2In vitro and in vivo activity of D-serine in combination with β-lactam antibiotics against methicillin-resistant Staphylococcus aureus显示文摘As D-amino acids play important roles in the physiological metabolism of bacteria,combination of D-amino acids with antibiotics may provide synergistic antibacterial activity. The aim of the study was to evaluate in vitro and in vivo activity of D-serine alone and in combination withβ-lactams against methicillin-resistant Staphylococcus aureus(MRSA) strains, and to explore the possible sensitization mechanisms. The activity of D-serine, β-lactams alone and in combinations was evaluated both in vitro by standard MICs, time–kill curves and checkerboard assays, and in vivo by murine systemic infection model as well as neutropenic thigh infection model. An in vitro synergistic effect was demonstrated with the combination of D-serine and β-lactams against MRSA standard and clinical strains.Importantly, the combinations enhanced the therapeutic efficacy in the animal models as compared toβ-lactam alone groups. Initial mechanism study suggested possible revision of D-alanine-D-alanine residue to D-alanine-D-serine in peptidoglycan by adding of D-alanine in the medium, which may cause decreased affinity to PBPs during transpeptidation. In conclusion, D-serine had synergistic activity in combination with β-lactams against MRSA strains both in vitro and in vivo. 3Investigation of hemostatic effect of Spleeninvigorating,Qi-replenishing and Bloodarresting Formula on simvastatin-induced zebrafish hemorrhage model显示文摘Blood-arresting Formula on simvastatin-induced zebrafish hemorrhage model compared with that of the Heat-clearing and Blood-cooling Formula.Methods:AB strain wild type zebrafish were treated with 0.5 mM simvastatin for 24 hours to establish the zebrafish hemorrhage model.Under strictly blind experimental conditions,the zebrafish were then treated with experimental agents in different concentration within the maximum non-lethal dose.The intervention effect of the Spleen-invigorating,Qi-replenishing and Blood-arresting Formula was comprehensively assessed by examining the main observational parameters such as the bleeding reduction rate and hemostatic rate while referring to the additional parameters such as blood flow,improvement rate of blood flow,velocity of movement,improvement rate of motion under spleen-qi deficiency.Results:Hemostatic effect:The bleeding rate and hemostatic rate in the zebrafish were respectively,30%,15%,45%,40%and 60%,80%,40%,47%when equal concentration of 500 mg/mL,1000 mg/mL were used.This showed that the experimental herbs B1 was superior to B2 in decreasing the bleeding rate and improving hemostatic effect in the zebrafish.Improvement of blood flow:With equal concentration,the experimental herb B1 was superior to B2 in increasing the blood flow and improvement rate of blood flow in hemorrhagic zebrafish.Promotion of the improvement of motion:With equal concentration of 500 mg/mL and 1000 mg/mL,the experimental herb B1 was superior to B2 in promoting the velocity of motion in the zebrafish and promoting the improvement rate of motion.Conclusion:The manifestations of simvastatin-induced zebrafish hemorrhage are basically similar to that of failure of the spleen to control blood in human beings.The Spleeninvigorating,Qi-replenishing and Blood-arresting Formula has good hemostatic effect on simvastatin-induced hemorrhage in the zebrafish.It also boasts the functions of improving blood flow and velocity of motion in the hemorrhagic zebrafish.Such a conclusion provides the experimental basis for the treatment of the syndrome of failure of the spleen to control blood by this formula.Yuting Chu Ling Zhang Xiaoyu Zhu Yayue Zhang Bo Xia Li Hou Rushun Song Tiantian Li Chunqi Li Qing Dong a Xinyi Chen 2016Journal of Traditional Chinese Medical Sciences2016,3,4:1
4In-situ TEM observation of the evolution of helium bubbles & dislocation loops and their interaction in Pd during He^(+) irradiation显示文摘The microstructural evolution of purity Pd under 30 keV He^(+)irradiation at 573 K was investigated by in-situ transmission electron microscopy.The nucleation,growth,merging,annihilation,size change,number density variation,and types of dislocation loops were analyzed under the influence of irradiation fluence and sample thickness.Both perfect dislocation loops with b=1/2<110>and faulted dislocation loops with b=1/3<111>were formed.However,at low irradiation fluence,most of the loops were 1/3<111>loops.The thickness of TEM foil obviously affected the ratio of 1/3<111>loop variants,the size and number density of dislocation loops,and the characteristics of bubble-loop complexes.With the increase of irradiation fluence,the size of dislocation loops increased,but loop volume number density remained almost constant until dislocation loops merged and evolved into dislocation network.There was an obvious interaction between dislocation loops and bubbles,indicating that 1/3<111>loop was first formed at the initial stage of irradiation,and when the loop grew to a certain size,obvious helium bubbles appeared inside its region.Qing Han Yipeng Li Guang Ran Xinyi Liu Lu Wu Yang Chen Piheng Chen Xiaoqiu Ye Yifan Ding Xiaoyong Wu 2021Journal of Materials Science & Technology2021,,28:1
5Sustainability of SARS-CoV-2 Induced Humoral Immune Responses in COVID-19 Patients from Hospitalization to Convalescence Over Six Months显示文摘Understanding the persistence of antibody in convalescent COVID-19 patients may help to answer the current major concerns such as the risk of reinfection,the protection period of vaccination and the possibility of building an active herd immunity.This retrospective cohort study included 172 COVID-19 patients who were hospitalized in Wuhan.A total of404 serum samples were obtained over six months from hospitalization to convalescence.Antibodies in the specimens were quantitatively analyzed by the capture chemiluminescence immunoassays(CLIA).All patients were positive for the anti-SARS-Co V-2 Ig M/Ig G at the onset of COVID-19 symptoms,and the Ig G antibody persisted in all the patients during the convalescence.However,only approximately 25%of patients can detect the Ig M antibodies,Ig M against N protein(NIg M)and receptor binding domain of S protein(RBD-Ig M)at the 27 th week.The titers of Ig M,N-Ig M and RBD-Ig M reduced to 16.7%,17.6%and 15.2%of their peak values respectively.In contrast,the titers of Ig G,N-Ig G and RBD-Ig G peaked at 4–5 th week and reduced to 85.9%,62.6%and 87.2%of their peak values respectively at the end of observation.Dynamic behavior of antibodies and their correlation in age,gender and severity groups were investigated.In general,the COVID-19 antibody was sustained at high levels for over six months in most of the convalescent patients.Only a few patients with antibody reducing to an undetectable level which needs further attention.The humoral immune response against SARS-Co V-2 infection in COVID-19 patients exhibits a typical dynamic of acquired immunity.Yang Zheng Qing Zhang Ashaq Ali Ke Li Nan Shao Xiaoli Zhou Zhiqin Ye Xiaomin Chen Shanshan Cao Jing Cui Juan Zhou Dianbing Wang Baidong Hou Min Li Mengmeng Cui Lihua Deng Xinyi Sun Qian Zhang Qinfang Yang Yong li Hui Wang Yake Lei Bo Yu Yegang Cheng Xiaolin Tong Dong Men Xian-En Zhang 2021Virologica Sinica2021,36,5:1
6JaponiconeA induces apoptosis of bortezomib-sensitive and -resistant myeloma cells in vitro and in vivo by targeting IKKβ显示文摘Objective:Multiple myeloma(MM)remains incurable with high rates of relapse.New therapeutic drugs are therefore urgently needed to improve the prognosis.JaponiconeA(JA),a natural product isolated from Inula japonica Thunb,has shown good anti-MM potential.A comprehensive study should therefore be conducted to identify both the in vitro and in vivo mechanisms of the anti-MM effects of JA.Methods:CCK8 assays and flow cytometry were used to detect the proliferation,apoptosis,and cell cycle of MM cell lines when treated with JA.In vivo experiments were conducted using subcutaneous xenograft mouse models.We also identified possible targets and the mechanism of JA using RNA-seq and c-Map databases,and identified the specific targets of JA in bortezomib-sensitive and-resistant MM cell lines using CETSA,DARTS,and rescue experiments.Furthermore,JA and bortezomib were used separately or together to characterize their possible synergistic effects.Results:In vitro,JA inhibited proliferation,and induced apoptosis and G2/M phase arrest in MM cell lines,and selectively killed primary CD138+MM cells.In vivo,JA also demonstrated a strong anti-tumor effect with no observable toxicity.In addition,JA showed synergetic effects in combination with bortezomib,and enhanced the anti-tumor effect of bortezomib in bortezomibresistant cells.CETSA and DARTS confirmed direct binding of JA to NF-κB inhibitor kinase beta(IKKβ),and overexpression of IKKβor knockdown of IκBαpartially rescued the apoptosis induced by JA.Conclusions:JA exhibited strong anti-tumor effects in MM.It sensitized myeloma cells to bortezomib and overcame NF-κB-induced drug resistance by inhibiting IKKβ,providing a new treatment strategy for MM patients.Zilu Zhang Chenjing Ye Jia Liu Wenbin Xu Chao Wu Qing Yu Xiaoguang Xu Xinyi Zeng Huizi Jin Yingli Wu Hua Yan 2022Cancer Biology & Medicine2022,19,5:1
7Manipulating nonlinear exciton polaritons in an atomically-thin semiconductor with artificial potential landscapes显示文摘Exciton polaritons in atomically thin transition-metal dichalcogenide microcavities provide a versatile platform for advancing optoelectronic devices and studying the interacting Bosonic physics at ambient conditions.Rationally engineering the favorable properties of polaritons is critically required for the rapidly growing research.Here,we demonstrate the manipulation of nonlinear polaritons with the lithographically defined potential landscapes in monolayer WS_(2)microcavities.The discretization of photoluminescence dispersions and spatially confined patterns indicate the deterministic on-site localization of polaritons by the artificial mesa cavities.Varying the trapping sizes,the polariton-reservoir interaction strength is enhanced by about six times through managing the polariton–exciton spatial overlap.Meanwhile,the coherence of trapped polaritons is significantly improved due to the spectral narrowing and tailored in a picosecond range.Therefore,our work not only offers a convenient approach to manipulating the nonlinearity and coherence of polaritons but also opens up possibilities for exploring many-body phenomena and developing novel polaritonic devices based on 2D materials.Yuan Luo Quanbing Guo Xinyi Deng Sanjib Ghosh Qing Zhang Hongxing Xu Qihua Xiong 2023Light(Science & Applications)2023,12,10:1

9The transcription factor KLF14 regulates macrophage glycolysis and immune function by inhibiting HK2 in sepsis显示文摘Sepsis is a heterogeneous syndrome induced by a dysregulated host response to infection.Glycolysis plays a role in maintaining the immune function of macrophages,which is crucial for severely septic patients.However,how the pathways that link glycolysis and macrophages are regulated is still largely unknown.Here,we provide evidence to support the function of KLF14,a novel Krüppel-like transcription factor,in the regulation of glycolysis and the immune function of macrophages during sepsis.KLF14 deletion led to significantly increased mortality in lethal models of murine endotoxemia and sepsis.Mechanistically,KLF14 decreased glycolysis and the secretion of inflammatory cytokines by macrophages by inhibiting the transcription of HK2.In addition,we confirmed that the expression of KLF14 was upregulated in septic patients.Furthermore,pharmacological activation of KLF14 conferred protection against sepsis in mice.These findings uncover a key role of KLF14 in modulating the inflammatory signaling pathway and shed light on the development of KLF14-targeted therapeutics for sepsis.Yuan Yuan Guangjian Fan Yuqi Liu Lu Liu Tong Zhang Pengfei Liu Qing Tu Xinyi Zhang Shiyuan Luo Liangfang Yao Feng Chen Jingbao Li 2022Cellular & Molecular Immunology2022,19,4:1
10Accurate modulation of photoprinting under stiffness imaging feedback for engineering ECMs with high-fidelity mechanical properties显示文摘Engineered extracellular matrices(ECMs)that replicate complex in-vivo features have shown great potential in tissue engineering.Biocompatible hydrogel microstructures have been widely used to replace these native ECMs for physiologically relevant research.However,accurate reproduction of the 3D hierarchical and nonuniform mechanical stffness inside one integrated microstructure to mimic the complex mechanical properties of native ECMs presents a major challenge.Here,by using digital holographic microscopy(DHM)-based stffness imaging feedback,we propose a novel closed-loop control algorithm to achieve high-accuracy control of mechanical properties for hydrogel microstructures that recapitulate the physiological properties of native ECMs with high fidelity.During photoprinting,the photocuring area of the hydrogel is divided into microscale grid areas to locally control the photocuring process.With the assistance of a motorized microfluidic channel,the curing thickness is controlled with layer-by-layer stacking.The DHM-based stiffness imaging feedback allows accurate adjustment of the photocuring degree in every grid area to change the crosslinking network density of the hydrogel,thus enabling large-span and high-resolution modulation of mechanical properties.Finally,the gelatin methacrylate was used as a typical biomaterial to construct the highfidelity biomimetic ECMs.The Young's modulus could be flexibly modulated in the 10 kPa to 50 kPa range.Additionally,the modulus gradient was accurately controlled to within 2.9 kPa.By engineering ECM with locally different mechanical properties,cell spreading along the stff areas was observed successfully.We believe that this method can regenerate complex biomimetic ECMs that closely recapitulate in-vivo mechanical properties for further applications in tissue engineering and biomedical research.Xin Li Huaping Wang Xinyi Dong Qing Shi Tao Sun Shingo Shimoda Qiang Huang Toshio Fukuda 2022Microsystems & Nanoengineering2022,8,3:0
11In-situ TEM investigation of dislocation loop reaction and irradiation hardening in H_(2)^(+)-He^(+) dual-beam irradiated Mo显示文摘Through in-situ TEM observation during 30 keV H_(2)^(+)-He^(+) dual-beam irradiation at 723 K,the reaction and transformation of dislocation loops in pure Mo were investigated,especially for<100>loops.Irradiation could directly cause the formation of 1/2<111>loops and<100>loops,but 1/2<111>loops were dominant.In-situ observation confirmed the formation mechanism of<100>loops,including direct irradiation induced mechanism,1/2<111>loop direct conversion mechanism,and reaction mechanism of two 1/2<111>loops.Meanwhile,the reaction of two 1/2<111>loops to produce<100>loop should not require the strict size similarity condition.The reaction between 1/2<111>loops could also produce 1/2<111>loop,which was essentially a process in which one loop absorbed another one.The yield strength increment caused by irradiation-induced loops was analyzed,and its saturation value reached0.48 GPa at 0.06 dpa.Compared with single He+irradiation,the number density and average diameter of loops increased significantly and more serious damage was caused under the synergistic effect of hydrogen and helium.The mechanism based on in-situ experimental observation was discussed in depth.Yipeng Li Guang Ran Xinyi Liu Qing Han Xiuyin Huang Yifan Ding 2022Journal of Materials Science & Technology2022,,12:0
12Constructing representative driving cycle for heavy duty vehicle based on Markov chain method considering road slope显示文摘Electrification of heavy duty vehicles(HDVs)is critical to realization of the target of carbon neutralization in the future.For most HDVs,the influence of road slope on vehicle power usually cannot be ignored due to significant road slope variation during long driving mileages.In order to design the powertrain system for electrified HDVs effectively,it is necessary to construct representative driving cycles with road slope information.There are two difficulties for this task.(1)Road slope measuring devices are usually costly.A cheaper yet effective method for measuring road slope needs to be developed.(2)A 3D(three dimension)Markov chain method is usually utilized for constructing cycles with velocity and road slope.This method is complex and time consuming,and needs to be improved.In this paper,a 2D(two dimension)Markov chain method for addressing these issues is proposed.A road slope observation is designed based on normal GPS(Global Positioning System)signals and a high order Butterworth filter.The effectiveness of the method is validated.Driving velocity and road slope are collected and observed for the area between Beijing and Zhangjiakou in northern China.Representative cycles with road slope are constructed using a 2D Markov chain method and a matching algorithm based on average speed.With the introduced technology,three representative driving cycles with road slope for urban,suburban and highway routes are designed.Statistic results on vehicle power show that,the representative driving cycles are effective with relative errors less than 4%compared to the real driving conditions.These driving cycles will be utilized in designing electric HDVs,such as hydrogen fuel cell vehicles in the future.Xinyi Jia Hewu Wang Liangfei Xu Qing Wang Hang Li Zunyan Hu Jianqiu Li Minggao Ouyang 2021Energy and AI2021,6,4:0

14Nano-capillary induced assemble of quantum dots on perovskite grain boundaries for efficient and stable perovskite solar cells显示文摘In recent years, perovskite solar cells(PSCs) have propelled into the limelight owing to rapid development of efficiency;however, the abundant defects at the perovskite grain boundaries result in unwanted energy loss and structural degradation. Here, the grain boundaries of perovskite polycrystalline films have been found to act as nanocapillaries for capturing perovskite quantum dots(PQDs), which enable the conformal assemble of PQDs at the top interspace between perovskite grains. The existence of PQDs passivated the surface defects, optimized the interfacial band alignments, and ultimately improved the power conversion efficiency from 19.27% to 22.47% in inverted PSCs. 18Design of a Minimally Actuated Lower Limb Exoskeleton with Mechanical Joint Coupling显示文摘Powered lower limb exoskeletons have traditionally used four or more powered joints to provide ambulation assistance for individuals with spinal cord injury.Exoskeletons with numerous powered joints commonly lost some excellent features of passive orthoses and further decreased utility due to added weight and increased control complexity.This work adopts joints coupling mechanism to design a powered exoskeleton to minimize the number of actuated joints and control complexity.Unlike conventional powered exoskeletons,the joint-coupled-powered exoskeleton only has a single motor-actuated joint for each exoskeleton leg in conjunction with a unique knee coupled system to enable their users to walk,sit,and stand.And two types of joint coupled systems are designed,respectively,hip-knee coupled and knee-ankle coupled.The joint-coupled-powered exoskeleton system allows a single actuator to power the hip motion,and allows activate knee motion through the coupled motions of the hip or ankle.More specifically,when the mechanical coupled system is activated,the knee joint is unlocked,resulting in synchronized hip-knee or ankle-knee flexion and extension.The coupling mechanism is switched on and off at specific phases of the gait(the stance phase and the swing phase)to generate the desired motions.The research work proves that minimal actuated robotic systems with joint coupled could achieve safe and natural walking.Hui Li Hongliu Yu Yingwei Chen Xinyi Tang Duojin Wang Qiaoling Meng Qing Du 2022Journal of Bionic Engineering2022,19,2:0
19A method for retrieving soil moisture from GNSS-R by using experiment data显示文摘Soil moisture is a key parameter in agricultural irrigation.The L band(1.58GHz)on board global position system(GPS) satellite is well suited for monitoring the change of soil moisture.In order to investigate the potential of retrieving soil moisture using the L-band GPS bistatic radar,this paper analyzed a retrieval method by using field experiment data.In order to investigate the relationship between the soil moisture(corresponding roughly to the 05cm top soil layer) and the signalto-noise ratio(Pr-SNR) to the direct GPS signal-to-noise ratio(Pd-SNR),an experiment was conducted in Hulunber grassland of China in 2009 and 2011.Six field sites in the soil moisture experiment were utilized to analyze the relationship between soil moisture and the ratio of Pr-SNR to Pd-SNR and the square of correlation coefficient was about 0.9 when the surface type was known and the elevation angle of the satellite ranged from 65 to 85 degrees approximately.The analysis shows that ratio of Pr-SNR to P(d-SNR) can be used to monitor the soil moisture,because the ratio of P(r-SNR) to P(d-SNR) maximized the elimination of the influence of different signals from different GPS satellites.The estimation accuracy could be improved if we make full use of the empirical knowledge on elevation angles of GPS satellites and ground roughness of different surface types.毛克彪 Ma Ying Shen Xinyi Xia Lang Tian Shiying Han Jiaqi Liu Qing 2015High Technology Letters2015,21,2:0
20CTCF organizes inter-A compartment interactions through RYBP-dependent phase separation显示文摘Chromatin is spatially organized into three-dimensional structures at different levels including A/B compartments, topologically associating domains and loops. The canonical CTCF-mediated loop extrusion model can explain the formation of loops. However, the organization mechanisms underlying long-range chromatin interactions such as interactions between A–A compartments are still poorly understood. Here we show that different from the canonical loop extrusion model, RYBP-mediated phase separation of CTCF organizes inter-A compartment interactions. Based on this model, we designed and verified an induced CTCF phase separation system in embryonic stem cells (ESCs), which facilitated inter-A compartment interactions, improved self-renewal of ESCs and inhibited their differentiation toward neural progenitor cells.
