1Potential and challenges of tannins as an alternative to in-feed antibiotics for farm animal production显示文摘Naturally occurring plant compounds including tannins, saponins and essential oils are extensively assessed as natural alternatives to in-feed antibiotics.Tannins are a group of polyphenolic compounds that are widely present in plant region and possess various biological activities including antimicrobial,anti-parasitic, anti-viral, antioxidant, anti-inflammatory, immunomodulation, etc.Therefore, tannins are the major research subject in developing natural alternative to in-feed antibiotics.Strong protein affinity is the well-recognized property of plant tannins, which has successfully been applied to ruminant nutrition to decrease protein degradation in the rumen, and thereby improve protein utilization and animal production efficiency.Incorporations of tannin-containing forage in ruminant diets to control animal pasture bloat, intestinal parasite and pathogenic bacteria load are another 3 important applications of tannins in ruminant animals.Tannins have traditionally been regarded as 'anti-nutritional factor' for monogastric animals and poultry, but recent researches have revealed some of them, when applied in appropriate manner, improved intestinal microbial ecosystem, enhanced gut health and hence increased productive performance.The applicability of plant tannins as an alternative to in-feed antibiotics depends on many factors that contribute to the great variability in their observed efficacies.Qianqian Huang Xiuli Liu Guoqi Zhao Tianming Hu Yuxi Wang 2018Animal Nutrition2018,,2:54

4Programmed Death Ligand-1 on Microglia Regulates Th1 Differentiation via Nitric Oxide in Experimental Autoimmune Encephalomyelitis显示文摘Microglia are considered to be potential antigen-presenting cells and have the ability to present antigen under pathological conditions. Nevertheless, whether and how microglia are involved in immune regulation are largely unknown. Here, we investigated the suppressive activity of microglia during experimental autoimmune encephalomyelitis(EAE) induced by myelin oligodendrocyte glycoprotein, with the goal of understanding their role in regulating the T cell reaction. Using flow cytometric analysis, we found that microglia were characterized by increased cell number and up-regulated programmed death ligand-1(PD-L1) at the peak phase of EAE. Meanwhile,both the CD^(4+)T cells and microglia that infiltrated the central nervous system expressed higher levels of PD1, the receptor for PD-L1, accompanied by a decline of Th1 cells.In an ex vivo co-culture system, microglia from EAE mice inhibited the proliferation of antigen-specific CD^(4+)T cells and the differentiation of Th1 cells, and this was significantly inhibited by PD-L1 blockade. Further,microglia suppressed Th1 cells via nitric oxide(NO), the production of which was dependent on PD-L1. 5The calcium-dependent protein kinase ZmCDPK7 functions in heat-stress tolerance in maize显示文摘Global warming poses a serious threat to crops.Calcium-dependent protein kinases(CDPKs)/CPKs play vital roles in plant stress responses,but their exact roles in plant thermotolerance remains elusive.Here,we explored the roles of heat-induced ZmCDPK7 in thermotolerance in maize.ZmCDPK7-overexpressing maize plants displayed higher thermotolerance,photosynthetic rates,and antioxidant enzyme activity but lower H2 O2 and malondialdehyde(MDA)contents than wild-type plants under heat stress.ZmCDPK7-knockdown plants displayed the opposite patterns.ZmCDPK7 is attached to the plasma membrane but can translocate to the cytosol under heat stress.ZmCDPK7 interacts with the small heat shock protein sHSP17.4,phosphorylates sHSP17.4 at Ser-44 and the respiratory burst oxidase homolog RBOHB at Ser-99,and up regulates their expression.Site-directed mutagenesis of sHSP17.4 to generate a Ser-44-Ala substitution reduced ZmCDPK7’s enhancement of catalase activity but enhanced ZmCDPK7’s suppression of MDA accumulation in heat-stressed maize protoplasts.sHSP17.4,ZmCDPK7,and RBOHB were less strongly upregulated in response to heat stress in the abscisic acid-deficient mutant vp5 versus the wild type.Pretreatment with an RBOH inhibitor suppressed sHSP17.4 and ZmCDPK7 expression.Therefore,abscisic acid-induced ZmCDPK7 functions both upstream and downstream of RBOH and participates in thermotolerance in maize by mediating the phosphorylation of sHSP17.4,which might be essential for its chaperone function.Yulong Zhao Hanwei Du Yankai Wang Huali Wang Shaoyu Yang Chaohai Li Ning Chen Hao Yang Yihao Zhang Yulin Zhu Luyao Yang Xiuli Hu 2021Journal of Integrative Plant Biology2021,63,3:6
6Preparation and evaluation of poly(L-histidine) based pH-sensitive micelles for intracellular delivery of doxorubicin against MCF-7/ADR cells显示文摘In this study, a p H-sensitive micelle self-assembled from poly(L-histidine) based triblock copolymers of poly(ethylene glycol)–poly(D,L-lactide)–poly(L-histidine)(mPEG-PLA-PHis) was prepared and used as the intracellular doxorubicin(Dox) delivery for cancer chemotherapy. Dox was loaded into the micelles by thin-film hydration method and a Box–Behnken design for three factors at three levels was used to optimize the preparations. The optimized mPEG-PLA-Phis/Dox micelles exhibited good encapsulation efficiency of 91.12%,a mean diameter of 45 nm and narrow size distribution with polydispersity index of 0.256.In vitro drug release studies demonstrated that Dox was released from the micelles in a p Hdependent manner. Furthermore, the cellular evaluation of Dox loaded micelles displayed that the micelles possessed high antitumor activity in vitro with an IC50 of 35.30 μg/ml against MCF-7/ADR cells. The confocal microscopy and flow cytometry experiments indicated that m PEG-PLA-Phis micelles mediated efficient cytoplasmic delivery of Dox with the aid of poly(Lhistidine) mediated endosomal escape. In addition, blank m PEG-PLA-Phis micelles were shown to be nontoxic to MCF-7/ADR cells even at a high concentration of 200 μg/ml. 7Investigation on experimental load spectrum for high and low cycle combined fatigue test显示文摘As the outfield load spectrum is so complicated that it cannot be used directly for test study in laboratory.This paper presents a method to determine load spectrum for high and low cycle combined fatigue of turbine mortise at elevated temperature through experimental and numerical method.First of all,the low cycle load spectrum with duration time is determined through cumulative damage rule.The rain flow counting method is applied to obtain main cycles and sub cycles,and then the stress cycle is converted into pulsation cycle(stress ratio=0)based on the S-N curve and the Goodman curve;Secondly,three groups of different amplitudes tests are established to determine the high cycle amplitude.Finally,another three groups of the high-low combined cycle fatigue(HLCCF)tests for turbine mortise of a certain type engine are carried out.The results show that the macro and micro failure modes are identical with outfield's,which verifies the accuracy of the conversion method.Rongqiao Wang Jiaming Wei Dianyin Hu Xiuli Shen Jiang Fan 2013Propulsion and Power Research2013,2,4:4
8Significant Inhibition of Porcine Epidemic Diarrhea Virus In Vitro by Remdesivir,Its Parent Nucleoside and β-D-N^(4)-hydroxycytidine显示文摘Porcine epidemic diarrhea(PED)caused by porcine epidemic diarrhea virus(PEDV)is widespread in the world.In recent years,the increased virulence of the virus due to viral variations,has caused great economic losses to the pig industry in many countries.It is always worthy to find effective therapeutic methods for PED.As an important class of antivirals,nucleoside drugs which target viral polymerases have been applied in treating human viral infections for half a century.Herein,we evaluated the anti-PEDV potential of three broad-spectrum antiviral nucleoside analogs,remdesivir(RDV),its parent nucleoside(RDV-N)andβ-D-N^(4)-hydroxycytidine(NHC).Among them,RDV-N was the most active agent in Vero E6 cells with EC_(50) of 0.31μmol/L,and more potent than RDV(EC_(50)=0.74μmol/L)and NHC(EC_(50)=1.17μmol/L).The activity of RDV-N was further confirmed using an indirect immuno-fluorescence assay.Moreover,RDV-N exhibited a good safety profile in cells and in mice.The high sequence similarity of the polymerase functional domains of PEDV with other five porcine coronaviruses indicated a broader antiviral spectrum for the three compounds.Generally,RDV-N is a promising broad-spectrum antiviral nucleoside,and it would be worthy to make some structural modifications to increse its oral bioavailability.Yuanchao Xie Xiaozhen Guo Tianwen Hu Daibao Wei Xiuli Ma Jiaqiang Wu Bing Huang Jingshan Shen 2021Virologica Sinica2021,36,5:3

11Dendron-polymer hybrid mediated anticancer drug delivery for suppression of mammary cancer显示文摘Dendron-polymer-based nanoscale and stimuli-responsive drug delivery systems have shown great promise in tumor-targeting accumulation without significant toxicity.Here we report a dendronized polymer-doxorubicin(DOX)hybrid(DPDH)with an improved in vivo drug delivery efficiency for cancer therapy compared with a linear polymer-DOX conjugate(LPDC).The in vitro drug release profile of DOX indicates that DPDH displays pH-responsive drug release due to cleavage of hydrazone bonds since a greater amount of DOX is released at pH 5.2 at a faster rate than at pH 7.4.DPDH efficiently enters 4 T1 cells and releases DOX to induce cytotoxicity and apoptosis.Owing to the dendronzied structure,DPDH has a significantly longer blood circulation time than LPDC.DPDH substantially enhances the therapeutic efficacy to suppress tumor growth in a 4 T1 mammary cancer model than LPDC as well as free drug,evidenced from tumor growth inhibition,TUNEL assessment and histological analysis.Biosafety of DPDH is also confirmed from hemolysis,body weight shifts during treatment and pathological analysis.This study demonstrates the use of dendronized polymer-DOX hybrids for specific drug molecules is a promising approach for drug delivery.Dayi Pan Xiuli Zheng Miao Chen Qianfeng Zhang Zhiqian Li Zhenyu Duan Qiyong Gong Zhongwei Gu Hu Zhang Kui Luo 2021Journal of Materials Science & Technology2021,,4:2
12Combining immune checkpoint blockade with ATP-based immunogenic cell death amplifier for cancer chemo-immunotherapy显示文摘Amplifying“eat me signal”during tumor immunogenic cell death(ICD)cascade is crucial for tumor immunotherapy.Inspired by the indispensable role of adenosine triphosphate(ATP,a necessary“eat me signal”for ICD),a versatile ICD amplifier was developed for chemotherapy-sensitized immunotherapy.Doxorubicin(DOX),ATP and ferrous ions(Fe^(2+))were co-assembled into nanosized amplifier(ADO-Fe)throughπ‒πstacking and coordination effect.Meanwhile,phenylboric acid-polyethylene glycol-phenylboric acid(PBA-PEG-PBA)was modified on the surface of ADO-Fe(denoted as PADO-Fe)by the virtue of d-ribose unit of ATP.PADO-Fe could display active targetability against tumor cells via sialic acid/PBA interaction.In acidic microenvironment,PBA-PEG-PBA would dissociate from amplifier.Moreover,high H_(2)O_(2)concentration would induce hydroxyl radical(·OH)and oxygen(O_(2))generation through Fenton reaction by Fe^(2+).DOX and ATP would be released from the amplifier,which could induce ICD effect and“ICD adjuvant”to amplify this process.Together with programmed death ligands 1(PD-L1)checkpoint blockade immunotherapy,PADO-Fe could not only activate immune response against primary tumor,but also strong abscopal effect against distant tumor.Our simple and multifunctional ICD amplifier opens a new window for enhancing ICD effect and immune checkpoint blockade therapy.Jiulong Zhang Xiaoyan Sun Xiufeng Zhao Chunrong Yang Menghao Shi Benzhuo Zhang Haiyang Hu Mingxi Qiao Dawei Chen Xiuli Zhao 2022Acta Pharmaceutica Sinica B2022,12,9:2
13Effect of surface ligand density on cytotoxicity and pharmacokinetic profile of docetaxel loaded liposomes显示文摘Various biotin-modified liposomes incorporated with docetaxel(DTX) were prepared to study the effect of surface biotin density on the pharmacokinetic profile of the liposome. Four types of liposomes such as PEG modified liposome(PDL), 0.5%(mol) biotin modified liposome(0.5 BDL), 1%(mol) biotin modified liposome(1 BDL) and 2%(mol) biotin modified liposome(2 BDL) were prepared using thin film dispersion method. The prepared liposomes were characterized by measuring encapsulation efficiency(EE), particle size, Zeta-potential, physical stability and drug release profiles in vitro. MTT assay was performed to elevate the cytotoxicity of liposomes on MCF-7 cells. In vivo evaluation was further performed to investigate the effect of biotin surface density on the pharmacokinetic profiles. All the prepared liposomes exhibited high encapsulation efficiency, small particle size, narrow particle distribution and sustained release profiles in vitro. In MTT assay, 0.5 BDL showed largest tumor cell toxicity, compared with DTX solution. All liposomes containing DTX showed prolonged blood circulation in vivo, and 0.5 BDL showed the longest circulation time among the biotin modified liposome. Surface modification of liposome had a negative impact on the circulation of liposomes in the blood, which needs to be considered when designing the ligand mediated targeting delivery systems.
