1Interleukin-8 associates with adhesion,migration,invasion and chemosensitivity of human gastric cancer cells显示文摘AIM:To investigate the relationship between Interleukin-8 (IL-8) and proliferation,adhesion,migration,invasion and chemosensitivity of gastric cancer (GC) cells.METHODS:The IL-8 cDNA was stably transfected into human GC cell line MKN-45 and selected IL-8-secreting transfectants.The expression of IL-8 in human GC cell line KATO-Ⅲ was inhibited by RNA interference.The expressions of mRNA and protein of IL-8 in GC cells were detected by real-time reverse transcriptionpolymerase chain reaction or enzyme-linked immunosorbent assay (ELISA).RESULTS:The overexpression of IL-8 resulted in an increased cell adhesion,migration and invasion,and a significant resistance to oxaliplatin in MKN-45 cells.Inhibition of IL-8 expression with small interfering RNA decreased the adhesion,migration and invasion functions and oxaliplatin resistance in KATO-Ⅲ cells.IL-8 increased NF-κB and Akt activities and adhesion molecules ICAM-1,VCAM-1,and CD44 expression in GC cells.CONCLUSION:Overexpression of IL-8 promotes the adhesion,migration,invasion,and chemoresistance of GC cells,indicating that IL-8 is an important therapeutic target in GC.Wen-Xia Kuai Qiong wang Xiao-Zhong Yang Yao Zhao Ren Yu Xiao-Jun Tang 2012World Journal of Gastroenterology2012,18,9:21
2Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy.Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou 2022Cell Research2022,32,6:10
3Protective effect of oxysophoridine on cerebral ischemia/reperfusion injury in mice显示文摘Oxysophoridine, a new alkaloid extracted from Sophora alopecuroides L., has been shown to have a protective effect against ischemic brain damage. In this study, a focal cerebral ischemia/reperfusion injury model was established using middle cerebral artery occlusion in mice. Both 62.5, 125, and 250 mg/kg oxysophoridine, via intraperitoneal injection, and 6 mg/kg nimodipine, via intragastric administration, were administered daily for 7 days before modeling. After 24 hours of reperfusion, mice were tested for neurological deficit, cerebral infarct size was assessed and brain tissue was collected. Results showed that oxysophoridine at 125, 250 mg/kg and 6 mg/kg nimodipine could reduce neurological deficit scores, cerebral infarct size and brain water content in mice. These results provided evidence that oxysophoridine plays a protective role in cerebral ischemia/reperfusion injury. In addition, oxysophoridine at 62.5, 125, and 250 mg/kg and 6 mg/kg nimodipine increased adenosine-triphosphate content, and decreased malondialdehyde and nitric oxide content. These compounds enhanced the activities of glutathione-peroxidase, superoxide dismutase, catalase, and lactate dehydrogenase, and decreased the activity of nitric oxide synthase. Protein and mRNA expression levels of N-methyl-D-aspartate receptor subunit NR1 were markedly inhibited in the presence of 250 mg/kg oxysophoridine and 6 mg/kg nimodipine. 14Alternations in the gut microbiota and metabolome with newly diagnosed unstable angina显示文摘Gut microbiota plays an important role in coronary heart disease, but its compositional and functional changes in unstable angina(UA) remain unexplored. We performed metagenomic sequencing of 133 newly diagnosed UA patients and 133 sex-and age-matched controls, and profiled the fecal and plasma metabolomes in 30 case-control pairs. The alpha diversity of gut microbiota was increased in UA patients:the adjusted odds ratios(ORs) per standard deviation increase in Shannon and Simpson indices were 1.30(95% confidence interval, 1.01-1.70) and 1.36(1.05-1.81), respectively. Two common species(depleted Klebsiella pneumoniae and enriched Streptococcus parasanguinis;P ≤ 0.002) and three rare species(depleted Weissella confusa, enriched Granulicatella adiacens and Erysipelotrichaceae bacterium 6_1_45;P ≤ 0.005) were associated with UA. The UA-associated gut microbiota was depleted in the pathway of Lphenylalanine degradation(P = 0.001), primarily contributed by Klebsiella pneumoniae. Consistently, we found increased circulating phenylalanine in UA patients(OR = 2.76 [1.17-8.16]). Moreover, Streptococcus parasanguinis was negatively correlated with fecal citrulline(Spearman's r_(s)=-0.470, P = 0.009), a metabolite depleted in UA patients(OR = 0.26 [0.08-0.63]). 16Dispersion and support dictated properties and activities of Pt/metal oxide catalysts in heterogeneous CO oxidation显示文摘The source of activity of metal/metal oxides has always been an interesting,important but highly challenging research topic in heterogeneous catalysis.In CO oxidation reaction,this work clarifies dispersion and support dictated activities of Pt including single-atom(Pt1),2.8 nm(PtNP-S)and 36 nm(PtNP-L)Pt supported on both reducible TiO_(2) and“inert”Al_(2)O_(3) supports.The X-ray absorption fine structure(XAFS)shows that chemical state of Pt is affected by both dispersion and support:Pt1 presents fully oxidized state in both Pt1/TiO_(2) and Pt1/Al_(2)O_(3);PtNP-S in PtNP-S/TiO_(2) appear nearly oxidized state while about half of Pt is metallic state in PtNP-S/Al_(2)O_(3);PtNP-L in both PtNP-L/TiO_(2) and PtNP-L/Al_(2)O_(3) exhibit metallic state.All the Pt species supported on TiO_(2) present much lower apparent activation barriers(Eapp)than that on Al_(2)O_(3).Moreover,Pt1/TiO_(2) possesses dozen times of efficiency than PtNP-S/TiO_(2) although they have similar Eapp values.A truth is finally made clear that a reducible metal oxide with low oxygen vacancy formation energy is critical to endow Pt/metal oxide a high activity and the single-atom dispersion of Pt is the way to maximize the active sites of Pt/metal oxide.Jiaojiao Song Yixuan Yang Shoujie Liu Lei Li Nan Yu Yuteng Fan Zhiming Chen Long Kuai Baoyou Geng 2021Nano Research2021,14,12:1
