3Development of single row automatic transplanting device for potted vegetable seedlings显示文摘In China,low degree of automation seriously affects the working efficiency and quality in vegetable transplanting.As one of the most important vegetables in China even in the world,tomato was taken as the research object in this study.An automatic single-row transplanting device was designed,based on the statistical analysis of the physical and mechanical properties of tomato seedlings of a typical variety.Based on the technology of mechatronics,the device integrated the functions of transporting seedling tray,automatic seedling extraction and mechanical planting.The kinematics orthogonality solution combined with the dynamic sequence solution method was used to optimize and analyze the kinematic parameters of the automatic seeding mechanism,and the“sickle”trajectory was obtained.According to the position and movement requirement for taking and dropping seedling,the mechanical conditions and the working parameters of key execution parts were obtained by using analytic drawing method to analyze the mechanical condition of seedling collecting mechanism.The transplanting experiment was conducted at room temperature of 25°C,and the age and moisture content of the seedlings were 40 d and 55%,respectively.The results showed that the highest success rate was 92.59%,and the lowest rate of leakage was 23.13%,when the transplanting frequency was 60 plants/min.The lowest success rate was 77.78%,and the highest rate of leakage was 38.75%,when transplanting frequency was 120 plants/min.When the transplanting frequency is between 60-90 plants/min,the device can meet the requirement of high speed transplanting for potted vegetable seedling.Xin Jin Daoyi Li Hao Ma Jiangtao Ji Kaixuan Zhao Jing Pang 2018International Journal of Agricultural and Biological Engineering2018,11,3:27

6Effect of electroacupuncture on expression of Ang/Tie-2 mRNA and protein in rats with acute cerebral infarction显示文摘OBJECTIVE: To identify the intervention mechanism of the effect of electroacupuncture on the expression of Ang/Tie-2 m RNA and protein in rats with acute cerebral infarction induced by middle cerebral artery occlusion(MCAO).METHODS: Altogether 120 Wistar rats were subjected to MCAO by inserting a nylon filament, andthen divided into 3 groups: control group, injured group and electroacupuncture group. The injured and electroacupuncture groups were further divided into the following 7 subgroups according to the time after MCAO: 3, 6, 12, 24 h, 3, 7 and 12 d, with 8 rats in each subgroup. The electroacupuncture group was given electroacupuncture treatment at Shuigou(GV 26) instantly after operation. 7Gualou Guizhi decoction promotes neurological functional recovery and neurogenesis following focal cerebral ischemia/reperfusion显示文摘Recovery following stroke involves neurogenesis and axonal remodeling within the ischemic brain. Gualou Guizhi decoction(GLGZD) is a Chinese traditional medicine used for the treatment of post-stroke limb spasm. GLGZD has been reported to have neuroprotective effects in cerebral ischemic injury. However, the effects of GLGZD on neurogenesis and axonal remodeling following cerebral ischemia remain unknown. In this study, a rat model of focal cerebral ischemia/reperfusion was established by middle cerebral artery occlusion. Neurological function was assessed immediately after reperfusion using Longa's 5-point scoring system. The rats were randomly divided into vehicle and GLGZD groups. Rats in the sham group were given sham operation. The rats in the GLGZD group were intragastrically administered GLGZD, once daily, for 14 consecutive days. The rats in the vehicle and sham groups were intragastrically administered distilled water. Modified neurological severity score test, balance beam test and foot fault test were used to assess motor functional changes. Nissl staining was performed to evaluate histopathological changes in the brain. Immunofluorescence staining was used to examine cell proliferation using the marker 5-bromo-2′-deoxyuridine(BrdU) as well as expression of the neural precursor marker doublecortin(DCX), the astrocyte marker glial fibrillary acidic protein(GFAP) and the axon regeneration marker growth associated protein-43(GAP-43). GLGZD substantially mitigated pathological injury, increased the number of BrdU, DCX and GFAP-immunoreactive cells in the subventricular zone of the ischemic hemisphere, increased GAP-43 expression in the cortical peri-infarct region, and improved motor function. 8Grape Seed Proanthocyanidin Extract Alleviates Arsenic-induced Oxidative Reproductive Toxicity in Male Mice显示文摘Objective To determine the ability of grape seed proanthocyanidin extract(GSPE) in alleviating arsenic-induced reproductive toxicity. Methods Sixty male Kunming mice received the following treatments by gavage: normal saline solution(control); arsenic trioxide(ATO; 4 mg/kg); GSPE(400 mg/kg); ATO+GSPE(100 mg/kg); ATO+GSPE(200 mg/kg) and ATO+GSPE(400 mg/kg). Thereafter, the mice were sacrificed and weighed, and the testis was examined for pathological changes. Nuclear factor(erythroid-derived 2)-like 2(Nrf2), heme oxygenase 1(HO1), glutathione S-transferase(GST), NAD(P)H dehydrogenase, and quinone 1(NQO1) expression in the testis was detected by real-time PCR. Superoxide dismutase(SOD), glutathione(GSH), total antioxidative capability(T-AOC), malondialdehyde(MDA), 8-hydroxydeoxyguanosine(8-OHd G), and reproductive indexes were analyzed. Results ATO-treated mice showed a significantly decreased sperm count and testis somatic index and activity levels of SOD, GSH, and T-AOC than control group. Compared to the ATO-treated group, ATO +GSPE group showed recovery of the measured parameters. Mice treated with ATO+high-dose GSPE showed the highest level of m RNA expression of Nrf2, HO, NQO1, and GST. 9Crop 3D a LiDAR based platform for 3D high-throughput crop phenotyping显示文摘With the growing population and the reducing arable land, breeding has been considered as an effective way to solve the food crisis.As an important part in breeding, high-throughput phenotyping can accelerate the breeding process effectively. Light detection and ranging(LiDAR) is an active remote sensing technology that is capable of acquiring three-dimensional(3 D) data accurately,and has a great potential in crop phenotyping. Given that crop phenotyping based on LiDAR technology is not common in China,we developed a high-throughput crop phenotyping platform, named Crop 3 D, which integrated LiDAR sensor, high-resolution camera, thermal camera and hyperspectral imager. Compared with traditional crop phenotyping techniques, Crop 3 D can acquire multi-source phenotypic data in the whole crop growing period and extract plant height, plant width, leaf length, leaf width, leaf area, leaf inclination angle and other parameters for plant biology and genomics analysis. In this paper, we described the designs,functions and testing results of the Crop 3 D platform, and briefly discussed the potential applications and future development of the platform in phenotyping. 10Effect of the cooling rate on plastic deformability of a Zr-based bulk metallic glass显示文摘The present work found the plastic deformability of Zr65Cu17.5Ni10Al7.5 BMG dependent on the cooling rate during the formation from the molten state alloy. The deformation behavior in the compression test of φ 2 mm Zr65Cu17.5Ni10Al7.5 BMGs as-cast or lathed from different sizes as-cast samples was characterized, and they exhibited different plastic strains. The compressive plastic strain increases with the decreasing diameter of the as-cast specimens, i.e. with increasing the cooling rate. 11Effect of caffeic acid phenethyl ester on proliferation and apoptosis of hepatic stellate cells in vitro显示文摘AIM: To investigate the role of nuclear factor-κB (NF-κB)inhibitor caffeic acid phenethy1 ester (CAPE) in the proliferation, collagen synthesis and apoptosis of hepatic stellate cells (HSCs) of rats. METHODS: The HSCs from rats were isolated and cultured in Dulbecco's Modified Eagle's Medium (DMEM) and treated with CAPE. The proliferation and collagen synthesis of HSCs were determined by 3H-TdR and 3H-proline incorporation respectively, and the expression of type Ⅰ, Ⅲ procollagen genes was further explored byin situ hybridization. Apoptosis cell indices (AIs) were examined using terminal deoxynucleotidyl transferase- mediated DIG-dUTP nick end labeling (TUNEL). RESULTS: Tn activated HSC in culture, CAPE significantly inhibited 3H-TdR and 3H-proline incorporation by HSCs at concentrations of 5 μmol/L and 10 μmol/L respectively. CAPE also reduced the type I procollagen gene expression (P<0.05)at higher concentration. Apoptosis of HSC was induced by CAPE and the AIs were time-and dose-dependently increased from 2.82+0.73 % to 7.66±1.25 % at 12 h (P<0.01) and from 3.15±0.88 % to 10.6L±2.88 % at 24 h (P<0.01). 12Retrospective study of Traditional Chinese Medicine treatment of type 2 diabetes mellitus显示文摘OBJECTIVE: To provide clinical evidence in support of Dahuang Huanglian Xiexin decoction(DHXD) to treat type 2 diabetes mellitus(T2DM) and to introduce a new treatment option for clinicians.METHODS: Retrospective analysis was used to evaluate DHXD for the treatment of T2DM by analyzing clinical records of 183 cases. Patients with T2DM who met the inclusion criteria between January 1,2013 and January 1, 2014 were enrolled. The effects of the treatment were evaluated by the changes in fasting blood-glucose(FBG), postprandial blood sugar(PBG), hemoglobin A1c(Hb Alc), blood lipid profiles and body mass index(BMI) at 1, 2, 3 and 6 months. The changes in main symptoms were alsoevaluated. The dosage of Huanglian(Rhizoma Coptidis) and related factors were analyzed.RESULTS: There was a significant improvement in mean Hb A1C at 3 and 6 months after DHXD treatment compared with the baseline level(P < 0.01).There were also significant improvements in FBG,PBG, blood lipid series and BMI. DHXD also improved the main symptoms of stomach and intestine excessive heat syndrome in patients with obese T2DM. Huanglian(Rhizoma Coptidis) was the most frequently used in 678 clinical visits, the dosage of Huanglian(Rhizoma Coptidis) was related to age, BMI, DM duration, the level of blood glucose,and use of Western hypoglycemic drugs.CONCLUSION: This study suggests that DHXD could decrease blood glucose and improve T2DM symptoms and reduce body weight. 13Buried straw layer and plastic mulching increase microflora diversity in salinized soil显示文摘Salt stress has been increasingly constraining crop productivity in arid lands of the world. In our recent study, salt stress was alleviated and crop productivity was improved remarkably by straw layer burial plus plastic film mulching in a saline soil. However, its impact on the microflora diversity is not well documented. Field micro-plot experiments were conducted from 2010 to 2011 using four tillage methods:(i) deep tillage with plastic film mulching(CK),(ii) straw layer burial at 40 cm(S),(iii) straw layer burial plus surface soil mulching with straw material(S+S), and(iv) plastic film mulching plus buried straw layer(P+S). Culturable microbes and predominant bacterial communities were studied; based on 16 S r DNA, bacterial community structure and abundance were characterized using denaturing gradient gel electrophoresis(DGGE) and polymerase chain reaction(PCR). Results showed that P+S was the most favorable for culturable bacteria, actinomyces and fungi and induced the most diverse genera of bacteria compared to other tillage methods. Soil temperature had significant positive correlations with the number of bacteria, actinomyces and fungi(P<0.01). However, soil water was poorly correlated with any of the microbes. Salt content had a significant negative correlation with the number of microbers, especially for bacteria and fungi(P<0.01). DGGE analysis showed that the P+S exhibited the highest diversity of bacteria with 20 visible bands followed by S+S, S and CK. Moreover, P+S had the highest similarity(68%) of bacterial communities with CK. The major bacterial genera in all soil samples were Firmicutes, Proteobacteria and Actinobacteria. 14Cardioprotective effects of ghrelin and des-octanoyl ghrelin on myocardial injury induced by isoproterenol in rats显示文摘<正> Aim:To determine the cardioprotective action of ghrelin and des-octanoyl ghrelinin rats with isoproterenol-induced myocardial injury.Methods:Rats were subcu-taneously injected with isoproterenol (ISO;20,10,and 5 mg/kg) on d 1,2 and 3,respectively,and then 3 mg/kg for the next 7d with or without ghrelin or des-octanoyl-ghrelin (100μg/kg,twice daily).Plasma ghrelin and growth hormonelevels were assayed using radioimmunoassay methods.Growth hormone secreta-gogue receptor (GHSR) and ghrelin mRNA were determined using RT-PCR.Themaximal binding capacity and the affinity for [~3H]ghrelin were determined by re-ceptor binding assays.Results:Compared with controls,ISO-treated rats showedsevere myocardial injury,cardiomegaly,infarction-like necrosis and massive fi-brosis with increases in irradiated-ghrelin (ir-ghrelin) content in plasma by 67%and myocardia by 66% and in the mRNA level in the myocardia by 93% (P<0.01).ISO-treated rats had 95% (P<0.01) higher GHSR mRNA levels in the myocardia.The maximal binding capacity of [~3H]ghrelin for myocardial sarcolemma was higherin ISO-treated rats than in controls.Ghrelin administration improved cardiac func-tion and ameliorated cardiomegaly and attenuated myocardial lipid peroxidationinjury and relieved cardiac fibrosis as compared with ISO treatment alone.Admin-istration of des-octanoyl ghrelin effectively antagonized ISO-induced myocardialinjury and improved all parameters measured.However,the therapeutic effect ofdes-octanoyl ghrelin was significantly weaker than that of ghrelin.The plasmagrowth hormone level increased markedly,by 1.5-fold (P<0.01),with ghrelinadministration as compared with that in controls,but was unaltered in the des-octanoyl ghrelin group.Conclusion:Myocardial ghrelin and GHSR were up-regulated during ISO-induced myocardial injury.The protective effect of ghrelinagainst ISO-induced cardiac function injury and fibrosis was more potent thanthat of des-octanoyl ghrelin,which suggests that ghrelin could be an endog-enous cardioprotective factor in ischemic heart disease,and that its effects in-clude growth hormone-dependent and-independent pathways.Lian LI Li-ke ZHANG Yong-zheng PANG Chun-shui PAN Yong-fen QI Li CHEN Xian WANG Chao-shu TANG Jing ZHANG 2006Acta Pharmacologica Sinica2006,27,5:9
15Drug targeting through platelet membrane-coated nanoparticles for the treatment of rheumatoid arthritis显示文摘The effective drug treatment of rheumatoid arthritis (RA) is hindered by poordelivery efficiency to the diseased site and the serious side effects caused bywide-spread drug distribution. Traditional drug-targeting strategies, such asligand modification, are complex, laborious, and inefficient. Inspired by theintrinsic relationship between platelets and RA, platelet-mimetic nanoparticles(PNPs) were developed for targeted drug delivery in RA. Through plateletreceptor-mediated adhesion, an intact platelet membrane was coated onto poly(lactic-co-glycolic add) nanopartides, endowing the resulting PNPs with variousfunctional receptors. By coating with platelet membranes, the nanoparticleswere stabilized and had a better circulation profile, providing a benefit forpassive targeting. In vitro binding of PNPs to inflamed endothelium, and in vivoaccumulation in joints of a collagen-induced arthritis (CIA) mouse model of RAwere significantly improved via P-selectin and GVPI recognition, indicating thatthe PNPs could effectively target to RA tissues through multiple mechanisms,similar to natural platelets. 17Risk of Active Pulmonary Tuberculosis among Patients with Coal Workers'Pneumoconiosis:A Case-control Study in China显示文摘The aim of this study was to estimate the association between occupational, environmental, behavioral risk factors, and active pulmonary tuberculosis(PTB) among coal workers' pneumoconiosis(CWP) patients. A matched case-control study was conducted in 86 CWP patients with active PTB and 86 CWP controls without TB. A standardized questionnaire was used for risk factors assessment. Conditioned logistic regression analysis was used to identify associations between the risk factors and active PTB among CWP patients. 19Hypoxic preconditioning reduces NLRP3 inflammasome expression and protects against cerebral ischemia/reperfusion injury显示文摘Hypoxic preconditioning can protect against cerebral ischemia/reperfusion injury. However, the underlying mechanisms that mediate this effect are not completely clear. In this study, mice were pretreated with continuous, intermittent hypoxic preconditioning;1 hour later, cerebral ischemia/reperfusion models were generated by middle cerebral artery occlusion and reperfusion. Compared with control mice, mice with cerebral ischemia/reperfusion injury showed increased Bederson neurological function scores, significantly increased cerebral infarction volume, obvious pathological damage to the hippocampus, significantly increased apoptosis;upregulated interleukin-1β, interleukin-6, and interleukin-8 levels in brain tissue;and increased expression levels of NOD-like receptor family pyrin domain containing 3(NLRP3), NLRP inflammasome-related protein caspase-1, and gasdermin D. However, hypoxic preconditioning significantly inhibited the above phenomena. Taken together, these data suggest that hypoxic preconditioning mitigates cerebral ischemia/reperfusion injury in mice by reducing NLRP3 inflammasome expression. 20The R2R3-MYB transcription factor GhMYB1a regulates flavonol and anthocyanin accumulation in Gerbera hybrida显示文摘Anthocyanins and flavonols have vital roles in flower coloration,plant development,and defense.Because anthocyanins and flavonols share the same subcellular localization and common biosynthetic substrates,these pathways may compete for substrates.However,the mechanism regulating this potential competition remains unclear.Here,we identified GhMYB1a,an R2R3-MYB transcription factor involved in the regulation of anthocyanin and flavonol accumulation in gerbera(Gerbera hybrida).GhMYB1a shares high sequence similarity with that of other characterized regulators of flavonol biosynthesis.In addition,GhMYB1a is also phylogenetically grouped with these proteins.The overexpression of GhMYB1a in gerbera and tobacco(Nicotiana tabacum)resulted in decreased anthocyanin accumulation and increased accumulation of flavonols by upregulating the structural genes involved in flavonol biosynthesis.We further found that GhMYB1a functions as a homodimer instead of interacting with basic helix-loop-helix cofactors.These results suggest that GhMYB1a is involved in regulating the anthocyanin and flavonol metabolic pathways through precise regulation of gene expression.The functional characterization of GhMYB1a provides insight into the biosynthesis and regulation of flavonols and anthocyanins.Chunmei Zhong Yi Tang Bin Pang Xukun Li Yuping Yang Jing Deng Chengyong Feng Lingfei Li Guiping Ren Yaqin Wang Jianzong Peng Shulan Sun Shan Liang Xiaojing Wang 2020Horticulture Research2020,7,1:7
