2The calcium-dependent protein kinase ZmCDPK7 functions in heat-stress tolerance in maize显示文摘Global warming poses a serious threat to crops.Calcium-dependent protein kinases(CDPKs)/CPKs play vital roles in plant stress responses,but their exact roles in plant thermotolerance remains elusive.Here,we explored the roles of heat-induced ZmCDPK7 in thermotolerance in maize.ZmCDPK7-overexpressing maize plants displayed higher thermotolerance,photosynthetic rates,and antioxidant enzyme activity but lower H2 O2 and malondialdehyde(MDA)contents than wild-type plants under heat stress.ZmCDPK7-knockdown plants displayed the opposite patterns.ZmCDPK7 is attached to the plasma membrane but can translocate to the cytosol under heat stress.ZmCDPK7 interacts with the small heat shock protein sHSP17.4,phosphorylates sHSP17.4 at Ser-44 and the respiratory burst oxidase homolog RBOHB at Ser-99,and up regulates their expression.Site-directed mutagenesis of sHSP17.4 to generate a Ser-44-Ala substitution reduced ZmCDPK7’s enhancement of catalase activity but enhanced ZmCDPK7’s suppression of MDA accumulation in heat-stressed maize protoplasts.sHSP17.4,ZmCDPK7,and RBOHB were less strongly upregulated in response to heat stress in the abscisic acid-deficient mutant vp5 versus the wild type.Pretreatment with an RBOH inhibitor suppressed sHSP17.4 and ZmCDPK7 expression.Therefore,abscisic acid-induced ZmCDPK7 functions both upstream and downstream of RBOH and participates in thermotolerance in maize by mediating the phosphorylation of sHSP17.4,which might be essential for its chaperone function.Yulong Zhao Hanwei Du Yankai Wang Huali Wang Shaoyu Yang Chaohai Li Ning Chen Hao Yang Yihao Zhang Yulin Zhu Luyao Yang Xiuli Hu 2021Journal of Integrative Plant Biology2021,63,3:6

4Association of assisted reproductive technology, germline de novo mutations and congenital heart defects in a prospective birth cohort study显示文摘Emerging evidence suggests that children conceived through assisted reproductive technology(ART)have a higher risk of congenital heart defects(CHDs)even when there is no family history.De novo mutation(DNM)is a well-known cause of sporadic congenital diseases;however,whether ART procedures increase the number of germline DNM(gDNM)has not yet been well studied.Here,we performed whole-genome sequencing of 1137 individuals from 160 families conceived through ART and 205 families conceived spontaneously.Children conceived via ART carried 4.59 more gDNMs than children conceived spontaneously,including 332 paternal and 1.26 maternal DNMs,after correcting for parental age at conception,cigarette smoking,alcohol drinking,and exercise behaviors.Paternal DNMs in offspring conceived via ART are characterized by C>T substitutions at CpG sites,which potentially affect protein-coding genes and are significantly associated with the increased risk of CHD.In addition,the accumulation of non-coding functional mutations was independently associated with CHD and 87.9% of the mutations were originated from the father.Among ART offspring,infertility of the father was associated with elevated paternal DNMs;usage of both recombinant and urinary follicle-stimulating hormone and high-dosage human chorionic gonadotropin trigger was associated with an increase of maternal DNMs.In sum,the increased gDNMs in offspring conceived by ART were primarily originated from fathers,indicating that ART itself may not be a major reason for the accumulation of gDNMs.Our findings emphasize the importance of evaluating the germline status of the fathers in families with the use of ART.Cheng Wang Hong Lv Xiufeng Ling Hong Li Feiyang Diao Juncheng Dai Jiangbo Du Ting Chen Qi Xi Yang Zhao Kun Zhou Bo Xu Xiumei Han Xiaoyu Liu Meijuan Peng Congcong Chen Shiyao Tao Lei Huang Cong Liu Mingyang Wen Yangqian Jiang Tao Jiang Chuncheng Lu Wei Wu Di Wu Minjian Chen Yuan Lin Xuejiang Guo Ran Huo Jiayin Liu Hongxia Ma Guangfu Jin Yankai Xia Jiahao Sha Hongbing Shen Zhibin Hu 2021Cell Research2021,31,8:2

6Population Genetics of SARS-CoV-2: Disentangling Effects of Sampling Bias and Infection Clusters显示文摘A novel RNA virus,the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is responsible for the ongoing outbreak of coronavirus disease 2019(COVID-19).Population genetic analysis could be useful for investigating the origin and evolutionary dynamics of COVID-19.However,due to extensive sampling bias and existence of infection clusters during the epidemic spread,direct applications of existing approaches can lead to biased parameter estimations and data misinterpretation.In this study,we first present robust estimator for the time to the most recent common ancestor(TMRCA)and the mutation rate,and then apply the approach to analyze 12,909 genomic sequences of SARS-CoV-2.The mutation rate is inferred to be 8.69×10^(−4) per site per year with a 95%confidence interval(CI)of[8.61×10^(−4),8.77×10^(−4)],and the TMRCA of the samples inferred to be Nov 28,2019 with a 95%CI of[Oct 20,2019,Dec 9,2019].The results indicate that COVID-19 might originate earlier than and outside of Wuhan Seafood Market.We further demonstrate that genetic polymorphism patterns,including the enrichment of specific haplotypes and the temporal allele frequency trajectories generated from infection clusters,are similar to those caused by evolutionary forces such as natural selection.Our results show that population genetic methods need to be developed to efficiently detangle the effects of sampling bias and infection clusters to gain insights into the evolutionary mechanism of SARS-CoV-2.Software for implementing VirusMuT can be downloaded at http://gffzzdbc7b6aaae734bddsqp9v95w9cxxv6bc6.ffgz.tsg.suse.edu.cn/biocode/tools/BT007081.Qi Liu Shilei Zhao Cheng-Min Shi Shuhui Song Sihui Zhu Yankai Su Wenming Zhao Mingkun Li Yiming Bao Yongbiao Xue Hua Chen 2020Genomics, Proteomics & Bioinformatics2020,18,6:1
7Experimental study on filtering mixed solid-liquid dust with a sliding granular bed filter显示文摘The filtration of mixed dust that included a small number of melted(liquid)particles was studied in an experimental granular bed filter(GBF).Results show that the collection efficiency of dust containing melted particles is higher than that of dust composed of solid particles but the pressure drop from the former is higher than that of the latter.The collection efficiency and pressure drop increase as the concentration of melted particles increases.A surface sliding GBF exhibits good comprehensive performance when filtering dust,especially dust containing a mix of solid and liquid particles.The effects of the gas temperature,gas velocity,sliding filter bed thickness,and sliding collector flux on filter performance were also examined.Based on the experimental results,correlations for the collection efficiency and pressure drop for mixed dust are presented.Yankai Peng Chuanping Liu Li Wang Shaowu Yin Lige Tong Zeyi Jiang 2021Particuology2021,19,5:1

9Profiles of metabolic gene expression in the white adipose tissue, liver and hypothalamus in leptin knockout(Lep^(△I14/△I14)) rats显示文摘Leptin deficiency is principally linked to metabolic disorders. Leptin knockout(Lep^(△I14/△I14) Sprague Dawley rats created by CRISPR/Cas9 is a new model to study metabolic disorders. We used a whole rat genome oligonucleotide microarray to obtain tissue-specific gene expression profiles of the white adipose tissue, liver and hypothalamus in Lep^(△I14/△I14))and wild-type(WT) rats. We found 1,651 differentially expressed(enriched) genes in white adipose tissue,916 in the liver, and 306 in the hypothalamus in the Lep^(△I14/△I14) rats compared to WT. Gene ontology category and KEGG pathway analysis of the relationships among differentially expressed genes showed that these genes were represented in a variety of functional categories, including fatty acid metabolism, molecular transducers and cellular processes. The reliability of the data obtained from microarray was verified by quantitative real-time PCR on 14 representative genes. 11Composite anti-disturbance resilient control for Markovian jump nonlinear systems with general uncertain transition rate显示文摘In this paper, the issue of disturbance observer based resilient control is addressed for Markovian jump nonlinear systems with multiple disturbances and general uncertain transition rates. The disturbances are divided into two parts: one has a bounded H2 norm, and the other is given by an exogenous system. The general uncertain transition rate matrix is composed of unknown elements and uncertain ones. The uncertain transition rate only has a known approximate range. First, the disturbance described by the exogenous system is estimated by a disturbance observer, and its estimation is used for the controller as feedforward compensation. Subsequently, by using the resilient control method, a composite anti-disturbance resilient controller is constructed to guarantee stochastic stability with L_2-L_∞ performance of the closed-loop systems. Subsequently, the Lyapunov stability method and linear matrix inequality technique are applied to obtain the controller gain. 12Abnormal miR-214/A20 expression might play a role in T cell activation in patients with aplastic anemia显示文摘Aberrant T cell activation is a major cause of aplastic anemia(AA)pathogenesis.Recent studies have shown that miRNAs regulate T cell activation and are involved in AA.A previous study found that miR-214 was significantly up-regulated upon T cell activation in a CD28-dependent fashion by targeting PTEN.However,the expression characteristics of miR-214 and its target genes in AA have not been defined.In this study,target genes for miR-214 were predicted and confirmed by bioinformatics and luciferase reporter assays.The expression levels of miR-214 and target genes were detected in 36 healthy individuals and 35 patients with AA in peripheral blood mononuclear cells by real-time quantitative reverse transcriptase-polymerase chain reaction.Bioinformatics and luciferase reporter assays identified that miR-214 could bind to the A2030 untranslated regions.Significantly increased miR-214 and the decreased A20 expression level were detected in the AA patients compared with the healthy group.In addition,significantly increased miR-214 was found in non-severe aplastic anemia compared with severe aplastic anemia patients.These results suggested that the A20 gene was a potential target of miR-214,and elevated miR-214 might medicate T cell activation at least in part by regulating A20 expression in AA.We firstly confirmed that miR-214 regulated A20 expression,and aberrant miR-214/A20 expression might contribute to immunopathology in AA.The miR-214 expression might be used as a potential biomarker that assisted in diagnosing AA severity.Zhi Yu Cunte Chen Yankai Xiao Xiaohui Chen Lixing Guo Guangxiao Tan Guixuan Huang Weifeng Luo Ming Zhou Yumiao Li Chen Lin Qi Shen Yuping Zhang Bo Li 2020Blood Science2020,2,3:1
13Neuroprotection of a cocktail therapy involving anti-intercellular adhesion molecule 1 antibody and insulin-like growth factor 1 in treatment of cerebral ischemia/reperfusion injury in cats Nuclear magnetic resonance examination and pathological validatio显示文摘BACKGROUND: The pathophysiological mechanisms of ischemia are extremely complicated. It is difficult to confirm and maintain the therapeutic effects if only one neuroprotective agent is used. It is hypothesized that a cocktail therapy involving a combined application of neuroprotective agents is feasible and offers excellent therapeutic potential. OBJECTIVE: To evaluate the neuroprotective effects of a cocktail therapy of insulin-like growth factor 1 (IGF1) combined with anti-intercellular adhesion molecule 1 (ICAM1) antibody in the treatment of cerebral ischemia/reperfusion injury using medical imaging, pathology, and functional neurological deficit scoring techniques. DESIGN, TIME AND SETTING: This randomized, controlled, neuroimaging analysis of function and pathological observation was performed at the Laboratory of Molecular Imaging, Second Hospital, Hebei Medical University between September 2006 and December 2007. MATERIALS: Transient middle cerebral artery occlusion (MCAO) was induced in 24 healthy adult cats. Anti-ICAM1 antibody and IGF1 were sourced from the Shanghai Kangcheng Biological Product Co., Ltd., China. Stereotaxic apparatus was purchased from the Center for Medical Apparatus and Instruments, Shandong Liaocheng People’s Hospital, China. The in situ apoptosis kit was provided by the Beijing Zhongshan Biotechnique Co., Ltd., China. METHODS: Twenty-four cat models of MCAO were randomly divided into four groups (n = 6): control, IGF1, anti-ICAM1 antibody and cocktail therapy. Following a 2-hour ischemia and subsequent lateral cerebral ventricular puncture, 100 μg IGF1(cerebral ventricular), 100 μg anti-ICAM1 antibody (i.v.), 50 μg IGF1(lateral cerebral ventricular) + 50 μg anti-ICAM1 antibody (i.v.), and 100 μg physiological saline (i.v.) were administered to the IGF1, anti-ICAM1 antibody, cocktail therapy and control groups, respectively. On the following day, the same administration was performed again. MAIN OUTCOME MEASURES: Pathological observation of the cerebral dura mater tissue surrounding the MCAO target site was performed by electron microscopy. At days 3 and 7 following MCAO induction, the volume of the cerebral infarction was measured by Philips functional neurological deficit scoring and nuclear magnetic resonance imaging. RESULTS: Pathological observation revealed that the control group exhibited a great number of swollen neurons, glial cells and vascular endothelial cells, and showed severely injured mitochondria with an absence of the double membrane structure. The same phenomena were partially alleviated in the IGF1 and anti-ICAM1 antibody groups, and the most obvious alleviation of injuries was in the cocktail therapy group. At day 6 following MCAO establishment, the cocktail therapy group exhibited the smallest cerebral infarction volumes among the four groups (F = 71.322, P < 0.01). At days 3 and 7 following MCAO induction, the F value of Philips functional neurological deficit scoring was 10.398 and 14.430, respectively (P < 0.01); however, the best neurological functional recovery was in the cocktail therapy group. 15A systematic review of the impacts of exposure to micro-and nano-plastics on human tissue accumulation and health显示文摘Micro-and nano-plastics(MNPs)pollution has become a pressing global environmental issue,with growing concerns regarding its impact on human health.However,evidence on the effects of MNPs on human health remains limited.This paper reviews the three routes of human exposure to MNPs,which include ingestion,inhalation,and dermal contact.It further discusses the potential routes of translocation of MNPs in human lungs,intestines,and skin,analyses the potential impact of MNPs on the homeostasis of human organ systems,and provides an outlook on future research priorities for MNPs in human health.There is growing evidence that MNPs are present in human tissues or fluids.Lab studies,including in vivo animal models and in vitro human-derived cell cultures,revealed that MNPs exposure could negatively affect human health.MNPs exposure could cause oxidative stress,cytotoxicity,disruption of internal barriers like the intestinal,the air–blood and the placental barrier,tissue damage,as well as immune homeostasis imbalance,endocrine disruption,and reproductive and developmental toxicity.Limitedly available epidemiological studies suggest that disorders like lung nodules,asthma,and blood thrombus might be caused or exacerbated by MNPs exposure.However,direct evidence for the effects of MNPs on human health is still scarce,and future research in this area is needed to provide quantitative support for assessing the risk of MNPs to human health.Yudong Feng Chen Tu Ruijie Li Di Wu Jie Yang Yankai Xia Willie J.G.M.Peijnenburg Yongming Luo 2023Eco-Environment & Health2023,2,4:0
16Genetic diversity,population structure and core collection analysis of Hunan tea plant germplasm through genotyping-by-sequencing显示文摘Tea is considered to be a well-known and widely consumed beverage and Hunan province is rich in tea plant germplasm.In order to better conserve and utilize Hunan tea plant resources,110 tea accessions from seven geographical origins were used to assess genetic diversity of Hunan tea plant germplasm through genotyping by sequencing(GBS)technology.As a result,a total of 311,044 high-quality single nucleotide polymorphism(SNP)markers were obtained.Population structure,phylogenetic relationships and principal component analysis(PCA)divided the entire accessions into three groups.The genetic diversity and population differentiation analysis showed that the mean observed heterozygosity(Ho)ranged from 0.16 to 0.24,while the mean polymorphic information content(PIC)ranged from 0.14 to 0.17,and mean minor allele frequency(MAF)ranged from 0.11 to 0.14.Analysis of molecular variance(AMOVA)indicated that 81.38%of the total variance was derived from within populations,which suggested a rich genetic diversity in Hunan tea germplasms.Furthermore,a core tea germplasm set was developed,which was comprised of 22 tea plant accessions and maintained the whole genetic diversity of the entire collection.This work should be valuable for conservation and utilization of tea germplasm in Hunan。Feiyi Huang Jihua Duan Yu Lei Zhen Liu Yankai Kang Yi Luo Yingyu Chen Youyong Li Shuoqian Liu Saijun Li Zhonghua Liu 2022Beverage Plant Research2022,2,1:0
17Cumulative live birth rates of in vitro fertilization/intracytoplasmic sperm injection after multiple complete cycles in China显示文摘There were few studies of cumulative live birth rates(CLBRs) based on multicenter reproductive clinical data from the general Chinese population.Here we report a retrospective cohort study,including 14 311 women with17 315 cycles,in three reproductive centers to evaluate two estimated parameters of CLBRs with multiple transfer cycles of in vitro fertilization(IVF)/intracytoplasmic sperm injection(ICSI) in a Chinese population.We found that CLBRs were related to female age and endometrial thickness.By the fourth transfer cycle,the conservative and optimal estimates of CLBRs were 52.95% and 77.30% in women under 30 years of age,and 18.17% and26.51% in those 37 years of age or older,respectively.The two estimates were 44.70% and 63.15% in women with endometrial thickness more than 7 mm,and 32.05% and 46.18% in those with less than 7 mm,respectively.In addition,body mass index(BMI),duration of infertility,and infertility diagnoses may also be related to CLBRs on certain conditions.The findings from this study on CLBRs after multiple transfer cycles of IVF/ICSI treatment on different conditions in the Chinese population should be beneficial to both infertile couples and clinicians.Lei Huang Qun Lu Jiangbo Du Hong Lv Shiyao Tao Shiyao Chen Xiuzhu Li Xiumei Han Kun Zhou Bo Xu Xiaoyu Liu Hongxia Ma Yankai Xia Guangfu Jin Hongbing Shen Xiufeng Ling Zhibin Hu Jichun Tan Feiyang Diao 2020The Journal of Biomedical Research2020,34,5:0
18Bioinformatics analysis of genes differentially expressed in autism and screening of hub genes in the occurrence and development of autism显示文摘Objective:to screen the genes differentially expressed in autism using bioinformatics methods,and to explore their functional enrichment,related signaling pathways and the tissue-specific expression of hub genes.Methods:the autism expression profile chip numbered GSE77103 in the Gene Expression Omnibus(GEO)database was selected for examination.R language and related R packages were used for the screening and visualization of the diflerentially expressed genes.Gene Ontology(GO)function enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)signaling pathway analysis of the differential genes were carried out using the relevant R package of R language.The database STRING was used to construct the interaction network of the proteins encoded by the differentially expressed genes,and the software Cytoscape was used to screen the hub genes in the network.The selected hub genes were imported into the BioGPS database to analyze the tissue-specific expression of the hub genes.Results:six hundred and sixty diflerentially expressed genes were screened out.Three hundred and seventy-three up-regulated genes and 287 down-regulated genes in the peripheral blood mononuclear cells of autistic children were compared with the peripheral blood mononuclear cells of healthy children.GO functional enrichment results showed that biological processes(BP)were mainly involved in viral response,negative regulation of viral genome replication,negative regulation of multiple biological processes,negative reg ulation reg ulation of viral life cycle,and defense responses to viruses.Cell components(CC)were involved in vesicles,lysosomal membranes,lysosomal lumen,etc.;molecular functions(MF)were involved in regulating glutathione transferase activity peroxidase activity,oxidoreductase activity,Glutathione peroxidase and transferase activity,etc.The results of KEGG signaling pathway analysis showed that the differentially expressed genes were related to the lysosomal pathway,the glutathione metabolism pathway and the arachidonic acid metabolism pathway.The hub genes screened by cytoHubba were:interferon regulatory factor 7(IRF7),interferon stimulated exonuclease gene 15(ISG15),XIAP associated factor 1(XAF1),MX dynamin like GTPase 1(MX1),interferon induced protein with tetratricopeptide repeats 1(IFIT1),interferon induced protein with tetratricopeptide repeats 5(IFIT5),29-55-oligoadenylate synthetase 3(OAS3),interferon induced protein 44(IFI44),HECT and the RLD domain containing E3 ubiquitin protein ligase 5(HERC5),interferon stimulated exonuclease gene 20(ISG20).Conclusion:there are genes that are diflerentially expressed in the peripheral blood mononuclear cells of autistic toddlers and healthy toddlers.IRF7,ISG15,XAF1,MX],IFIT1,IFIT5,OAS3,IFI44,HERC5,ISG20 are the hub regulatory genes of autism.Manli Li Xiaoli Ma Chendi Mai Zhiru Fan Yankai Ren 2022Journal of Translational Neuroscience2022,7,1:0

20Crosstalk between glioblastoma and tumor microenvironment drives proneural–mesenchymal transition through ligand-receptor interactions显示文摘Glioblastoma(GBM)is the most common intrinsic and aggressive primary brain tumor in adults,with a median survival of approximately 15 months.GBM heterogeneity is considered responsible for the treatment resistance and unfavorable prognosis.Proneural-mesenchymal transition(PMT)represents GBM malignant progression and recurrence,which might be a breakthrough to understand GBM heterogeneity and overcome treatment resistance.PMT is a complicated process influenced by crosstalk between GBM and tumor microenvironment,depending on intricate ligand-receptor interactions.In this review,we summarize the autocrine and paracrine pathways in the GBM microenvironment and related ligand-receptor interactions inducing PMT.We also discuss the current therapies targeting the PMT-related autocrine and paracrine pathways.Together,this review offers a comprehensive understanding of the failure of GBM-targeted therapy and ideas for future tendencies of GBM treatment.Yancheng Lai Xiaole Lu Yankai Liao Pei Ouyang Hai Wang Xian Zhang Guanglong Huang Songtao Qi Yaomin Li 2024Genes & Diseases2024,11,2:0
