2Antioxidant and Anti-inflammatory Capacity of Ferulic Acid Released from Wheat Bran by Solid-state Fermentation of Aspergillus niger显示文摘Objective A strain of Aspergillus niger(A. niger), capable of releasing bound phenolic acids from wheat bran, was isolated. This strain was identified by gene sequence identification. The antioxidant and anti-inflammatory capacity of ferulic acid released from wheat bran by this A. niger strain(FA-WB) were evaluated. Methods Molecular identification techniques based on PCR analysis of specific genomic sequences were conducted; antioxidant ability was examined using oxygen radical absorbance capacity(ORAC), cellular antioxidant activity(CAA) assays, and erythrocyte hemolysis assays. RAW264.7 cells were used as a model to detect anti-inflammatory activity. Results The filamentous fungal isolate was identified to be A. niger. ORAC and CAA assay showed that FA-WB had better antioxidant activity than that of the ferulic acid standard. The erythrocyte hemolysis assay results suggested that FA-WB could attenuate AAPH-induced oxidative stress through inhibition of reactive oxy gen species(ROS) generation. FA-WB could significantly restore the AAPH-induced increase in intracellular antioxidant enzyme activities to normal levels as well as inhibit the intracellular malondialdehyde formation. TNF-?, IL-6, and NO levels indicated that FA-WB can inhibit the inflammation induced by lipopolysaccharide(LPS). 3The Performance of Whole Genome Amplification Methods and Next-Generation Sequencing for Pre-Implantation Genetic Diagnosis of Chromosomal Abnormalities显示文摘Reliable and accurate pre-implantation genetic diagnosis(PGD) of patient's embryos by next-generation sequencing(NGS) is dependent on efficient whole genome amplification(WGA) of a representative biopsy sample. However, the performance of the current state of the art WGA methods has not been evaluated for sequencing. Using low template DNA(15 pg) and single cells, we showed that the two PCR-based WGA systems Sure Plex and MALBAC are superior to the REPLI-g WGA multiple displacement amplification(MDA) system in terms of consistent and reproducible genome coverage and sequence bias across the 24 chromosomes, allowing better normalization of test to reference sequencing data. When copy number variation sequencing(CNV-Seq) was applied to single cell WGA products derived by either Sure Plex or MALBAC amplification, we showed that known disease CNVs in the range of 3e15 Mb could be reliably and accurately detected at the correct genomic positions. 4Endoscopic management of biliary complications after orthotopic liver transplantation显示文摘BACKGROUND: Biliary complications are a serious problem in patients after liver transplantation and often require reoperation. This study was conducted to summarize the endoscopic diagnosis and management of biliary complications after orthotopic liver transplantation (OLT). METHODS: From December 2000 to November 2003, twelve endoscopic retrograde cholangiopancreatographies(ERCPs) were performed in 7 patients after OLT at Digestive Endoscopic Center of Changhai Hospital in Shanghai, China. The therapeutic maneuvers included endoscopic sphincterotomy (EST), biliary stent placement, balloon and basket extraction, irrigation, and nasobiliary tube placement. A retrospective study was made to determine the types of biliary tract complications after OLT. The success of ERCP and therapeutic maneuvers was also evaluated. RESULTS: Biliary tract complications including biliary stricture, biliary leak, biliary sludge, and stump leak of the cyst duct were treated respectively by endoscopic sphincterotomy with sludge extraction, stricture dilation or endoscopic retrograde biliary drainage. Two of the 3 patients with proximal common bile duct stricture were successfully treated with ERCP and stent placement. Four patients with anastomotic stricture and/without bile leak were treated successfully by dilation and stent placement or endoscopic nosobiliary drainage. No severe ERCP-related complications occurred. 5Efficacy of chitosan and sodium alginate scaffolds for repair of spinal cord injury in rats显示文摘Spinal cord injury results in the loss of motor and sensory pathways and spontaneous regeneration of adult mammalian spinal cord neurons is limited. Chitosan and sodium alginate have good biocompatibility, biodegradability, and are suitable to assist the recovery of damaged tissues, such as skin, bone and nerve. Chitosan scaffolds, sodium alginate scaffolds and chitosan-sodium alginate scaffolds were separately transplanted into rats with spinal cord hemisection. Basso-Beattie-Bresnahan locomotor rating scale scores and electrophysiological results showed that chitosan scaffolds promoted recovery of locomotor capacity and nerve transduction of the experimental rats. Sixty days after surgery, chitosan scaffolds retained the original shape of the spinal cord. Compared with sodium alginate scaffolds-and chitosan-sodium alginate scaffolds-transplanted rats, more neurofilament-H-immunoreactive cells(regenerating nerve fibers) and less glial fibrillary acidic protein-immunoreactive cells(astrocytic scar tissue) were observed at the injury site of experimental rats in chitosan scaffold-transplanted rats. Due to the fast degradation rate of sodium alginate, sodium alginate scaffolds and composite material scaffolds did not have a supporting and bridging effect on the damaged tissue. 8Pathophysiological mechanisms and therapeutic approaches in obstructive sleep apnea syndrome显示文摘Obstructive sleep apnea syndrome(OSAS)is a common breathing disorder in sleep in which the airways narrow or collapse during sleep,causing obstructive sleep apnea.The prevalence of OSAS continues to rise worldwide,particularly in middle-aged and elderly individuals.The mechanism of upper airway collapse is incompletely understood but is associated with several factors,including obesity,craniofacial changes,altered muscle function in the upper airway,pharyngeal neuropathy,and fluid shifts to the neck.The main characteristics of OSAS are recurrent pauses in respiration,which lead to intermittent hypoxia(IH)and hypercapnia,accompanied by blood oxygen desaturation and arousal during sleep,which sharply increases the risk of several diseases.This paper first briefly describes the epidemiology,incidence,and pathophysiological mechanisms of OSAS.Next,the alterations in relevant signaling pathways induced by IH are systematically reviewed and discussed.For example,IH can induce gut microbiota(GM)dysbiosis,impair the intestinal barrier,and alter intestinal metabolites.These mechanisms ultimately lead to secondary oxidative stress,systemic inflammation,and sympathetic activation.We then summarize the effects of IH on disease pathogenesis,including cardiocerebrovascular disorders,neurological disorders,metabolic diseases,cancer,reproductive disorders,and COVID-19.Finally,different therapeutic strategies for OSAS caused by different causes are proposed.Multidisciplinary approaches and shared decision-making are necessary for the successful treatment of OSAS in the future,but more randomized controlled trials are needed for further evaluation to define what treatments are best for specific OSAS patients.Renjun Lv Xueying Liu Yue Zhang Na Dong Xiao Wang Yao He Hongmei Yue Qingqing Yin 2023Signal Transduction and Targeted Therapy2023,8,6:8
9Distribution and characteristic of PAHs in snow of Fildes Peninsula显示文摘Polycyclic aromatic hydrocarbons (PAHs) investigation in different matrices has been reported largely,whereas reports on snow samples were limited.Snow,as the main matrix in the polar region,has an important study meaning.PAHs in snow samples were analyzed to investigate the distribution and contamination status of them in the Antarctic,as well as to provide some references for global migration of PAHs.Snow samples collected in Fildes Peninsula were enriched and separated by solid-phase membrane disks and eluted by methylene dichloride,then quantified by gas chromatography/mass spectrometry.All types of PAHs were detected except for Benzo(a)pyrene.Principal component analysis method was applied to characterize them.Three factors (Naphthalene,Fluorene and Phenanthrene) accounted for 60.57%,21.61% and 9.80%,respectively.The results showed that the major PAHs sources maybe the atmospheric transportation,and the combustion of fuel in Fildes Peninsula.The comparison of concentration and types of PAHs between accumulated snow and fresh snow showed that the main compound concentrations in accumulated snow samples were higher than those in fresh ones.The risk assessment indicated that the amount of PAHs in the snow samples would not lead to ecological risk.Guangshui Na Chunyang Liu Zhen Wang Linke Ge Xindong Ma Ziwei Yao 2011Journal of Environmental Sciences2011,23,9:8
10Functional analysis of the ABA-responsive protein family in ABA and stress signal transduction in Arabidopsis显示文摘The phytohormone abscisic acid(ABA)plays an important role in plant growth and development,for example in seed dormancy and germination,as well as in plant responses to environmental stresses,such as drought and high salinity.Previous studies have shown that ABA regulates the expression of genes with an ABA-responsive element(ABRE)and their corresponding physiological responses.Bioinformatics analysis identified a GRAM domain-containing gene family that has a multiple ABRE cis-element,which was termed the ABA-responsive protein(ABR)family.To analyze the function of the ABR family,we identified homozygous T-DNA insertion mutants and constructed abr1,2,3 double mutants and triple mutant.The abr1,abr2 and abr3single mutants showed a normal phenotype;however,the germination of seeds of the double mutants and triple mutant were insensitive to ABA,NaCl,mannitol and glucose.ABR1-GFP was distributed as a punctate structure in the cytosol and may be localized in the endomembrane system.The ABR2-GFP and ABR3-GFP proteins localized in the cytoplasm.In addition,ABR1,ABR2 and ABR3 were expressed in various tissues,and could be induced by several abiotic stresses,especially by ABA.The expressions of these genes were significantly suppressed in aba2,abi1 and abi2 null mutants.These results suggested that the ABR family may act downstream of ABI1 and ABI2 in the ABA signal transduction process in plants.LIU LingYun LI Na YAO ChunPeng MENG SaSa SONG ChunPeng 2013Chinese Science Bulletin2013,58,31:7
11Biodegradation of pendimethalin by Bacillus subtilis Y3显示文摘A bacterium strain Y3,capable of efficiently degrading pendimethalin,was isolated from activated sludge and identified as Bacillus subtilis according to its phenotypic features and 16 S rRNA phylogenetic analysis.This strain could grow on pendimethalin as a sole carbon source and degrade 99.5%of 100 mg/L pendimethalin within 2.5 days in batch liquid culture,demonstrating a greater efficiency than any other reported strains.Three metabolic products,6-aminopendimethalin,5-amino-2-methyl-3-nitroso-4-(pentan-3-ylamino) benzoic acid,and 8-amino-2-ethyl-5-(hydroxymethyl)-1,2-dihydroquinoxaline-6-carboxylic acid,were identified by HPLC-MS/MS,and a new microbial degradation pathway was proposed.A nitroreductase catalyzing nitroreduction of pendimethalin to 6-aminopendimethalin was detected in the cell lysate of strain Y3.The cofactor was nicotinamide adenine dinucleotide phosphate(NADPH) or more preferably nicotinamide adenine dinucleotide(NADH).The optimal temperature and pH for the nitroreductase were 30℃ and 7.5,respectively.Hg^(2+),Ni^(2+),Pb^(2+),Co^(2+),Mn^(2+) Cu^(2+),Ag^+,and EDTA severely inhibited the nitroreductase activity,whereas Fe^(2+),Mg^(2+),and Ca^(2+) enhanced it.This study provides an efficient pendimethalin-degrading microorganism and broadens the knowledge of the microbial degradation pathway of pendimethalin.Haiyan Ni Li Yao Na Li Qin Cao Chen Dai Jun Zhang Qin He Jian He 2016Journal of Environmental Sciences2016,28,3:7

13A global empirical model for mapping zenith wet delays onto precipitable water vapor using GGOS Atmosphere data显示文摘The importance of water vapor in research of global climate change and weather forecast cannot be over emphasized; therefore substantial efforts have been made in exploring the optimal methods to measure water vapor. It is well-established that with a conversion factor, zenith wet delays can be mapped onto precipitable water vapor(PWV). However, the determination of the exact conversion factor depends heavily on the accurate calculation of a key variable, weighted mean temperature of the troposphere(T_m). As a critical parameter in Global Positioning System(GPS) meteorology, T_m has recently been modeled into a global grid known as GWMT. The GWMT_model only requires the location and the day of year to calculate T_m. Despite the advantages that the GWMT_model offers, anomalies still exist in oceanic areas due to low sampling resolution. In this study, we refine the GWMT_model by incorporating the global T_m grid from Global Geodetic Observing System(GGOS) and obtain an improved model, GWMT-G. 15Ti-Zr-Be-Fe quaternary bulk metallic glasses designed by Fe alloying显示文摘A series of Ti-Zr-Be-Fe bulk metallic glasses(BMGs)with good glass-forming ability(GFA)and high specific strength have been developed.With different alloying routes and content of Fe,it is found that these alloys exhibit different GFA and mechanical properties.The effects of Fe addition on the GFA and mechanical properties of Ti-Zr-Be alloy are systemically investigated.The possible mechanisms for the improvement or damage to the GFA by addition of Fe can be interpreted in view of the mixing enthalpy,atomic size differences and electronegativity differences of the alloys,while the mechanical properties strongly depend on the Poisson’s ratio and free volume concentration.The experimental results also show that alloying technology is an effective method to improve the GFA and mechanical properties of Ti-Zr-Be glassy alloy.GONG Pan WANG Xin SHAO Yang CHEN Na YAO KeFu 2013Science China(Physics,Mechanics & Astronomy)2013,56,11:6
16High performance hydroxyapatite ceramics and a triply periodic minimum surface structure fabricated by digital light processing 3D printing显示文摘High performance hydroxyapatite(HA)ceramics with excellent densification and mechanical properties were successfully fabricated by digital light processing(DLP)three-dimensional(3D)printing technology.It was found that the sintering atmosphere of wet C02 can dramatically improve the densification process and thus lead to better mechanical properties.HA ceramics with a relative density of 97.12%and a three-point bending strength of 92.4 MPa can be achieved at a sintering temperature of 1300℃,which makes a solid foundation for application in bone engineering.Furthermore,a relatively high compressive strength of 4.09 MPa can be also achieved for a DLP-printed p-cell triply periodic minimum surface(TPMS)structure with a porosity of 74%,which meets the requirement of cancellous bone substitutes.A further cell proliferation test demonstrated that the sintering atmosphere of wet CO2 led to improve cell vitality after 7 days of cell culture Moreover,with the possible benefit from the bio-inspired structure,the 3D-printed TPMS structure significantly improved the cell vitality,which is crucial for early osteogenesis and osteointegration.Yongxia YAO Wei QIN Bohang XING Na SHA Ting JIAO Zhe ZHAO 2021Journal of Advanced Ceramics2021,10,1:5
17β-Arrestin 2 mediates arginine vasopressin-induced IL-6 induction via the ERK1/2-NF-κB signal pathway in murine hearts显示文摘Evidence to date suggests thatβ-arrestins act beyond their role as adapter proteins.Arginine vasopressin(AVP)may be a factor in inflammation and fibrosis in the pathogenesis of heart failure.In the present study we investigated the effect of AVP on inflammatory cytokine IL-6 production in murine hearts and the impact ofβ-arrestin 2-dependent signaling on AVP-induced IL-6 production.We found that administration of AVP(0.5 U/kg,iv)markedly increased the levels of IL-6 mRNA in rat hearts with the maximum level occurred at 6 h.Inβ-arrestin 2 KO mouse hearts,deletion ofβ-arrestin 2 decreased AVP-induced IL-6 mRNA expression.We then performed in vitro experiments in adult rat cardiac fibroblasts(ARCFs).We found that AVP(10?9–10?6 M)dose-dependently increased the expression of IL-6 mRNA and protein,activation of NF-κB signaling and ERK1/2 phosphorylation,whereas knockdown ofβ-arrestin 2 blocked AVP-induced IL-6 increase,NF-κB activation and ERK1/2 phosphorylation.Pharmacological blockade of ERK1/2 using PD98059 diminished AVP-induced NF-κB activation and IL-6 production.The selective V1A receptor antagonist SR49059 effectively blocked AVP-induced NF-κB phosphorylation and activation as well as IL-6 expression in ARCFs.In AVP-treated mice,pre-injection of SR49059(2 mg/kg,iv)abolished AVP-induced NF-κB activation and IL-6 production in hearts.The above results suggest that AVP induces IL-6 induction in murine hearts via the V1A receptor-mediatedβ-arrestin2/ERK1/2/NF-κB pathway,thus reveal a novel mechanism of myocardial inflammation in heart failure involving the V1A/β-arrestin 2/ERK1/2/NF-κB signaling pathway.Shu-zhen Sun Hong Cao Na Yao Ling-ling Zhao Xiao-fang Zhu Er-an Ni Qi Zhu Wei-zhong Zhu 2020Acta Pharmacologica Sinica2020,41,2:5
18The “Traditional Chinese medicine regulating liver regeneration” treatment plan for reducing mortality of patients with hepatitis B-related liver failure based on real-world clinical data显示文摘On the basis of real-world clinical data,the study aimed to explore the effect and mechanisms of the treatment plan of“traditional Chinese medicine(TCM)regulating liver regeneration.”A total of 457 patients with HBV-related liver failure were retrospectively collected.The patients were divided into three groups:the modern medicine control group(MMC group),patients treated with routine medical treatment;the control group combining traditional Chinese and Western medicine(CTW),patients treated with routine medical treatment plus the common TCM formula;and the treatment group of“TCM regulating liver regeneration”(RLR),patients treated with both routine medical treatment and the special TCM formula of RLR.After 8 weeks of treatment,the mortality of patients in the RLR group(12.31%)was significantly lower than those in the MMC(50%)and CTW(29.11%)groups.Total bilirubin level significantly decreased and albumin increased in the RLR group when compared with the MMC and CTW groups(P<0.05).In addition,there were significant differences in the expression of several cytokines related to liver regeneration in the RLR group compared with the MMC group.RLR treatment can decrease jaundice,improve liver function,and significantly reduce the mortality in patients with HBV-related liver failure.The mechanism may be related to the role of RLR treatment in influencing cytokines related to liver regeneration.Ling Dai Xiang Gao Zhihua Ye Hanmin Li Xin Yao Dingbo Lu Na Wu 2021Frontiers of Medicine2021,15,3:4
19Alleviation of acute pancreatitis-associated lung injury by inhibiting the p38 mitogen-activated protein kinase pathway in pulmonary microvascular endothelial cells显示文摘BACKGROUND Previous reports have suggested that the p38 mitogen-activated protein kinase signaling pathway is involved in the development of severe acute pancreatitis(SAP)-related acute lung injury(ALI).Inhibition of p38 by SB203580 blocked the inflammatory responses in SAP-ALI.However,the precise mechanism associated with p38 is unclear,particularly in pulmonary microvascular endothelial cell(PMVEC)injury.AIM To determine its role in the tumor necrosis factor-alpha(TNF-α)-induced inflammation and apoptosis of PMVECs in vitro.We then conducted in vivo experiments to confirm the effect of SB203580-mediated p38 inhibition on SAP-ALI.METHODS In vitro,PMVEC were transfected with mitogen-activated protein kinase kinase 6(Glu),which constitutively activates p38,and then stimulated with TNF-α.Flow cytometry and western blotting were performed to detect the cell apoptosis and inflammatory cytokine levels,respectively.In vivo,SAP-ALI was induced by 5%sodium taurocholate and three different doses of SB203580(2.5,5.0 or 10.0 mg/kg)were intraperitoneally injected prior to SAP induction.SAP-ALI was assessed by performing pulmonary histopathology assays,measuring myeloperoxidase activity,conducting arterial blood gas analyses and measuring TNF-α,interleukin(IL)-1βand IL-6 levels.Lung microvascular permeability was measured by determining bronchoalveolar lavage fluid protein concentration,Evans blue pulmonary cells was confirmed by performing a terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling analysis and examining the Bcl2,Bax,Bim and cle-caspase3 levels.The proteins levels of P-p38,NFκB,IκB,P-signal transducer and activator of transcription-3,nuclear factor erythroid 2-related factor 2,HO-1 and Myd88 were detected in the lungs to further evaluate the potential mechanism underlying the protective effect of SB203580.RESULTS In vitro,mitogen-activated protein kinase(Glu)transfection resulted in higher apoptotic rates and cytokine(IL-1βand IL-6)levels in TNF-α-treated PMVECs.In vivo,SB2035080 attenuated lung histopathological injury,decreased inflammatory activity(TNF-α,IL-1β,IL-6 and myeloperoxidase)and preserved pulmonary function.Furthermore,SB203580 significantly reversed changes in the bronchoalveolar lavage fluid protein concentration,Evans blue accumulation,terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling-positive cell numbers,apoptosis-related proteins(cle-caspase3,Bim and Bax)and endothelial microstructure.Moreover,SB203580 significantly reduced the pulmonary P-p38,NFκB,P-signal transducer and activator of transcription-3 and Myd88 levels but increased the IκB and HO-1 levels.CONCLUSION p38 inhibition may protect against SAP-ALI by alleviating inflammation and the apoptotic death of PMVECs.Xiao-Xin Zhang Hao-Yang Wang Xue-Fei Yang Zi-Qi Lin Na Shi Chan-Juan Chen Lin-Bo Yao Xin-Min Yang Jia Guo Qing Xia Ping Xue 2021World Journal of Gastroenterology2021,27,18:4
20铜离子改性的凹凸棒石粘土在预防和治疗由沙门氏菌引起的腹泻中的应用(英文)显示文摘目的:研究开发一种抗生素替代品,应用于动物细菌性腹泻病的治疗。创新点:将凹凸棒石的吸附作用、肠道粘膜保护作用和铜离子的杀菌作用巧妙的结合在一起,能够替代抗生素,在动物细菌性腹泻的预防和治疗中发挥作用。方法:用硝酸铜和凹凸棒石粘土(Pal)制备铜离子改性的凹凸棒石粘土(CM-Pal)。通过比较添加CM-Pal前后沙门氏菌数量的变化检测抗菌活性。体内试验,将70只小鼠分为7组,其中5组感染沙门氏菌后分别给予Pal、CM-Pal、蒙脱石散、庆大霉素和生理盐水;1组在感染沙门氏菌之前给予CM-Pal,作为预防组;1组不感染沙门氏菌,作为未感染组。通过粪便形态、粪便计分、生长性能和肠绒毛高度等指标判断对沙门氏菌引起的小鼠腹泻的预防和治疗效果。结论:体外抑菌试验显示,CM-Pal与沙门氏菌作用2 h后抗菌率达100%。体内试验也显示,CM-Pal组小鼠24 h后腹泻症状消失,所有小鼠恢复正常。但是其他组小鼠(Pal组、蒙脱石散组和庆大霉素组)分别在48、48和96 h后才恢复正常。CM-Pal治疗组小鼠肠绒毛的高度在感染3 d后恢复正常,但是其他组小鼠肠绒毛高度恢复正常至少需要5 d。CM-Pal预防组小鼠增重高于其他组小鼠。本研究表明,CM-Pal在预防和治疗由沙门氏菌引起的腹泻方面能够替代抗生素具有较好的效果。Da-wei YAO Ze-zhong YU Na LI Yu-nong HOU Jia-rong XU De-ji YANG 2017Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2017,18,6:4
