| 1 | A universal pipeline for mobile mRNA detection and insights into heterografting advantages under chilling stress显示文摘Heterografting has long been used to enhance the chilling tolerance of temperature-sensitive crops,including watermelon,whose mechanism is known to involve bidirectional long-distance mRNA movements.Despite several studies reporting on mobile mRNA(mb-mRNA)profiles in plants,accurate identification of mb-mRNAs is challenging owing to an array of technical problems.Here,we developed a bioinformatical pipeline that took most of the known technical concerns into consideration and is considered to be a universal tool for mb-mRNA detection in heterografts.By applying this pipeline to a commercial watermelon–bottle gourd heterografting system,we detected 130 and 1144 mb-mRNAs upwardly and 167 and 1051 mb-mRNAs downwardly transmitted under normal and chilling-stress conditions,respectively.Quantitative real-time PCR indicated a high accuracy rate(88.2%)of mb-mRNA prediction with our pipeline.We further revealed that the mobility of mRNAs was not associated with their abundance.Functional annotation and classification implied that scions may convey the stress signal to the rootstock,subsequently triggering energy metabolism reprogramming and abscisic acid-mediated stress responses by upward movement of effective mRNAs,ultimately leading to enhanced chilling tolerance.This study provides a universal tool for mb-mRNA detection in plant heterografting systems and novel insights into heterografting advantages under chilling stress. | Ying Wang Lingping Wang Nailin Xing Xiaohua Wu Xinyi Wu Baogen Wang Zhongfu Lu Pei Xu Ye Tao Guojing Li Yuhong Wang | 2020 | Horticulture Research2020,7,1: | 3 |

| 5 | JaponiconeA induces apoptosis of bortezomib-sensitive and -resistant myeloma cells in vitro and in vivo by targeting IKKβ显示文摘Objective:Multiple myeloma(MM)remains incurable with high rates of relapse.New therapeutic drugs are therefore urgently needed to improve the prognosis.JaponiconeA(JA),a natural product isolated from Inula japonica Thunb,has shown good anti-MM potential.A comprehensive study should therefore be conducted to identify both the in vitro and in vivo mechanisms of the anti-MM effects of JA.Methods:CCK8 assays and flow cytometry were used to detect the proliferation,apoptosis,and cell cycle of MM cell lines when treated with JA.In vivo experiments were conducted using subcutaneous xenograft mouse models.We also identified possible targets and the mechanism of JA using RNA-seq and c-Map databases,and identified the specific targets of JA in bortezomib-sensitive and-resistant MM cell lines using CETSA,DARTS,and rescue experiments.Furthermore,JA and bortezomib were used separately or together to characterize their possible synergistic effects.Results:In vitro,JA inhibited proliferation,and induced apoptosis and G2/M phase arrest in MM cell lines,and selectively killed primary CD138+MM cells.In vivo,JA also demonstrated a strong anti-tumor effect with no observable toxicity.In addition,JA showed synergetic effects in combination with bortezomib,and enhanced the anti-tumor effect of bortezomib in bortezomibresistant cells.CETSA and DARTS confirmed direct binding of JA to NF-κB inhibitor kinase beta(IKKβ),and overexpression of IKKβor knockdown of IκBαpartially rescued the apoptosis induced by JA.Conclusions:JA exhibited strong anti-tumor effects in MM.It sensitized myeloma cells to bortezomib and overcame NF-κB-induced drug resistance by inhibiting IKKβ,providing a new treatment strategy for MM patients. | Zilu Zhang Chenjing Ye Jia Liu Wenbin Xu Chao Wu Qing Yu Xiaoguang Xu Xinyi Zeng Huizi Jin Yingli Wu Hua Yan | 2022 | Cancer Biology & Medicine2022,19,5: | 1 |

| 13 | Application Research of Detailed Nursing of Tuberculous Meningitis Patients显示文摘Objective:To explore the application effect of detailed nursing in tuberculous meningitis nursing.Methods:A total of 52 patients with tuberculous meningitis who were admitted to our hospital from May 2019 to February 2022 were selected as the research objects,and divided into 25 patients in the control group and 27 patients in the study group according to the random table method,and the compliance,quality of life,recurrence,nursing satisfaction,depression,anxiety and PSQI scores of the two groups were observed and analyzed.Results:In the study group,the number of patients with complete compliance,partial compliance and non-compliance were 13,11 and 3,respectively,and the compliance rate was 88.89%.In the control group,the number of patients with complete compliance,partial compliance and non-compliance were 11,5 and 9,respectively,and the compliance rate was 67.86%,and P<0.05 was considered statistically significant.The quality-of-life score of the control group was 48.36±2.24,which was lower than that of the study group,50.23±2.12,(P<0.05).The rate of recurrence of the control group was 36.00%,which was significantly higher than 7.41%in the study group(P<0.05).There was no significant statistical difference between the SAS,SDS,and PSQI scores of the two groups before nursing,(P>0.05).After nursing,both groups showed a decrease in SDS and SAS scores,with the study group showing significantly lower scores than the control group(P<0.001).Besides,the PSQI dimension scores and total score in the study group were lower than those in the control group(P<0.001).Moreover,the nursing satisfaction of the patients in the study group was significantly higher than that in the control group(93.02%>71.79%),(P<0.05).Conclusion:A full range of detailed care is not only helpful for accurate diagnosis and treatment of patients,but also helps to create a satisfactory hospital environment for patients.Results from this study showed that detailed nursing care for patients with tuberculous meningitis shows a significant positive effect and should be applied in clinical practices. | Wei Yuan Dandan Yang Zhangying Li Chuo Guo Xinyi Ye Min Zheng | 2022 | Journal of Clinical and Nursing Research2022,6,5: | 0 |

| 16 | Genome sequencing reveals the role of rare genomic variants in Chinese patients with symptomatic intracranial atherosclerotic disease显示文摘Objectives The predisposition of intracranial atherosclerotic disease(ICAD)to East Asians over Caucasians infers a genetic basis which,however,remains largely unknown.Higher prevalence of vascular risk factors(VRFs)in Chinese over Caucasian patients who had a stroke,and shared risk factors of ICAD with other stroke subtypes indicate genes related to VRFs and/or other stroke subtypes may also contribute to ICAD.Methods Unrelated symptomatic patients with ICAD were recruited for genome sequencing(GS,60-fold).Rare and potentially deleterious single-nucleotide variants(SNVs)and small insertions/deletions(InDels)were detected in genome-wide and correlated to genes related to VRFs and/or other stroke subtypes.Rare aneuploidies,copy number variants(CNVs)and chromosomal structural rearrangements were also investigated.Lastly,candidate genes were used for pathway and gene ontology enrichment analysis.Results Among 92 patients(mean age at stroke onset 61.0±9.3 years),GS identified likely ICAD-associated rare genomic variants in 54.3%(50/92)of patients.Forty-eight patients(52.2%,48/92)had 59 rare SNVs/InDels reported or predicted to be deleterious in genes related to VRFs and/or other stroke subtypes.None of the 59 rare variants were identified in local subjects without ICAD(n=126).31 SNVs/InDels were related to conventional VRFs,and 28 were discovered in genes related to other stroke subtypes.Our study also showed that rare CNVs(n=7)and structural rearrangement(a balanced translocation)were potentially related to ICAD in 8.7%(8/92)of patients.Lastly,candidate genes were significantly enriched in pathways related to lipoprotein metabolism and cellular lipid catabolic process.Conclusions Our GS study suggests a role of rare genomic variants with various variant types contributing to the development of ICAD in Chinese patients. | Mengmeng Shi Xinyi Leng Ying Li Zihan Chen Ye Cao Tiffany Chung Bonaventure YM Ip Vincent HL Ip Yannie OY Soo Florence SY Fan Sze Ho Ma Karen Ma Anne Y Y Chan Lisa WC Au Howan Leung Alexander Y Lau Vincent CT Mok Kwong Wai Choy Zirui Dong Thomas W Leung | 2022 | Stroke & Vascular Neurology2022,7,3: | 0 |
