2Identification of mecciRNAs and their roles in the mitochondrial entry of proteins显示文摘Mammalian mitochondria have small genomes encoding very limited numbers of proteins.Over one thousand proteins and noncoding RNAs encoded by the nuclear genome must be imported from the cytosol into the mitochondria.Here,we report the identification of hundreds of circular RNAs(mecciRNAs)encoded by the mitochondrial genome.We provide both in vitro and in vivo evidence to show that mecciRNAs facilitate the mitochondrial entry of nuclear-encoded proteins by serving as molecular chaperones in the folding of imported proteins.Known components involved in mitochondrial protein and RNA importation,such as TOM40 and PNPASE,interact with mecciRNAs and regulate protein entry.The expression of mecciRNAs is regulated,and these transcripts are critical for the adaption of mitochondria to physiological conditions and diseases such as stresses and cancers by modulating mitochondrial protein importation.mecciRNAs and their associated physiological roles add categories and functions to the known eukaryotic circular RNAs and shed novel light on the communication between mitochondria and the nucleus.Xu Liu Xiaolin Wang Jingxin Li Shanshan Hu Yuqi Deng Hao Yin Xichen Bao Qiangfeng Cliff Zhang Geng Wang Baolong Wang Qinghua Shi Ge Shan 2020Science China(Life Sciences)2020,63,10:15
3Profiles of muscular amino acids,fatty acids,and metabolites in Shaziling pigs of different ages and relation to meat quality显示文摘Pork meat is closely related to physicochemical alterations during growth and development,resulting in differences in nutritional value and meat flavor.This study aimed to evaluate the composition of amino acids,fatty acids,and metabolic profiles in the longissimus thoracis muscle(LM)of Shaziling pigs aged 30,90,150,210,and 300 days.The results showed that the predominant fatty acids identified in the LM of Shaziling pigs were C16:0,C16:1,C18:0,C18:1n9c,and C18:2n6c.An opposite correlation was observed for C18:2n6c and n6/n3 polyunsaturated fatty acids(P<0.05).Alanine,aspartate,glutamate,Dglutamine,and D-glutamate metabolism were the main metabolic pathways for the Shaziling pig meat flavor(P<0.05).Moreover,the correlation coefficients revealed that the contents of anserine,C16:0,C16:1,and C18:1n9c were positively correlated with intramuscular fat and/or p H24hand were negatively correlated with the values of L^(*)(lightness)and b^(*)(yellowness)(P<0.05).In conclusion,age greatly affected the meat quality of Shaziling pigs,and the contents of muscular anserine,C16:0,C16:1,and C18:1n9c might be promising indicators for better meat quality.Yehui Duan Changbing Zheng Jie Zheng Lu Ma Xinran Ma Yinzhao Zhong Xichen Zhao Fengna Li Qiuping Guo Yulong Yin 2023Science China(Life Sciences)2023,66,6:2

5Starch supplementation improves the reproductive performance of sows in different glucose tolerance status显示文摘This study was to evaluate the effects of glucose tolerance status,maternal starch supplementation and soybean substitution in diets on the performance of dams and their offspring.Eighty-eight pregnant sows(Landrace x Large White)were selected from an initial total of 120 sows,based on blood glucose test values,and assigned to 4 experimental treatments in a 2×2 factorial design.The factors were glucose tolerance status(glucose intolerant[GIT]vs.normal glucose tolerant[NGT])or dietary treatments(corn starch diet[CS]vs.soybean substitution diet[SSI]).A higher area under the curve(AUC)for post-meal glucose was observed(P<0.05)in the GIT group than in the NGT group on d 109 of gestation.The CS group had a lower value of homeostasis model assessment-insulin resistance than the SS group(P<0.05)on d 109 of gestation.Corn starch supplementation for sows decreased the stillbirth rate(P<0.05),regardless of the sows’glucose tolerance status.The villus height of the jejunum and the villus height to crypt depth ratio of the ileum were greater in normal birth weight piglets from the CS group than from the SS group(P<0.01),and so was the activity of sucrase in the jejunum and ileum(P<0.01).Compared with the SS group,the CS group showed a reduction in pre-weaning mortality rate,an increase in the number of high-birth-weight piglets,and a decrease in the number of low-birth-weight piglets(P<0.05)under GIT status.In conclusion,sows fed CS decreased stillbirth rate and improved insulin resistance,as well as improving the intestinal morphology and digestive enzyme activities of their progeny,regardless of glucose tolerance status.Additionally,the CS group improved birth weight distribution and decreased pre-weaning mortality rate of piglets under GIT status.Yunyu Yang Ming Deng Jianzhao Chen Xichen Zhao Kaili Xiao Wenliang He Xinggang Qiu Yanzhen Xu Yulong Yin Chengquan Tan 2021Animal Nutrition2021,,4:2

7A novel detection method of Cryptosporidium parvum infection in cattle based on Cryptosporidium parvum virus 1显示文摘Cryptosporidium parvum is an important zoonotic parasite that causes significant economic loss in the animal husbandry industry,especially the cattle industry.As there is no specific vaccine or drug against Cryptosporidium,a rapid and accurate method for the detection of C.parvum is of great significance.In this study,colloidal gold strips were developed based on Cryptosporidium parvum virus 1 (CSpV1) for the detection of C.parvum infection in cattle fecal samples.The colloidal gold solution was prepared by reducing trisodium citrate and the CSpV1 #5 monoclonal antibody was labeled with colloidal gold.A polyclonal antibody against the CSpV1 capsid protein and an anti-mouse IgG antibody were coated on the colloidal gold strips for use in the test and control lines,respectively.Our results showed that the detection sensitivity in fecal samples was up to a 1:64 dilution.There was no cross-reaction with Cryptosporidium andersoni or Giardia in the fecal samples.The different preservation conditions (room temperature,4℃,and 37℃) and preservation time (7,30,60,and 90 days) were analyzed.The data showed that the strips could be preserved for 90 days at 4℃ and for 60 days at room temperature or 37℃.The colloidal gold strips were used to detect the samples of 120 clinical fecal in Changchun,China.The results indicated that the rate of a positive test was 5%(6/120).This study provides a rapid and accurate method for detecting C.parvum infection in cattle and humans.Lixin Tai Jianhua Li Jigang Yin Nan Zhang Ju Yang He Li Zhengtao Yang Pengtao Gong Xichen Zhang 2019Acta Biochimica et Biophysica Sinica2019,51,1:1
8Zeolite-encaged Ultrasmall Pt-Zn Species with Trace Amount of Pt for Efficient Propane Dehydrogenation显示文摘Propane dehydrogenation(PDH)has become a globe-welcoming technology to meet the massive demand for propylene,but the most commonly used Pt-based catalysts suffer from quick sintering,poor selectivity for propylene,and unsatisfied Pt utilization.Herein,a series of Silicalite-1(S-1)zeolite-encaged ultrasmall Pt-Zn clusters with a trace amount of Pt[40—180 ppm(parts per million)]were developed by using a one-pot ligand-protected direct H_(2) reduction method.Interestingly,the extremely low amount of Pt can significantly promote the activity of zeolite-encaged Zn catalysts in PDH reactions.Thanks to the high Pt dispersion,the synergy between Pt and Zn species,and the confinement effect of zeolites,the optimized PtZn@S-1 catalyst with 180 ppm Pt and 1.88%(mass fraction)Zn,exhibited an extraordinarily high propane conversion(33.9%)and propylene selectivity(99.5%)at 550℃with a weight hourly space velocity(WHSV)of 8 h^(-1),affording an extremely high propylene formation rate of 340.7 mol_(C_(3)H_(6))·g_(Pt^(-1))·h^(-1).This work provides a reference for the preparation of zeolite-encaged metal catalysts with high activity and noble metal utilization in PDH reactions.QU Ziqiang ZHANG Tianjun YIN Xichen ZHANG Junyi XIONG Xiaoyun SUN Qiming 2023Chemical Research in Chinese Universities2023,39,6:0
9Piglet growth performance improved by dietary supplementation of porous or nano particles of zinc oxide may be related to the gut microbiota显示文摘Previous studies on porous or nano particles zinc oxide(ZnO)in the piglets have mainly focused on growth performance and intestinal inflammation,but have scarcely explored the efficacy on gut microbiota.In addition,the efficacy of nano particles ZnO,which is related to its product quality,remains undefined.This study aimed to determine the efficacy of dietary 500 mg/kg porous or nano particles ZnO on the growth performance and gut microbiota of the weaned piglets.A total of 128 weaned piglets were randomly assigned to the dietary groups:NC(basal diet),PC(basal diet+3,000 mg/kg conventional ZnO),500HiZ(basal diet+500 mg/kg porous particles ZnO),and 500ZNP(basal diet+500 mg/kg nano particles ZnO).Compared with the NC diet group,both 500HiZ and 500ZNP increased(P<0.05)average daily feed intake(1 to 28 d)and average daily gain(1 to 28 d),and the 500ZNP tended to decrease feed to gain ratio(F:G ratio,1 to 28 d)(P=0.09).Both 500HiZ and 500ZNP decreased crypt depth of the ileum and increased claudin-2 in the duodenum and zonula occludens-1 in the ileum(P<0.05).Moreover,both 500HiZ and 500ZNP decreased IL-1βand tumor necrosis factor-a(TNF-a)in the jejunum and decreased TNF-a and IL-6 in the ileum(P<0.05).Both 500HiZ and 500ZNP increased microbialβ-diversity index in the ileum and microbial a-diversity indices in the colon of piglets(P<0.05).The probiotic genera Coprococcus(500ZNP)and Blautia(500HiZ)were positively correlated with the F:G ratio(1to 28 d)in colon of piglets(P<0.05).In addition,500HiZ promoted mitochondrial fusion protein 1(MFN1)and zinc transporter-1(ZnT-1)in the jejunum(P<0.05),whilst 500ZNP decreased MFN1 in the jejunum and ZnT-1 in the ileum(P<0.05).In summary,both 500HiZ and 500ZNP improved the growth performance of piglets,which is likely via the genera Blautia and Coprococcus,respectively.Both 500HiZ and 500ZNP improved barrier function and inflammation of the intestine,and 500HiZ achieved better efficacy than 500ZNP on intestine mitochondrial functions.Lina Long Xichen Zhao Jie Chen Zixi Wang Yanfang Tang Jian Huang Yulong Yin 2023Animal Nutrition2023,,4:0
