1A brief history of the discovery of PXR and CAR as xenobiotic receptors显示文摘The nuclear receptors pregnane X receptor(PXR) and constitutive androstane receptor(CAR) were cloned and/or established as xenobiotic receptors in 1998.Due to their activities in the transcriptional regulation of phase I and phase II enzymes as well as drug transporters,PXR and CAR have been defined as the master regulators of xenobiotic responses.The discovery of PXR and CAR provides the essential molecular basis by which drugs and other xenobiotic compounds regulate the expression of xenobiotic enzymes and transporters.This article is intended to provide a historical overview on the discovery of PXR and CAR as xenobiotic receptors.Jiong Yan Wen Xie 2016Acta Pharmaceutica Sinica B2016,6,5:9

3Marine spatio-temporal process semantics and its applications-taking the El Nio Southern Oscilation process and Chinese rainfall anomaly as an example显示文摘Spatio-temporal semantics based on 'object views' or 'event views' has few abilities to represent and model the continuity and gradual oceanic phenomena or objects,which seriously limits the specific marine applications and knowledge discovery and data mining,so this paper proposes a hierarchical abstraction semantics with 'marine spatio-temporal process → life span phases → evolution sequences → state units' and process objects included by level with 'marine process objects → phase objects → sequence object → state objects' with the oceanic process characteristics into the marine process semantics.In addition,this paper designs the storage and representation of marine process objects using the backus normal forms (BNF) and abstract data type (ADT).Base on El Nin o Southern Oscilation (ENSO) index and Chinese rain gauging station data,this paper also gives a case of study.The spatio-temporal analysis between ENSO process and Chinese rainfall anomalies shows that the marine spatio-temporal semantics not only can illustrate the spatial distribution of Chinese rainfall anomalies in different time scales at ENSO process,life span phases and state units,but also analyze the dynamic changes of Chinese rainfall anomalies in different life span phases or state units within ENSO evolution.XUE Cunjin DONG Qing XIE Jiong 2012Acta Oceanologica Sinica2012,31,2:4

6Kupffer cells contribute to concanavalin A-induced hepatic injury through a Th1 but not Th17 type response-dependent pathway in mice显示文摘BACKGROUND:Increasing evidence suggests that a close interaction of Kupffer cells with T cells plays a central role in concanavalin A-induced hepatic injury in mice,but the underlying mechanisms remain obscure.The present study aimed to determine the relative roles of Th1 and Th17 type responses in concanavalin A-induced hepatic injury in mice, and to investigate whether or not Kupffer cells contribute to hepatic injury via a Th1 or Th17 type response-dependent pathway. METHODS:Immune-mediated hepatic injury was induced in C57BL/6 mice by intravenous injection of concanavalin A. Kupffer cells were inactivated by pretreatment with gadolinium chloride 24 hours before the concanavalin A injection.The interferon-gamma(IFN-γ)and interleukin-17(IL-17)pathways were blocked by specific neutralizing antibodies.Hepatic injury was assessed using serum transferase activity and pathological analysis.Expression of inflammatory cytokines within the liver was detected by real-time polymerase chain reaction and immunohistochemistry. 7Characteristics of β-oxidative and reductive metabolism on the acyl side chain of cinnamic acid and its analogues in rats显示文摘Cinnamic acid and its analogues (pyragrel and ozagrel) undergo chain-shortened (β-oxidative) and reductive metabolism on acyl side chain. In this study, we characterized the β-oxidative and reductive metabolism on acyl side chain of cinnamic acid and its analogues using primary rat hepatocytes, hepatic mitochondrial, and microsomal systems. A compartmental model including parent compounds and metabolites was developed to characterize in vivo β-oxidative and reductive metabolism following an intravenous dose of parent compounds to rats. The ?tted total in vivo clearance values were further compared with the in vitro values predicted by the well-stirred model. We showed that hepatic microsomal CYP450s did not catalyze β-oxidative or reductive metabolism of the three compounds. Similar to β-oxidation of fatty acids,β-oxidative metabolism on their acyl side chain occurred mainly in mitochondria, which was highly dependent on ATP, CoA and NAD+. Fatty acids and NADH inhibited the β-oxidative metabolism. Reductive metabolism occurred in both mitochondria and microsomes. Reduction in mitochondria was ATP-, CoA-, and NAD(P)H-dependent and reversible, which was suppressed by enoyl reductase inhibitor triclosan. Reduction in microsomes was ATP-, CoA-, and NADPH-dependent but little affected by triclosan. Both plasma concentrations of β-oxidative metabolites and reductive metabolites were successfully ?tted using the compartmental model. The estimated total in vivo clearance values were consistent with those predicted from hepatocytes and organelles, implicating signi?cance of in vitro kinetics. 10Assessment of biological functions for C3A cells interacting with adverse environments of liver failure plasma显示文摘Background:For its better differentiated hepatocyte phenotype,C3A cell line has been utilized in bioar-tificial liver system.However,up to now,there are only a few of studies working at the metabolic alter-nations of C3A cells under the culture conditions with liver failure plasma,which mainly focus on car-bohydrate metabolism,total protein synthesis and ureagenesis.In this study,we investigated the effects of acute liver failure plasma on the growth and biological functions of C3A cells,especially on CYP450 enzymes.Methods:C3A cells were treated with fresh DMEM medium containing 10%FBS,fresh DMEM medium containing 10%normal plasma and acute liver failure plasma,respectively.After incubation,the C3A cells were assessed for cell viabilities,lactate dehydrogenase leakage,gene transcription,protein levels,albu-min secretion,ammonia metabolism and CYP450 enzyme activities.Results:Cell viabilities decreased 15%,and lactate dehydrogenase leakage had 1.3-fold elevation in acute liver failure plasma group.Gene transcription exhibited up-regulation,down-regulation or stability for different hepatic genes.In contrast,protein expression levels for several CYP450 enzymes kept constant,while the CYP450 enzyme activities decreased or remained stable.Albumin secretion reduced about 48%,and ammonia accumulation increased approximately 41%.Conclusions:C3A cells cultured with acute liver failure plasma showed mild inhibition of cell viabilities,reduction of albumin secretion,and increase of ammonia accumulation.Furthermore,CYP450 enzymes demonstrated various alterations on gene transcription,protein expression and enzyme activities.Zu-Hong Li Zhong-Yang Xie Xiao-Xi Ouyang Kai-Zhou Huang Xiao-Peng Yu Ya-Lei Zhao Yan-Hong Zhang Dan-Hua Zhu Jiong Yu Lan-Juan Li 2020Hepatobiliary & Pancreatic Diseases International2020,19,2:1

15Deciphering the roles of the constitutive androstane 'eceptor in energy metabolism显示文摘The constitutive androstane receptor (CAR) is initially defined as a xenobiotic nuclear receptor that protects the liver from injury. Detoxification of damaging chemicals iS achieved by CAR-mediated induction of drug-metabolizing enzymes and transporters. More recent research has implicated CAR in energy metabolism, suggesting a therapeutic potential for CAR in metabolic diseases, such as type 2 diabetes and obesity. A better understanding of the mechanisms by which CAR regulates energy metabolism will allow us to take advantage of its effectiveness while avoiding its side effects. Both the boundaries and the cracks are modeled by distributed dislocations. This method results in a system of singular integral equations with Cauchy kernels which can be solved by Gauss-Chebyshev quadrature method. 19Detecting benign uterine tumors by autofuorescence lifetime imaging microscopy through adjacent healthy cervical tissues显示文摘The endogenous fluorophores such as reduced nicotinarmide adenine dinucleotide(phosphate)(NAD(P)H)and favin adenine dinucleotide(FAD)in cells and tissues can be imaged by fuo-rescence lifetime imaging microscopy(FLIM)to show the tisute morphology features,as well as the biomolecular changes in microenvironment.The two important coenzymes in cellular metabolism,NAD(P)H and FAD,can be used to monitor the cellular metabolic status.This work proposed a novel method to study the uterine metabolism at the adjacent site of healthy cervix.It was found that the benign uterine tumors such as leiomyomas and adenomyosis with abnormal cell growth can be detected by measuring the fluorescence lifetime of NAD(P)H and FAD in adjacent healthy cervical tisues.This method opened a novel strategy for alicted women to undergo the cervical biopsies instead of hysterectomies for detecting tumors,which can preserve the fertility of patients.The FLIM studying on NAD(P)H and FAD indicated the correlation between metabolism and some diseases,including diabetes,hyperthyroidism and obesity.It was also suggested that the metabolic level might be quite different for a patient with a malignant tumnor history.Maojia Huang Zixiao Zhang Xinyi Wang Yonghui Xie Yiyan Fei Jiong Ma Jing Wang Li Chen Lan Mi Yulan Wang 2019Journal of Innovative Optical Health Sciences2019,,5:0
