| 1 | Effects of Saikosaponin-D on syndecan-2,matrix metalloproteinases and tissue inhibitor of metalloproteinases-2 in rats with hepatocellular carcinoma显示文摘OBJECTIVE:To investigate effects of Saikosaponin D(SSd) on syndecan-2,matrix metalloproteinases(MMPs) and tissue inhibitor of metalloproteinases-2(TIMP-2) in livers of rat with hepatocellular carcinoma(HCC).METHODS:Male SD rats were divided into control(n=10),model(n=20) and SSd(n=20) groups,and model and SSd groups given intragastric 0.2%(w/v) N-diethylnitrosamine to induce HCC.SSd group received 0.03%(w/v) SSd in saline.Liver samples were analysed immunohistochemically for syndecan-2,MMP-2,MMP-13 and TIMP-2 at 16 weeks.RESULTS:The model group had more malignant nodules than the SSd group;all model-group HCC cells were grade III;SSd-group HCC cells were grades I-II.Controls showed normal hepatic cell phenotypes and no syndecan-2 + staining.Syndecan-2 + staining was greater in the model group(35.2%,P≤0.001) than in controls or the SSd group(16.5%,P ≤ 0.001).The model group had more intense MMP-2 + staining than controls(0.37 vs 0.27,P≤0.01) or the SSd group(0.31 vs 0.37,P≤0.05);and higher MMP-13 + staining(72.55%) than in controls(12.55%,P≤0.001) and SSd group(20.18%,P≤0.01).The model group also had more TIMP-2 + staining(57.2%) than controls(20.9%,P≤0.001) and SSd group(22.7%,P≤0.001).Controls and SSd group showed no difference in TIMP-2 + rates.CONCLUSION:SSd inhibited HCC development,and downregulated expression of syndecan-2,MMP-2,MMP-13 and TIMP-2 in rat HCC liver tissue. | Xiaoli Jia Shuangsuo Dang Yanan Cheng Xin Zhang Mei Li Yaping Li Siyuan Li | 2012 | Journal of Traditional Chinese Medicine2012,32,3: | 12 |
| 2 | Anlotinib suppresses lymphangiogenesis and lymphatic metastasis in lung adenocarcinoma through a process potentially involving VEGFR-3 signaling显示文摘Objective:Lymphatic metastasis is one of the leading causes of malignancy dispersion in various types of cancer.However,few anti-lymphangiogenic drugs have been approved for clinical use to date.Therefore,new therapies to block lymphangiogenesis are urgently required.Methods:Immunohistochemistry,immunofluorescence,Western blot,migration assays,and lymphangiogenesis and lymphatic metastasis assays were used.Results:Anlotinib,a receptor tyrosine kinase inhibitor,suppressed the rate of new metastatic lesions(31.82%in the placebo arm and 18.18%in the anlotinib arm)in patients with advanced lung adenocarcinoma who were enrolled in our ALTER-0303 study.D2-40+-lymphatic vessel density was strongly correlated with disease stage,metastasis,and poor prognosis in 144 Chinese patients with lung adenocarcinoma.In mice bearing A549EGFP tumors,tumor lymphatic vessel density,tumor cell migration to lymph nodes,and the number of distant metastatic lesions were lower in the anlotinib group than in the controls.Anlotinib inhibited the growth and migration of human lymphatic endothelial cells(hLECs)and lymphangiogenesisin vitro andin vivo.Treatment of hLECs with anlotinib downregulated phosphorylated vascular endothelial growth factor receptor 3(VEGFR-3).Conclusions:Anlotinib inhibits lymphangiogenesis and lymphatic metastasis,probably through inactivating VEGFR-3 phosphorylation.The results indicate that anlotinib may be beneficial for treatment in avoiding lymphangiogenesis and distant lymphatic metastasis in lung adenocarcinoma.(Trial registration:ALTER0303;NCT02388919;March 17,2015.) | Tingting Qin Zhujun Liu Jing Wang Junling Xia Shaochuan Liu Yanan Jia Hailin Liu Kai Li | 2020 | Cancer Biology & Medicine2020,17,3: | 9 |
| 3 | Comparison of the protective effects of resveratrol and pterostilbene against intestinal damage and redox imbalance in weanling piglets显示文摘Background: Evidence indicates that early weaning predisposes piglets to intestinal oxidative stress and increases the risk of intestinal dysfunction;however, there are minimal satisfactory treatment strategies for these conditions.This study investigated the potential of resveratrol and its analog, pterostilbene, as antioxidant protectants for regulating intestinal morphology, barrier function, and redox status among weanling piglets.Methods: A total of 144 piglets were selected at 21 days of age and randomly allocated into one of four treatment groups, each of which included six replicates. Piglets in a sow-reared control group were suckling normally between ages 21 and 28 days, while those in weaned groups were fed a basal diet, supplemented with either 300 mg/kg of resveratrol or with 300 mg/kg of pterostilbene. Parameters associated with intestinal injury and redox status were analyzed at the end of the feeding trial.Results: Early weaning disrupted the intestinal function of young piglets, with evidence of increased diamine oxidase activity and D-lactate content in the plasma, shorter villi, an imbalance between cell proliferation and apoptosis, an impaired antioxidant defense system, and severe oxidative damage in the jejunum relative to suckling piglets. Feeding piglets with a resveratrol-supplemented diet partially increased villus height(P = 0.056) and tended to diminish apoptotic cell numbers(P = 0.084) in the jejunum compared with those fed a basal diet. Similarly, these beneficial effects were observed in the pterostilbene-fed piglets. Pterostilbene improved the feed efficiency of weanling piglets between the ages of 21 and 28 days;it also resulted in diminished plasma diamine oxidase activity and D-lactate content relative to untreated weaned piglets(P < 0.05). Notably, pterostilbene restored jejunal antioxidant capacity, an effect that was nearly absent in the resveratrol-fed piglets. | 5 | Hypoxia-stressed cardiomyocytes promote early cardiac differentiation of cardiac stem cells through HIF-1α/Jagged1/Notch1 signaling显示文摘Hypoxia is beneficial for the differentiation of stem cells transplanted for myocardial injury,but mechanisms underlying this benefit remain unsolved. Here, we report the impact of hypoxia-induced Jagged1 expression in cardiomyocytes(CMs) for driving the differentiation of cardiac stem cells(CSCs).Forced hypoxia-inducible factor 1α(HIF-1α) expression and physical hypoxia(5% O_2) treatment could induce Jagged1 expression in neonatal rat CMs. Pharmacological inhibition of HIF-1α by YC-1 attenuated hypoxia-promoted Jagged1 expression in CMs. An ERK inhibitor(PD98059), but not inhibitors of JNK(SP600125), Notch(DAPT), NF-κB(PTDC), JAK(AG490), or STAT3(Stattic) suppressed hypoxiainduced Jagged1 protein expression in CMs. c-Kit^+ CSCs isolated from neonatal rat hearts using a magnetic-activated cell sorting method expressed GATA4, SM22α or vWF, but not Nkx2.5 and cTnI.Moreover, 87.3% of freshly isolated CSCs displayed Notch1 receptor expression. Direct co-culture of CMs with BrdU-labeled CSCs enhanced CSCs differentiation, as evidenced by an increased number of BrdU^+/Nkx2.5^+ cells, while intermittent hypoxia for 21 days promoted co-culture-triggered differentiation of CSCs into CM-like cells. | 6 | Micro RNA profiles and potential regulatory pattern during the early stage of spermatogenesis in mice显示文摘Spermatogenesis is a complicated and poorly understood process that relies on the precise regulation of the self-renewal and differentiation of spermatogonia. In many organisms, micro RNAs(mi RNAs) are involved in multiple developmental processes as critical regulators of transcriptional and post-transcriptional gene silencing. This study investigated the expression pattern of mi RNAs in type B spermatogonia cells(BSc) and primary spermatocytes(PSc) of mice, using a high-throughput small RNA sequencing system. The results revealed that the expression levels of Let-7 family mi RNAs were remarkably high in both cell types. Furthermore, the expression levels of mi R-21, mi R-140-3p, mi R-103, mi R-30 a, mi R-101 b and mi R-99 b were decreased during the transformation from BSc to PSc. These mi RNAs target vital genes that participate in apoptosis, cell proliferation and differentiation, junction assembly and cell cycle regulation. | 7 | A study on JA-and BTH-induced resistance of Rosa rugosa‘Plena’to powdery mildew(Sphaerotheca pannosa)显示文摘Different concentrations of jasmonic acid(JA)and benzothiadiazole(BTH) were sprayed on 2-year-old Rosa rugosa‘Plena’ seedlings. The induced resistance of JA and BTH to Sphaerotheca pannosa(Wallr.) and the changes of their related physiological indices were investigated. Results showed that JA and BTH treatments had inhibitory impacts on S. pannosa infection. The optimal concentration of JA and BTH was 0.5 mmol/L for the disease-resistance induction of the leaves, its inductive effect was up to 66.36% for BTH and 54.49% for JA. Our results confirmed that exogenous JA and BTH significantly improved R. rugose ‘Plena’ resistance to S. pannosa. When treated with JA and BTH, activities of the three defense enzymes(POD, PPO, and PAL) increased significantly.Contents of total phenolics, flavonoids, and lignin also increased significantly. | 8 | Efficient inversions and duplications of mammalian regulatory DNA elements and gene clusters by CRISPR/Cas9显示文摘The human genome contains millions of DNA regulatory elements and a large number of gene clusters,most of which have not been tested experimentally.The clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated nuclease 9(Cas9)programed with a synthetic single-guide RNA(sgRNA)emerges as a method for genome editing in virtually any organisms.Here we report that targeted DNA fragment inversions and duplications could easily be achieved in human and mouse genomes by CRISPR with two sgRNAs.Specifically,we found that,in cultured human cells and mice,efficient precise inversions of DNA fragments ranging in size froma few tens of bp to hundreds of kb could be generated.In addition,DNA fragment duplications and deletions could also be generated by CRISPR through trans-allelic recombination between the Cas9-induced double-strand breaks(DSBs)on two homologous chromosomes(chromatids).Moreover,junctions of combinatorial inversions and duplications of the protocadherin(Pcdh)gene clusters induced by Cas9 with four sgRNAs could be detected.In mice,we obtained founders with alleles of precise inversions,duplications,and deletions of DNA fragments of variable sizes by CRISPR.Interestingly,we found that very efficient inversions were mediated by microhomology-mediated end joining(MMEJ)through short inverted repeats.We showed for the first time that DNA fragment inversions could be transmitted through germlines in mice.Finally,we applied this CRISPR method to a regulatory element of the Pcdha cluster and found a new role in the regulation of members of the Pcdhg cluster.This simple and efficient method should be useful in manipulating mammalian genomes to study millions of regulatory DNA elements as well as vast numbers of gene clusters. | Jinhuan Li Jia Shou YaGuo Yuanxiao Tang YonghuWu Zhilian Jia Yanan Zhai Zhifeng Chen Quan Xu Qiang Wu | 2015 | Journal of Molecular Cell Biology2015,7,4: | 4 |
| 9 | Resveratrol and its derivative pterostilbene ameliorate intestine injury in intrauterine growth-retarded weanling piglets by modulating redox status and gut microbiota显示文摘Background:Intestinal disorder is an important factor contributing to growth lag and high rates of morbidity and mortality of piglets with intrauterine growth retardation(IUGR).Resveratrol(RSV)and its derivative pterostilbene(PT)are natural stilbenes possessing various bioactivities,such as antioxidative and anti-inflammatory effects.This study compared the protective potential of RSV and PT on the intestinal redox status and gut microbiota in weanling piglets with IUGR.Methods:Eighteen male piglets of normal body weight(NBW)and 54 same-sex IUGR piglets were chosen according to their birth and weaning weights.The NBW piglets accepted a basal diet,while the IUGR piglets were allotted to one of three groups according to their body weight at weaning and received a basal diet,an RSV-supplemented diet(300 mg/kg),or a PT-supplemented diet(300 mg/kg),respectively.Results:Compared with IUGR piglets,both RSV and PT improved the IUGR-associated decrease in jejunal villus height and increases in plasma diamine oxidase activity and D-lactate level and jejunal apoptosis of piglets(P<0.05).Administering RSV and PT also enhanced jejunal superoxide dismutase activity and the mRNA and protein expression of superoxide dismutase 2 of IUGR piglets by promoting nuclear factor erythroid 2-related factor 2(Nrf2)nuclear translocation(P<0.05).Comparatively,PT was more effective than RSV in elevating the villus height/crypt depth ratio and occludin mRNA and protein levels in the jejunum of IUGR piglets(P<0.05).PT was also superior to RSV in increasing Nrf2 nuclear translocation and inhibiting malondialdehyde accumulation in the jejunum of IUGR piglets(P<0.05).Additionally,RSV modulated the composition of cecal microbiota of IUGR piglets,as evidenced by increasing the prevalence of the phylum Bacteroidetes and the genera Prevotella,Faecalibacterium,and Parabacteroides and inhibiting the growth of the phylum Proteobacteria and its genera Escherichia and Actinobacillus(P<0.05).Moreover,RSV significantly increased the butyrate concentration in the cecum of IUGR piglets(P<0.05).Conclusion:PT is more potent than RSV to prevent intestinal oxidative stress,while RSV has a stronger capacity to regulate gut microbiota compared to PT. | Yanan Chen Hao Zhang Yueping Chen Peilu Jia Shuli Ji Yuying Zhang Tian Wang | 2021 | Journal of Animal Science and Biotechnology2021,12,4: | 3 |
| 10 | Bevacizumab promotes active biological behaviors of human umbilical vein endothelial cells by activating TG Fpi pathways via off-VEGF signaling显示文摘Objective:Bevacizumab is a recombinant humanized monoclonal antibody that blocks vascular endothelial growth factor(VEGF)with clear clinical benefits.However,overall survival of some cancer types remains low owing to resistance to bevacizumab therapy.While resistance is commonly ascribed to tumor cell invasion induced by hypoxia-inducible factor(HIF),less attention has been paid to the potential involvement of endothelial cells(ECs)in vasculature activated by anti-angiogenic drugs.Methods:Human umbilical vein ECs(HUVECs),bEnd.3 cells,and mouse retinal microvascular ECs(MRMECs)were treated with bevacizumab under conditions of hypoxia and effects on biological behaviors,such as migration and tube formation,examined.Regulatory effects on TGFpi and CD 105(endoglin)were established via determination o f protein and mRNA levels.We further investigated whether the effects of bevacizumab could be reversed using the receptor tyrosine kinase inhibitor anlotinib.Results:Bevacizumab upregulated TGFpi as well as CD 105,a component o f the TGFP receptor complex and an angiogenesis promoter.Elevated CD 105 induced activation of Sm adl/5,the inflammatory pathway and endothelial-mesenchymal transition.The migration ability of HUVECs was enhanced by bevacizumab under hypoxia.Upregulation o f CD 105 was abrogated by anlotinib,which targets multiple receptor tyrosine kinases including VEGFR2/3,FGFR1-4,PD G FRα/β,C-Kit,and RET.Conclusions:Bevacizumab promotes migration and tube formation of HUVECs via activation of the TGFβi pathway and upregulation of CD105 expression.Anlotinib reverses the effects of bevacizumab by inhibiting the above signals. | Xiaoling Zhang Yan Zhang Yanan Jia Tingting Qin Cuicui Zhang Yueya Li Chengmou Huang Zhujun Liu Jing Wang Kai Li | 2020 | Cancer Biology & Medicine2020,17,2: | 3 |
| 11 | Femtosecond laser-induced periodic surface structures on lithium niobate crystal benefiting from sample heating显示文摘Periodic surface structures were fabricated by irradiating lithium niobate(LN) crystals with femtosecond laser pulses at sample temperatures ranging from 28℃ to 800℃. Carrier density and conductivity of the samples were increased via heating LN, which inhibited coulomb explosion to obtain a uniform periodic surface structure. The periodic surface structures cover an area of 8 mm × 8 mm and have an average spacing of 174 5 nm.Meanwhile, the absorption of the processed sample is about 70% in the spectral range of 400–1000 nm, which is one order of magnitude higher than that of pure LN. | 12 | Effect of external surface of HZSM-5 zeolite on product distribution in the conversion of methanol to hydrocarbons显示文摘The external surface of HZSM-5 zeolite was passivated by liquid siliceous deposition and by acidic sites poisoning with lepidine, respectively.Then methanol-to-hydrocarbons(MTH) reaction was investigated over the above as-prepared catalysts and the dissoluble coke on these used catalysts was analyzed by GC-MS, to study the role of the external surface of HZSM-5 in the catalytic reaction. Comparison with the experimental results based on parent ZSM-5 showed that the product distribution of MTH reaction was obviously influenced by the external surface.Evidences were listed as follows:(1) the final product on parent HZSM-5 showed higher aromatic selectivity, lower olefin selectivity, lower ratio of C2/C3+aliphatics and higher ratio of C3/C4+aliphatics than the reaction mixture produced by the sole catalysis of acidic sites in HZSM-5channel;(2) a little of pentamethylbenzene and hexamethylbenzene in the product on parent HZSM-5, was produced via multi-methylation of methylbenzene on the external surface. | 13 | Prediction of Polycyclic Aromatic Hydrocarbon Bioaccessibility to Earthworms in Spiked Soils by Composite Extraction with Hydroxypropyl-β-Cyclodextrin and Organic Acids显示文摘Traditional exhaustive extraction methods often overestimate the risk of polycyclic aromatic hydrocarbon(PAH) bioaccessibility to biota. Therefore, reliable assessment methods need to be established. In this study, a composite extraction with hydroxypropyl-β-cyclodextrin(HPCD) and three low-molecular-weight organic acids, oxalic acid(OA), malic acid(MA), and citric acid(CA), was used to predict the PAH bioaccessibility to earthworms, subjecting to two soils(red soil and yellow soil) spiked with selected PAHs,phenanthrene, pyrene, chrysene, benzo(a)anthracene, benzo(b)fluoranthene, benzo(k)fluoranthene, and benzo(a)pyrene. For both soils,concentrations of PAHs by composite extraction using HPCD-OA(R^2= 0.89–0.92, slope = 1.89–2.03; n = 35), HPCD-MA(R^2=0.92–0.96, slope = 1.43–1.67; n = 35), and HPCD-CA(R^2= 0.92–0.96, slope = 1.26–1.56; n = 35) were significantly correlated with PAH accumulation in the Eisenia fetida earthworms. Moreover, the HPCD-CA-and HPCD-MA-extracted PAH concentrations were closer to the earthworm-accumulated PAH concentration than the extraction using just HPCD. | 15 | Stable confinement of Fe/Fe_(3)C in Fe,N-codoped carbon nanotube towards robust zinc-air batteries显示文摘Catalytic oxygen reduction reaction(ORR)and oxygen evolution reaction(OER)have garnered great attention as the key character in metal-air batteries.Herein,we developed a superior nonprecious bifunctional oxygen electrocatalyst,fabricated through spatial confinement of Fe/Fe_(3)C nanocrystals in pyridinic N and Fe-Nx rich carbon nanotubes(Fe/Fe_(3)C-N-CNTs).During ORR,the resultant electrocatalyst exhibits positive onset pote ntial of 1.0 V(vs.RHE),large half-wave potentials of 0.88 V(vs.RHE),which is more positive than Pt/C(0.98 V and 0.83 V,respectively).Remarkably,Fe/Fe_(3)C-N-CNTs exhibits outstanding durability and great methanol tolerance,exceeding Pt/C and most reported nonprecious metal-based oxygen reduction electrocatalysts.Moreover,Fe/Fe_(3)C-N-CNTs show a markedly low potential at j=10 mA/cm^(2),small Tafel slopes and extremely high stability for OER.Impressively,the Fe/Fe_(3)C-N-CNTs-based Zn-air batteries demonstrate high power density of 183 mW/cm^(2)and robust charge/discharge stability.It is revealed that the spatial confinement effect can impede the aggregation and corrosion of Fe/Fe_(3)C nanocrystals.Meanwhile,Fe/Fe_(3)C and Fe-Nx play synergistic effect on boosting the ORR/OER activity,which provides an important guideline for construction of inexpensive nonprecious metal-carbon hybrid nanomaterials. | Lingbo Zong Xin Chen Shuming Dou Kaicai Fan Zumin Wang Wenjun Zhang Yunmei Du Jie Xu Xiaofei Jia Qi Zhang Xia Li Yida Deng Yanan Chen Lei Wang | 2021 | Chinese Chemical Letters2021,32,3: | 2 |
| 16 | High wave transmittance and low thermal conductivity Y-a-SiAlON porous ceramics for high-temperature radome applications显示文摘With the development of hypersonic vehicle technology,ceramic-based radome materials are highly demanded due to their high operating temperatures,good dielectric properties,and high mechanical properties.Althoughα-SiAlON is an ideal material for radome applications,its intrinsic low transmittance and high thermal conductivity limit its applications.Herein,we prepared Y-α-SiAlON porous ceramics through tert-butanol(TBA)gel-casting using the self-synthesizedα-SiAlON powder.The Y-α-SiAlON porous ceramics exhibited a uniform micron-level connected pore structure with the porosity(P)of 44.2%-58.6%.The real part of permittivity(s')was 3.13-4.18(8.2-12.4 GHz),which decreased significantly with the increasing porosity.The wave transmittance(|T|^(2))of the sample with porosity of 58.6%could exceed 80%in the thickness range of 6-10 mm.The thermal conductivity was maintained at a low level of 1.38-2.25 W·m^(-1)·K^(-1) owing to the introduction of the pore structure.The flexural strength was 44.73-88.33 MPa,which may be increased by rod-likeα-SiAlON grains.The results indicate that the prepared Y-α-SiAlON porous ceramics meet the requirements of high-temperature wave-transmitting materials for radome applications. | Runwu Yang Jie Xu Jia Guo Xuanyu Meng Ping Zhang Fengying Fan Yanan Qu Feng Gao | 2023 | Journal of Advanced Ceramics2023,12,6: | 1 |
