Hyaluronic acid facilitates bone repair effects of calcium phosphate cement by accelerating osteogenic expression显示文摘Calcium phosphate cements(CPC)are widely anticipated to be an optimum bone repair substitute due to its satisfied biocompatibility and degradability, suitable to be used in minimally invasive treatment of bone defects. However the clinical application of CPC is still not satisfied by its poor cohesiveness and mechanical properties,in particular its osteoinductivity.Hyaluronic acid reinforced calcium phosphate cements(HA/CPC)showed extroadinary potential not only enhancing the compressive strength of the cements but also significantly increasing its osteoinductivity. In our study, the compressive strength of HA/CPC increased significantly when the cement was added 1%hyaluronic acid(denoted as 1-HA/CPC).In the meantime, hyaluronic acid obviously promoted ALP activity, osteogenic related protein and mRNA expression of hBMSCs(human bone marrow mesenchymal stem cells)in vitro, cement group of HA/CPC with 4%hyaluronic acid adding(denoted as 4-HA/CPC)showed optimal enhancement in hBMSCs differentiation. After being implanted in rat tibial defects,4-HA/CPC group exhibited better bone repair ability and bone growth promoting factors, comparing to pure CPC and 1-HA/CPC groups. The underlying biological mechanism of this stimulation for HA/CPC may be on account of higher osteogenic promoting factors secretion and osteogenic genes expression with hyaluronic acid incorporation. These results indicate that hyaluronic acid is a highly anticipated additive to improve physicochemical properties and osteoinductivity performance of CPCs for minimally invasive healing of bone defects. | Xu Cui Chengcheng Huang Zhizhen Chen Meng Zhang Chunyu Liu Kun Su Jianyun Wang Li Li Renxian Wang Bing Li Dafu Chen Changshun Ruan Deping Wang William W.Lu Haobo Pan | 2021 | Bioactive Materials2021,6,11: | 6
Formation and migration of space charge packet in low density polyethylene显示文摘Pressure wave propagation (PWP) method was utilized to observe the space charges packet-like behavior in sheet samples of low-density polyethylene (LDPE) under the relative low electric field. The experimental results show that space charge packets are always generated at the anode and move across the sample to the cathode regard- less of the polarity of DC voltage. This paper discusses the migration of space charge packets at 30―40 ℃ , the possible charge resource of space charge packets carriers and trap level of the space charge. | ZHENG Feihu 1 , ZHANG Yewen 1 , GONG Bin 1 , ZHU Jianwei 1 & WU Changshun 2 1. | 4 | Strontium modulates osteogenic activity of bone cement composed of bioactive borosilicate glass particles by activating Wnt/β-catenin signaling pathway显示文摘There is a need for synthetic grafts to reconstruct large bone defects using minimal invasive surgery. Our previous study showed that incorporation of Sr into bioactive borate glass cement enhanced the osteogenic capacity in vivo. However, the amount of Sr in the cement to provide an optimal combination of physicochemical properties and capacity to stimulate bone regeneration and the underlying molecular mechanism of this stimulation is yet to be determined. In this study, bone cements composed of bioactive borosilicate glass particles substituted with varying amounts of Sr(0 mol%to 12 mol%SrO)were created and evaluated in vitro and in vivo. The setting time of the cement increased with Sr substitution of the glass.Upon immersion in PBS,the cement degraded and converted more slowly to HA(hydroxyapatite)with increasing Sr substitution. The released Sr2+modulated the proliferation, differentiation, and mineralization of hBMSCs(human bone marrow mesenchymal stem cells)in vitro.Osteogenic characteristics were optimally enhanced with cement(designated BG6Sr)composed of particles substituted with 6mol%SrO.When implanted in rabbit femoral condyle defects,BG6Sr cement supported better peri-implant bone formation and bone-implant contact, comparing to cements substituted with 0mol%or 9mol%SrO.The underlying mechanism is involved in the activation of Wnt/β-catenin signaling pathway in osteogenic differentiation of hBMSCs.These results indicate that BG6Sr cement has a promising combination of physicochemical properties and biological performance for minimally invasive healing of bone defects. | Xu Cui Yadong Zhang Jianyun Wang Chengcheng Huang Yudong Wang Hongsheng Yang Wenlong Liu Ting Wang Deping Wang Guocheng Wang Changshun Ruan Dafu Chen William W.Lu Wenhai Huang Mohamed N.Rahaman Haobo Pan | 2020 | Bioactive Materials2020,5,2: | 3
CUL4B facilitates HBV replication by promoting HBx stabilization显示文摘Objective:Hepatitis B virus(HBV)infection is a major public health problem worldwide. However, the regulatory mechanisms underlying HBV replication remain unclear.Cullin 4 B-RING ubiquitin E3 ligase(CRL4 B)is involved in regulating diverse physiological and pathophysiological processes. In our study,we aimed to explain the role of CUL4 B in HBV infection.Methods:Cul4 b transgenic mice or conditional knockout mice,as well as liver cell lines with CUL4 B overexpression or knockdown, were used to assess the role of CUL4 B in HBV replication.Immunoprecipitation assays and immunofluorescence staining were performed to study the interaction between CUL4 B and HBx.Cycloheximide chase assays and in vivo ubiquitination assays were performed to evaluate the half-life and the ubiquitination status of HBx.Results:The hydrodynamics-based hepatitis B model in Cul4 b transgenic or conditional knockout mice indicated that CUL4 B promoted HBV replication(P<0.05).Moreover,the overexpression or knockdown system in human liver cell lines validated that CUL4 B increased HBV replication in an HBx-dependent manner.Importantly, immunoprecipitation assays and immunofluorescence staining showed an interaction between CUL4 B and HBx.Furthermore,CUL4 B upregulated HBx protein levels by inhibiting HBx ubiquitination and proteasomal degradation(P<0.05).Finally,a positive correlation between CUL4 B expression and HBV pg RNA level was observed in liver tissues from HBV-positive patients and HBV transgenic mice.Conclusions:CUL4 B enhances HBV replication by interacting with HBx and disrupting its ubiquitin-dependent proteasomal degradation.CUL4 B may therefore be a potential target for anti-HBV therapy. | Haixia Shan Bo Wang Xiaodong Zhang Hui Song Xi Li Yongxin Zou Baichun Jiang Huili Hu Hao Dou Changshun Shao Lifen Gao Chunhong Ma Xiaoyun Yang Xiaohong Liang Yaoqin Gong | 2022 | Cancer Biology & Medicine2022,19,1: | 2

Simultaneous determination of four Sudan dyes in rat blood by UFLC–MS/MS and its application to a pharmacokinetic study in rats显示文摘A rapid and sensitive method based on ultrafast liquid chromatography–tandem mass spectrometry was developed and validated for simultaneous determination of Sudan I, Sudan II, Sudan III, and Sudan IV levels in rat whole blood. Cleanert C18 mixed-mode polymeric sorbent was used for effective solid-phase extraction cleanup. Separation was carried out on a reversed-phase C18column(100 mm 2.1 mm,1.8 μm) using 0.1%(v/v) formic acid in water/0.1%(v/v) formic acid in acetonitrile as the mobile phase in gradient elution. Quanti fi cation was performed by an electrospray ionization source in the positive multiple reaction monitoring mode using D5-Sudan I as the internal standard. Calibration curves showed good linearity between 0.2 and 20.0 μg/L, with correlation coef fi cients higher than 0.9990. The average recovery rates were between 93.05% and 114.98%. The intra- and inter-day relative standard deviations were within 6.2%. The lower limit of quanti fi cation was 0.2 μg/L. All the analytes were found to be stable in a series of stability studies. The proposed method was successfully applied to a pharmacokinetic study of four Sudan dyes after oral administration to rats. | Hao Zhu Yijun Chen Changshun Huang Yangyang Han Yiwei Zhang Shucan Xie Xiaohong Chen Micong Jin | 2015 | Journal of Pharmaceutical Analysis2015,5,4: | 1

Multiple walking human recognition based on radar micro-Doppler signatures显示文摘The recognition of human movements based on radar m-D(micro-Doppler) signatures attracts great interest in the field of radar research on automatic target recognition. Because there are multiple frequency components overlapping seriously in the radar echoes from walking humans, it is a very difficult work to recognize walking humans based on radar echoes. In this paper, a recognition method of walking humans based on radar m-D signatures is proposed. In this method, the m-D spectrum is generated by generalized S transform first, and then the entropy segmentation is used to segment the interesting region from the original spectrum. Next, the m-D features are extracted from the m-D region. Lastly, the support vector machine is used to recognize different walking human targets. The simulation experiments considering two factors of height and velocity are also conducted to test the performance of this proposed method. | SUN ZhongSheng WANG Jun ZHANG YaoTian SUN JinPing YUAN ChangShun BI YanXian | 2015 | Science China Chemistry2015,58,12: | 1

Small-molecule amines:a big role in the regulation of bone homeostasis显示文摘Numerous small-molecule amines(SMAs)play critical roles in maintaining bone homeostasis and promoting bone regeneration regardless of whether they are applied as drugs or biomaterials. On the one hand,SMAs promote bone formation or inhibit bone resorption through the regulation of key molecular signaling pathways in osteoblasts/osteoclasts;on the other hand, owing to their alkaline properties as well as their antioxidant and anti-inflammatory features, most SMAs create a favorable microenvironment for bone homeostasis. However, due to a lack of information on their structure/bioactivity and underlying mechanisms of action, certain SMAs cannot be developed into drugs or biomaterials for bone disease treatment. In this review,we thoroughly summarize the current understanding of SMA effects on bone homeostasis, including descriptions of their classifications, biochemical features, recent research advances in bone biology and related regulatory mechanisms in bone regeneration. In addition,we discuss the challenges and prospects of SMA translational research. | Qian Zhang Jirong Yang Nan Hu Juan Liu Huan Yu Haobo Pan Di Chen Changshun Ruan | 2023 | Bone Research2023,11,3: | 0
