1Isoliquiritigenin inhibits TGF-β1-induced fibrogenesis through activating autophagy via PI3K/AKT/mTOR pathway in MRC-5 cells显示文摘Isoliquiritigenin(ISL),a natural flavonoid derived from the root of liquorice,has been reported to possess anti-inflammatory and antioxidant activities.Previous studies have found that ISL plays a crucial role in anti-fibrosis of adipose tissue and renal tissue;however,its effect on pulmonary fibrogenesis has not been demonstrated.In this study,we aimed to explore the roles and the underlying mechanisms of ISL in TGF-β1-induced fibrogenesis using human lung fibroblast-derived MRC-5 cells.Cell proliferation and migration were determined by MTT and wound healing assay,respectively.The expression levels of alpha-smooth muscle actin(α-SMA),collagen type I alpha 1(COLIA1)and fibronectin(FN),microtubule-associated protein light chain 3(LC3)and related signaling molecules were detected by quantitative real-time PCR,western blot and immunofluorescence assay,correspondingly.EGFP-LC3 transfection was used for autophagy analysis.The results showed that ISL inhibited the TGF-β1-induced proliferation and migration,and down-regulated the expressions ofα-SMA,COLIA1 and FN.ISL treatment led to up-regulation of LC3 in TGF-β1-treated MRC-5 cells,accompanied by significant decrease in the phosphorylation levels of phosphatidylinositol 3-kinase(PI3K),protein kinase B(AKT),and mammalian target of rapamycin(mTOR).In addition,the inhibitory effects of ISL on TGF-β1-induced fibrogenic features in MRC-5 cells were enhanced by pretreatment with autophagy activator Rapmycin and PI3K/AKT inhibitor LY294002 and reversed by autophagy inhibitor 3-methyladenine and PI3K/AKT activator IGF-1.Taken together,our results demonstrated that ISL could attenuate the fibrogenesis of TGF-β1-treated MRC-5 cells by activating autophagy via suppressing the PI3K/AKT/mTOR pathway.Therefore,ISL holds a great potential to be developed as a novel therapeutic agent for the treatment of pulmonary fibrosis.Jinjuan He Hao Peng Meifang Wang Ying Liu Xingrong Guo Bin Wang Longjun Dai Xueqin Cheng Zhongji Meng Leyong Yuan Fenglin Cai Yijun Tang 2020Acta Biochimica et Biophysica Sinica2020,52,8:16
2Study on Agricultural Intellectual Property Protection and High-Quality Development of Traditional Chinese Medicine of Ginseng显示文摘Agricultural intellectual property has the functions of carrying forward agricultural cultural heritage,encouraging agricultural scientific and technological innovation,cultivating the brand of agricultural products and so on. The protection and application of agricultural intellectual property rights can enhance the vitality,innovation and competitiveness of agricultural economy,and promote the strategy of high-quality agricultural development and rural revitalization. Ginseng is a traditional medicinal variety with superior characteristics in Northeast China,and it is one of the most famous ' three treasures' in Northeast China. This paper introduces ginseng( Panax ginseng C. A. 3The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 4PagGRF12a interacts with PagGIF1b to regulate secondary xylem development through modulating PagXND1a expression in Populus alba×P.glandulosa显示文摘Growth-regulating factors(GRFs)are important regulators of plant development and growth,but their possible roles in xylem development in woody plants remain unclear.Here,we report that Populus alba×Papulus glandulosa PagGRF12a negatively regulates xylem development in poplar.PagGRF12a is expressed in vascular tissues.Compared to non-transgenic control plants,transgenic poplar plants overexpressing PagGRF12a exhibited reduced xylem width and plants with repressed expression of PagGRF12a exhibited increased xylem width.Xylem NAC domain 1(XND1)encodes a NAC domain transcription factor that regulates xylem development and transcriptional analyses revealed that PagXND1a is highly upregulated in PagGRF12a-overexpressing plants and downregulated in PagGRF12a-suppressed plants,indicating that PagGRF12a may regulate xylem development through PagXND1a.Transient transcriptional assays and chromatin immunoprecipitation-polymerase chain reaction assays confirmed that PagGRF12a directly upregulates PagXND1a.In addition,PagGRF12a interacts with the GRF-Interacting Factor(GIF)PagGIF1b,and this interaction enhances the effects of PagGRF12a on PagXND1a.Our results indicate that PagGRF12a inhibits xylem development by upregulating the expression of PagXND1a.Jinnan Wang Houjun Zhou Yanqiu Zhao Cheng Jiang Jihong Li Fang Tang Yingli Liu Shutang Zhao Jianjun Hu Xueqin Song MengZhu Lu 2021Journal of Integrative Plant Biology2021,63,10:3

9Environmental selection and evolutionary process jointly shape genomic and functional profiles of mangrove rhizosphere microbiomes显示文摘Mangrove reforestation with introduced species has been an important strategy to restore mangrove ecosystem functioning.However,how such activities affect microbially driven methane(CH4),nitrogen(N),and sulfur(S)cycling of rhizosphere microbiomes remains unclear.To understand the effect of environmental selection and the evolutionary process on microbially driven biogeochemical cycles in native and introduced mangrove rhizospheres,we analyzed key genomic and functional profiles of rhizosphere microbiomes from native and introduced mangrove species by metagenome sequencing technologies.Compared with the native mangrove(Kandelia obovata,KO),the introduced mangrove(Sonneratia apetala,SA)rhizosphere microbiome had significantly(p<0.05)higher average genome size(AGS)(5.8 vs.5.5 Mb),average 16S ribosomal RNA gene copy number(3.5 vs.3.1),relative abundances of mobile genetic elements,and functional diversity in terms of the Shannon index(7.88 vs.7.84)but lower functional potentials involved in CH4 cycling(e.g.,mcrABCDG and pmoABC),N2 fixation(nifHDK),and inorganic S cycling(dsrAB,dsrC,dsrMKJOP,soxB,sqr,and fccAB).Similar results were also observed from the recovered Proteobacterial metagenome-assembled genomes with a higher AGS and distinct functions in the introduced mangrove rhizosphere.Additionally,salinity and ammonium were identified as the main environmental drivers of functional profiles of mangrove rhizosphere microbiomes through deterministic processes.This study advances our understanding of microbially mediated biogeochemical cycling of CH_(4),N,and S in the mangrove rhizosphere and provides novel insights into the influence of environmental selection and evolutionary processes on ecosystem functions,which has important implications for future mangrove reforestation.Xiaoli Yu Qichao Tu Jihua Liu Yisheng Peng Cheng Wang Fanshu Xiao Yingli Lian Xueqin Yang Ruiwen Hu Huang Yu Lu Qian Daoming Wu Ziying He Longfei Shu Qiang He Yun Tian Faming Wang Shanquan Wang Bo Wu Zhijian Huang Jianguo He Qingyun Yan Zhili He 2023mLife2023,2,3:0

These y-CD derivatives form stable 1:2 ternary complexes with AC in aqueous ammonia solutions and the head-tohead photodimers 3 and 4 were greatly enhanced in the presence of these diguanidio-γ-CDs. 11Study on Element Contents in Tobacco Leaves from Different Positions of Tobacco CB-1显示文摘[Objectives]This study was conducted to clarify the differences and abundance or deficiency of various elements in tobacco leaves from different positions of CB-1 in Zhangping City,so as to guide the production of precision fertilization.[Methods]The element contents of CB-1 were determined by an X-ray fluorescence spectrometer.[Results]In terms of the contents of carbon,the upper leaves(45.73%)were significantly higher than the cutters(41.53%),and the lower leaves(43.15%)had no significant differences from both of them.For the content of oxygen element,there was no significant difference between the cutters(44.77%)and the lower leaves(43.40%),both of which were significantly higher than the upper leaves(40.75%).The contents of nitrogen,phosphorus,sulfur,boron and nickel all increased with the increase of the position,and they were significantly higher in the upper leaves than in the cutters and lower leaves.The contents of potassium,calcium,magnesium,chlorine and silicon all decreased with the increase of the position,and the lower leaves were significantly higher than the cutters and upper leaves.The contents of aluminum,titanium and chromium increased significantly with the increase of the position.The contents of strontium,zirconium and arsenic decreased significantly with the increase of the position.The element contents of tobacco leaves had significant or extremely significant differences between different parts of CB-1 in Zhangping City mostly.The contents of nitrogen,potassium,calcium,sulfur,chlorine,molybdenum and nickel in tobacco leaves were appropriate,and the contents of manganese and iron were too high,while the contents of phosphorus and copper were slightly low.The contents of magnesium,silicon,boron and sodium were too low.[Conclusions]This study can guide targeted fertilization and regulation of element contents in the tobacco leaves from different parts in production,and is of great significance to improving the production,quality and industrial availability of CB-1.Caijun XIE Shilei ZHANG Deqing CHEN Penghui LYU Wensheng LIN Jianrong ZHENG Jinchai YU Qiuying CAO Siyi CHENG Xueqin ZHANG Yongxin FENG Zenghan XU 2022Agricultural Biotechnology2022,11,6:0
12Effects of the structure of the branches on the two-photon absorption properties for the multi-branched molecules with nitrogen (N) as coupling center显示文摘In order to investigate the effects of the structure of branches on the TPA properties for multi-branched molecules, the TPA cross section is calculated by using ZINDO/SOS method. The investigated mole- cules have different branches (chomorfores based on stilbene, dithienothiophene and flourene) with nitrogen(N) as coupling center. The results show that the cooperative enhancement in multi-branched molecules depends on the structures of the branches and the structures of branches play an important role in the enhancement of the TPA cross section. 13G protein-coupled receptor 35 attenuates nonalcoholic steatohepatitis by reprogramming cholesterol homeostasis in hepatocytes显示文摘Nonalcoholic fatty liver disease(NAFLD)is the most common chronic liver disease worldwide.Fat accumulation“sensitizes”the liver to insult and leads to nonalcoholic steatohepatitis(NASH).G protein-coupled receptor 35(GPR35)is involved in metabolic stresses,but its role in NAFLD is unknown.We report that hepatocyte GPR35 mitigates NASH by regulating hepatic cholesterol homeostasis.Specifically,we found that GPR35 overexpression in hepatocytes protected against high-fat/cholesterol/fructose(HFCF)diet-induced steatohepatitis,whereas loss of GPR35 had the opposite effect.Administration of the GPR35 agonist kynurenic acid(Kyna)suppressed HFCF diet-induced steatohepatitis in mice.Kyna/GPR35 induced expression of StAR-related lipid transfer protein 4(STARD4)through the ERK1/2 signaling pathway,ultimately resulting in hepatic cholesterol esterification and bile acid synthesis(BAS).The overexpression of STARD4 increased the expression of the BAS rate-limiting enzymes cytochrome P450 family 7 subfamily A member 1(CYP7A1)and CYP8B1,promoting the conversion of cholesterol to bile acid.The protective effect induced by GPR35 overexpression in hepatocytes disappeared in hepatocyte STARD4-knockdown mice.STARD4 overexpression in hepatocytes reversed the aggravation of HFCF diet-induced steatohepatitis caused by the loss of GPR35 expression in hepatocytes in mice.Our findings indicate that the GPR35–STARD4 axis is a promising therapeutic target for NAFLD.Xiaoli Wei Fan Yin Miaomiao Wu Qianqian Xie Xueqin Zhao Cheng Zhu Ruiqian Xie Chongqing Chen Menghua Liu Xueying Wang Ruixue Ren Guijie Kang Chenwen Zhu Jingjing Cong Hua Wang Xuefu Wang 2023Acta Pharmaceutica Sinica B2023,13,3:0
