterol continues to transport to downstream organelles including the ER for specific structural and functional needs.Peroxisomes are recently found to receive cholesterol from lysosomes through lysosomeperoxisome membrane contacts.However,whether and how cholesterol is conveyed from peroxisomes to the ER remain unknown.Here,by combining high-resolution microscopic analyses and in vitro reconstitution of highly purified organelles or artificial liposomes,we demonstrate that peroxisomes form membrane contacts with the ER through the interaction between peroxisomal PI(4,5)P2 and ER-resident extended synaptotagmin-1,2 and 3(E-Syts).Depletion of peroxisomal PI(4,5)P2 or ESyts markedly decreases peroxisome-ER membrane contacts and induces cholesterol accumulation in lysosomes.Furthermore,we show that cholesterol is delivered from 3H-labeled peroxisomes or PI(4,5)P2-containing liposomes to the ER in vitro,and that the presence of peroxisomes augments cholesterol transfer from lysosomes to the ER.Together,our study reveals a new cholesterol transport pathway along the lysosome-peroxisome-ER membrane contacts in the cell. | Jian Xiao Jie Luo Ao Hu Ting Xiao Meixin Li Zekai Kong Luyi Jiang Zimu Zhou Yacheng Liao Chang Xie Beibei Chu Honghua Miao Boliang Li Xiongjie Shi Bao-Liang Song | 2019 | Science China(Life Sciences)2019,62,9: | 8 |
