2Analysis of structural components of decellularized scaffolds in renal fibrosis显示文摘Chronic kidney disease has been recognized as a major public health problem worldwide and renal fibrosis is a common pathological process occurring in chronic renal failure.It is very promising to find the strategies to slow or even prevent the progression of fibrosis.This study focused on whether renal fibrosis decellularized scaffolds has the potential to be a model of cellular mechanisms of tissue fibrosis or donors for tissue engineering.In order to evaluate the feasibility of decellularized scaffolds derived from pathological kidneys,histology,proteomics and ELISA will be used to analysis the changes in the structure and main components of fibrotic tissue.The fibrosis model in this paper was induced by adenine-fed and the results showed that the structure of fibrotic scaffold was changed and some protein were up-regulated or down-regulated,but the cytokines associated with renal regeneration after injury were remained.In cell experiments,endothelial progenitor cells proliferated well,which proved that the fibrotic scaffolds have non-cytotoxic.All these conclusions indicate that the renal fibrosis decellularized scaffolds model has the ability to study fibrosis mechanism and the potential to be engineering donors as well as normal scaffolds.Rui Zhang Junqun Jiang Yaling Yu Fangfang Wang Niuniu Gao Yingjie Zhou Xinlong Wan Zhibin Wang Peng Wei Jin Mei 2021Bioactive Materials2021,6,7:5
3High-sensitivity self-powered temperature/pressure sensor based on flexible Bi-Te thermoelectric film and porous microconed elastomer显示文摘Electronic skins are artificial skin-type multifunctional sensors,which hold great potentials in intelligent robotics,limb prostheses and human health monitoring.However,it is a great challenge to independently and accurately read various physical signals without power supplies.Here,a self-powered flexible temperature-pressure bimodal sensor based on high-performance thermoelectric films and porous microconed conductive elastic materials is presented.Through introducing flexible heat-sink design and harvesting body heat energy,the thin-film thermoelectric device could not only precisely sense temperature signal but also drive the pressure sensor for detecting external tactile stimulus.The integration of Bi-Te based thermoelectric film with high stability in wide temperature range enables the sensor to sense the ambient temperature with high resolution(<0.1 K)as well as excellent sensitivity(3.77 mV K^(-1)).Meanwhile,the porous microconed elastomer responds to pressure variation with low-pressure detection(16 Pa)and a high sensitivity of 37 kPa^(-1).Furthermore,the bimodal sensor could accurately and simultaneously monitor human wrist pulse and body temperature in real time,which demonstrates promising applications in self-powered electronic skins for human health monitoring systems.Yaling Wang Wei Zhu Yuan Deng Pengcheng Zhu Yuedong Yu Shaoxiong Hu Ruifeng Zhang 2022Journal of Materials Science & Technology2022,,8:4

6Correlation of genomic and expression alterations of AS3 with esophageal squamous cell carcinoma显示文摘Androgen-induced proliferation shutoff gene AS3, also known as APRIN, is a growth inhibitory gene that is initially implicated in prostate cancer. This gene is required for androgen-dependent growth arrest and is a primary target for 1,25(OH)2D3 and androgens. Alle- lic loss at AS3 locus has been linked to a variety of cancers. However, the correlation of genomic and expression alterations of AS3 with esophageal squamous cell carcinoma (ESCC) is not well established. In this study, the genomic and expression alterations of AS3 in ESCC and their clinical significance are evaluated. Loss of heterozygosity (LOH) analysis using an AS3 intragenic microsatellite marker D13S171 revealed 72% allelic loss at AS3 locus in ESCC, which is significantly correlated with higher pathological grade (P=0.042). RT-PCR examination showed that AS3 mRNA obviously decreased in 44% tumors and its down-regulation was correlated with the sex of patients (P=0.03). Furthermore, the correlation between genomic and expression alterations of AS3 gene was analyzed in 18 ESCC specimens, which indicated that the consistency between allelic loss and decreased mRNA expression of AS3 was relatively poor. 16Neuroprotective effect of high-dose hyperbaric oxygenation on rats with acute cerebral infarction in super-early stage:Curative comparison between 9-hour and 18-hour therapeutic protocols显示文摘BACKGROUND:Previously,only single short-time low-dose hyperbaric oxygenation(HBO) protocol was administrated to treat acute ischemic stroke in early stage and the conflicting results were obtained. There are few studies to report the outcome of administering long-time(can cover all the natural pathologic progression period) high-dose HBO to treat the disease. OBJECTIVE:To evaluate the therapeutic effect between two kinds of high-dose hyperbaric oxygenation on super-early stage of acute permanent middle cerebral artery occlusion(MCAO) in rats. DESIGN:A randomized controlled experimental study. SETTING:Beijing Tiantan Hospital,Capital Medical University;Beijing Research Institute of Neurosurgery. MATERIALS:Seventy-four male SD rats,aged 2.5 months old,weighing(280±20)g,were provided by the Animal Institute,Chinese Academy of Medical Sciences. Hyperbaric oxygenation device was hyperbaric air cabin in which there was a self-made pure oxygen animal experimental cabin(made in China). METHODS:This experiment was carried out in the municipal laboratory of Beijing Tiantan Hospital affiliated to Capital Medical University and Beijing Research Institute of Neurosurgery. ① Experimental intervention:All the rats were developed into models of permanent MCAO by suture embolism. Then,they were randomly divided into two HBO groups(9 hours and 18 hours) and control group,with 24 rats in each as well as 3-hour ultrastructure control group,with 2 rats. After being modeled for 3 hours,rats in the two HBO groups stayed in the hyperbaric cabin for 9 hours and 18 hours,separately. Rats in the 9-hour HBO group inhaled pure oxygen at hours 1,3,5,7 and 9,and hyperbaric air at hours 2,4,6 and 8. Rats in the 18-hour HBO group inhaled pure oxygen at hours 1,3,5,7,9,11,13,15 and 17,and hyperbaric air at hours 2,4,6,8,10 12,14,16 and 18. After being created into models,rats in the control group and 3-hour ultrastructure control group breathed room air. ② Experimental evaluation:Neurologic functions of rat models in the 9-hour and 18-hour HBO groups as well as control group were scored by Bederson and Garica two neurological grading systems at hours 14 and 28 and on day 5;Infarct volume of rat models in the two HBO groups and control group was measured at hour 24 and on day 5 with NIH image processing software Image J;The pathological changes of brain tissue in the brain infarct region and its opposite region of rat models in the two HBO groups and 3-hour ultrastructure control group were observed with a Philips EM 208S transmission electron microscope. MAIN OUTCOME MEASURES:① Neurobehavioral outcome. ② Rat brain infarct volume. ③ Ultrastructure of brain tissue in the ischemic penumbra of infarct models at the different time points RESULTS:① Neurobehavioral outcome:After treatment,Garica score in the 9-hour and 18-hour HBO groups was significantly higher than that in the control group(P < 0.01). Bederson score on day 5 after modeling in the 9-hour and 18-hour HBO groups was significantly lower than that in the control group(P < 0.01). ② Cerebral infarct volume:Cerebral infarct volume in the 9-hour and 18-hour HBO groups was significantly smaller than that in the control group at hour 24 and on day 5 after modeling(P < 0.01). In the 18-hour HBO group,infarct volume on day 5 after modeling was significantly larger than that at hour 24 after modeling(P < 0.05). ③In the 3-hour ultrastructure control group,astrocyte edema and neuron damage around the capillary in the infarct cerebral tissue significantly relieved in the rats which were subjected to HBO. 18Muscle-inspired soft robots based on bilateral dielectric elastomer actuators显示文摘Muscle groups perform their functions in the human body via bilateral muscle actuation,which brings bionic inspiration to artificial robot design.Building soft robotic systems with artificial muscles and multiple control dimensions could be an effective means to develop highly controllable soft robots.Here,we report a bilateral actuator with a bilateral deformation function similar to that of a muscle group that can be used for soft robots.To construct this bilateral actuator,a low-cost VHB 4910 dielectric elastomer was selected as the artificial muscle,and polymer films manufactured with specific shapes served as the actuator frame.By end-to-end connecting these bilateral actuators,a gear-shaped 3D soft robot with diverse motion capabilities could be developed,benefiting from adjustable actuation combinations.Lying on the ground with all feet on the ground,a crawling soft robot with dexterous movement along multiple directions was realized.Moreover,the directional steering was instantaneous and efficient.With two feet standing on the ground,it also acted as a rolling soft robot that can achieve bidirectional rolling motion and climbing motion on a 2°slope.Finally,inspired by the orbicularis oris muscle in the mouth,a mouthlike soft robot that could bite and grab objects 5.3 times of its body weight was demonstrated.The bidirectional function of a single actuator and the various combination modes among multiple actuators together allow the soft robots to exhibit diverse functionalities and flexibility,which provides a very valuable reference for the design of highly controllable soft robots.Yale Yang Dengfeng Li Yanhua Sun Mengge Wu Jingyou Su Ying Li Xinge Yu Lu Li Junsheng Yu 2023Microsystems & Nanoengineering2023,9,5:0
19A direct,sensitive and high-throughput genus and species-specific molecular assay for large-scale malaria screening显示文摘Background:Infectious disease diagnostics often requires sensitive molecular assays that identify at both genus and species levels.For large scale screening,such as malaria screening for elimination,diagnostic assay can be a challenge,as both the throughput and cost of the assay must be considered.The requirement of nucleic acid extraction hampers the throughput of most molecular assays.Co-amplification of multiple species or multiplex identification either can result in missed diagnosis or are too costly for large-scale screening.A genus-and species-specific diagnostic assay with simplified procedure,high sensitivity and throughput is still needed.This study aimed to develop a sensitive and high-throughput approach for large-scale infectious disease screening.Methods:We developed multi-section Capture and Ligation Probe PCR(mCLIP-PCR)for the direct detection of RNA without extraction and reverse transcription.Multiple tailed sandwich hybridization probes were used to bind at genus-and species-specific sections of the target RNA to cooperatively capture the target onto a 96-well plate.After enzymatic ligation of the bound probes,a single-stranded DNA formed at each section with distinct tail sequence at the ends.They were separately PCR-amplified with primers corresponding to tail sequences for genus or species identification.We applied the method to the active screening ofPlasmodium infections of 4,580 asymptomatic dried blood spot samples collected in malaria endemic areas and compared the results with standard qPCR using linear regression.Results:With multi-section cooperative capture but separate amplification strategy,we accurately identified genusPlasmodium and speciesP.falciparum andP.vivax without RNA extraction,with favorable sensitivities among the published reports.In the active screening,our method identified all 53 positive infections including two mixed infections,and twoP.vivax infections that were missed by standard qPCR.Conclusions:mCLIP-PCR provides a sensitive and high-throughput approach to large-scale infectious disease screening with low cost and labor,making it a valuable tool for malaria elimination in endemic region.Yaling Zhao Ye Zhao Yu Sun Lihua Fan Duoquan Wang Heng Wang Xiaodong Sun Zhi Zheng 2022Infectious Diseases of Poverty2022,11,2:0
20Long-chain branched/cyclic alkanes in recent sediment of Lake Fuxian and their environmental implications显示文摘Lake Fuxian is a large, deep, and oligotrophic freshwater lake. In recent sediment of this lake, a series of branched and cyclic alkanes, including branched alkanes with one or two quaternary carbon atoms(C2ncompounds),alkylcycloheptane, and alkylcyclohexane, were detected in high abundance. For each homolog of long-chain branched/cyclic alkanes, molecules with only an odd or even carbon number were present in the sediment. The abundance variation of each compound homolog corresponded to those of other compound homolog in the section, indicating the same origin of these compounds. The concentrations of long-chain branched/cyclic alkanes in the sediment profile exhibited variations similar to those of phytoplankton biomarkers,such as 3-methyl-alkanes, short-chain odd carbon n-alkanes,hopene, polyunsaturated fatty acids, phytol, sterols, and long-chain alkyl diols, but slightly different from those of long-chain odd carbon n-alkanes, which are ubiquitous biomarkers for higher plants.
