| 1 | In vitro culture and differentiation of rat embryonic midbrain-derived neural stem cells显示文摘BACKGROUND:Midbrain-derived neural stem cells (mNSCs) can differentiate into functional mature dopaminergic neurons. The mNSCs are considered the ideal choice for cell therapy of Parkinson’s disease. OBJECTIVE: To isolate rat embryonic mNSCs and to observe the differentiation characteristics of mNSCs induced by cell growth-promoting factors. DESIGN,TIME AND SETTING: An in vitro cell culture study based on the molecular biology of nerve cells was carried out at the Institute of Clinical Medicine,China-Japan Friendship Hospital (China) from March to November 2007. MATERIALS: Sprague Dawley rats at embryonic day 14 were used in this study. Nestin antibody,β-Ⅲ tubulin antibody,glial fibrillary acidic protein (GFAP) antibody and cyclic nucleotide 3’-phosphohydrolase (CNPase) antibody were provided by Abcam; DMEM/F12 medium and N2 supplement were provided by In-vitrogen; epidermal growth factor (EGF) and fibroblast growth factor-2 (FGF2) were provided by R&D Sys-tems. METHODS: The ventral mesencephalon was dissected from embryonic day 14 rat embryos. By trypsin di-gestion and mechanical separation,the brain tissue was triturated into a fine single-cell suspension. The cells were cultured in 5 mL serum-free medium containing DMEM/F12,1% N2 supplement,20 ng/mL EGF and FGF2. The mNSCs at the third generation were coated with 10 μg/mL polylysine and induced to differenti-ate in the DMEM/F12 supplemented with 1% fetal bovine serum and 1% N2. MAIN OUTCOME MEASURES: The neural spheres of the third passage were identified by nestin im-munofluorescence; at the same time,the cells were induced to differentiate,and the types of differentiated cell were identified by immunofluorescence for βⅢ tubulin,GFAP and CNPase. RESULTS: Seven days after primary culture,a great many neurospheres could be obtained by successive pasage. Immunofluorescence assays showed that the neurospheres were nestin positive,and after differentia-tion,the cells expressed GFAP,CNPase and β-Ⅲ-tubulin. | 2 | Direct observation of ultrafast plasmonic hot electron transfer in the strong coupling regime显示文摘Achieving strong coupling between plasmonic oscillators can significantly modulate their intrinsic optical properties.Here,we report the direct observation of ultrafast plasmonic hot electron transfer from an Au grating array to an MoS_(2) monolayer in the strong coupling regime between localized surface plasmons(LSPs)and surface plasmon polaritons(SPPs).By means of femtosecond pump-probe spectroscopy,the measured hot electron transfer time is approximately 40 fs with a maximum external quantum yield of 1.65%.Our results suggest that strong coupling between LSPs and SPPs has synergetic effects on the generation of plasmonic hot carriers,where SPPs with a unique nonradiative feature can act as an‘energy recycle bin’to reuse the radiative energy of LSPs and contribute to hot carrier generation.Coherent energy exchange between plasmonic modes in the strong coupling regime can further enhance the vertical electric field and promote the transfer of hot electrons between the Au grating and the MoS_(2) monolayer.Our proposed plasmonic strong coupling configuration overcomes the challenge associated with utilizing hot carriers and is instructive in terms of improving the performance of plasmonic opto-electronic devices. | Hangyong Shan Ying Yu Xingli Wang Yang Luo Shuai Zu Bowen Du Tianyang Han Bowen Li Yu Li Jiarui Wu Feng Lin Kebin Shi Beng Kang Tay Zheng Liu Xing Zhu Zheyu Fang | 2019 | Light(Science & Applications)2019,8,1: | 7 |

| 4 | Evaluation of coronary plaque and stent deployment by intravascular optical coherence tomography in elderly patients with unstable angina and non-ST-elevation myocardial infarction显示文摘Objective To evaluate the feasibility and efficacy of intravascular optical coherence tomography (OCT) in the assessment of plaque characteristics and drug eluting stent deployment quality in the elderly patients with unstable angina (UA) and non-ST segment elevation myocardial infarction (NSTEMI). Methods OCT was used in elderly patients undergoing percutaneous coronary interventions. Fifteen patients, 9 males and 6 females with mean age of 72.6±5.3 years (range 67-92 years) were enrolled in the study. Images were obtained before initial balloon dilatation and following stent deployment. The plaque characteristics before dilation, vessel dissection, tissue prolapse, stent apposition and strut distribution after stent implantation were evaluated. Results Fifteen lesions were selected from 32 angiographic lesions as study lesions for OCT imaging after diagnostic coronary angiography. There were 7 lesions in the left anterior descending artery, 5 lesions in the right coronary artery and 3 lesions in the left circumflex coronary artery. Among them, 12 (80.0%) were lipid-rich plaques, and 10 (66.7%) were vulnerable plaques with fibrous cap thickness 54.2±7.3 μm. Seven ruptured culprit plaques (46.7%) were found; 4 in UA patients and 3 in NSTEMI patients. Tissue prolapse was observed in 11 lesions (73.3%). Irregular stent strut distribution was detected in 8 lesions (53.3%). Vessel dissections were found in 5 lesions (33.3%). Incomplete stent apposition was observed in 3 stents (20%) with mean spacing between the struts and the vessel wall 172±96 mm (range 117-436 mm). | 5 | C-reactive protein as a predictor for cardiac events in Chinese elderly patients with coronary heart disease显示文摘Background and objective To assess the predictive value of C-reactive protein(CRP) for major adverse cardiac events and the association between CRP level and the coronary lesion morphology and extent in patients with coronary heart disease (CHD).Methods CRP was measured on admission in 177 consecutive elderly (age≥60 years) patients with CHD who underwent coronary angiography. Patients were divided into high CRP group (CRP≥3mg/L) and normal CRP group (CRP <3mg/L). The association between CRP levels and the coronary lesion features, including severity of stenosis (mild, moderate, severe), extent of lesion (diffused or nondiffused), eccentricity of the plaque (eccentric or non-eccentric) were analyzed. Patients were followed up for a mean of 8 months for the occurrences of major adverse cardiac events (MACE). Results Compared with patients in normal CRP group, patients in high CRP group were more frequently to have unstable angina, multi-vessel, diffuse, eccentric lesions, positive remodeling, and non-smooth plaques (P<0.01). | 6 | Brain-derived neurotrophic factor gene transfection promotes neuronal repair and neurite regeneration after diffuse axonal injury显示文摘This study sought to assess the potential of brain-derived neurotrophic factor (BDNF) to promote neuronal repair and regeneration in rats with diffuse axonal injury, and to examine the accompanying neurobiological changes. BDNF gene transfection reduced the severity of the pathological changes associated with diffuse axonal injury in cortical neurons of the frontal lobe and increased neurofilament protein expression. | 12 | Allosteric probe-modified liposome loading bufalin-fluorouracil complex for targeted colorectal cancer therapy显示文摘Objective Bufalin,the main active anti-tumor monomer of toad venom,is crucial in cancer treatment.However,intrinsic issues,such as poor solubility and systematic toxicity,have considerably mitigated its anticancer functions and caused unwanted side effects.It is essential to develop innovative targeting systems to precisely and efficiently deliver anticancer drugs to achieve satisfying therapeutic efficiency.Methods This work established a novel and more efficient system for simultaneously detecting and killing colorectal cancer cells.The proposed method designed two allosteric probes,a report probe and a recognize probe.The method exhibited high sensitivity towards cell detection via the recognizing probe identifying target cancer cells and the report probe’s signal report.Combining bufalin and fluorouracil endowed better tumor cell inhibition.Results We observed significantly enhanced fluorescence dots surrounding the HCT-116 cell membranes.No fluorescence increments in the other three cells were identified,indicating that the established liposome complex could specifically bind with target cells.In addition,the best ratio of bufalin to fluorouracil was 0.15 and 0.5,respectively.This improved the anti-tumor effects and achieved more than 60%tumor cell inhibition.Conclusion This method will provide new opportunities for intracellular biomolecule detection and targeted cancer cell therapy. | Fajiang Mao Xingli Wu Chenyue Yuan Haiyan Huang Yanyan Qiu Jianlin Ren Peihao Yin | 2022 | Oncology and Translational Medicine2022,8,5: | 0 |
