1MicroRNA-24 promotes pancreatic beta cells toward dedifferentiation to avoid endoplasmic reticulum stress-induced apoptosis显示文摘Current research indicates that beta cell loss in type 2 diabetes may be attributed to beta cell dedifferentiation rather than apoptosis;however,the mechanisms by which this occurs remain poorly understood.Our previous study demonstrated that elevation of microRNA-24(miR-24)in a diabetic setting caused beta cell dysfunction and replicative deficiency.In this study,we focused on the role of miR-24 in beta cell apoptosis and dedifferentiation under endoplasmic reticulum(ER)stress conditions.We found that miR-24 overabundance protected beta cells from thapsigargin-induced apoptosis at the cost of accelerating the impairment of glucosestimulated insulin secretion(GSIS)and enhancing the presence of dedifferentiation markers.Ingenuity?Pathway Analysis(IPA)revealed that elevation of miR-24 had an inhibitory effect on XBP1 and ATF4,which are downstream effectors of two key branches of ER stress,by inhibiting its direct target,Irela.Notably,elevated miR-24 initiated another pathway that targeted Mafa and decreased GSIS function in surviving beta cells,thus guiding their dedifferentiation under ER stress conditions.Our results demonstrated that the elevated miR-24,to the utmost extent,preserves beta cell mass by inhibiting apoptosis and inducing dedifFerentiation.This study not only provides a novel mechanism by which miR-24 dominates beta cell turnover under persistent metabolic stress but also offers a therapeutic consideration for treating diabetes by inducing dedifferentiated beta cells to re-differentiation.Yunxia Zhu Yi Sun Yuncai Zhou Yan Zhang Tao Zhang Yating Li Weiyan You Xiaoai Chang Li Yuan Xiao Han 2019Journal of Molecular Cell Biology2019,11,9:9

3Sensitive electrochemical DNA sensor for the detection of HIV based on a polyaniline/graphene nanocomposite显示文摘A nanocomposite of polyaniline/graphene(PAN/GN)was prepared using reverse-phase polymerization.The nanocomposite material was dropcast onto a glassy carbon electrode(GCE).Then,a single-stranded DNA(ssDNA)probe for HIV-1 gene detection was immobilized on the modified electrode,and the negative charged phosphate backbone of the HIV-1 was bound to the modified electrode surface via p-p*stacking interactions.The hybridization between the ssDNA probe and the target HIV-1 formed doublestranded DNA(dsDNA),and the electron transfer resistance of the electrode was measured using impedimetric studies with a[Fe(CN)6]^(3-/4-)redox couple.Under the optimized experimental conditions,the change of the impedance value was linearly related to the logarithm of the concentration of HIV genes in the range from 5.0×10^(-16)M to 1.0×10^(-10)M(R=0.9930),and the HIV sensor exhibited a lower detection limit of 1.0×10^(-16)M(S/N=3).The results show that this biosensor presented wonderful selectivity,sensitivity and specificity for HIV-1 gene detection.Thus,this biosensor provides a new method for the detection of HIV gene fragments.Qiaojuan Gong Haixia Han Haiying Yang Meiling Zhang Xiaoling Sun Yunxia Liang Zhaorong Liu Wenchan Zhang Jinli Qiao 2019Journal of Materiomics2019,5,2:5

5Effects of adding sodium dichloroacetate to low-protein diets on nitrogen balance and amino acid metabolism in the portaldrained viscera and liver of pigs显示文摘Background:Identifying regulatory measures to promote glucose oxidative metabolism while simultaneously reducing amino acid oxidative metabolism is one of the foremost challenges in formulating low-protein(LP)diets designed to reduce the excretion of nitrogen-containing substances known to be potential pollutants.In this study,we investigated the effects of adding sodium dichloroacetate(DCA)to a LP diet on nitrogen balance and amino acid metabolism in the portal-drained viscera(PDV)and liver of pigs.To measure nitrogen balance,18 barrows(40±1.0 kg)were fed one of three diets(n=6 per group):18%crude protein(CP,control),13.5%CP(LP),and 13.5%CP+100 mg DCA/kg dry matter(LP-DCA).To measure amino acid metabolism in the PDV and liver,15 barrows(40±1.0 kg)were randomly assigned to one of the three diets(n=5 per group).Four essential amino acids(Lys,Met,Thr,and Trp)were added to the LP diets such that these had amino acid levels comparable to those of the control diet.Results:The LP-DCA diet reduced nitrogen excretion in pigs relative to that of pigs fed the control diet(P<0.05),without any negative effects on nitrogen retention(P>0.05).There were no differences between the control and LP-DCA groups with respect to amino acid supply to the liver and extra-hepatic tissues in pigs(P>0.05).The net release of ammonia into the portal vein and production rate of urea in the liver of pigs fed the LP-DCA diet was reduced relative to that of pigs fed the control and LP diets(P<0.05).Conclusion:The results indicated that addition of DCA to a LP diet can efficiently reduce nitrogen excretion in pigs and maximize the supply of amino acids to the liver and extra-hepatic tissues.Weizhong Sun Yunxia Li Zhiru Tang Huiyuan Chen Ke Wan Rui An Liuting Wu Zhihong Sun 2020Journal of Animal Science and Biotechnology2020,11,3:2

9Effects of confined space and near vision stimulation on refractive status and vitreous chamber depth in adolescent rhesus monkeys显示文摘This study aimed to investigate the effects of sustained near vision stimulation,on the refractive development and elongation of the vitreous chamber in adolescent rhesus monkeys.A total of 12 adolescent rhesus monkeys(1.5-2.0 years old) were randomly assigned to 3 groups.In groups A(n=4) and B(n=4),monkeys were reared in close-vision cages for 8 and 4 h d-1,respectively;tiny granules were added on the cage floor to avoid visual deprivation and to encourage near gaze.In group C(n=4),monkeys were reared in open-vision cages,with non-granule food as a control.Vitreous chamber depth,refractive status,and corneal refractive power were assessed over 18 months.Paired t-test was used to compare the differences and a P-value<0.05 was considered to be statistically significant.In group A,vitreous chamber depth and optical axis elongated significantly,and refractive error shifted towards myopia during the observation period.In group B,vitreous chambers and optical axis elongated but the refractive power did not show significant changes.In group C,there was no significant elongation in vitreous chambers and optical axis,and the refractive power changed slightly towards hypermetropia.There were no significant changes in corneal refractive power in each group.Sustained near vision can promote vitreous chamber growth and induce myopic shifts in refractive power in adolescent monkeys.Our results demonstrate the potential for a primate model of near-work-related myopia.LENG YunXia,LAN WeiZhong,YU KeMing,LIU BingQian,YANG ZhiKuan,LI Zheng,ZHONG XingWu,ZHANG ShaoChong & GE Jian* State Key Laboratory of Ophthalmology,Zhongshan Ophthalmic Center,Sun Yat-sen University,Guangzhou 510060,China 2010Science China(Life Sciences)2010,53,12:1

13Phenotype,Physiology,and Gene Expression of Barley Seedlings in Response to Nano Zinc Oxide Stress显示文摘In recent years,zinc oxide nanoparticles(ZnO NPs)have been widely used as zinc fertilizers and pesticides.The use of ZnO NPs in this way can provide benefits to humans,but also has potential risks.ZnO NPs inevitably enter the environment during their production and use,which affects the ecological environment and crop growth.In order to investigate the phenotype,physiology,and gene expression of barley(Hordeum vulgare L.)seedlings under ZnO NPs stress,the barely cultivars ZJU3(P21),Golden Promise(GP)and L23 were chosen for study.Different ZnO NPs concentrations were applied to compare the physiological and biochemical indexes of the barley seedlings and the responses of six stress-related genes,when seedlings were cultured to the two-leaf stage through hydroponics.The results showed that the density of brown spots on the leaf surface increased with increasing ZnO NPs concentration.ZnO NPs stress inhibited the root growth of barley seedlings,and P21 was the most sensitive.Furthermore,ZnO NPs stress could stimulate plants to produce a large number of reactive oxygen species(ROS),resulting in an imbalance between the production and removal of ROS and membrane lipid peroxidation in plants.This imbalance inhibited the growth and development of the barley seedlings.With increasing ZnO NPs concentration,the activity of superoxide dismutase was gradually increased,the activity of catalase was progressively decreased,and the contents of malondialdehyde and proline were increased.Compared with the control,among six stress-related genes,the expression levels of five genes were downregulated and one gene was upregulated in the experimental group.This study preliminarily revealed the toxic effect of ZnO NPs on seedlings and the effect on the expression of stress-related resistance genes in different barley varieties.Mengyuan Dong Rong Sun Qianhui Yang Lantian Zhang Yangying Yong Yunxia Fang Xian Zhang Xiaoqin Zhang Dawei Xue 2021Phyton-International Journal of Experimental Botany2021,90,6:0

15The aging behavior,microstructure and mechanical properties of AlN/AZ91 composite显示文摘Microstructure evolution and mechanical properties of the aging treated AlN/AZ91 composites were systematically investigated by optical microscopy(OM),high resolution scanning electron microscopy(HRSEM)with an energy dispersive spectrum(EDS),and high-angle annular dark field scanning transmission electron microscopy(HAADF-STEM).The results show that the higher fracture elongation(14±1%)and ultimate tensile strength(275±6 MPa)were simultaneously obtained in the peak-aged AlN/AZ91 composites.Comparied with AZ91 matrix alloy,the strength was increased by about 44%and the elongation was approximately five times higher,which mainly attributed to the precipitation of nano-sizedγ-Mg_(17)Al_(12)phase and the activation of non-basal slip systems induced by in-situ AlN particles at room temperature.However,the in-situ formation of AlN reinforcements consumed part of Al element in the matrix alloy,which resulted into the volume fraction decreasing ofγ-Mg_(17)Al_(12)precipitates,and then the age hardening and strengthening efficiency were reduced in the AlN/AZ91 composites.On the other hand,the mismatch of thermal expansion coefficient between AlN particles and AZ91 matrix generated high density dislocations around AlN particles,which promoted the precipitation ofγ-Mg_(17)Al_(12)phase,and then the peak aging time and temperature were decreased.Yunxia Sun Changlin Yang Bin Zhang Jianfeng Fan Hongxiang Li Tianhao Zhao Jing Li 2023Journal of Magnesium and Alloys2023,11,7:0
16Putting aniline radical cations in a bottle显示文摘Salts containing aniline radical cations have been isolated and characterized by electron paramagnetic resonance(EPR)spectroscopy, UV-Vis absorption spectroscopy and single crystal X-ray diffraction. The EPR spectra and theoretical calculations indicate the unpaired electron is delocalized on phenyl rings and nitrogen atoms. Both radical cations feature a quinoidal geometry with a partially double C–N bond, but are distinct in that the C–N bond is coplanar to the phenyl plane in one cation while deviates from the plane in the other due to steric crowding. 17BRSK2 in pancreatic β cells promotes hyperinsulinemia-coupled insulin resistance and its genetic variants are associated with human type 2 diabetes显示文摘Brain-specific serine/threonine-protein kinase 2(BRSK2)plays critical roles in insulin secretion andβ-cell biology.However,whether BRSK2 is associated with human type 2 diabetes mellitus(T2DM)has not been determined.Here,we report that BRSK2 genetic variants are closely related to worsening glucose metabolism due to hyperinsulinemia and insulin resistance in the Chinese population.BRSK2 protein levels are significantly elevated inβcells from T2DM patients and high-fat diet(HFD)-fed mice due to enhanced protein stability.Mice with inducibleβ-cell-specific Brsk2 knockout(βKO)exhibit normal metabolism with a high potential for insulin secretion under chow-diet conditions.Moreover,βKO mice are protected from HFD-induced hyperinsulinemia,obesity,insulin resistance,and glucose intolerance.Conversely,gain-of-function BRSK2 in matureβcells reversibly triggers hyperglycemia due toβ-cell hypersecretion-coupled insulin resistance.Mechanistically,BRSK2 senses lipid signals and induces basal insulin secretion in a kinase-dependent manner.The enhanced basal insulin secretion drives insulin resistance andβ-cell exhaustion and thus the onset of T2DM in mice fed an HFD or with gain-of-function BRSK2 inβcells.These findings reveal that BRSK2 links hyperinsulinemia to systematic insulin resistance via interplay betweenβcells and insulin-sensitive tissues in the populations carrying human genetic variants or under nutrient-overload conditions.Rufeng Xu Kaiyuan Wang Zhengjian Yao Yan Zhang Li Jin Jing Pang Yuncai Zhou Kai Wang Dechen Liu Yaqin Zhang Peng Sun Fuqiang Wang Xiaoai Chang Tengli Liu Shusen Wang Yalin Zhang Shuyong Lin Cheng Hu Yunxia Zhu Xiao Han 2023Journal of Molecular Cell Biology2023,15,5:0
18The Influence of Differential Leadership on Employees’Affective Commitment:A Perspective of the Insiders and Outsiders显示文摘From the perspective of the insiders and outsiders,this study explores the influence of differential leadership on employees’affective commitment and the moderating effect of leader’s self-enhancing humor and individual traditionality.The results show that the differential leadership has a positive impact on the organizational affective commitment of employees,the leader’s self-enhancing humor and the employees’traditionality play a positive regulatory role respectively.Moreover,compared with the outsiders,the low traditionality has a stronger influence on the relationship between differential leadership and organizational affective commitment of the insiders.This paper enriches the research on the influence of leadership style on employee’s affective commitment,proposes and verifies the moderation of leader’s self-enhancing humor and employee’s traditionality,which complements the boundary conditions for the effectiveness of differential leadership style.Yunxia Su Minggui Sun 2020Journal of Business Administration Research2020,3,3:0
19Magnetic NiFe_(2)O_(4)/Polypyrrole nanocomposites with enhanced electromagnetic wave absorption显示文摘NiFe_(2)O_(4)/polypyrrole(NiFe_(2)O_(4)/PPy)nanocomposites are prepared by a simple surface-initiated polymerization method and demonstrate negative permittivity in the low frequency regions.These nanocomposites also exhibit significantly enhanced electromagnetic wave(EMW)absorption property in the high frequency regions.Compared with pure PPy,the enhanced negative permittivity is observed in the NiFe_(2)O_(4)/PPy nanocomposites with a NiFe_(2)O_(4)loading of 5.0,10.0,20.0 and 40.0 wt%,indicating the formation of metal-like electrical conducting network in NiFe_(2)O_(4)/PPy nanocomposites.Moreover,the negative permittivity could be tuned by changing the NiFe_(2)O_(4)loading.The minimum reflection loss(RL)of-40.8 dB is observed in the 40.0 wt%NiFe_(2)O_(4)/PPy composites with a thickness of only 1.9 mm.The effective absorption bandwidth below-10.0 and-20.0 dB reaches 6.08 and 2.08 GHz,respectively.The enhanced EMW absorption performance benefits from the improved independence matching,EMW attenuation capacity,and synergistic effects of conduction loss,dielectric loss(interfacial and dipole polarizations)and magnetic loss(exchange and natural resonances).This research work provides a guidance for the fabrication of nanocomposites with an excellent EMW absorption.Jiang Guo Xu Li Zhuoran Chen Jianfeng Zhu Xianmin Mai Renbo Wei Kai Sun Hu Liu Yunxia Chen Nithesh Naik Zhanhu Guo 2022Journal of Materials Science & Technology2022,,13:0
20Screening Coarctation of Aorta with Clinical and Echocardiographic Profiles in Infants:A Pilot Study显示文摘Aim:To determine the profiles of clinical features including four-limb blood pressure(BP),saturations of peripheral oxygen(SpO2),and echocardiographic features in infants with coarctation of aorta(CoA)to facilitate congenital heart diseases screening.Methods:The charts of infants with CoA were retrospectively reviewed.All in-hospital infants suspected of congenital heart diseases by clinical teams were prospectively measured of four limbs BPs and SpO2 in a regional cardiac transferring center during 2013 and 2019.Echocardiography as a gold standard test was followed within 2 days after suspicion.All infants were divided into non-significant CoA group or significant CoA group based on the difference of BPs between right arm and lower limbs.Predictors of nonsignificant CoA were determined with multivariable logistic regression.Results:One hundred thirty-three infants with CoA were identified.The BPs on upper limbs were higher than those on lower limbs(P=0.001).No statistical difference in SpO2 was found between four limbs.Fifty-three(39.8%)infants presented with significant CoA.Thirty-four infants presented with low SpO2 and 26 of 34 presented with non-significant CoA.Small ascending aorta diameter[0.070(95%CI:0.005–0.136),P=0.036]was an independent risk factors for non-significant CoA.Eighteen(13.5%)infants with CoA didn’t present with any of the BP difference,low SpO2,murmur,or weak femoral pulse.Conclusion:Less than half of the infants with CoA presented with a significant BP difference.Another one fifth presented with low SpO2.Small ascending aorta diameter was an independent factor for non-significant CoA.Ruikun Zou Yifei Wang Chengcheng Pang Yunxia Sun Chen Chen Jian Zhuang 2021Congenital Heart Disease2021,16,2:0
