| 1 | Pharmacological effects of Astragaloside IV: a literature review显示文摘OBJECTIVE: To review the pharmacological effects and mechanisms of action of Astragaloside IV in Huangqi (Radix Astragali Mongolici). METHODS: Aticles focusing on Astragaloside IV in English and Chinese in databases were collected and reviewed in order to summarize the latest extraction separation, pharmacokinetics, and the pharmacological effects of astrageloside IV. RESULTS: Protective effects of Astrageloside IV on the cardiovascular system, immune, digestive, nervous system were identified, and the action mechanisms were associated with regulation of the calcium balance, anti-oxydant, antiapoptosis, antivirus, and so on. | 5 | Eco-environment range in the source regions of the Yangtze and Yellow rivers显示文摘Based on geographical and hydrological extents delimited, four principles are identified, as the bases for delineating the ranges of the source regions of the Yangtze and Yellow rivers in the paper. According to the comprehensive analysis of topographical characteristics, climate conditions, vegetation distribution and hydrological features, the source region ranges for eco-environmental study are defined. The eastern boundary point is Dari hydrological station in the upper reach of the Yellow River. The watershed above Dari hydrological station is the source region of the Yellow River which drains an area of 4.49×10 4 km 2 . Natural environment is characterized by the major topographical types of plateau lakes and marshland, gentle landforms, alpine cold semi-arid climate, and steppe and meadow vegetation in the source region of the Yellow River. The eastern boundary point is the convergent site of the Nieqiaqu and the Tongtian River in the upstream of the Yangtze River. The watershed above the convergent site is the source region of the Yangtze River, with a watershed area of 12.24×10 4 km 2 . | 6 | Influence of Gd_2O_3 and Yb_2O_3 Co-doping on Phase Stability,Thermo-physical Properties and Sintering of 8YSZ显示文摘The role of multicomponent rare earth oxides in phase stability, thermophysical properties and sintering for ZrO2-based thermal barrier coatings (TBCs) materials is investigated. 8YSZ codoped with 3 mol% Gd2O3 and 3 mol% Yb2O3 (GYb-YSZ) powders are synthesized by solid state reaction for 24 h at various temperatures. As temperature increases, stabilizers are dissolved into zirconia matrix gradually. Synthesized at 1 500 °C, GYb-YSZ is basically composed of cubic phase. GYb-YSZ exhibits excellent phase stability and sinters lower than 8YSZ by nearly three times. The thermal conductivity of GYb-YSZ is much lower than that of 8YSZ, and the thermal expansion coefficient of GYb-YSZ is comparable to that of 8YSZ. | 13 | Unique characteristics of the genetics epidemiology of amyotrophic lateral sclerosis in China显示文摘Continual discoveries of new genes and unraveling the genetic etiology in amyotrophic lateral sclerosis(ALS) have provided greater insight into the underlying pathogenesis in motor neuron degeneration, as well as facilitating the disease modeling and the testing of targeted therapeutics. While, the genetic etiology accounted for two-thirds of FALS and approximately 11% of SALS in Caucasians. However, the contributions of these causative genes to ALS vary among different populations. Furthermore, the prominent difference between Chinese population and other ethnics remains a source of ongoing debate. We systemically reviewed genetics literature of Chinese ALS populations and updated the mutation frequencies of the main ALS-implicated genes aiming to determine the genetic features of ALS in Chinese population. We also reviewed the associations between ALSimplicated single nucleotide polymorphisms(SNPs) and the risk of ALS in Chinese population. A total of 116 studies were included in this analysis(86 gene mutation study articles and 30 SNPs study articles). The results showed that the overall gene mutation rates of ALS-related causative genes were 55.0% in familial ALS(FALS) and 11.7% in sporadic ALS(SALS) in Chinese population. In Chinese FALS, the highest mutation frequency was found in SOD1 gene(25.6%), followed by FUS(5.8%), TARDBP(5.8%), DCTN1(3.6%) and C9 orf 72(3.5%). In Chinese SALS, the highest mutation frequency was also identified in SOD1 gene(1.6%), followed by ANXA11(1.4%), FUS(1.3%), SQSTM1(1.0%), OPTN(0.9%) and CCNF(0.8%).The associations between several SNPs and risk of ALS were also reported in Chinese population. The genetic features of ALS in Chinese population are significantly different from those in Caucasian population, indicating an association between genetic susceptibility and origin of population. | 14 | Reduning injection for fever, rash, and ulcers in children with mild hand, foot, and mouth disease: a randomized controlled clinical study显示文摘OBJECTIVE: To assess the efficacy and safety of Reduning injection for fever, rash, and ulcers in children with mild hand, foot, and mouth disease(HFMD). METHODS: A stratified-block randomized, double-blind, parallel-controlled, and multicenter clinical trial was conducted with 360 patients in five hospitals across China: Quanzhou Children's Hospital, Shijiazhuang No. 5 Hospital, Shanghai Public Health Centre, Hunan Provincial Children's Hospital, and Kaifeng Children's Hospital. Patients were randomized into three groups with 120 in each. Group A was treated with Western Medicine, group B with Reduning injection, a Chinese herbal medicine, and group C with both Reduning injection and Western Medicine. Results were compared for treatment efficacy and safety on HFMD. The clinical outcomes were observed as follows: fever and onset time of antifebrile effect(time to bring the body temperature down ≥0.5℃ after medication); cumulative time for fever recovery(body temperature recovering to normal and lasting more than 24 h without medication); cumulative time for rash disappearance(without new rashes or ulcers appearing and the original ones fading away); and cumulative time for mouth ulcer disappearance. RESULTS: For the onset time of the antifebrile effect, there was no statistical difference between groups A and B(P>0.05) and groups B and C(P> 0.05). However, there was a statistical difference between groups A and C(P<0.05), and the effect in group C was the best. For the cumulative time for rash disappearance, there was no statistical difference between groups A and B(P>0.05). There were statistical differences between groups A and C, and groups B and C(P<0.05), and the effect in group C was the best. For the cumulative time for mouth ulcers disappearance, there were no statistical differences among the three groups(P>0.05). CONCLUSION: Reduning injection with Western Medicine for symptomatic treatment is most effective for mild HFMD. | 18 | INTERVENTIONAI DIAGNOSIS AND TREATMENT OF VASCULOGENEIC PULSATILE TINNITUS显示文摘Objective To retrospectively study clinical features and diagnostic imaging of vasculogeneic pulsatile tinnitus, and the feasibility and efficacy of transvascular interventional treatment for this condition. Methods Data from 82 cases of arterial or venous pulsatile tinnitus were reviewed. DSA characteristics and possible pathophysiological mechanisms of pulsatile tinnitus in these cases were studied. Diagnoses in this group included intracranial arterovenous fistula(AVF)(n=3), spontaneous skull base dural AVF(n=16), traumatic carotid-cavernous sinus fistula(n=5), subclavian artery stenosis(n=2), internal carotid artery stenosis(n=3), intracranial arterial stenosis(n=1), kinked and/or elongated vertebrobasilar artery(n=2), venous sinus diverticulum(n=2), venous sinus stenosis on the dominant drainage side(n=46) and occipital sinus stenosis(n=2).Treatments included embolization and stenting using coils, NBCA glue, Balt balloons, self-expansion stents and intracranial micro-stents via either the femoral artery or femoral vein. Results Procedures were successful in all cases with no surgery-related complications. Tinnitus disappeared within 2 days after the procedure in all cases. Follow up duration was 5-36 months. Recurrence occurred in 4 cases of arterial tinnitus within 3 months following the initial procedure, which improved after revision embolization or symptom management. There was no recurrence in venous tinnitus cases following stent plastic or stent-coiling embolization treatments. Conclusions Endovascular intervention provides a new approach to the diagnosis and treatment of intractable pulsatile tinnitus. In this model,deformations of the bulk pad substrate and the asperities were considered. The deformations of the bulk pad sub- strate and the asperity layer,as well as the contact pressure and fluid pressure,were revealed with nu- merical methods. Numerical simulation results show a counterintuitive phenomenon: a diverging clear- ance is formed in the leading region of the wafer and thereby it gives rise to a suction pressure (subambi- ent pressure). A high stress concentration is pre- sented at the wafer edge and thereby over polishing can be introduced. | 20 | Genome-wide CRISPR screen identifies synthetic lethality between DOCK1 inhibition and metformin in liver cancer显示文摘Metformin is currently a strong candidate anti-tumor agent in multiple cancers.However,its anti-tumor effectiveness varies among different cancers or sub-populations,potentially due to tumor heterogeneity.It thus remains unclear which hepatocellular carcinoma(HCC)patient subpopulation(s)can benefit from met-formin treatment.Here,through a genome-wide CRISPR-Cas9-based knockout screen,we find that DOCK1 levels determine the anti-tumor effects of met-formin and that DOCK1 is a synthetic lethal target of metformin in HCC.Mechanistically,metformin promotes DOCK1 phosphorylation,which activates RAC1 to facilitate cell survival,leading to metformin resistance.The DOCK1-selective inhibitor,TBOPP,potentiates anti-tumor activity by metformin in vitro in liver cancer cell lines and patient-derived HCC organoids,and in vivo in xenografted liver cancer cells and immunocompetent mouse liver cancer models.Notably,metformin improves overall survival of HCC patients with low DOCK1 levels but not among patients with high DOCK1 expression.This study shows that metformin effective-ness depends on DOCK1 levels and that combining metformin with DOCK1 inhibition may provide a promising personalized therapeutic strategy for met-formin-resistant HCC patients. | Junru Feng Hui Lu Wenhao Ma Wenjing Tian Zhuan Lu Hongying Yang Yongping Cai Pengfei Cai Yuchen Sun Zilong Zhou Jiaqian Feng Jiazhong Deng Ying Shu Kun Qu Weidong Jia Ping Gao Huafeng Zhang | 2022 | Protein & Cell2022,13,11: | 5 |
