2Cell–cell contact with proinflammatory macrophages enhances the immunotherapeutic effect of mesenchymal stem cells in two abortion models显示文摘Mesenchymal stem cells(MSCs),which are pluripotent cells with immunomodulatory properties,have been considered good candidates for the therapy of several immune disorders,such as inflammatory bowel diseases,concanavalin A-induced liver injury,and graft-versus-host disease.The embryo is a natural allograft to the maternal immune system.A successful pregnancy depends on the timely extinction of the inflammatory response induced by embryo implantation,followed by the switch to a tolerant immune microenvironment in both the uterus and the system.Excessive infiltration of immune cells and serious inflammatory responses are triggers for embryo rejection,which results in miscarriage.Here,we demonstrated that adoptive transfer of MSCs could prevent fetal loss in a lipopolysaccharide(LPS)-induced abortion model and immune response-mediated spontaneous abortion model.The immunosuppressive MSCs alleviated excessive inflammation by inhibiting CD4+T cell proliferation and promoting the decidual macrophage switch to M2 in a tumor necrosis factor-stimulated gene-6(TSG-6)-dependent manner.Cell-tocell contact with proinflammatory macrophages increased the TSG-6 production by the MSCs,thereby enhancing the suppressive regulation of T cells and macrophages.Moreover,proinflammatory macrophages in contact with the MSCs upregulated the expression of CD200 on the stem cells and facilitated the reprogramming of macrophages towards an anti-inflammatory skew through the interaction of CD200 with CD200R on proinflammatory macrophages.Therefore,the results demonstrate that a TSG-6-mediated paracrine effect,reinforced by cell-to-cell contact between MSCs and proinflammatory macrophages,is involved in the mechanism of MSC-mediated abortion relief through the induction of immune tolerance.Our study also indicates the potential application of MSCs in clinical recurrent miscarriages.Yanhong Li Di Zhang Ling Xu Lin Dong Ji Zheng Yikong Lin Jiefang Huang Yanyun Zhang Yu Tao Xingxing Zang Dajin Li Meirong Du 2019Cellular & Molecular Immunology2019,16,12:23
3Doping bioactive elements into a collagen scaffold based on synchronous self-assembly/mineralization for bone tissue engineering显示文摘Pure collagen is biocompatible but lacks inherent osteoinductive,osteoimmunomodulatory and antibacterial activities.To obtain collagen with these characteristics,we developed a novel methodology of doping bioactive elements into collagen through the synchronous self-assembly/mineralization(SSM)of collagen.In the SSM model,amorphous mineral nanoparticles(AMN)(amorphous SrCO3,amorphous Ag3PO4,etc.)stabilized by the polyampholyte,carboxymethyl chitosan(CMC),and collagen molecules were the primary components under acidic conditions.As the pH gradually increased,intrafibrillar mineralization occurred via the self-adaptive interaction between the AMNs and the collagen microfibrils,which were self-assembling;the AMNs wrapped around the microfibrils became situated in the gap zones of collagen and finally transformed into crystals.Srdoped collagen scaffolds(Sr-CS)promoted in vitro cell proliferation and osteogenic differentiation of rat bone marrow mesenchymal stromal cells(rBMSCs)and synergistically improved osteogenesis of rBMSCs by altering the macrophage response.Ag-doped collagen scaffolds(Ag-CS)exhibited in vitro antibacterial effects on S.aureus,as well as cell/tissue compatibility.Moreover,Sr-CS implanted into the calvarial defect of a rat resulted in improved bone regeneration.Therefore,the SSM model is a de novo synthetic strategy for doping bioactive elements into collagen,and can be used to fabricate multifunctional collagen scaffolds to meet the clinical challenges of encouraging osteogenesis,boosting the immune response and fighting severe infection in bone defects.Huanhuan Liu Mingli Lin Xue Liu Ye Zhang Yuyu Luo Yanyun Pang Haitao Chen Dongwang Zhu Xue Zhong Shiqing Ma Yanhong Zhao Qiang Yang Xu Zhang 2020Bioactive Materials2020,5,4:6

8Risk factors for recurrence of colorectal conventional adenoma and serrated polyp显示文摘Background Removal of colorectal polyps during screening could reduce the incidence of colorectal cancer(CRC).However,there is a lack of data on risk factors associated with recurrence of polyps,including conventional adenomas and serrated polyps(SPs).This study aimed to determine risk factors for recurrence of colorectal polyps and their subtypes based on the characteristics of the patients and polyps.Methods A total of 1,165 patients diagnosed with conventional adenoma or SP in the Sixth Affiliated Hospital of Sun Yat-sen University between January 2013 and December 2019 were enrolled in this study,including 668 cases with conventional adenomas,385 with SPs,and 112 with coexistence of adenomas and SPs.Univariate analysis and multivariate logistic regression were used to identify potential risk factors for polyp recurrence.A nomogram was established according to risk factors and the performance was evaluated using calibration plots.Results During a median follow-up of 24 months,recurrent polyps were observed in 531(45.6%)cases.Male,age≥50 years,body mass index(BMI)≥24 kg/m^(2),at least three polyps,smoking,alcohol consumption,family history of polyps,and family history of CRC were independent risk factors for polyp recurrence.The Harrell’s C-index of the nomogram developed with these parameters was 0.69 and the calibration plots showed good agreement between actual polyp recurrence and nomogram-predicted recurrence probability.In the subtype analyses,conventional adenomas had the same risk factors for recurrence as all polyps,while smoking,alcohol consumption,family history of polyps,and family history of CRC were not risk factors for SP recurrence.Conclusions We identified several risk factors for recurrence of colorectal polyps and found that some of them could increase the risk of adenoma recurrence but not SP recurrence,including smoking,alcohol consumption,and family history of polyps/CRC,which might help us to understand different etiology and biology between conventional adenomas and SPs.Zengjie Chi Yanyun Lin Juanni Huang Min-Yi Lv Junguo Chen Xijie Chen Bin Zhang Yufeng Chen Jiancong Hu Xiaosheng He Ping Lan 2022Gastroenterology Report2022,10,1:1

11Wild pink bayberry free phenolic extract induces mitochondria-dependent apoptosis and G0/G1 cell cycle arrest through p38/MAPK and PI3K/Akt pathway in MDA-MB-231 cancer cells显示文摘Polyphenol-rich foods have been shown to be good for cancer prevention as powerful antioxidants. In this study, the mechanisms of wild pink bayberry free phenolic extract(WPBFE)inhibiting the proliferation and inducing apoptotic of MDA-MB-231 breast cancer cells was examined. The main phenolic acids and flavonols in WPBFE were gallic acid((18.83 ± 0.44)μg/g FW)and myricetin((1.52 ± 0.05)μg/g FW), respectively. The maximum inhibition rate of WPBFE at non-cytotoxicity dose(below 80 mg/mL)was 81%. Western blotting analysis showed that WPBFE could cause the arrest of cell cycle in G0/G1 phase by down-regulating expression levels of PCNA, CDK4, cyclin D1 and up-regulating the expression level of p21. Meanwhile, WPBFE induced apoptosis through initiating the mitochondrial death pathway by up-regulating cleaved caspase-3 and enhancing the ratio of Bax/Bcl-2, with the maximum expression levels of 1.29 and 2.03 folds that of control group, respectively. Further study of the upstream protein, we found that WPBFE down-regulated TRAF2, while upregulated p-ASK1, p-p38 and p-p53. Furthermore, WPBFE could down-regulate the expression of p-PI3K and p-Akt. 13QTL Mapping for Plant Height of Wheat under Different Irrigation Modes显示文摘In order to mine gene loci associated with drought resistance and water use efficiency,and provide guidance for the breeding of drought-resistant wheat varieties,quantitative trait loci(QTLs)for plant height(PH)and stem height(SH)of wheat were evaluated using a population of 302 recombinant inbred lines(RILs)derived from a cross between drought-tolerant cultivar Luohan 2 and water-sensitive cultivar Weimai 8 under different water regimes in six environments.Totally 17 additive QTLs for PH and SH were identified and distributed on chromosomes 2B,3A,3B,4A,5A,5D,6B,7A and 7B,respectively,which explained3.3%to 26.95%phenotypic variations,with additive effect of 1.919 6 to 5.382 8 cm.Among them,14 QTLs,24 times and 5 QTLs,7 times were detected in three irrigation environments and three drought stress environments,respectively.Both the number of QTLs for PH and the times detected in drought stress environments were significantly less than those in irrigation environments.Of all the 17 QTLs for PH,12 were detected only in irrigation environment,three QTLs were detected only in drought stress environment,and two QTLs were detected in both environments,suggesting that the expression of the genes is greatly influenced by environmental condition.QPh-WL-7A.3 detected between maker Xbarc049 and Xgwm273 on chromosome 7A in all the three drought stress environments,can enhance2.481 5-3.697 2 cm of PH,explain 8.6%-10.0%additive effect,so it is useful for the genetic improvement of drought tolerance and molecular marker-assisted selection in wheat.Yanyun REN Ling HUANG Minmin SHAO Leiming SUN Kai ZHAO Jinping WANG Xingke XU Weiying FENG Jifeng WANG Lu YAN Shichong WANG Lin WANG 2017Agricultural Biotechnology2017,6,4:0
14Numerical studies on the radiation uniformity of Z-pinch dynamic hohlraum显示文摘Radiation uniformity is important for Z-pinch dynamic hohlraum driven fusion. In order to understand the radiation uniformity of Z-pinchdynamic hohlraum, the code MULTI-2D with a new developed magnetic field package is employed to investigate the related physical processeson Julong-I facility with drive current about 7e8 MA. Numerical simulations suggest that Z-pinch dynamic hohlraum with radiation temperaturemore than 100 eV can be created on Julong-I facility. Although some X-rays can escape out of the hohlraum from Z-pinch plasma and electrodes, the radiation field near the foam center is quite uniform after a transition time. For the load parameters used in this paper, the transitiontime for the thermal wave transports from r = 1 mm to r = 0 mm is about 2.0 ns. Implosion of a testing pellet driven by cylindrical dynamichohlraum shows that symmetrical implosion is hard to achieve due to the relatively slow propagation speed of thermal wave and the compressionof cylindrical shock in the foam. 15E3 ligase MG53 suppresses tumor growth by degrading cyclin D1显示文摘Due to the essential role of cyclin D1 in regulating transition from G1 to S phase in cell cycle,aberrant cyclin D1 expression is a major oncogenic event in many types of cancers.In particular,the dysregulation of ubiquitination-dependent degradation of cyclin D1 contributes to not only the pathogenesis of malignancies but also the refractory to cancer treatment regiments with CDK4/6 inhibitors.Here we show that in colorectal and gastric cancer patients,MG53 is downregulated in more than 80%of tumors compared to the normal gastrointestinal tissues from the same patient,and the reduced MG53 expression is correlated with increased cyclin D1 abundance and inferior survival.Mechanistically,MG53 catalyzes the K48-linked ubiquitination and subsequent degradation of cyclin D1.Thus,increased expression of MG53 leads to cell cycle arrest at G1,and thereby markedly suppresses cancer cell proliferation in vitro as well as tumor growth in mice with xenograft tumors or AOM/DSS induced-colorectal cancer.Consistently,MG53 deficiency results in accumulation of cyclin D1 protein and accelerates cancer cell growth both in culture and in animal models.These findings define MG53 as a tumor suppressor via facilitating cyclin D1 degradation,highlighting the therapeutic potential of targeting MG53 in treating cancers with dysregulated cyclin D1 turnover.Meng Fang Hong-Kun Wu Yumeng Pei Yan Zhang Xiangyu Gao Yanyun He Gengjia Chen Fengxiang Lv Peng Jiang Yumei Li Wenwen Li Peng Jiang Lin Wang Jiafu Ji Xinli Hu Rui-Ping Xiao 2023Signal Transduction and Targeted Therapy2023,8,8:0
