1Multiple genetic alterations and behavior of cellular biology in gastric cancer and other gastric mucosal lesions:H.pylori infection,histological types and staging显示文摘AIM To investigate the expression of multiplegenes and the behavior of cellular biology ingastric cancer(GC)and other gastric mucosallesions and their relations to Helicobacter pylori(H.pylori)infection,tumor staging andhistological subtypes.METHODS Three hundred and twenty-sevenspecimens of gastric mucosa obtained viaendoscopy or surgical resection,and ABCimmunohistochemical staining were used todetect the expression of p53,p16,Bcl-2 andCOX-2 proteins.H.pylori was determined byrapid urea test combined with pathologicalstaining or14C urea breath test.Cellular image analysis was performed in 66 patients withintestinal metaplasia(IM)and/or dysplasia(Dys).In 30 of them,both cancer and theparacancerous tissues were obtained at the timeof surgery.Histological pattern,tumor staging,lymph node metastasis,grading ofdifferentiation and other clinical data werestudied in the medical records.RESULTS p16 expression of IM or Dys wassignificantly lower in positive H.pylori chronicatrophic gastritis(CAG)than those withnegative H.pylori(CAG:54.8% vs 88.0%,IM:34.4% vs 69.6%,Dys:23.8% vs 53.6%,allP<0.05),Bcl-2 or COX-2 expression of IM orDys in positive H.pylori cases was significantlyhigher than that without H.pylori(Bcl-2:68.8%vs23.9%,90.5% vs 60.7%;COX-2:50.0% vs10.8%,61.8% vs 17.8%;all P<0.05).Themean number of most parameters of cellularimage analysis in positive H.pylori group wassignificantly higher than that in negative H.pylori group(Ellipser:53±14,40±12μm,Area1:748±572,302±202 μm2,Area2:3050±1661,1681±1990 μm2,all P<0.05;Ellipseb:79±23,58±15 μm,Ratio1:22%±5%,13%±4%,Ratio2:79%±17%,53%±20%,all P<0.01).There was significant correlation between Bcl-2and histologic pattern of gastric carcinoma,andbetween COX-2 and tumor staging or lymph nodemetastasis(Bcl-2:75.0% vs 16.7%;COX-2:76.0% vs 20.0%,79.2% vs 16.7%;allP<0.05).CONCLUSION p1l6, Bcl-2, and COX-2 but not p53 gene may play a role in the early genesis/ progression of gastric carcinoma and are associated with H. pylori infection. p53 gene is relatively late event in gastric tumorigenesis and mainly relates to its progression. There is more cellular-biological behavior of malignant tumor in gastric mucosal lesions with H. pylori infection. Aberrant Bcl-2 protein expression appears to be preferentially associated with the intestinal type cancer. 3Forming limits under multi-index constraints in NC bending of aluminum alloy thin-walled tubes with large diameters显示文摘With increasing diameters of aluminum alloy thin-walled tubes (AATTs), the tube forming limits, i.e. the minimum bending factors, and their predictions under multi-index constraints including wrinkling, thinning and flattening have been being a key problem to be urgently solved for improving tube forming potential in numerical control (NC) bending processes of AATTs with large diameters. Thus in this paper, a search algorithm of the forming limits is put forward based on a 3D elastic-plastic finite element (FE) model and a wrinkling energy prediction model for the bending processes under axial compression loading (ACL) or not. This algorithm enables to be considered the effects of process parameter combinations including die, friction parameters on the multi-indices. Based on this algorithm, the forming limits of the different size tubes are obtained, and the roles of the process parameter combinations in enabling the limit bending processes are also revealed. The followings are found: the first, within the appropriate ranges of friction and clearances between the different dies and the tubes enabling the bending processes with smaller bending factors, the ACL enables the tube limit bending processes after a decrease of the mandrel ball thickness and diameters; then, without considering the effects of the tube geometry sizes on the tube constitutive equations, the forming limits will be decided by the limit thinning values for the tubes with diameters smaller than 80 mm, while the wrinkling for the tubes with diameters no less than 80 mm. 6Morinda officinalis oligosaccharides increase serotonin in the brain and ameliorate depression via promoting 5-hydroxytryptophan production in the gut microbiota显示文摘Morinda officinalis oligosaccharides(MOO) are an oral drug approved in China for the treatment of depression in China. However, MOO is hardly absorbed so that their anti-depressant mechanism has not been elucidated. Here, we show that oral MOO acted on tryptophan → 5-hydroxytryptophan(5-HTP) → serotonin(5-HT) metabolic pathway in the gut microbiota. MOO could increase tryptophan hydroxylase levels in the gut microbiota which accelerated 5-HTP production from tryptophan;meanwhile, MOO inhibited 5-hydroxytryptophan decarboxylase activity, thus reduced 5-HT generation,and accumulated 5-HTP. The raised 5-HTP from the gut microbiota was absorbed to the blood, and then passed across the blood-brain barrier to improve 5-HT levels in the brain. Additionally, pentasaccharide,as one of the main components in MOO, exerted the significant anti-depressant effect through a mechanism identical to that of MOO. 9Efficacy and safety of secukinumab in Chinese patients with moderate-to-severe plaque psoriasis: a real-life cohort study显示文摘Background:There have been few real-life dose-comparing studies on the efficacy and safety of secukinumab in Chinese patients with plaque psoriasis.We conducted a real-life cohort study to investigate the efficacy and safety of secukinumab 150 and 300 mg in Chinese patients with moderate-to-severe plaque psoriasis.Methods:A total of 106 patients with moderate-to-severe plaque psoriasis were included in this study.Patients received either secukinumab 150 mg or secukinumab 300 mg according to patients’weights and severity of psoriasis.The treatment continued for at least 24 weeks.The efficacy was evaluated by improvement in the psoriasis area and severity index(PASI)scores.The safety was also analyzed.Results:Fifty-nine patients(55.7%)were treated with secukinumab 300 mg and 47 patients(44.3%)were treated with secukinumab 150 mg.After 12-week treatment,PASI75/90/100 responses were achieved in 100%,97.8%,and 95.7%of patients,respectively,in secukinumab 150 mg group,and the efficacy was maintained to week 24.In secukinumab 300 mg group,PASI75/90/100 responses were achieved in 93.2%,81.4%,and 76.3%of patients,respectively,at week 12.In this group,PASI75/90/100 responses reached 91.5%,86.4%,and 79.9%,respectively,at week 24.Biologic-experienced patients had lower responses than biologic-naïve patients.Secukinumab 150 and 300 mg were well tolerated.Five patients discontinued treatment due to poor response,adverse event,or economic reasons.Conclusions:This real-life study demonstrated that high PASI 90 and PASI 100 responses were achieved in Chinese psoriasis patients receiving secukinumab 150 or 300 mg.Biologic-naïve was associated with better clinical efficacy.Yan Zhao Lin Cai Xiao-Yang Liu Heng Zhang Jian-Zhong Zhang 2021Chinese Medical Journal2021,,11:10

11MicroTom Metabolic Network: Rewiring Tomato Metabolic Regulatory Network throughout the Growth Cycle显示文摘Tomato(Solanum lycopersicum)is a major horticultural crop worldwide and has emerged as a preeminent model for metabolic research.Although many research efforts have focused on the analysis of metabolite differences between varieties and species,the dynamics of metabolic changes during the tomato growth cycle and the regulatory networks that underlie these changes are poorly understood.In this study,we integrated high-resolution spatio-temporal metabolome and transcriptome data to systematically explore the metabolic landscape across 20 major tomato tissues and growth stages.In the resulting MicroTom Metabolic Network,the 540 detected metabolites and their co-expressed genes could be divided into 10 distinct clusters based on their biological functions.Using this dataset,we constructed a global map of the major metabolic changes that occur throughout the tomato growth cycle and dissected the underlying regulatory network.In addition to verifying previously well-established regulatory networks for important metabolites,we identified novel transcription factors that regulate the biosynthesis of important secondary metabolites such as steroidal glycoalkaloids and flavonoids.Our findings provide insights into spatiotemporal changes in tomato metabolism and generate a valuable resource for the study of metabolic regulatory processes in model plants.Yan Li Yang Chen Lu Zhou Shengjie You Heng Deng Ya Chen Saleh Alseekh Yong Yuan Rao Fu Zixin Zhang Dan Su Alisdair R.Fernie Mondher Bouzayen Tao Ma Mingchun Liu Yang Zhang 2020Molecular Plant2020,13,8:10

13Classification of microseismic events in high stress zone显示文摘For the purpose of having a better understanding of failure mechanisms of rock fracturing in mines, the equivalent point source models of tensile, shear and explosive seismic events were established, and the relationship between far-field seismic displacements of the waves and the corresponding equivalent forces were analyzed as well. Based on the results of a microseismic monitoring carried out in the mining progress of 9202 working face under the upper remnant coal pillar in Sanhejian Mine, the waveform features of the seismic events associated with different failure modes were further analyzed. The results show that the signals corresponding to different failure mechanisms have different radiation patterns of the seismic displacements, and different characteristics in waveform features, such as dominant frequency, energy released, the ratio of S- to P-wave energy, and so on. In addition, the rock burst happened in the high stress zone is mainly the result of the strong shear fracturing in the mining process. 15Characterization of the aerosol chemical composition during the COVID-19 lockdown period in Suzhou in the Yangtze River Delta,China显示文摘To control the spread of COVID-19,rigorous restrictions have been implemented in China,resulting in a great reduction in pollutant emissions.In this study,we evaluated the air quality in the Yangtze River Delta during the COVID-19 lockdown period using satellite and ground-based data,including particle matter(PM),trace gases,water-soluble ions(WSIs) and black carbon(BC).We found that the impacts of lockdown policy on air quality cannot be accurately assessed using MODIS aerosol optical depth(AOD) data,whereas the tropospheric nitrogen dioxide(NO_(2)) vertical column density can well reflect the influences of these restrictions on human activities.Compared to the pre-COVID period,the PM_(2.5),PM_(10),NO_(2),carbon monoxide(CO),BC and WSIs during the lockdown in Suzhou were observed to decrease by 37.2%,38.3%,64.5%,26.1%,53.3% and 58.6%,respectively,while the sulfur dioxide(SO_(2)) and ozone(O_(3)) increased by 1.5% and 104.7%.The WSIs ranked in the order of NO_(3)~->NH_(4)^(+)>SO_(4)^(2-)>Cl~->Ca^(2+)>K^(+)>Mg^(2+)>Na + during the lockdown period.By comparisons with the ion concentrations during the pre-COVID period,we found that the ions NO 3-,NH 4 +,SO_(4)^(2-),Cl~-,Ca^(2+),K^(+) and Na^(+) decreased by 66.3%,48.8%,52.9%,56.9%,57.9% and 76.3%,respectively,during the lockdown,in contrast to Mg^(2+),which increased by 30.2%.The lockdown policy was found to have great impacts on the diurnal variations of Cl~-,SO_(4)^(2-),Na^(+) and Ca^(2+).Honglei Wang Qing Miao Lijuan Shen Qian Yang Yezheng Wu Heng Wei Yan Yin Tianliang Zhao Bin Zhu Wen Lu 2021Journal of Environmental Sciences2021,33,4:8
16Gasless laparoscopy for benign gynecological diseases using an abdominal wall-lifting system显示文摘Objectives: The use of gasless laparoscopy with an abdominal wall-lifting device for benign gynecological diseases was compared to conventional laparoscopy with CO2 pneumoperitoneum. Methods: From February 2007 to July 2007,76 women with uterine and/or adnexal benign diseases and candidates for laparoscopic surgery were recruited in this study. Thirty-two women underwent gasless laparoscopic surgery and 44 women underwent pneumoperitoneum laparoscopic surgery. Results: Diverse pathologies,including adnexal cyst,uterine myoma and ectopic pregnancy,were treated successfully with gasless laparoscopic surgery. Compared with the patients in the pneumoperitoneum group,the similar hospital stay (P=0.353) and intraoperative blood loss (P=0.157) were observed. However,the mean operative time in the gasless group was significantly longer than that in the pneumoperitoneum group (P=0.003). No severe intraoperative or postoperative complications were found in either group,except for one case of laparotomic conversion in the pneumoperitoneum group due to dense pelvic adhesions. The total hospital charges were significantly less in the gasless group than in the pneumoperitoneum group (P=0.001). In 38 cases of ovarian cyst resection,the mean operative time in the gasless group remained longer than that in the pneumoperitoneum group (P=0.017). The total hospital charges were also significantly less in the gasless group than in the pneumoperitoneum group (P<0.001). Conclusion: Our preliminary results demonstrated that the laparoscopic procedure using the gasless technique was a safe,effective method to treat benign gynecological diseases. Moreover,it was easy to master. 18Chemical cleaning of fouled PVC membrane during ultrafiltration of algal-rich water显示文摘Cleaning of hollow-fibre polyvinyl chloride (PVC) membrane with different chemical reagents after ultrafiltration of algal-rich water was investigated. Among the tested cleaning reagents (NaOH, HCl, EDTA, and NaClO), 100 mg/L NaClO exhibited the best performance (88.4% ± 1.1%) in removing the irreversible fouling resistance. This might be attributed to the fact that NaClO could eliminate almost all the major foulants such as carbohydrate-like and protein-like materials on the membrane surface, as confirmed by Fourier transform infrared spectroscopy analysis. However, negligible irreversible resistance (1.5% ± 1.0%) was obtained when the membrane was cleaning by 500 mg/L NaOH for 1.0 hr, although the NaOH solution could also desorb a portion of the major foulants from the fouled PVC membrane. Scanning electronic microscopy and atomic force microscopy analyses demonstrated that 500 mg/L NaOH could change the structure of the residual foulants on the membrane, making them more tightly attached to the membrane surface. This phenomenon might be responsible for the negligible membrane permeability restoration after NaOH cleaning. 19Improve access to the EU market by identifying French consumer preference for fresh fruit from China显示文摘Most previous studies have investigated country-of-origin effect for agricultural products from the economically developed country, but few have focused on the impact of country-of-origin on agricultural products from economically developing countries. Using a national online survey in France, we determine French consumers' perceptions and preferences for fresh fruit from China. Results of ordered logit models show that fresh fruit from China was perceived as low quality and unsafe. Respondents who care about country-of-origin were less likely to purchase fresh fruit from China, while those who care about brand were more likely to buy Chinese fresh fruit. Risk perception and quality perception both had a significant impact on the purchase of Chinese fresh fruit. Results of latent class models obtained with key consumer demographics show that heterogeneous preferences exist among different consumer groups.
