As the most frequently mutated RAS isoform, KRAS is intensively studied in the past years. Despite its well-recognized importance in cancer malignancy,continuous efforts in the past three decades failed to develop approved therapies for KRAS mutant cancer. KRAS has thus long been considered to be undruggable. | 5 | BMAL1 knockout macaque monkeys display reduced sleep and psychiatric disorders显示文摘Circadian disruption is a risk factor for metabolic, psychiatric and age-related disorders, and non-human primate models could help to develop therapeutic treatments. Here, we report the generation of BMAL1 knockout cynomolgus monkeys for circadian-related disorders by CRISPR/Cas9 editing of monkey embryos. These monkeys showed higher nocturnal locomotion and reduced sleep, which was further exacerbated by a constant light regimen. Physiological circadian disruption was reflected by the markedly dampened and arrhythmic blood hormonal levels. Furthermore, BMAL1-deficient monkeys exhibited anxiety and depression, consistent with their stably elevated blood cortisol, and defective sensory processing in auditory oddball tests found in schizophrenia patients. Ablation of BMAL1 up-regulated transcriptional programs toward inflammatory and stress responses, with transcription networks associated with human sleep deprivation, major depressive disorders, and aging. | 6 | Cloning of a gene-edited macaque monkey by somatic cell nuclear transfer显示文摘Cloning of macaque monkeys by somatic cell nucleus transfer(SCNT) allows the generation of monkeys with uniform genetic backgrounds that are useful for the development of non-human primate models of human diseases. Here, we report the feasibility of this approach by SCNT of fibroblasts from a macaque monkey(Macaca fascicularis), in which a core circadian transcription factor BMAL1 was knocked out by clustered regularly interspaced short palindromic repeat/Cas9 gene editing(see accompanying paper).Out of 325 SCNT embryos transferred into 65 surrogate monkeys, we cloned five macaque monkeys with BMAL1 mutations in both alleles without mosaicism, with nuclear genes identical to that of the fibroblast donor monkey. Further peripheral blood m RNA analysis confirmed the complete absence of the wild-type BMAL1 transcript. | 7 | An Estimation of Internal Soliton Forces on a Pile in the Ocean显示文摘Internal soliton forces on oil-platform piles in the ocean are estimated with the Morison Formula. Different from sur- face wave forces, which change only in magnitude along a pile, internal soliton forces can be distributed over the entire pile in the water and they change not only in magnitude but also in direction with depth. Our calculations show that the maximum total force caused by a soliton with its associated current of 2.1 m s-1 is nearly equal to the maximum total force exerted by a surface wave with a wavelength of 300 m and a wave-height of 18 m. The total internal soliton force is large enough to affect the operations of marine oil platforms and other facilities. | 9 | Detailed sedimentary facies of a sandstone reservoir in the eastern zone of the Sulige gas field, Ordos Basin显示文摘Given sustaining exploration, the eastern zone of the Sulige gas field may soon become a key area of exploitation. In order to explore its genesis, types and distribution of the reservoir sandstones in the eastern zone of this gas field, we focused in our study on the provenance and detailed sedimentary facies of sandstone of the He8 (the eighth part of the Shihezi formation, Permian system) and Shanxi1 (the first part of the Shanxi formation, Permian system) members, based on core observations, analyses in petrography, granularity and logging. The results show that: 1) the sandstone provenance of Shanxi1 and He8 in the eastern zone of the Sulige gas field is from the north of the Ordos Basin, characterized by dual directions from the north and northeast. 2) The He8 and Shanxi1 members were deposited in a fluvial-delta sedimentary system. The He8 was mainly deposited in braided rivers, in-cluding braided channels, channel bars, levee and floodplain sub-environments, whereas the Shanxi1 Member was deposited in braided rivers and deltas, including braided channels, channel bars, floodplains, tributaries and inter-tributary sub-environments. 3) Sedimentary facies bands migrated in drastic fashion towards the basin from the Shanxi1 to the He8 Member. Base levels of sedi-mentation generally present a trend of small increases in-amplitude, large decreasing amplitudes and slow and gradual Increases. 4) The continuity of the reservoir sandbodies along the source direction is better than that perpendicular to the direction. | 11 | Alterations in phosphoproteome under salt stress in Thellungiella roots显示文摘High salinity stress is a major environmental factor that limits plant's distribution and productivity. An Arabidopsis-related halophyte,Thellungiella halophila,is an emerging model system used for plant abiotic stress tolerance research. Previous studies have suggested that protein phosphorylation has a crucial role in the high salinity response in plants. However,the phosphoproteome differential expression under high salinity stress in halophytes has not been well studied. In this report,phosphoproteome differential expression was analyzed under high salinity stress in Thellungiella roots. Twenty-six putative phosphoproteins were found to have changed expression pattern at the post-translational level. Twenty of these were identified by mass spectrometric analysis,including 18 upregulated and two downregulated phosphoproteins. These proteins were involved in a variety of cellular processes,such as signal transduction,ROS detoxification,energy pathway,protein synthesis and protein folding. While most of these salt-responsive putative phosphoproteins are known salt-stress-related proteins,some of them have not been previously reported. | 12 | Development and validation of an endoscopic images-based deep learning model for detection with nasopharyngeal malignancies显示文摘Background:Due to the occult anatomic location of the nasopharynx and frequent presence of adenoid hyperpla-sia,the positive rate for malignancy identification during biopsy is low,thus leading to delayed or missed diagnosis for nasopharyngeal malignancies upon initial attempt.Here,we aimed to develop an artificial intelligence tool to detect nasopharyngeal malignancies under endoscopic examination based on deep learning.Methods:An endoscopic images-based nasopharyngeal malignancy detection model(eNPM-DM)consisting of a fully convolutional network based on the inception architecture was developed and fine-tuned using separate training and validation sets for both classification and segmentation.Briefly,a total of 28,966 qualified images were collected.Among these images,27,536 biopsy-proven images from 7951 individuals obtained from January 1st,2008,to December 31st,2016,were split into the training,validation and test sets at a ratio of 7:1:2 using simple randomiza-tion.Additionally,1430 images obtained from January 1st,2017,to March 31st,2017,were used as a prospective test set to compare the performance of the established model against oncologist evaluation.The dice similarity coef-ficient(DSC)was used to evaluate the efficiency of eNPM-DM in automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images,by comparing automatic segmentation with manual segmenta-tion performed by the experts.Results:All images were histopathologically confirmed,and included 5713(19.7%)normal control,19,107(66.0%)nasopharyngeal carcinoma(NPC),335(1.2%)NPC and 3811(13.2%)benign diseases.The eNPM-DM attained an overall accuracy of 88.7%(95%confidence interval(CI)87.8%-89.5%)in detecting malignancies in the test set.In the prospective comparison phase,eNPM-DM outperformed the experts:the overall accuracy was 88.0%(95%CI 86.1%-89.6%)vs.80.5%(95%CI 77.0%-84.0%).The eNPM-DM required less time(40 s vs.110.0±5.8 min)and exhibited encouraging performance in automatic segmentation of nasopharyngeal malignant area from the background,with an average DSC of 0.78±0.24 and 0.75±0.26 in the test and prospective test sets,respectively.Conclusions:The eNPM-DM outperformed oncologist evaluation in diagnostic classification of nasopharyngeal mass into benign versus malignant,and realized automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images. | Chaofeng Li Bingzhong Jing Liangru Ke Bin Li Weixiong Xia Caisheng He Chaonan Qian Chong Zhao Haiqiang Mai Mingyuan Chen Kajia Cao Haoyuan Mo Ling Guo Qiuyan Chen Linquan Tang Wenze Qiu Yahui Yu Hu Liang Xinjun Huang Guoying Liu Wangzhong Li Lin Wang Rui Sun Xiong Zou Shanshan Guo Peiyu Huang Donghua Luo Fang Qiu Yishan Wu Yijun Hua Kuiyuan Liu Shuhui Lv Jingjing Miao Yanqun Xiang Ying Sun Xiang Guo Xing Lv | 2018 | Cancer Communications2018,38,1: | 9 |
| 13 | 30-W supercontinuum generation based on ZBLAN fiber in an all-fiber configuration显示文摘We report an all-fiberized 30-W supercontinuum(SC) generation in a piece of ZrF4-BaF2-LaF3-AlF3-NaF(ZBLAN) fiber. The pump source is a thulium-doped fiber amplifier(TDFA) with broadband output spectrum spanning the 1.9 to 2.6 μm region. The used ZBLAN fiber has a core diameter of 10 μm, and was directly fusion-spliced to the pigtail of the TDFA without using a traditional mode field adapter(MFA) or a piece of transition fiber. Such a low-loss and robust fusion splice joint, together with a robust AlF3-fiber-based endcap,enables efficient and high-power SC generation in the ZBLAN fiber. An SC with an average power up to 30.0 W and a spectral coverage of 1.9–3.35 μm with 20-dB bandwidth of 1.92–3.20 μm was obtained. Moreover, an SC with a broader spectrum was achieved by raising the pump pulse peak power(via reducing the duty ratio of the pump laser pulse). An SC with an output power of 27.4 W and a spectral coverage of 1.9–3.63 μm(with 20-dB bandwidth of 1.92–3.47 μm) was obtained, as well as an SC with output power of 24.8 W and a spectral coverage of 1.9–3.70 μm(with 20-dB bandwidth of 1.93–3.56 μm). The power conversion efficiency was measured as>69%. | 16 | Effect of a cyclonic flotation column on the separation of magnesium from phosphate ore显示文摘This study was performed to investigate the effects of the cyclonic separation mechanism on the removal of magnesium from phosphate ore. An analysis of the particle-bubble collision mechanism in a cyclonic force field was used to design two different pulp circulation systems for a cyclonic-static micro-bubble flotation column (FCSMC). Experiments comparing the two conditions were then conducted. Size analysis of the feed and separated products was conducted with a BXF cyclone size analyzer. The results show that about 34.60% of the total MgO content in the feed exists in the 10 lm fraction. This demonstrates that magnesium removal from phosphate ore has the characteristics of a micro-fine particle flotation problem. Under conditions of cyclonic circulation the MgO level of the concentrate is 1.74%, which is lower than that obtained from the column operated in direct-flow circulation by 0.34%. These results indicate that the cyclonic circulation mechanism of the FCSMC can promote removal of micro-fine particles containing magnesium. | 18 | An expression atlas of miRNAs in Arabidopsis thaliana显示文摘MicroRNAs(miRNAs) are small non-coding RNAs that regulate a variety of biological processes. miRNA expression often exhibits spatial and temporal specificity. However, genome-wide miRNA expression patterns in different organs during development of Arabidopsis thaliana have not yet been systemically investigated. In this study, we sequenced small RNA libraries generated from 27 different organ/tissue types, which cover the entire life cycle of Arabidopsis. Analysis of the sequencing data revealed that most miRNAs are ubiquitously expressed, whereas a small set of miRNAs display highly specific expression patterns. In addition, different miRNA members within the same family have distinct spatial and temporal expression patterns. Moreover, we found that some miRNAs are produced from different arms of their hairpin precursors at different developmental stages. | 20 | Semantic-Oriented Sentiment Classification for Chinese Product Reviews: An Experimental Study of Book and Cell Phone Reviews显示文摘Sentiment classification is an automatic opinion classification method to classify the product reviews on web into positive or negative opinions to help consumers or sellers to understand the opinions and evaluations from existing customers. Semantic-oriented approach is one of the recent developments in sentiment classification. Up to now, most research of sentiment classification is on English reviews,and little work has been done on Chinese reviews using sentiment classification. The detailed techniques used in English review cannot be applied directly to Chinese reviews due to the different characteristics between these two languages. This study modified and improved the semantic-oriented approach to a 6-step process for Chinese review, focusing on the modification and improvement on the text segmentation and reference words pairs (RWPs) identification. Two experiments were conducted on book reviews and cell phone reviews.
