1Predictive value of immune cell counts and neutrophil-to-lymphocyte ratio for 28-day mortality in patients with sepsis caused by intra-abdominal infection显示文摘Background:The current study aimed to evaluate the value of immune cell counts and neutrophilto-lymphocyte ratio(NLR)when attempting to predict 28-day mortality.Methods:We conducted an observational retrospective study that included consecutive septic patients.Severity scores on the first day and peripheral circulating immune cell counts(at day 1,day 3,day 5 and day 7 of admission)were collected during each patient’s emergency intensive care unit stay.We assessed the associations of peripheral circulating immune cell counts and NLR with the severity of illness.The relationships between 28-day mortality and peripheral circulating immune cell counts and NLR with were evaluated using Cox proportional cause-specific hazards models.Results:A total of 216 patients diagnosed with sepsis caused by IAI were enrolled.The lymphocyte counts(days 1,3,5 and 7)and monocyte counts(days 3,5 and 7)were significantly lower in nonsurvivors(n=72)than survivors(n=144).The NLR values at each time point were significantly higher in non-survivors.The day 1 lymphocyte counts,as well as the monocyte counts,were significantly lower in the highest-scoring group,when stratified by the Acute Physiology and Chronic Health Evaluation II and Sequential Organ Failure Assessment scores,than in the other groups(p<0.05).The day 1 NLR was significantly higher in the highest-scoring group than in the other groups(p<0.05).The day 5 and day 7 lymphocyte counts,day 3 and day 7 monocyte counts and day 7 NLR were significant predictors of 28-day mortality in the Cox proportional hazards models(day 5 lymphocyte count:hazard ratio,0.123(95%CI,0.055–0.279),p<0.001;day 7 lymphocyte count:hazard ratio,0.115(95%CI,0.052–0.254),p<0.001;day 3 monocyte count:hazard ratio,0.067(95%CI,0.005–0.861),p=0.038;day 7 monocyte count:hazard ratio,0.015(95%CI,0.001–0.158),p<0.001;day 7 NLR:hazard ratio,0.773(95%CI,0.659–0.905),p=0.001).Conclusions The results showed that circulating lymphocytes and monocytes were dramatically decreased within 7 days in non-survivors following sepsis from an IAI.Lymphocyte counts,monocyte counts and NLR appeared to be associated with the severity of illness,and they may serve as independent predictors of 28-day mortality in septic patients with IAIs.Shuangqing Liu Yuxuan Li Fei She Xiaodong Zhao Yongming Yao 2021Burns & Trauma2021,9,1:6

3High mobility group box 1 protein (HMGB1) as an immune-modulating factor for polarization of human T lymphocytes显示文摘Objective This study was performed to investigate the effect of high mobility group box-1 protein (HMGB1) on immune function of human T lymphocytes in vitro and explore its potential role in cell-mediated immune dysfunction. Methods Fresh blood was obtained from healthy adult volunteers and peripheral blood mononuclear cells (PBMCs) were isolated, then rhHMGB1 was added to PBMCs. Four-color flow cytometric (FCM) analysis was used for the measurement of intracellular cytokine including interleukin IL-4 and interferon IFN- ? ELISA kits were employed for the determination of IL-2 and sIL-2R protein levels in cell culture supernatants. Results (1) Different stimulating time and dosage of rhHMGB1 did not alter the number of IFN- αpositive cells (Th1). rhHMGB1 stimulation provoked a dose-dependent and time-dependent increase in Th2 subset and decrease in ratio of Th1 to Th2. (2) Compared with the untreated cells, when the cells were coincubated with rhHMGB1 (10-100ng/ml) for 12 hrs, protein release of IL-2 and sIL-2R were significantly up-regulated. At 48 hrs, in contrast, protein production was relatively lower in cells after exposure to 100-1000 ng/ ml rhHMGB1. 4Gas sensing capabilities of TiO_(2) porous nanoceramics prepared through premature sintering显示文摘Pure and noble metal(Pt,Pd,and Au)doped TiO_(2)nanoceramics have been prepared from TiO_(2)nanoparticles through traditional pressing and sintering.For those samples sintered at 550℃,a typical premature sintering occurred,which led to the formation of a highly porous microstructure with a Brunauer-Emmett-Teller(BET)specific surface area of 23 m^(2)/g.At room temperature,only Pt-doped samples showed obvious response to hydrogen,with sensitivities as high as~500 for 1000 ppm H_(2)in N_(2);at 300℃,all samples showed obvious responses to CO,while the responses of noble metal doped samples were much higher than that of the undoped ones.The mechanism for the observed sensing capabilities has been discussed,in which the catalytic effect of Pt for hydrogen is believed responsible for the room-temperature hydrogen sensing capabilities,and the absence of glass frit as commonly used in commercial thick-film metal oxide gas sensors is related to the high sensitivities.It is proposed that much attention should be paid to metal oxide porous nanoceramics in developing gas sensors with high sensitivities and low working temperatures.Yao XIONG Zilong TANG Yu WANG Yongming HU Haoshuang GU Yuanzhi LI Helen Lai Wah CHAN Wanping CHEN 2015Journal of Advanced Ceramics2015,4,2:3

6Combination therapy of thiamine,vitamin C and hydrocortisone in treating patients with sepsis and septic shock:a meta-analysis and trial sequential analysis显示文摘Background:The objective of this study was to evaluate the clinical efficacy of thiamine and vitamin C with or without hydrocortisone coadministration on the treatment of sepsis and septic shock.Methods:MEDLINE,EMBASE and CENTRAL databases were searched for randomized controlled trials(RCTs)that made a comparative study between the combination therapy of vitamin C and thiamine with or without hydrocortisone and the administration of placebo in patients with sepsis or septic shock.Two reviewers independently performed study selection,data extraction and quality assessment.Both short-term mortality and change in the sequential organ failure assessment(SOFA)score from baseline(delta SOFA)were set as the primary outcomes.Secondary endpoints included intensive care unit(ICU)mortality,new onset of acute kidney injury,total adverse events,ICU and hospital length of stay,duration of vasopressor usage and ventilator-free days.Meanwhile,trial sequential analysis was conducted for primary outcomes.Results:Eight RCTs with 1428 patients were included in the current study.The results showed no significant reduction of short-term mortality in sepsis and septic shock patients who received combination therapy of vitamin C and thiamine with or without hydrocortisone compared to those with placebo{risk ratio(RR),1.02[95%confidence interval(CI),0.87 to 1.20],p=0.81,I2=0%;risk difference(RD),0[95%CI,−0.04 to 0.05]}.Nevertheless,the combination therapy was associated with significant reduction in SOFA score[mean difference(MD),−0.63,(95%CI,−0.96 to−0.29,p<0.001,I^(2)=0%]and vasopressors duration(MD,−22.11[95%CI,−30.46 to−13.77],p<0.001,I2=6%).Additionally,there were no statistical differences in the pooled estimate for other outcomes.Conclusions:In the current meta-analysis,the combination therapy of vitamin C and thiamine,with or without hydrocortisone had no impact on short-term mortality when compared with placebo,but was associated with significant reduction in SOFA score among patients with sepsis and septic shock.Renqi Yao Yibing Zhu Yue Yu Zhixuan Li Lixue Wang Liyu Zheng Jingyan Li Huibin Huang Guosheng Wu Feng Zhu Zhaofan Xia Chao Ren Yongming Yao 2021Burns & Trauma2021,9,1:2

8A distributed decision method for missiles autonomous formation based on potential game显示文摘The distributed cooperative decision problems of missiles autonomous formation with network packet loss are investigated by using the potential game based on formation principles.In particular,a dynamic target allocation method for missiles formation is provided based on the potential game and formation principles,after the introduction of cooperative guidance and control system of the missiles formation.Then we seek the optimization of a global utility function through autonomous missiles that are capable of making individually rational decisions to optimize their own utility functions.The first important aspect of the problem is to design an individual utility function considering the characteristics of the missiles formation,with which the objective of the missiles are localized to each missile yet aligned with the global utility function.The second is to equip the missiles with an appropriate coordination mechanism with each missile pursuing the optimization of its own utility function.We present the design procedure for the utility,and present a coordination mechanism based on spatial adaptive play and then introduce the idea of“cyclical selected spatial adaptive play”and“negotiation based on time division multiple address(TDMA)protocol formation support network”.Finally,we present simulations for the distributed dynamic target allocation on the comprehensive digital simulation system,and the results illustrate the effectiveness and engineering applicability of the method.JIA Xiang WU Sentang WEN Yongming YAO Zheng 2019Journal of Systems Engineering and Electronics2019,30,4:2
9A Vaccine Based on the Receptor-Binding Domain of the Spike Protein Expressed in Glycoengineered Pichia pastoris Targeting SARS-CoV-2 Stimulates Neutralizing and Protective Antibody Responses显示文摘In 2020 and 2021,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),a novel coronavirus,caused a global pandemic.Vaccines are expected to reduce the pressure of prevention and control,and have become the most effective strategy to solve the pandemic crisis.SARS-CoV-2 infects the host by binding to the cellular receptor angiotensin converting enzyme 2(ACE2)via the receptor-binding domain(RBD)of the surface spike(S)glycoprotein.In this study,a candidate vaccine based on a RBD recombinant subunit was prepared by means of a novel glycoengineered yeast Pichia pastoris expression system with characteristics of glycosylation modification similar to those of mammalian cells.The candidate vaccine effectively stimulated mice to produce high-titer anti-RBD specific antibody.Furthermore,the specific antibody titer and virus-neutralizing antibody(NAb)titer induced by the vaccine were increased significantly by the combination of the double adjuvants Al(OH)_(3) and CpG.Our results showed that the virus-NAb lasted for more than six months in mice.To summarize,we have obtained a SARS-CoV-2 vaccine based on the RBD of the S glycoprotein expressed in glycoengineered Pichia pastoris,which stimulates neutralizing and protective antibody responses.A technical route for fucose-free complex-type N-glycosylation modified recombinant subunit vaccine preparation has been established.Bo Liu Ying Yin Yuxiao Liu Tiantian Wang Peng Sun Yangqin Ou Xin Gong Xuchen Hou Jun Zhang Hongguang Ren Shiqiang Luo Qian Ke Yongming Yao Junjie Xu Jun Wu 2022Engineering2022,8,6:1
10A wheat integrative regulatory network from large-scale complementary functional datasets enables trait-associated gene discovery for crop improvement显示文摘Gene regulation is central to all aspects of organism growth,and understanding it using large-scale functional datasets can provide a whole view of biological processes controlling complex phenotypic traits in crops.However,the connection between massive functional datasets and trait-associated gene discovery for crop improvement is still lacking.In this study,we constructed a wheat integrative gene regulatory network(wGRN)by combining an updated genome annotation and diverse complementary functional datasets,including gene expression,sequence motif,transcription factor(TF)binding,chromatin accessibility,and evolutionarily conserved regulation.wGRN contains 7.2 million genome-wide interactions covering 5947 TFs and 127439 target genes,which were further verified using known regulatory relationships,condition-specific expression,gene functional information,and experiments.We used wGRN to assign genome-wide genes to 3891 specific biological pathways and accurately prioritize candidate genes associated with complex phenotypic traits in genome-wide association studies.In addition,wGRN was used to enhance the interpretation of a spike temporal transcriptome dataset to construct high-resolution networks.We further unveiled novel regulators that enhance the power of spike phenotypic trait prediction using machine learning and contribute to the spike phenotypic differences among modern wheat accessions.Finally,we developed an interactive webserver,wGRN(http://gffzzaeec8df70e7c4b21hb5kn6uuqkqcw6v9q.ffgz.tsg.suse.edu.cn/wGRN),for the community to explore gene regulation and discover trait-associated genes.Collectively,this community resource establishes the foundation for using large-scale functional datasets to guide trait-associated gene discovery for crop improvement.Yongming Chen Yiwen Guo Panfeng Guan Yongfa Wang Xiaobo Wang Zihao Wang Zhen Qin Shengwei Ma Mingming Xin Zhaorong Hu Yingyin Yao Zhongfu Ni Qixin Sun Weilong Guo Huiru Peng 2023Molecular Plant2023,16,2:1

14Non-pharmaceutical interventions for COVID-19 reduced the incidence of infectious diseases:a controlled interrupted time-series study显示文摘Background Non-pharmaceutical interventions(NPIs)have been implemented worldwide to suppress the spread of coronavirus disease 2019(COVID-19).However,few studies have evaluated the effect of NPIs on other infectious diseases and none has assessed the avoided disease burden associated with NPIs.We aimed to assess the effect of NPIs on the incidence of infectious diseases during the COVID-19 pandemic in 2020 and evaluate the health economic benefits related to the reduction in the incidence of infectious diseases.Methods Data on 10 notifiable infectious diseases across China during 2010–2020 were extracted from the China Information System for Disease Control and Prevention.A two-stage controlled interrupted time-series design with a quasi-Poisson regression model was used to examine the impact of NPIs on the incidence of infectious diseases.The analysis was first performed at the provincial-level administrative divisions(PLADs)level in China,then the PLAD-specific estimates were pooled using a random-effect meta-analysis.Results A total of 61,393,737 cases of 10 infectious diseases were identified.The implementation of NPIs was associated with 5.13 million(95%confidence interval[CI]3.45‒7.42)avoided cases and USD 1.77 billion(95%CI 1.18‒2.57)avoided hospital expenditures in 2020.There were 4.52 million(95%CI 3.00‒6.63)avoided cases for children and adolescents,corresponding to 88.2%of total avoided cases.The top leading cause of avoided burden attributable to NPIs was influenza[avoided percentage(AP):89.3%;95%CI 84.5‒92.6].Socioeconomic status and population density were effect modifiers.Conclusions NPIs for COVID-19 could effectively control the prevalence of infectious diseases,with patterns of risk varying by socioeconomic status.These findings have important implications for informing targeted strategies to prevent infectious diseases.Wenyi Zhang Yao Wu Bo Wen Yongming Zhang Yong Wang Wenwu Yin Shanhua Sun Xianyu Wei Hailong Sun Zhijie Zhang Shanshan Li Yuming Guo 2023Infectious Diseases of Poverty2023,12,2:1

16A highly effective polycyclic aromatic hydrocarbon-degrading bacterium,Paracoccus sp.HPD-2,shows opposite remediation potential in two soil types显示文摘Bioaugmentation is an efficient and eco-friendly strategy for the bioremediation of polycyclic aromatic hydrocarbons(PAHs).Since the degrading abilities of soils can greatly alter the abilities of PAH-degrading bacteria,illustrating the potential and mechanism of highly efficient degrading bacteria in different soil environments is of great importance for bioremediation.A PAH-degrading bacterium,Paracoccus aminovorans HPD-2,and two soil types,red and paddy soils,with distinct PAH-degrading abilities,were selected for this study.A soil microcosm experiment was performed by adding pyrene(PYR)and benzo[a]pyrene(B[a]P).Illumina sequencing was used to examine bacterial community structure.The results showed that inoculation with HPD-2 significantly elevated PYR and B[a]P degradation rates by 44.7%and 30.7%,respectively,in the red soil,while it only improved the degradation rates by 1.9%and 11%,respectively,in the paddy soil.To investigate the underlying mechanism,the fate of strain HPD-2 and the response of the indigenous bacterial communities were determined.Strain HPD-2 occupied certain niches in both soils,and the addition of the bacterium changed the native community structure more noticeably in the red soil than in the paddy soil.The addition of PAHs and strain HPD-2 significantly changed the abundances of 7 phyla among the 15 detected phyla in the red soil.In the paddy soil,5 of the 12 dominant phyla were significantly affected by PAHs and the inoculation of HPD-2,while 6new phyla were detected in the low-abundance phyla(<0.1%).The abundances of Massilia,Burkholderia,and Rhodococcus genera with PAH degradation efficiency were significantly increased by the inoculation of HPD-2 in the red soil during 42 d of incubation.Meanwhile,in the paddy soil,the most dominant effective genus,Massilia,was reduced by HPD-2 inoculation.This research revealed the remediation ability and inherent mechanism of the highly effective PAH-degrading strain HPD-2 in two different soil types,which would provide a theoretical basis for the application of degrading bacteria in different soils.Wei CHEN Ying TENG Wenjie REN Yongming LUO Yao YU 2022Pedosphere2022,32,5:1
