| 3 | Development and characterization of a clinical strain of Coxsackievirus A16 and an eGFP infectious clone显示文摘Coxsackievirus A16(CA16) is one of the major causes of hand, foot, and mouth disease(HFMD) worldwide, which is a common illness that affects children. The frequent occurrence of HFMD outbreaks has become a serious public health problem in Asia. Therefore, it is important to understand the pathogenesis and replication of CA16. In this study, a stable infectious c DNA clone of an epidemic strain of Coxsackievirus A16(CA16) was assembled, and subsequently a reporter virus(e GFP-CA16) was constructed by inserting the e GFP gene between the 5'-UTR and the N-terminus of VP4, with the addition of a 2A protease cleavage site(ITTLG) at its C-terminus. This was transfected into Vero cells to generate infectious recombinant viruses. The growth characteristics and plaque morphology, in vitro, in mammalian cells were found to be indistinguishable between the parental and recombinant viruses. Although the e GFP-CA16 showed smaller plaque size as compared to recombinant CA16, both were found to exhibit similar growth trends and EC50 of NITD008. In summary, this stable infectious c DNA clone should provide a valuable experimental system to study CA16 infection and host response. | 4 | Trans Complementation of Replication-defective Omsk Hemorrhagic Fever Virus for Antiviral Study显示文摘Omsk hemorrhagic fever virus(OHFV) is a tick-borne flavivirus classified as a biosafety level-4(BSL4) pathogen. Studies of OHFV are restricted to be conducted within BSL4 laboratories. Currently, no commercial vaccines or antiviral drugs are available against OHFV infection. In this study, we recovered a replication-deficient OHFV with an NS1 deletion(OHFVDNS1) and reporter virus replacing NS1 with the Gaussia luciferase(Gluc)(OHFV-ΔNS1-Gluc). Both the defective OHFVDNS1 and OHFV-ΔNS1-Gluc virus could only replicate efficiently in the BHK21 cell line expressing NS1(BHK21NS1) but not in na?ve BHK21 cells. The Gluc reporter gene of OHFV-ΔNS1-Gluc virus was maintained stably after serial passaging of BHK21NS1 cells and was used to surrogate the replication of OHFV. Using NITD008, OHFV-ΔNS1-Gluc virus was validated for antiviral screening, and high-throughput screening parameters were optimized in a 96-well plate format with a calculated Z0 value above 0.5. | 5 | Experimental study on freezing of liquids under static magnetic field显示文摘Freezing processes of several liquids under static magnetic field(SMF) less than 50 mT were investigated. Central temperature of liquid samples held in glass test tubes immersed in a liquid bath was measured and collected. Nucleation temperature and phase transition time were obtained from freezing curves. Normality tests were performed for nucleation temperature of these liquids with/without magnetic field and normality distributions were justified. Analysis of variances was carried out for nucleation temperature of these liquids with magnetic field flux density as the influencing factor. Results showed that no significant difference was found for deionized water with or without SMF. However, differences exist in 0.9% NaCl solution and 5% ethylene glycol solution with and without SMF. Nucleation temperature of 0.9% NaCl with SMF is lower than that without SMF, while its phase transition time is shorter than that without SMF. In this work, Sn02 nanoparticles with the particle size of 200 nm uniformly anchored on the surface of graphene oxide (GO) was prepared by combination of the ultrasonic method and the following calcination process. The Sn02/GO composite with the weight ratio of Sn02 to GO at 4:1 exhibits excellent electrochemical performance, which originates from the synergistic effects between GO and Sn02 nanoparticles. A high discharge capacity of 492 mA·h·g^-1 can be obtained after 100 cycles at 0.2C, and after cycling at higher current densities of 1C and 2C, a discharge capacity of 641 mA·h·g^-1 can be restored when the current density goes back to 0.1C. | 10 | In vitro comparison of the hemocompatibility of diamond-like carbon and carbon nitride coatings with different atomic percentages of N显示文摘Carbon nitride (CNx) and diamond-like carbon (DLC) coatings were prepared by dc magnetron sputtering at room temperature. Different partial pressures of N2 were used to synthesize CNx to evaluate the relationship between the atomic percentage of nitrogen and hemocompatibility. Auger electron spectroscopy and atomic force microscopy indicated atomic percentages of N of 0.12 and 0.22 and that the CNx coatings were smooth. An in vitro study of the hemocompatibility of the coatings revealed that both CNx coatings had better anticoagulant properties and lower platelet adhesion than DLC. Compared with CN0.12, the CN0.22 coating showed longer dynamic clotting time (about 42 min), static clotting time (23.6 min) and recalcification time (45.6 s), as well as lower platelet adhesion (102 cells μm 2), aggregation, and activation. | 13 | Double Position Servo Synchronous Drive System Based on Cross-Coupling Integrated Feedforward Control for Broacher显示文摘Synchronization errors directly deteriorate the machining accuracy of metal parts and the existed method cannot keep high synchronization precision because of external disturbances. A new double position servo synchronous driving scheme based on semi-closed-loop crosscoupling integrated feedforward control is proposed. The scheme comprises a position error cross-coupling feedforward control and a load torque identification with feedforward control. A digital integrated simulation system for the dual servo synchronous drive system is established.Using a 20 t servo broacher, performance analysis of the scheme is conducted based on this simulation system and the simulation results show that systems with traditional parallel or single control have problems when the worktable works with an unbalanced load. However, the system with proposed scheme shows good synchronous performance and positional accuracy. Broaching tests are performed and the experimental results show that the maximum dual axis synchronization error of the system is only 8 μm during acceleration and deceleration processes and the error between the actual running position and the given position is almost zero. | 15 | Chaos Particle Swarm Optimization Combined with Circular Median Filtering for Geophysical Parameters Retrieval from WindSat显示文摘This paper established a geophysical retrieval algorithm for sea surface wind vector, sea surface temperature, columnar atmospheric water vapor, and columnar cloud liquid water from WindSat, using the measured brightness temperatures and a matchup database. To retrieve the wind vector, a chaotic particle swarm approach was used to determine a set of possible wind vector solutions which minimize the difference between the forward model and the WindSat observations. An adjusted circular median filtering function was adopted to remove wind direction ambiguity. The validation of the wind speed, wind direction, sea surface temperature, columnar atmospheric water vapor, and columnar liquid cloud water indicates that this algorithm is feasible and reasonable and can be used to retrieve these atmospheric and oceanic parameters. Compared with moored buoy data, the RMS errors for wind speed and sea surface temperature were 0.92 m s^(-1) and 0.88℃, respectively. The RMS errors for columnar atmospheric water vapor and columnar liquid cloud water were 0.62 mm and 0.01 mm, respectively, compared with F17 SSMIS results. In addition, monthly average results indicated that these parameters are in good agreement with AMSR-E results. Wind direction retrieval was studied under various wind speed conditions and validated by comparing to the Quik SCAT measurements, and the RMS error was 13.3?. | 17 | A novel method of periodic orbit computation in circular restricted three-body problem显示文摘Periodic orbits are fundamental keys to understand the dynamical system of circular restricted three-body problem, and they play important roles in practical deep-space exploration. Current methods of periodic orbit computation need a high-order analytical approximate solution to start the iteration process, thus making the computation complicated and limiting the types of periodic orbits that can be obtained. By utilizing the symmetry of the restricted three-body problem, a special kind of flow function is constructed, so as to map a state on the plane of symmetry to another state that also lies in this plane. Based on this flow function, a new method of periodic orbit computation is derived. This method needs neither a starting analytic approximation nor the state transition matrix to be computed, so it can be conveniently implemented on a computer. Besides, this method is unaffected by the nonlinearity of the dynamical system, allowing a large set of periodic orbits which have x-z plane symmetry to be computed numerically. As examples, some planar periodic orbits (e.g. Lyapunov orbit) and spatial periodic orbits (e.g. Halo orbit) are computed. By further combining with a differential correction process, the method introduced here can be used to design resonant orbits that can jump between different resonant frequencies. | 20 | Impact of Taihu Lake on City Ozone in the Yangtze River Delta显示文摘The lake-breeze at Taihu Lake generates a different specific heat capacity between the water body and the surrounding land. Taihu Lake has a significant impact on the atmospheric conditions and the air quality in the Yangtze River Delta. This phenomenon is referred to as the Taihu Lake effect. In this study, two simulations were conducted to determine the impact of the Taihu Lake effect in the reference experiment(R-E) and sensitivity experiments(NO TH). The control simulations demonstrated that the meteorological field and the spatial distribution of ozone(O3) concentrations over Taihu lake obviously changed once the land-use type of water body was substituted by cropland. The surface temperature of Taihu Lake was reduced under the impact of Taihu Lake, and a huge temperature difference caused a strong lake-breeze effect. The results also showed that the difference in the average concentrations of O3 between the R-E and NO TH experiments reached 12 ppbv in most areas of Taihu Lake, all day, on 20 May 2014. During daytime(0800–1600 LST, LST=UTC+8), the influence of the Taihu Lake effect on O3 in the Suzhou region was not significant. However, the influence of the Taihu Lake effect on O3 in the Suzhou region was obvious during nighttime(1800–2400 LST).
