1Antidiabetic Effects of Gegen Qinlian Decoction via the Gut Microbiota Are Attributable to Its Key Ingredient Berberine显示文摘Gegen Qinlian Decoction(GQD),a traditional Chinese medicine(TCM)formula,has long been used for the treatment of common metabolic diseases,including type 2 diabetes mellitus.However,the main limitation of its wider application is ingredient complexity of this formula.Thus,it is critically important to identify the major active ingredients of GQD and to illustrate mechanisms underlying its action.Here,we compared the effects of GQD and berberine,a hypothetical key active pharmaceutical ingredient of GQD,on a diabetic rat model by comprehensive analyses of gut microbiota,short-chain fatty acids,proinflammatory cytokines,and ileum transcriptomics.Our results show that berberine and GQD had similar effects on lowering blood glucose levels,modulating gut microbiota,inducing ileal gene expression,as well as relieving systemic and local inflammation.As expected,both berberine and GQD treatment significantly altered the overall gut microbiota structure and enriched many butyrate-producing bacteria,including Faecalibacterium and Roseburia,thereby attenuating intestinal inflammation and lowering glucose.Levels of short-chain fatty acids in rat feces were also significantly elevated after treatment with berberine or GQD.Moreover,concentration of serum proinflammatory cytokines and expression of immune-related genes,including Nfkb1,Stat1,and Ifnrg1,in pancreatic islets were significantly reduced after treatment.Our study demonstrates that the main effects of GQD can be attributed to berberine via modulating gut microbiota.The strategy employed would facilitate further standardization and widespread application of TCM in many diseases.Xizhan Xu Zezheng Gao Fuquan Yang Yingying Yang Liang Chen Lin Han Na Zhao Jiayue Xu Xinmiao Wang Yue Ma Lian Shu Xiaoxi Hu Na Lyu Yuanlong Pan Baoli Zhu Linhua Zhao Xiaolin Tong Jun Wang 2020Genomics, Proteomics & Bioinformatics2020,18,6:29
2Pollen indication to source plants in the eastern desert of China显示文摘In order to investigate the indication of major pollen types to source plants, 39 surface pollen samples are collected from 39 plant communities. The results show that diversity of pollen assemblages presents in different commu-nities. Chenopodiaceae pollen percentages are highest in Kalidium communities and Ceratoides communities (higher than 45%, average is 63.4%), but high Ephedra pollen per-centages (higher than 3.3%, average is 16.5%)in Ceratoides communities make it distinguished from Kalidium communi-ties. Artemisia pollen percentages (more than 50%) are much higher than Chenopodiaceae (less than 35%) in Artemisia Communities. Nitraria pollen percentages (higher than 2%, average is 13.6%) are highest in Nitraria communities com-pared with those in other community patterns. Tamariceae pollen percentages (higher than 2%, average is 8.6%) are highest both in Tetraena and Reaumuria communities com-pared with those in other community patterns. But in Reau-muria communities, Artemisia pollen percentages are higher than Chenopodiaceae with some other pollen types such as Gramineae, Compositae, Leguminosae; on the contrary, in Tetraena communities, Artemisia pollen percentages are less than Chenopodiaceae with few other pollen types. Calli-gonum pollen only appears in Calligonum communities (higher than 2%,average is 5%). The results of cluster analysis (CLA) and detrended correspondence analysis (DCA) show that the pollen indication to source plants can be reflected by association indices (A), over-representation in-dices (O), under-representation indices (U), R-value, the av-erage pollen percentages where parent plants are absent (Xa) and the average pollen percentages where parent plants are present (Xp). Zygophyllum, Calligonum, Tetraena, Tribulus and Tamariceae can indicate source plants very well, owing to high A scores. Artemisia, Chenopodiaceae and Gramineae have high R-values, and Nitraria, Cruciferae and sedum have high O scores, so they are over-represented types, but the higher Xp and the lower Xa values make that these pollen types can also indicate source plants moderately. Allium, Leguminosae, Roseceae, Pedicularis and Compositae have low A scores (less than 0.5), high O and/or U scores, so they cannot indicate source plants clearly. Some other pollen types, such as Pinus, Picea, Betula, Quercus, Ulmus, Ephedra, Elaegnaceae, Ranunculaceae, Caryophyllaceae, Polygona-ceae, Saxifragaceae, only appear in pollen assemblages, so their indication cannot be decided. Just like the pollen per-centage ratios of Artemisia and Chenopodiaceae (A/C), the ratios of Ephedra and Artemisia (E/A), Nitraria and Ephedra (N/E) can also indicate the aridity of environment. In typical desert communities, A/C ratios are less than 0.5, E/A ratios are more than 0.1, and N/E ratios are less than 1. 3Palaeoecological and palaeoenvironmental significance of someimportant spores and micro-algae in Quaternary deposits显示文摘Spores of ferns and allied plants, and micro-algae frequently occur in abundance in Quaternary deposits, but their palaeoenvironmental significance is seldom evaluated. This paper presents morphological descriptions of spores of Selaginella sinensis (Desv.) Spring, 1843 (Selaginellaceae) and Ceratopteris cf. thalictroides (L.) Brongniart, 1821 (Parkeriaceae) and the algae Pediastrum boryanum (Turpin) Meneghini, 1840, P. simplex Meyen, 1829, P. integrum N geli, 1849 (all Hydrodictyaceae), Spiniferites Mantell, 1850, emend. Sarjeant, 1970 (Spiniferitaceae) and Concentricystes Rossignal, 1962, emend. Jiabo, 1978 (systematic position unclear), and discusses their occurrence in mostly Quaternary sedimentary successions. All are closely associated with aquatic habitats. Extant Selaginella sinensis often colonizes fairly wet hillsides and gaps between rocks, and shady sides of ravines and slopes; it is also found within thickets of trees and shrubs (boscages) and in forests with calcareous soils. Ceratopteris thalictroides, Pediastrum and Concentricystes inhabit freshwater lakes and both natural and artificial wetlands, such as paddy fields and ditches. Spiniferites is a salt-water genus and usually associated with the marine realm. 4Occurrence, spatial distribution and ecological risks of antibiotics in soil in urban agglomeration显示文摘Antibiotics in soil environment are regarded as emerging pollutants and have introduced increasing risks to soil ecosystem and human health in rapid urbanization areas. Identifying the occurrence and spatial variability of antibiotics in soils is an urgent issue in sustaining soil security. In this study, antibiotics in soils were investigated and analyzed in BeijingTianjin-Hebei urban agglomeration. The occurrence, spatial distribution, and related affecting factors of antibiotics in soils were identified and ecological risks of antibiotics in soil environment were assessed. Results showed that(1) The mean concentration of soil antibiotics in Beijing-Tianjin-Hebei urban agglomeration was 21.79 μg/kg. Land use substantially affected the occurrence and concentration of antibiotics in soils. Concentrations of antibiotics in cropland and orchard soils were 2-3 times higher than the other land use types.(2)The concentrations of antibiotics in soils in Beijing-Tianjin-Hebei urban agglomeration presented a spatial pattern of high values in southeast, and low values in northwest. Spatial variability of antibiotics in soils was closely related to the application of organic fertilizer and wastewater irrigation as well as topographical features. Furthermore, soil properties and land management policy had substantial influences on soil antibiotics, and soil heavy metals may aggravate the accumulation of antibiotics in soils.(3) Ecological risks assessment of antibiotics in soils demonstrated that erythromycin(ERY), sulfamethoxazole(SMX), and doxycycline(DOX) may introduce high risks to soil ecosystem health, and more attention should be paid to the areas with intensive human activities that had potential high risk to soil ecosystem health. 6Memory stem T cells generated by Wnt signaling from blood of human renal clear cell carcinoma patients显示文摘Objective: Memory stem T cells(Tscm) have attracted attention because of their enhanced self-renewal, multipotent capacity, and anti-tumor capacities. However, little is known about Tscm in patients with renal clear cell carcinoma(RCC) and the role of Wnt signaling in these cells. We evaluated Tscm from RCC patients concerning their activation of Wnt signaling in vitro and explored the mechanism of preferential survival.Methods: Flow cytometry identified surface markers and cytokines produced from accumulated Tscm in the presence of the glycogen synthase kinase beta inhibitor TWS119. Apoptosis was evaluated after induction using tumor necrosis factor-alpha.Immunofluorescence and Western blot analyses were used to investigate the activation of the nuclear factor-kappa B(NF-КB)pathway.Results: RCC patients had a similar percentage of CD4^+ and CD8^+ Tscm as healthy donors. Activation of Wnt signaling by TWS119 resulted in the accumulation of Tscm in activated T cells, but reversal of differentiated T cells to Tscm was not achieved.Preferential survival of Tscm was associated with increased anti-apoptotic ability mediated downstream of the NF-КB activation pathway.Conclusions: The finding that Tscm can accumulate by Wnt signaling in vitro in blood from RCC patients will help in devising new cancer therapy strategies of Tscm-based adoptive immunotherapy, such as dendritic cell-stimulated Tscm, and T cell receptor or chimeric antigen receptor-engineered Tscm.Cihui Yan Jingjing Chang Xinmiao Song Fan Yan Wenwen Yu Yang An Feng Wei Lili Yang Xiubao Ren 2019Cancer Biology & Medicine2019,16,1:4
71 T moderate intensity static magnetic field affects Akt/mTOR pathway and increases the antitumor efficacy of m TOR inhibitors in CNE-2Z cells显示文摘Static magnetic field(SMF) has been known to affect cell proliferation in a cell-type-dependent manner,while the mechanism still remains unclear. We found that 1T moderate intensity SMF inhibits cell proliferation of nasopharyngeal carcinoma CNE-2Z cells and the Akt/m TOR signaling pathway, which is upregulated in many cancers. m TOR inhibitors are potential chemodrugs, but their clinical effects are limited by the feedback reactivation of other signaling components such as EGFR and Akt.We showed that 1 T SMF increases the antitumor efficacy of m TOR inhibitor Torin 2. In addition, 1 T SMF increases the inhibition efficiency on m TOR substrates phosphorylation and represses the m TOR inhibitor-induced feedback reactivation of EGFR and Akt. 8Biting force and tongue muscle strength as useful indicators for eating and swallowing capability assessment among elderly patients显示文摘Weakened capability of eating and swallowing is common among elderly population,affecting negatively on their health and well-being.This work measured occlusal force and tongue muscle strength of elderly patients and then assessed whether correlations existed between oral physiological properties and the eating and swallowing capability.A total of 26 elderly patients with potential swallow disorder were recruited in this work.Participants’denture status and daily diet texture were determined by a pre-questionnaire analysis.Their swallowing capability was assessed and rated by water drinking test.Maximum tongue pressure,incisor occlusal force and hand gripping force were measured for each subject.It was observed that participating subjects have on average a significantly lower biting force and maximum tongue pressure than that of normal adults.The swallowing grades assessed by water drinking test showed significant correlations with both the biting force(r=−0.498,P<0.05)and the tongue muscle strength(r=−0.544,P<0.05).The biting force of dysphagia participants correlates significantly with dental conditions(P<0.05).The observed correlation between tongue pressure and diet texture indicates that tongue muscle strength could be used as a reliable indicator of the eating and swallowing capability for elderly people.Xinmiao Wang Gangying Zheng Mingsong Su Yanqiu Chen Hua Xie Weijia Han Qing Yang Jianqin Sun Jianshe Chen 2019Food Science and Human Wellness2019,8,2:4

11A reversed phase/hydrophilic interaction/ion exchange mixed-mode stationary phase for liquid chromatography显示文摘A reversed phase (RP)/hydrophilic interaction (HILIC)/ion exchange (IEX) mixed tri-mode stationary phase (TMSP) has been prepared via a divergent synthesis scheme starting from propylamine on silica then by amine-epoxy reactions with 1,4-butanedioldiglycidyl ether and tertiary amines (N,Ndimethyldecylamine, DMDA). Its retention mechanism was found to follow RP/HILIC/IEX mixed-mode.The stop-flow test revealed that TMSP had good compatibility with 100% aqueous mobile phase. 16Precision Methylome and In Vivo Methylation Kinetics Characterization of Klebsiella pneumoniae显示文摘Klebsiella pneumoniae(K.pneumoniae)is an important pathogen that can cause severe hospital-and community-acquired infections.To systematically investigate its methylation features,we determined the whole-genome sequences of 14 K.pneumoniae strains covering varying serotypes,multilocus sequence types,clonal groups,viscosity/virulence,and drug resistance.Their methylomes were further characterized using Pacific Biosciences single-molecule real-time and bisulfite technologies.We identified 15 methylation motifs[13 N6-methyladenine(6mA)and two 5-methylcytosine(5mC)motifs],among which eight were novel.Their corresponding DNA methyltransferases were also validated.Additionally,we analyzed the genomic distribution of GATC and CCWGG methylation motifs shared by all strains,and identified differential distribution patterns of some hemi-/un-methylated GATC motifs,which tend to be located within intergenic regions(IGRs).Specifically,we characterized the in vivo methylation kinetics at single-base resolution on a genome-wide scale by simulating the dynamic processes of replication-mediated passive demethylation and MTase-catalyzed re-methylation.The slow methylation of the GATC motifs in the replication origin(oriC)regions and IGRs implicates the epigenetic regulation of replication initiation and transcription.Our findings illustrate the first comprehensive dynamic methylome map of K.pneumoniae at single-base resolution,and provide a useful reference to better understand epigenetic regulation in this and other bacterial species.Jing Fu Ju Zhang Li Yang Nan Ding Liya Yue Xiangli Zhang Dandan Lu Xinmiao Jia Cuidan Li Chongye Guo Zhe Yin Xiaoyuan Jiang Yongliang Zhao Fei Chen Dongsheng Zhou 2022Genomics, Proteomics & Bioinformatics2022,20,2:1
17Epigenome-wide DNA methylation study of whole blood in patients with sporadic amyotrophic lateral sclerosis显示文摘Background:Epigenetics,and especially DNA methylation,contributes to the pathogenesis of sporadic amyotrophic lateral sclerosis(SALS).This study aimed to investigate the role of DNA methylation in SALS using whole blood of SALS patients.Methods::In total,32 SALS patients and 32 healthy controls were enrolled in this study.DNA was isolated from whole blood collected from the participants.DNA methylation profiles were generated using Infinium MethylationEPIC BeadChip.Results:We identified 34 significant differentially methylated positions(DMPs)in whole blood from SALS patients,compared with the healthy controls.Of these DMPs,five were hypermethylated and 29 were hypomethylated;they corresponded to 13 genes.For the DMPs,ATAD3B and BLK were hypermethylated,whereas DDO,IQCE,ABCB1,DNAH9,FIGN,NRP1,TMEM87B,CCSAP,ST6GALNAC5,MYOM2,and RUSC1-AS1 were hypomethylated.We also identified 12 differentially methylated regions(DMRs),related to 12 genes(NWD1,LDHD,CIS,IQCE,TNF,PDE1C,LGALS1,CSNK1E,LRRC23,ENO2,ELOVL2,and ELOVL2-AS1).According to data from the Kyoto Encyclopedia of Genes and Genomes database,DNAH9 and TNF are involved in the amyotrophic lateral sclerosis(ALS)pathway.Correlation analysis between clinical features and DNA methylation profiling indicated that the methylation level of ELOVL2 and ARID1B was positively associated with the age of onset(r=0.86,adjust P=0.001)and disease duration(r=0.83,adjust P=0.01),respectively.Conclusions:We found aberrant methylation in DMP-and DMR-related genes,implying that many epigenetic alterations,such as the hypomethylation of DNAH9 and TNF,play important roles in ALS etiology.These findings can be helpful for developing new therapeutic interventions.Zhengyi Cai Xinmiao Jia Mingsheng Liu Xunzhe Yang Liying Cui 2022Chinese Medical Journal2022,,12:1
