2Molecular screening of patients with nonsyndromic hearing loss from Nanjing city of China显示文摘Hearing loss is the most frequent sensory disorder involving a multitude of factors,and at least 50% of cases are due to genetic etiology.To further characterize the molecular etiology of hearing loss in the Chinese population,we recruited a total of 135 unrelated patients with nonsyndromic sensorineural hearing loss (NSHL) for mutational screening of GJB2,GJB3,GJB6,SLC26A4,SLC26A5 IVS2-2A>G and mitochondrial 12SrRNA,tRNA Ser(UCN) by PCR amplification and direct DNA sequencing.The carrier frequencies of deafness-causing mutations in these patients were 35.55% in GJB2,3.70% in GJB6,15.56% in SLC26A4 and 8.14% in mitochondrial 12SrRNA,respectively.The results indicate the necessity of genetic screening for mutations of these causative genes in Chinese population with nonsyndromic hearing loss.Yajie Lu Dachun Dai Zhibin Chen Xin Cao Xingkuan Bu Qinjun Wei Guangqian Xing 2011The Journal of Biomedical Research2011,25,5:30
3Phytochemistry and pharmacology of genus Ephedra显示文摘The genus Ephedra of the Ephedraceae family contains more than 60 species of nonflowering seed plants distributed throughout Asia, America, Europe, and North Africa. These Ephedra species have medicinal, ecological, and economic value. This review aims to summarize the chemical constituents and pharmacological activities of the Ephedra species to unveil opportunities for future research. Comprehensive information on the Ephedra species was collected by electronic search(e.g., GoogleScholar, Pubmed, Sci Finder, and Web of Science) and phytochemical books. The chemical compounds isolated from the Ephedra species include alkaloids, flavonoids, tannins, polysaccharides, and others. The in vitro and in vivo pharmacological studies on the crude extracts, fractions and few isolated compounds of Ephedra species showed anti-inflammatory, anticancer, antibacterial, antioxidant, hepatoprotective, anti-obesity, antiviral, and diuretic activities. 4Depth of tumor invasion and tumor-occupied portions of stomach are predictive factors of intra-abdominal metastasis显示文摘Objective:Diagnostic laparoscopy is recommended for the pretherapeutic staging of gastric cancer to detect any unexpected or unconfirmed intra-abdominal metastasis.The aim of this study was to evaluate the role and indications of diagnostic laparoscopy in the detection of intra-abdominal metastasis.Methods:Standard diagnostic laparoscopy with peritoneal cytology examination was performed prospectively on patients who were clinically diagnosed with primary local advanced gastric cancer(c T≥2M0).We calculated the rate of intra-abdominal metastases identified by diagnostic laparoscopy,and examined the relationship between peritoneal dissemination(P)and cytology results(CY).Split-sample method was applied to find clinical risk factors for intra-abdominal metastasis.Multivariate logistic regression analysis and receiver-operator characteristic(ROC)analysis were performed in training set to find out risk factors of intra-abdominal metastasis,and then validate it in testing set.Results:Out of 249 c M0 patients,51(20.5%)patients with intra-abdominal metastasis were identified by diagnostic laparoscopy,including 20(8.0%)P1CY1,17(6.8%)P0CY1 and 14(5.6%)P1CY0 patients.In the training set,multivariate logistic regression analysis and ROC analysis showed that the depth of tumor invasion on computer tomography(CT)scan≥21 mm and tumor-occupied≥2 portions of stomach are predictive factors of metastasis.In the testing set,when diagnostic laparoscopy was performed on patients who had one or two of these risk factors,the sensitivity and positive predictive value for detecting intra-abdominal metastasis were 90.0%and32.1%,respectively.Conclusions:According to our results,depth of tumor invasion and tumor-occupied portions of stomach are predictive factors of intra-abdominal metastasis.Ziyu Li Zhemin Li Shuqin Jia Zhaode Bu Lianhai Zhang Xiaojiang Wu Shuangxi Li Fei Shan Xin Ji Jiafu Ji 2017Chinese Journal of Cancer Research2017,29,2:14
5Construction and external validation of a nomogram that predicts lymph node metastasis in early gastric cancer patients using preoperative parameters显示文摘Objective: To create a nomogram to predict the incidence of lymph node metastasis(LNM) in early gastric cancer(EGC) patients and to externally validate the nomogram.Methods: To construct the nomogram,we retrospectively analyzed a primary cohort of 272 EGC patients.Univariate analysis and a binary logistic regression were performed.A nomogram predicting the incidence of LNM in EGC patients was created.The discrimination ability of the nomogram was measured using the concordance index(c-index),and the nomogram was also calibrated.Then,another prospective cohort of 81 patients was analyzed to validate the nomogram.Results: In the primary cohort,LNM was pathologically confirmed in 37(13.6%) patients.In multivariate analysis,the presence of an ulcer,the maximum lesion diameter observed via gastroscopy,the thickness of the lesion observed via endoscopic ultrasonography,and the presence of enlarged lymph nodes on computed tomography(CT) were independent risk factors for LNM.A nomogram was then created based on the regression model with the c-index of 0.905,and the calibration curve of the nomogram fell approximately on the ideal 45-degree line.The cut-off score of the nomogram was 110,and the sensitivity,specificity,positive predictive and negative predictive values of the nomogram in the primary cohort were 81.1%,86.0%,47.6% and 96.7%,respectively,and in the prospective validation cohort were 75.0%,91.0%,60.0% and 95.5%,respectively.The calibration curve of the external validation cohort was almost on the 45-degree line.Conclusions: We developed an effective nomogram predicting the incidence of LNM for EGC patients.Yinan Zhang Yiqiang Liu Ji Zhang Xiaojiang Wu Xin Ji Tao Fu Ziyu Li Qi Wu Zhaode Bu Jiafu Ji 2018Chinese Journal of Cancer Research2018,30,6:12
6Comorbidity Burden of Dementia:A Hospital-Based Retrospective Study from 2003 to 2012 in Seven Cities in China显示文摘Dementia is increasing dramatically and imposes a huge burden on society. To date, there is a lack of data on the health status of patients with dementia in China. In an attempt to investigate the comorbidity burden of dementia patients in China at the national level, we enrolled 2,938 patients with Alzheimer's disease(AD), vascular dementia(Va D), or other types of dementia, who were admitted to tertiary hospitals in seven regions of China from January2003 to December 2012. The Charlson Comorbidity Index(CCI) was used to evaluate the comorbidity burden of the patients with dementia. Among these patients, 53.4% had AD, 26.3% had Va D, and 20.3% had other types of dementia. The CCI was 3.0 ± 1.9 for all patients,3.4 ± 1.8 for those with Va D, and 3.0 ± 2.1 for those with AD. The CCI increased with age in all patients, andthe length of hospital stay and daily expenses rose with age and CCI. Males had a higher CCI and a longer stay than females. Moreover, patients admitted in the last 5 years of the study had a higher CCI than those admitted in the first 5 years. We found that the comorbidity burden of patients with dementia is heavy. 7Microstructures and tensile properties of Mg-Zn-(Gd)-Zr alloys extruded at various temperatures显示文摘The microstructures and tensile properties of Mg-6.0 Zn-0.6 Zr(ZK60) and ZK60 + 1.8 Gd(ZEK620(RE = Gd)) alloys extruded at 290, 305, 320, 340 and360 ℃ were investigated. The results indicate that Gd addition can refine the microstructures of ZK60 alloy and the extrusion parameters such as the extrusion temperature will influence the tensile properties of both as-extruded ZK60 and ZEK620 alloys, although it is much slighter for ZK60 alloy. And the sensitivity is closely related to the textures developed during extrusion. In addition, Gd addition will also clearly improve the tensile strength at room temperature(24 ℃) and particularly at high temperatures(150 and 200 ℃), due to the much finer grains and smaller intermetallic particles. The highest strength is achieved in the as-extruded ZEK620 alloy with extrusion temperature of 320 ℃, with ultimate tensile strength(UTS) and tensile yield strength(TYS) of 405 and375 MPa, respectively, which are improved by approximately 9% and 11% than those of the as-extruded ZK60 alloy which owns the highest strength at extrusion temperature of 290 ℃.Xin Qiu Qiang Yang Kai Guan Fanqiang Bu Zhan-Yi Cao Yong-Bing Liu Jian Meng 2017Rare Metals2017,36,12:9
8Gangba sheep in the Tibetan plateau:Validating their unique meat quality and grazing factor analysis显示文摘Gangba sheep are known for having typical sensory characteristics attributed to free range conditions and grazing on wild plants.The genuine Gangba mutton was selected as the experimental group,and the commercial Tibetan mutton was selected as the control group,the nutritive composition of basic chemical components,amino acids and fatty acids in mutton were investigated to correlate its unique meat quality and eating satisfaction.The results showed that fatty acids were significantly higher(P<0.05)in Gangba mutton than in commercial mutton,and the higher content of flavoring amino acids(glutamic acid and aspartame)were primarily responsible for the taste attributes umami of meat juices.Moreover,the trace elements analysis in mutton and grazing factors(forage,water source and soil)were conducted,to explain the source of essential trace elements in mutton.The concentrations of essential trace elements show that the Gangba mutton was a valuable source for highly available Cu and Zn in human nutrition,and well managed with few detected of toxicity metal.The concentrations of essential trace elements in mutton are closely related to the trace elements in environmental grazing factors.In conclusion,the congenital grazing conditions(a highly mineralized water resource,natural forages and clean soils)were shown to contribute to the unique meat characteristics of Gangba sheep.Qiangying Zhang Ming Que Wei Li Shuang Gao Xin Tan Duo Bu 2021Journal of Environmental Sciences2021,33,3:8

10The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 11Treatment results and prognostic factors of patients undergoing postoperative radiotherapy for laryngeal squamous cell carcinoma显示文摘Postoperative radiotherapy (PRT) is widely advocated for patients with squamous cell carcinomas of the head and neck that are considered to be at high risk of recurrence after surgical resection. The aims of this study were to evaluate the treatment outcomes of PRT for patients with laryngeal carcinoma and to identify the value of several prognostic factors. We reviewed the records of 256 patients treated for laryngeal squamous cell carcinoma between January 1993 and December 2005. Disease-free survival (DFS) and overall survival (OS) were estimated using the Kaplan-Meier method. Log-rank test was employed to identify significant prognostic factors for DFS and OS. The Cox proportional hazards model was applied to identify covariates significantly associated with the aforementioned endpoints. Our results showed the 3-, 5-, and 10-year DFS for all patients were 69.9%, 59.5%, and 34.9%, respectively. The 3-, 5-, and 10-year OS rates were 80.8%, 68.6%, and 38.8%, respectively. Significant prognostic factors for both DFS and OS on univariate analysis were grade, primary site, T stage, N stage, overall stage, lymph node metastasis, overall treatment times of radiation, the interval between surgery and radiotherapy, and radiotherapy equipment. Favorable prognostic factors for both DFS and OS on multivariate analysis were lower overall stage, no cervical lymph node metastasis, and using 60Co as radiotherapy equipment. 15A Landscape Pattern Analysis Method Based on Boundaries and Nodes:A Case Study in Upper Minjiang River,China显示文摘Traditionally,patch-based analysis at the landscape scale fragmentation has been used in the study of landscape ecology while the study of boundary and node has not been considered as much detail until recently.This study investigated the possibility of applying boundary-and node-based methods in landscape pattern analysis to the upper reaches of the Minjiang River in the southwestern China.Boundary-based and node-based landscape indices were selected to be used in analyzing changes in landscape patterns,and the results were compared with analysis using traditional pattern indices.We compared the responses of patch-area-based,boundary-length-based and node-number-based indices,and concluded that boundary-based and node-based indices are more sensitive to disturbance than patch-based indices with various patterns,and node-based indices are even more sensitive than boundary-based ones.Thus,the results suggest that boundary-based and node-based pattern analysis methods provide helpful supplementary information to traditional patch-based pattern analysis methods.The results about pattern dynamics of landscapes in the upper reaches of the Minjiang River based on boundaries and nodes showed that with human disturbance,the dominance of forest landscape was weakened by other landscape types;thus the landscape pattern of the study area became more homogeneous and the boundary network became more complex.These changes further augmented disturbance interfaces in the landscape and increased the possibilities of further landscape fragmentation.WEN Qingchun LI Xiuzhen HE Hongshi HU Yuanman CHEN Xin CHANG Yu WANG Wei BU Rencang 2014Chinese Geographical Science2014,24,1:3
16Diagnostic value of negative enrichment and immune fluorescence in situ hybridization for intraperitoneal free cancer cells of gastric cancer显示文摘Objective:To explore the intraperitoneal free cancer cell(IFCC)detection value of negative enrichment and immune fluorescence in situ hybridization(NEimFISH)on chromosomes(CEN)8/17.Methods:To verify the reliability of NEimFISH,29 gastric cancer tumors,their adjacent tissues and greater omental tissues were tested.Our study then included 105 gastric cancer patients for IFCC.We defined patients as IFCC-positive if a signal was detected,regardless of the detailed cancer cell numbers.A comparison of clinicopathological features was conducted among IFCC groups.We also compared the diagnosis value and peritoneal recurrence predictive value among different detection methods.The comparison of IFCC number was also conducted among different groups.Results:A cutoff of 2.5 positive cells could distinguish all benign tissue samples and 97%of malignant tissue samples in our study.Compared to intestinal gastric cancer,patients with diffuse gastric cancer tended to have more IFCCs(6 vs.4,P=0.002).The IFCC counts were often higher in the lymphovascular invasion positive group than negative group(3 vs.1,P=0.022).All IFCC samples that were considered positive using conventional cytology were also found to be positive using NEimFISH.When compared to conventional cytology and paraffin pathology,NEimFISH had a higher IFCC positive rate(68.9%)and higher one-year peritoneal recurrence predictive value with area under the curve(AUC)of 0.922.Conclusions:Gastric cancer could be effectively diagnosed by NEimFISH.The IFCC number found using NEimFISH on CEN8/17 is closely associated with Lauren type and vascular invasion of cancer.NEimFISH is a reliable detection modality with a higher positive detection rate,higher one-year peritoneal recurrence predictive value and quantitative features for IFCC of gastric cancer.Anqiang Wang Zhongwu Li Qian Wang Yali Bai Xin Ji Tao Fu Ke Ji Yanwen Xue Tingxu Han Xiaojiang Wu Ji Zhang Yingjie Yang Guobin Xu Zhaode Bu Jiafu Ji 2019Chinese Journal of Cancer Research2019,31,6:3
17Natural variations in grain length 10(GL10)regulate rice grain size显示文摘Grain size is an important determinant of grain weight and yield in rice.Although several genes related to grain size have been identified,natural variations in these genes that affect grain size are poorly characterized.Here,we describe the grain length QTL GL10,encoding MADS56,which positively regulates grain length and grain weight.A natural allelic variation of NIL-gl10,containing an~1.0-kb deletion in the first exon that abolishes its transcription,results in shorter grain length,lower grain weight and delayed flowering in gl10 plants.The knockout of GL10 in the HJX74 background leads to grain phenotypes similar to that of NIL-gl10,while overexpression of GL10 results in increased grain length and weight and earlier heading date.GL10 regulates grain length by promoting greater longitudinal cell growth in the grain glume.Additionally,GL10 participates in the regulation of gibberellic acid(GA)signaling pathway genes in young panicle tissues.Analysis of GL10 haplotypes shows obvious divergence between the japonica and indica lineages.Our findings reveal an allelic variation of GL10 that may explain differences in grain length among modern cultivars and could be used to breed rice varieties with optimized grain shape.Penglin Zhan Shuaipeng Ma Zhili Xiao Fangping Li Xin Wei Shaojun Lin Xiaoling Wang Zhe Ji Yu Fu Jiahao Pan Mi Zhou Yue Liu Zengyuan Chang Lu Li Suhong Bu Zupei Liu Haitao Zhu Guifu Liu Guiquan Zhang Shaokui Wang 2022Journal of Genetics and Genomics2022,49,5:3
18Current sustained delivery strategies for the design of local neurotrophic factors in treatment of neurological disorders显示文摘Although therapeutic potential of neurotrophic factors(NTFs)has been well recognized for over two decades,attempts to translate that potential to the clinic have been disappointing,largely due to significant obstacles in delivery,including inadequate protein dose/kinetics released at target sites.Considerable efforts have been made to improve the therapeutic performance of NTFs.This articles reviews recent developments in localized delivery systems of NTFs for the neurological disorders treatments with a main focus on sustained delivery strategies.Different non-covalent binding approaches have been employed to immobilize proteins in hydrogels,microspheres,electrospun nanofibers,and their combined systems,which serve as depots for sustained local release of NTFs.The challenges associated with current NTFs delivery systems and how these systems can be applied to neurological diseases and disorders have been discussed in the review.In conclusion,optimal delivery systems for NTFs will be needed for reliable and meaningful clinical benefits;ideally,delivering a time and dose-controlled release of bioactive multiNTFs at different individual optimal kinetics to achieve multi-functions in target tissues is significant preferred.Hongzhuo Liu Yanyan Zhou Shichao Chen Meng Bu Jiayu Xin Sanming Li 2013Asian Journal of Pharmaceutical Sciences2013,8,5:2
19Selective sensing of Cr^(VI) and Fe^(Ⅲ) ions in aqueous solution by an exceptionally stable Tb^(Ⅲ)-organic framework with an AIE-active ligand显示文摘We herein report a new lanthanide metal-organic framework(MOF) that exhibits excellent chemical stability,especially in the aqueous solution over a wide pH range from 1 to 14.In contrast to many reported lanthanide MOFs,this Tb-based MOF emits cyan fluorescence inherited from the integrated AIEactive ligand,rather than Ln3+ ions.More remarkably,its fluorescence signal features a highly selective and sensitive 'turn-off' response toward CrO_(4)^(2-),Cr_(2)O_(7)^(2- )and Fe^(3+) ions,highlighted with the low detection limits down to 68.18,69.85 and 138.8 ppm,respectively.Thus,the exceptional structural stability and sensing performance render this material able to be a superior luminescent sensor for heavy metal ions in wastewater.Jing-Jing Pang Rui-Huan Du Xin Lian Zhao-Quan Yao Jian Xu Xian-He Bu 2021Chinese Chemical Letters2021,32,8:2
20A durable 4-1BB-based CD19 CAR-T cell for treatment of relapsed or refractory non-Hodgkin lymphoma显示文摘Objective:Previous studies reported that 4-1BB-based CD19 chimeric antigen receptor(CAR)-T cells were more beneficial for the clinical outcomes than CD28-based CAR-T cells,especially the lower incidence rate of severe adverse events.However,the median progression-free survival(mPFS)of 4-1BB-based product Kymriah was shorter than that of CD28-based Yescarta(2.9 months vs.5.9 months),suggesting that Kymriah was limited in the long-term efficacy.Thus,a safe and durable 4-1BB-based CD19 CAR-T needs to be developed.Methods:We designed a CD19-targeted CAR-T(named as IM19)which consisted of an FMC63 scFv,4-1BB and CD3ζintracellular domain and was manufactured into a memory T-enriched formulation.A phase I/II clinical trial was launched to evaluate the clinical outcomes of IM19 in relapsed or refractory(r/r)B cell non-Hodgkin lymphoma(B-NHL).Dose-escalation investigation(at a dose of 5×10^(5)/kg,1×10^(6)/kg and 3×106/kg)was performed in 22 r/r B-NHL patients.All patients received a single infusion of IM19 after 3-day conditional regimen.Results:At month 3,the overall response rate(ORR)was 59.1%,the complete response rate(CRR)was 50.0%.The mPFS was 6 months and the 1-year overall survival rate was 77.8%.Cytokine release syndrome(CRS)occurred in 13 patients(59.1%),with 54.5%of grade 1−2 CRS.Only one patient(4.5%)experienced grade 3 CRS and grade 3 neurotoxicity.Conclusions:These results demonstrated the safety and durable efficacy of a 4-1BB-based CD19 CAR-T,IM19,which is promising for further development and clinical investigation.Zhitao Ying Ting He Shanzhao Jin Xiaopei Wang Wen Zheng Ningjing Lin Meifeng Tu Yan Xie Lingyan Ping Weiping Liu Lijuan Deng Yanping Ding Xuelian Hu Bing Bu Xin’an Lu Yuqin Song Jun Zhu 2022Chinese Journal of Cancer Research2022,34,1:2
