| 1 | Identification of a novel coronavirus causing severe pneumonia in human:a descriptive study显示文摘Background:Human infections with zoonotic coronaviruses(CoVs),including severe acute respiratory syndrome(SARS)-CoV and Middle East respiratory syndrome(MERS)-CoV,have raised great public health concern globally.Here,we report a novel batorigin CoV causing severe and fatal pneumonia in humans.Methods:We collected clinical data and bronchoalveolar lavage(BAL)specimens from five patients with severe pneumonia from Wuhan Jinyintan Hospital,Hubei province,China.Nucleic acids of the BAL were extracted and subjected to next-generation sequencing.Virus isolation was carried out,and maximum-likelihood phylogenetic trees were constructed.Results:Five patients hospitalized from December 18 to December 29,2019 presented with fever,cough,and dyspnea accompanied by complications of acute respiratory distress syndrome.Chest radiography revealed diffuse opacities and consolidation.One of these patients died.Sequence results revealed the presence of a previously unknownβ-CoV strain in all five patients,with 99.8%to 99.9%nucleotide identities among the isolates.These isolates showed 79.0%nucleotide identity with the sequence of SARS-CoV(GenBank NC_004718)and 51.8%identity with the sequence of MERS-CoV(GenBank NC_019843).The virus is phylogenetically closest to a bat SARS-like CoV(SL-ZC45,GenBank MG772933)with 87.6%to 87.7%nucleotide identity,but is in a separate clade.Moreover,these viruses have a single intact open reading frame gene 8,as a further indicator of bat-origin CoVs.However,the amino acid sequence of the tentative receptor-binding domain resembles that of SARS-CoV,indicating that these viruses might use the same receptor.Conclusion:A novel bat-borne CoV was identified that is associated with severe and fatal respiratory disease in humans. | Li-Li Ren Ye-Ming Wang Zhi-Qiang Wu Zi-Chun Xiang Li Guo Teng Xu Yong-Zhong Jiang Yan Xiong Yong-Jun Li Xing-Wang Li Hui Li Guo-Hui Fan Xiao-Ying Gu Yan Xiao Hong Gao Jiu-Yang Xu Fan Yang Xin-Ming Wang Chao Wu Lan Chen Yi-Wei Liu Bo Liu Jian Yang Xiao-Rui Wang Jie Dong Li Li Chao-Lin Huang Jian-Ping Zhao Yi Hu Zhen-Shun Cheng Un-Lin Liu Zhao-Hui Qian Chuan Qin Qi Jin Bin Cao Jian-Wei Wang | 2020 | Chinese Medical Journal2020,,9: | 99 |

| 5 | BMAL1 regulates mitochondrial fission and mitophagy through mitochondrial protein BNIP3 and is critical in the development of dilated cardiomyopathy显示文摘Dysregulation of circadian rhythms associates with cardiovascular disorders.It is known that deletion of the core circadian gene Bma/1 in mice causes dilated car-diomyopathy.However,the biological rhythm regulation system in mouse is very different from that of humans.Whether BMAL1 plays a role in regulating human heart function remains unclear.Here we generated a BMAL1 knockout human embryonic stem cell(hESC)model and further derived human BMAL1 deficient cardiomy-ocytes.We show that BMAL1 deficient hESC-derived cardiomyocytes exhibited typical phenotypes of dilated cardiomyopathy including attenuated contractility,cal-cium dysregulation,and disorganized myofilaments.In addition,mitochondrial fission and mitophagy were suppressed in BMAL1 deficient hESC-cardiomyocytes,which resulted in significantly attenuated mitochondrial oxidative phosphorylation and compromised cardiomy-ocyte function.We also found that BMAL1 binds to the E-box element in the promoter region of BNIP3 gene and specifically controls BNIP3 protein expression.BMAL1 knockout directly reduced BNIP3 protein level,causing compromised mitophagy and mitochondria dysfunction and thereby leading to compromised cardiomyocyte function.Our data indicated that the core circadian gene S/VMLf is critical for normal mitochondria activities and cardiac function.Circadian rhythm disruption may directly link to compromised heart function and dilated cardiomyopathy in humans. | Ermin Li Xiuya Li Jie Huang Chen Xu Qianqian Liang Kehan Ren Aobing Bai Chao Lu Ruizhe Qian Ning Sun | 2020 | Protein & Cell2020,11,9: | 17 |
| 6 | Clinical profiles and outcomes of acute type A aortic dissection and intramural hematoma in the current era: lessons from the first registry of aortic dissection in China显示文摘Background::Acute type A aortic dissection(ATAAD)and acute type A intramural hematoma(ATAIMH)are life-threatening diseases with high mortality.To better understand their clinical features in the Chinese population,we analyzed the data from the first Registry of Aortic Dissection in China(Sino-RAD)to promote the understanding and management of the diseases.Methods::All patients with ATAAD and ATAIMH enrolled in Sino-RAD from January 1,2012 to December 31,2016 were involved.The data of patients’selection,history,symptoms,management,outcomes,and postoperation complications were analyzed in the study.The continuous variables were compared using the Student’s t test for normal distributions and the Mann-Whitney U test for non-normal distributions.Categorical variables were compared using the Chi-square test or Fisher exact test.Results::A total of 1582 patients with ATAAD and 130 patients with ATAIMH were included.The mean age of all patients was 48.4 years.Patients with ATAAD were significantly younger than patients with ATAIMH(48.9 years vs.55.6 years,P<0.001).For the total cohort,males were dominant,but the male ratio of patients with ATAAD was significantly higher compared to those with ATAIMH(P=0.01).The time range from the onset of symptom to hospitalization was 2.0 days.More patients of ATAIMH had hypertension than that of ATAAD(82.3%vs.67.6%,P<0.05).Chest and back pain were the most common clinical symptoms.Computerized tomography(CT)was the most common initial diagnostic imaging modality.84.7%received surgical treatment and in-hospital mortality was 5.3%.Patients with ATAAD mainly received surgical treatment(89.6%),while most patients with ATAIMH received medical treatment(39.2%)or endovascular repair(35.4%).Conclusions::Our study suggests that doctors should comprehensively use clinical examination and genetic background screening for patients with ATAAD and ATAIMH and further shorten the time range from symptoms onset to intervention,achieving early diagnosis and treatment,thereby reducing the mortality rate of patients with aortic dissection in China.We should standardize the procedures of aortic dissection treatment and improve people’s understanding.Meanwhile,the curing and transferring efficiency should also be improved. | Wei-Xun Duan Wei-Guang Wang Lin Xia Chao Xue Bo Yu Kai Ren Wei Yi Hong-Liang Liang Xiao-Chao Dong Jian Zuo Jin-Cheng Liu Shi-Qiang Yu Ding-Hua Yi | 2021 | Chinese Medical Journal2021,,8: | 15 |

| 9 | The electrochemical hydrogen storage performances of Si-added La–Mg–Ni–Co-based A_2B_7-type electrode alloys显示文摘In order to improve the electrochemical cycle stability of the RE–Mg–Ni-based A2B7-type electrode alloys, a small amount of Si has been added into the alloys.The casting and annealing technologies were adopted to fabricate the La0.8Mg0.2Ni3.3Co0.2Six(x = 0–0.2) electrode alloys. The impacts of the addition of Si and annealing treatment on the structures and electrochemical performances of the alloys were investigated systematically. The results obtained by XRD and SEM show that all the as-cast and annealed alloys are of a multiphase structure, involving two main phases(La, Mg)2Ni7and La Ni5 as well as a residual phase La Ni3. Both adding Si and the annealing treatment lead to an evident change in the phase abundance and cell parameters of(La, Mg)2Ni7and La Ni5 major phases of the alloy without altering its main phase component. Moreover, the annealing treatment has the composition of the alloy distributed more homogeneously overall and simultaneously causes the grain of the alloy to be coarsened obviously. The electrochemical measurements indicate that adding Si and the annealing treatment give a significant rise to the influence on the electrochemical performances of the alloys. In brief, the cycle stability of the as-cast and annealed alloys evidently increases with the rising of Si content, while their discharge capacities obviously decrease under the same circumstances. Furthermore, the electrochemical kineticproperties of the electrode alloys, including the high rate discharge ability, the limiting current density(IL), hydrogen diffusion coefficient(D), and the charge-transfer resistance, first augment and then decline with the rising of Si content. | 11 | Comparative analysis on grain quality and yield of different panicle weight indica-japonica hybrid rice(Oryza sativa L.) cultivars显示文摘Indica-japonica hybrid rice(Oryza sativa L.) cultivars showed high yield potential and poor tasting quality when compared with common japonica rice cultivars. Large panicle is a prominent factor of high yield for indica-japonica hybrid rice cultivars, and the panicle weight varies greatly among different indica-japonica hybrid rice cultivars. It is important to research on yield and grain quality of different panicle weight indica-japonica hybrid rice cultivars. In this study, two different panicle types indica-japonica hybrid cultivars were used to research on the relation of yield and grain quality. The yields of two heavy panicle weights indica-japonica hybrid cultivars were significantly higher than that of two medium panicle weight rice cultivars. The cooking and eating quality and starch properties of different panicle type cultivars were evaluated. Yongyou 6715(medium panicle) and Yongyou 1852(heavy panicle) got the relatively higher cooking and eating quality. Rice cultivars with medium panicle weight had more large starch granules and higher relative crystallinity than cultivars with heavy panicle weight. Transition temperature and retrogradation enthalpy(ΔHret) of medium panicle type cultivars were significantly higher than that of heavy panicle type cultivars. There was no significant difference in amylose content among different panicle type cultivars. Protein content of heavy panicle type cultivar was higher than that of medium panicle type cultivar, and protein content is the main factor affect cooking and eating quality in this study. The cultivar Yongyou 6715 got the highest taste value with the lowest protein content. Thus, it is suggested that the emphasis on improving rice cooking and eating quality of indica-japonica hybrid rice cultivars is how to reduce the protein content in rice grain. | 12 | Characterising the nature, evolution and origin of detachment fault in central depression belt, Qiongdongnan Basin of South China Sea: evidence from seismic reflection data显示文摘Using regional geological, newly acquired 2D and 3D seismic, drilling and well log data, especially 2D long cable seismic profiles, the structure and stratigraphy in the deep-water area of Qiongdongnan Basin are interpreted. The geometry of No.2 fault system is also re-defined, which is an important fault in the central depression belt of the deep-water area in the Qiongdongnan Basin by employing the quantitative analysis techniques of fault activity and backstripping. Furthermore, the dynamical evolution of the No.2 fault system and its controls on the central depression belt are analyzed. This study indicates that the Qiongdongnan Basin was strongly influenced by the NW-trending tensile stress field during the Late Eocene. At this time, No.2 fault system initiated and was characterized by several discontinuous fault segments, which controlled a series small NE-trending fault basins. During the Oligocene, the regional extensional stress field changed from NW-SE to SN with the oceanic spreading of South China Sea, the early small faults started to grow along their strikes, eventually connected and merged as the listric shape of the No.2 fault system as observed today. No.2 fault detaches along the crustal Moho surface in the deep domain of the seismic profiles as a large-scale detachment fault. A large-scale rollover anticline formed in hanging wall of the detachment fault. There are a series of small fault basins in both limbs of the rollover anticline, showing that the early small basins were involved into fold deformation of the rollover anticline. Structurally, from west to east, the central depression belt is characterized by alternatively arranged graben and half-graben. The central depression belt of the Qiongdongnan Basin lies at the extension zone of the tip of the V-shaped northwestern ocean sub-basin of the South China Sea, its activity period is the same as the development period of the northwestern ocean sub-basin, furthermore the emplacement and eruption of magma that originated from the mantle below the Moho surface occurred at the region between Songnan-Baodao and Changchang sags, from east to west with the early-stage spreading of the South China Sea. | 13 | Spatial and Temporal Evolution of Surface Subsidence in Tianjin from 2015 to 2020 Based on SBAS-InSAR Technology显示文摘Tianjin is one of the inland cities with the most severe cases of subsidence hazard in China.The majority of the existing studies have mainly focused on Beijing-Tianjin-Hebei,and little attention has been given to Tianjin.In addition,these existing studies are short-term investigations,lacking long-term monitoring of surface subsidence.In the present study,we use the Small Baseline Subset Interferometric Synthetic Aperture Radar(SBAS-InSAR)technique to monitor the subsidence process in Tianjin between 2015 and 2020 and reveal its spatial and temporal variation.We divided the 44-view Sentinel-1A image data into three periods to avoid the effect of temporal and spatial decoherence by extracting the surface deformation field in Tianjin.We finally verified the accuracy and reliability of the inversion results using second-order leveling data.Results showed that the correlation coefficient r between the two reached 0.89,and the root mean square error was 4.84 mm/y.Obvious subsidence funnels exist in Tianjin,mainly in the towns of Wangqingtuo and Shengfang.These subsidence funnels have a subsidence deformation rate of-136.2 mm/y and a maximum cumulative settlement of-346.3 mm within the study period.The subsidence area tends to extend to the southwest.The analysis of annual rainfall,groundwater resource extraction,spatial location distribution of industrial areas combined with SBAS-InSAR inversion results indicates that overextraction of groundwater resources is the main cause of land subsidence in the area.Therefore,strict control of groundwater extraction is the main approach to mitigate land subsidence effectively. | Lyu ZHOU Yizhan ZHAO Zilin ZHU Chao REN Fei YANG Ling HUANG Xin LI | 2022 | Journal of Geodesy and Geoinformation Science2022,5,1: | 8 |
| 14 | One Stone Four Birds:A Novel Liposomal Delivery System Multi-functionalized with Ginsenoside Rh2 for Tumor Targeting Therapy显示文摘Liposomes hold great potential in anti-cancer drug delivery and the targeting treatment of tumors.However,the clinical therapeutic efficacy of liposomes is still limited by the complexity of tumor microenvironment(TME)and the insufficient accumulation in tumor sites.Meanwhile,the application of cholesterol and polyethylene glycol(PEG),which are usually used to prolong the blood circulation and stabilize the structure of liposomes respectively,has been questioned due to various disadvantages.Herein,we developed a ginsenoside Rh2-based multifunctional liposome system(Rh2-lipo)to effectively address these challenges once for all.Different with the conventional’wooden’liposomes,Rh2-lipo is a much more brilliant carrier with multiple functions.In Rh2-lipo,both cholesterol and PEG were substituted by Rh2,which works as membrane stabilizer,long-circulating stealther,active targeting ligand,and chemotherapy adjuvant at the same time.Firstly,Rh2 could keep the stability of liposomes and avoid the shortcomings caused by cholesterol.Secondly,Rh2-lipo showed a specifically prolonged circulation behavior in the blood.Thirdly,the accumulation of the liposomes in the tumor was significantly enhanced by the interaction of glucose transporter of tumor cells with Rh2.Fourth,Rh2-lipo could remodel the structure and reverse the immunosuppressive environment in TME.When tested in a 4T1 breast carcinoma xenograft model,the paclitaxel-loaded Rh2-lipo realized high efficient tumor growth suppression.Therefore,Rh2-lipo not only innovatively challenges the position of cholesterol as a liposome component,but also provides another innovative potential system with multiple functions for anti-cancer drug delivery. | Chao Hong Jianming Liang Jiaxuan Xia Ying Zhu Yizhen Guo Anni Wang Chunyi Lu Hongwei Ren Chen Chen Shiyi Li Dan Wang Huaxing Zhan Jianxin Wang | 2020 | Nano-Micro Letters2020,12,10: | 8 |

| 17 | Risk factors for gastric cancer:a large-scale,population-based case-control study显示文摘Background:Early detection of gastric cancer(GC)has been the topic of major efforts in China.This study aimed to explore the risk factors associated with GC and to provide evidence for the selection of a high-risk population of GC.Methods:Based on the cancer screening cohort of the National Cancer Screening Program in Urban China,GC patients diagnosed by endoscopy and pathological examinations constituted the case group,and controls were 1:3 matched by sex and age(±5 years)individually.The variables were selected by univariable analysis of factors such as body mass index(BMI),dietary habits,lifestyle,stomach disease history,and family history of GC;and multivariable logistic regression was used to analyze the influencing factors of GC and to calculate the odds ratio(OR)of related factors and its 95%confidence interval(CI).Results:A total of 215 GC cases and 645 matched healthy controls were included in the final analysis,with a median age of 61 years for the case and control groups.Overall analysis showed that high educational level(above primary school)(OR=0.362,95%CI=0.219–0.599,P<0.001),overweight/obesity(BMI≥24 kg/m2;OR=0.489,95%CI=0.329–0.726,P<0.001),cigarette smoking(OR=3.069,95%CI=1.700–5.540,P<0.001),alcohol consumption(OR=1.661,95%CI=1.028–2.683,P=0.038),history of stomach disease(OR=6.917,95%CI=4.594–10.416,P<0.001),and family history of GC in first-degree relatives(OR=4.291,95%CI=1.661–11.084,P=0.003)were significantly correlated with the occurrence of GC.Subgroup analyses by age and gender indicated that GC risk was still increased in the presence of a history of stomach disease.A history of chronic gastritis,gastric ulcer,or gastric polyposis was positively associated with GC,with adjusted ORs of 4.155(95%CI=2.711–6.368),1.839(95%CI=1.028–3.288),and 2.752(95%CI=1.197–6.326).Conclusions:Subjects who smoke,drink,with history of stomach disease and family history of GC in first-degree relatives are the high-risk populations for GC.Therefore,attention should be paid to these subjects for GC screening. | Rui Zhang He Li Ni Li Ju-Fang Shi Jiang Li Hong-Da Chen Yi-Wen Yu Chao Qin Jian-Song Ren Wan-Qing Chen Jie He | 2021 | Chinese Medical Journal2021,,16: | 7 |
