New eolian red clay sequence on the western Chinese Loess Plateau linked to onset of Asian desertification about 25 Ma ago显示文摘The expansion of inland Asia deserts has considerably influenced the environmental, social and economic activities in Asia. Aridification of inland Asia, especially timing of the initiation of Asian desertification, is a contentious topic in paleoclimatology. Late Cenozoic eolian loess-red clay sequences on the Chinese Loess Plateau, which possess abundant paleoclimatic and paleo-environmental information, can be regarded as an indicator of inland Asia desertification. Here we present a detailed magnetostratigraphic investigation of a new red clay sequence about 654 m in Zhuanglang located at the western Chinese Loess Plateau. Sedimentological, geochemical, mineralogical, and quartz morphological lines of evidence show that the red clay is of eolian origin. Magnetostratigraphic correlations indicate that this core sequence spans from 25.6 to 4.8 Ma, and typical eolian red clay appears as early as 25 Ma. This extends the lower limit of the red clay on the Chinese Loess Plateau from the previously thought early Miocene back into the late Oligocene. This new red clay record further implies that the inland Asia desertification was initiated at least by the late Oligocene. This sequence provides a unique high-resolution geological record for understanding the inland Asia desertification process since the late Oligocene. | QIANG XiaoKe AN ZhiSheng SONG YouGui CHANG Hong SUN YouBin LIU WeiGuo AO Hong DONG JiBao FU ChaoFeng WU Feng LU FengYan CAI YanJun ZHOU WeiJian CAO JunJi XU XinWen AI Li | 2011 | Science China Earth Sciences2011,54,1: | 73
Effects of natural covers on soil evaporation of the shelterbelt along the Tarim Desert Highway显示文摘The control of soil evaporation is one of important approaches to save water. The artificially simulated evaporation experiments have been conducted in the hinterland of the Taklimakan Desert to reveal the effects of the natural covers on the soil evaporation of the Tarim Desert Highway shelterbelt as well as provide some insights in the efficient utilization of water resources and optimization of irrigation systems. The results showed that (1) All the covers, including the sand deposit, the salt crust, the litter, the sand-litter mixed layer and so on, can significantly inhibit the soil water evaporation. Specifically, the daily evaporation, the total evaporation, and the evaporation rate in covered sands were much smaller than that of sands without cover. The cover inhibition effects increased with the cover thickness. Particularly, the soil evaporation of the covered sands was less affected by external and internal factors than that of the bare sands. Moreover, the variation of daily evaporation of covered sands was smaller than that of bare sands. The cumulative evaporation varied linearly with time in the covered sands whereas it varied logarithmically in the bare sands. In addition, the soil evaporation in the bare sands showed significantly different characteristics in the early and late stages of the evaporation. (2) All the covers exhibited the significant inhibiting effect on the soil evaporation, and the inhibition efficiency increased logarithmically with the cover thickness. However, as the cover thickness was above a certain value, the increase in the inhibition efficiency was slow. Particularly, at a cover thickness of 2 cm, there was no obvious difference in the inhibition efficiency among all kinds of covers. The maximum inhibition efficiency as calculated from the daily evaporation on the first day of irrigation was: sand-litter mixed layer (79.92%) > litter layer (78.96%) > salt crust (75.58%) > sand bed (74.11%), whereas the average inhibiting efficiency as calculated from the cumulative soil evaporation at the end of an irrigation cycle (the fourth day) was: salt crust (67.78%) > sand-litter mixed layer (66.72%) > sand deposit (63.28%) > litter layer (61.74%). | ZHOU HongWei LI ShengYu SUN ShuGuo XU XinWen LEI JiaQiang LIU Shang DU WenYi YAN Zeng WANG YongChang | 2008 | Chinese Science Bulletin2008,53,S2: | 11
Ecological stability of Tarim Desert Highway shelterbelt显示文摘The Tarim Desert Highway shelterbelt, located in hinterland of Taklimakan Desert, is irrigated by underground saline water, with three to thirty gram per litter mineral degrees. The sustain-ability and stability are affected by multifarious stress. The structural and functional characteristics of shelterbelt are studied to probe into correlation between environment and shelterbelt. On basis, decision analysis is applied to study ecological stability of the Tarim Desert Highway shelterbelt, to screen out limited factors, to establish general index system, and to evaluate the stability of the shelterbelt nowadays. Finally, the concept of ecological stability is utilized to manage the artificial ecosystem. The results show that the artificial ecosystem is relatively flimsy, whose stability can be increased by adjusting stand structure and improving the nutrient cycle. | ZHOU Zhibin XU Xinwen LEI Jiaqiang LI Shengyu | 2006 | Chinese Science Bulletin2006,51,A01: | 8
Host metabolism dysregulation and cell tropism identification in human airway and alveolar organoids upon SARS-CoV-2 infection显示文摘The coronavirus disease 2019(COVID-19)pandemic is caused by infection with the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),which is spread primary via respiratory droplets and infects the lungs.Currently widely used cell lines and animals are unable to accurately mimic human physiological conditions because of the abnormal status of cell lines(transformed or cancer cells)and species differences between animals and humans.Organoids are stem cell-derived selforganized three-dimensional culture in vitro and model the physiological conditions of natural organs. Here we showed that SARS-CoV-2 infected and extensively replicated in human embryonic stem cells(hESCs)-derived lung organoids, including airway and alveolar organoids which covered the complete infection and spread route for SARS-CoV-2 within lungs. The infected ceils were ciliated, club, and alveolar type 2(AT2)cells,which were sequentially located from the proximal to the distal airway and terminal alveoli,respectively.Additionally,RNA-seq revealed early cell response to virus infection including an unexpected downregulation of the metabolic processes, especially lipid metabolism,in addition to the well-known upregulation of immune response.Further,Remdesivir and a human neutralizing antibody potently inhibited SARS-CoV-2 replication in lung organoids. Therefore, human lung organoids can serve as a pathophysiological model to investigate the underlying mechanism of SARS-CoV-2 infection and to discover and test therapeutic drugs for COVID-19. | Rongjuan Pei Jianqi Feng Yecheng Zhang Hao Sun Lian Li Xuejie Yang Jiangping He Shuqi Xiao Jin Xiong Ying Lin Kun Wen Hongwei Zhou Jiekai Chen Zhili Rong Xinwen Chen | 2021 | Protein & Cell2021,12,9: | 7
Productive HBV infection of well-differentiated, hNTCP-expressing human hepatoma-derived(Huh7) cells显示文摘Feasible and effective cell models for hepatitis B virus(HBV) infection are required for investigating the complete lifecycle of this virus, including the early steps of viral entry. Resistance to dimethyl sulfoxide/polyethylene glycol(DMSO/PEG), h NTCP expression, and a differentiated state are the limiting factors for successful HBV infection models. In the present study, we used a hepatoma cell line(Hu7^(hDNTCPh)) to overcome these limiting factors so that it exhibits excellent susceptibility to HBV infection. To achieve this goal, different hepatoma cell lines were tested with 2.5% DMSO/4%PEG8000, and one resistant cell line(Huh7 D) was used to construct a stable h NTCP-expressing cell line(Hu7^(hDNTCPh)) using a recombinant lentivirus system. Then, the morphological characteristics and differentiation molecular markers of Hu7^(hDNTCPh) cells with or without DMSO treatment were characterized. Finally, the susceptibility of Hu7^(hDNTCPh) cells to HBV infection was assessed. Our results showed that Huh7 D cells were resistant to 2.5% DMSO/4% PEG8000, whereas the others were not. Hu7^(hDNTCPh) cells were established to express a high level of h NTCP compared to liver extracts, and Hu7^(hDNTCPh) cells rapidly transformed into a non-dividing, well-differentiated polarized phenotype under DMSO treatment. Hu7^(hDNTCPh) cells fully supported the entire lifecycle of HBV infection. This cell culture system will be useful for the analysis of host-virus interactions, which should facilitate the discovery of antiviral drugs and vaccines. | Ming Zhou Kaitao Zhao Yongxuan Yao Yifei Yuan Rongjuan Pei Yun Wang Jizheng Chen Xue Hu Yuan Zhou Xinwen Chen Chunchen Wu | 2017 | Virologica Sinica2017,32,6: | 7
Site type classification for the shelter-forest ecological project along the Tarim Desert Highway显示文摘Site types of the afforestation region of the shelter-forest ecological project along the Tarim Desert Highway were classified based on the natural conditions and windblown sand damages. The extremely severe environment, the irrigation with saline water, and large-scale linear project makes this classification of site types most unique and significant. It adopted a three-level classification system integrating the dominant factors and restrictive factors in regard to their impacts on plant survival and growth as well as on the protective property. Six site type districts were classified based on the medium-scale geomorphic unit, the windblown sand damages, and the major production facilities; 21 site type groups were obtained according to the small-scale geomorphic type, terrain, and wind regime; 36 site types were further classified based on the salt contents of the underground water and soil types. Especially, in this study, spatial distribution of the six site type districts along the desert highway is continuous, which is unique and different from that of most other classifications. In addition, the salt-stress tolerance threshold of the main afforestation plant species to underground water have been set to 8 g/L and 15 g/L according to selective breeding tests and the salinity spatial distribution of the underground water. Thus, the underground water with salinity lower than 8 g/L is defined as light saline water in this area. | LI BingWen XU XinWen LEI JiaQiang QIU YongZhi XU Bo ZHOU HongWei WANG Qiang WANG BO SU Wei | 2008 | Chinese Science Bulletin2008,53,S2: | 6
The temporal and spatial fluctuation of the groundwater level along the Tarim Desert Highway显示文摘In this work, a groundwater monitor section along the Tarim Desert Highway was set up, and the groundwater level data were recorded. The temporal and spatial fluctuations of the groundwater level were analyzed based on the groundwater level measurements, hydrologic data, the groundwater/surface water interconversion theory and the groundwater dynamics. The spatial distribution of groundwater is mostly affected by sand dune height and landform variation. The four primary temporal influence factors of groundwater level can be ordered in decreasing significance as: shelterbelt pumping > vertical leakage > river flow change > runoff replenishment. The runoff replenishment has the biggest influence range, and the shelterbelt pumping has the smallest influence range. The groundwater level fluctuates annually in a natural pattern. | FAN JingLong XU XinWen LEI JaQiang ZHAO JingFeng LI ShengYu WANG HaiFeng ZHANG JianGuo ZHOU HongWei | 2008 | Chinese Science Bulletin2008,53,S2: | 6
Phosphatidylserine-Specific Phospholipase A1 is the Critical Bridge for Hepatitis C Virus Assembly显示文摘The phosphatidylserine-specific phospholipase A1(PLA1A)is an essential host factor in hepatitis C virus(HCV)assembly. In this study,we mapped the E2,NS2 and NS5A involved in PLA1A interaction to their lumenal domains and membranous parts, through which they form oligomeric protein complexes to participate in HCV assembly.Multiple regions of PLA1A were involved in their interaction and complex formation.Furthermore, the results represented structures with PLA1A and E2 in closer proximity than NS2 and NS5A,and strongly suggest PLA1 A-E2,s physical interaction in cells.Meanwhile,we mapped the NS5A sequence which participated in PLA1A interaction with the C-terminus of domain 1.Interestingly,these amino acids in the sequence are also essential for viral RNA replication.Further experiments revealed that these four proteins interact with each other.Moreover,PLA1A expression levels were elevated in livers from HCV-infected patients. In conclusion,we exposed the structural determinants of PLA1A,E2,NS2 and NS5A proteins which were important for HCV assembly and provided a detailed characterization of PLA1A in HCV assembly. | Qi Yang Min Guo Yuan Zhou Xue Hu Yun Wang Chunchen Wu Min Yang Rongjuan Pei Xinwen Chen Jizheng Chen | 2019 | Virologica Sinica2019,34,5: | 4
Heat treatment bimetallic PdAu nanocatalyst for oxygen reduction reaction显示文摘Pd-based nanocatalyst is a potential oxygen reduction oxidation(ORR)catalyst because of its high activity in alkaline medium and low cost.In this work, bimetallic Pd Au nanocatalysts are prepared by one-pot hydrothermal method using triblock pluronic copolymers, poly(ethylene oxide)-poly(propylene oxide)-poly(ethylene oxide)(PEO19-PPO69-PEO19)(P123)as reducer and stabilizer, and heat-treatment method is applied to regulate catalyst structure and improve catalyst activity. The results show that the heat treatment can agglomerate the catalyst to a certain extent, but effectively improve the crystallinity and alloying degree of the catalyst. The ORR performance of the Pd Au nanocatalysts obtained under different heat treatment conditions is systematically investigated.Compared with commercial Pd black and Pd Au catalyst before heat treatment, the ORR performance of Au Pd nanocatalyst obtained after heat treatment for one hour at 500℃ has been enhanced. The Pd Au nanocatalysts after heat treatment also display enhanced anti-methanol toxicity ability in acidic medium. | Qingyun Hu Wei Zhan Yifei Guo Laiming Luo Ronghua Zhang Di Chen Xinwen Zhou | 2020 | Journal of Energy Chemistry2020,29,1: | 3
Host HDAC4 regulates the antiviral response by inhibiting the phosphorylation of IRF3显示文摘Class II HDACs, such as HDAC4, are critical regulators of the immune response in various immune cells;however, its role in innate immunity remains largely unknown. Here, we report that the overexpression of HDAC4 suppresses the production of type I interferons triggered by pattern-recognition receptors (PRRs). HDAC4 repressed the translocation of transcription factor IRF3 to the nucleus, thereby decreasing IRF3-mediated IFN-β expression. In particular, we also determined that HDAC4 can be phosphorylated and simultaneously block the phosphorylation of IRF3 at Ser386 and Ser396 by TBK1 and IKKε, respectively, by interacting with the kinase domain of TBK1 and IKKε. Furthermore, IFN-β may stimulate the expression of HDAC4. Our findings suggest that HDAC4 acts as a regulator of PRR signaling and is a novel mechanism of negative feedback regulation for preventing an overreactive innate immune response. | Qi Yang Jielin Tang Rongjuan Pei XiaoXiao Gao Jing Guo Chonghui Xu Yun Wang QianWang Chunchen Wu Yuan Zhou Xue Hu He Zhao Yanyi Wang Xinwen Chen Jizheng Chen | 2019 | Journal of Molecular Cell Biology2019,11,2: | 2
Porous carbon coupled with an interlaced MoP–MoS2 heterojunction hybrid for efficient hydrogen evolution reaction显示文摘The design and development of electrocatalysts composed of non-noble-metal catalysts with both large surface area and high electrical conductivities are crucial for the hydrogen evolution reaction(HER).Here,a xylose-based porous carbon is coupled with a MoS2-Mo P heterojunction(MoS2-Mo P/FPC)hybrid and used as a promising catalyst for HER.The hybrid is prepared by immobilizing petal-like MoS2 nanosheets on porous carbon(MoS2/FPC),followed by controlling the phosphidation in Ar/H2 to form MoS2-Mo P/FPC.Red phosphorus provides the P species that can induce the construction of the heterojunction under the reducing atmosphere, along with the generation of a Mo P phase and the splitting of the MoS2 phase. The as-prepared MoS2-Mo P/FPC catalyst offers a low overpotential of 144 mV at a current density of 10 m A cm^-2 and a small Tafel slope of 41 m V dec^-1 for the HER in acidic media,as well as remarkable stability.Apart from the active nature of the hybrid, its outstanding activity is attributed to the MoS2-Mo P heterojunction, and the good charge/mass-transfer ability of porous carbon. This strategy provides a new method to develop and design low-cost and high-performance catalysts for the HER. | Qiusheng Zhou Jianrui Feng Xinwen Peng Linxin Zhong Runcang Sun | 2020 | Journal of Energy Chemistry2020,29,6: | 2
Nascent Proteome and Glycoproteome Reveal the Inhibition Role of ALG1 in Hepatocellular Carcinoma Cell Migration显示文摘Asparagine-linked glycosylation protein 1 homolog(ALG1)participates in the initial stage of protein N-glycosylation and N-glycosylation has been implicated in the process of hepatocellular carcinoma(HCC)progression. However, whether ALG1 plays a role in human HCC remains unknown. In this study, the expression profile of ALG1 in tumorous and corresponding adjacent non-tumor tissues was analyzed. The relationship of ALG1 expression with clinical features and prognosis of HCC patients was also evaluated using immuno-histochemical method. Here we found ALG1 decreased in HCC tissues compared with adjacent normal liver tissues, which predicted an unfavorable prognosis.Combined with RNA interference, nascent proteome and glycoproteome were determined systematically in Huh7 cell line.Bioinformatics analysis indicated that the differentially expressed proteins participating in the response of ALG1 knockdown were most significantly associated with cell-cell adhesion.Functional studies confirmed that knockdown of ALG1 reduced cell adhesion capacity, and promoted cell migration.Furthermore,down-regulation of H8N2(on N-glycosite N651)and H5N4S2F1(on N-glycosite N692)from N-cadherin was identified as a feature of ALG1 knockdown. Our findings revealed that ALG1 controlled the expression of glycosylated N-cadherin and played a role in HCC migration, with implications for prognosis. | Xinyi Cao Yuyin Shao Peiyi Meng Zhao Cao Guoquan Yan Jun Yao Xinwen Zhou Chao Liu Lei Zhang Hong Shu Haojie Lu | 2022 | Phenomics2022,2,4: | 2
Histone Deacetylase 3 Inhibitor Suppresses Hepatitis C Virus Replication by Regulating Apo-A1 and LEAP-1 Expression显示文摘Histone deacetylase (HDAC) inhibitors show clinical promise for the treatment of cancers, including hepatocellular carcinoma (HCC). In this study, we investigated the effect of HDAC inhibitor treatment on hepatitis C virus (HCV)replication in Huh7 human liver cells and in a mouse model of HCV infection. Viral replication was markedly suppressed by the HDAC3 inhibitor at concentrations below 1 mmol/L, with no cellular toxicity. This was accompanied by upregulation of liver-expressed antimicrobial peptide 1 (LEAP-1) and downregulation of apolipoprotein-A1 (Apo-A1), as determined by microarray and quantitative RT-PCR analyses. Moreover, HDAC3 was found to modulate the binding of CCAAT-enhancer-binding protein a (C/EBPa), hypoxia-inducible factor 1 a (HIF1 a), and signal transducer and activator of transcription 3 (STAT3) to the LEAP-1 promoter. HDAC3 inhibitor treatment also blocked HCV replication in a mouse model of HCV infection. These results indicate that epigenetic therapy with HDAC3 inhibitor may be a potential treatment for diseases associated with HCV infection such as HCC. | Yuan Zhou Qian Wang Qi Yang Jielin Tang Chonghui Xu Dongwei Gai Xinwen Chen Jizheng Chen | 2018 | Virologica Sinica2018,33,5: | 2

The dynamics variation of soil moisture of shelterbelts along the Tarim Desert Highway显示文摘We studied the variation of soil moisture as well as its regularity over the irrigation cycle at shelterbelts along the Tarim Desert Highway at different site types and different planting years. The results show that: (1) There is an obvious temporal variation of soil moisture within a typical irrigation period in shelterbelts along the Tarim Desert Highway, and the soil water storage varied linearly with the number of days after irrigation. Along the direction perpendicular to the soil top, the soil profile can be divided into four layers and each shows different dynamics of soil moisture variation, including the quickly changing layer (0—20 cm), the active layer (20—60 cm), the weakly layer (60—100 cm), and the regulated layer (under 100 cm). (2) Both the soil moisture and soil water content decreased gradually with the number of planting year, while the soil water deficit increased. It indicates that shelterbelts along the Tarim Desert Highway can retain the water accumulated from previous years. (3) The soil water storage of harden sand is the maximum among all types of sites. Specifically, it is about 1.58 times higher than that of longitudinal dune, 1.15 times higher than clay, and 1.43 times higher than flat sand. Its soil water deficit was over 900 mm. | WANG YongDong XU XinWen LEI JiaQiang LI ShengYu ZHOU ZhiBin CHANG Qing WANG LuHai GU Feng QIU YongZhi XU Bo | 2008 | Chinese Science Bulletin2008,53,S2: | 2
Development of the novel ACLY inhibitor 326E as a promising treatment for hypercholesterolemia显示文摘Hepatic cholesterol accumulation is an important contributor to hypercholesterolemia, which results in atherosclerosis and cardiovascular disease(CVD).ATP-citrate lyase(ACLY)is a key lipogenic enzyme that converts cytosolic citrate derived from tricarboxylic acid cycle(TCA cycle)to acetyl-CoA in the cytoplasm. Therefore,ACLY represents a link between mitochondria oxidative phosphorylation and cytosolic de novo lipogenesis. In this study,we developed the small molecule 326E with an enedioic acid structural moiety as a novel ACLY inhibitor, and its CoA-conjugated form 326E-CoA inhibited ACLY activity with an IC_(50)=5.31±1.2μmol/L in vitro.326E treatment reduced de novo lipogenesis, and increased cholesterol efflux in vitro and in vivo.326E was rapidly absorbed after oral administration, exhibited a higher blood exposure than that of the approved ACLY inhibitor bempedoic acid(BA)used for hypercholesterolemia.Chronic 326E treatment in hamsters and rhesus monkeys resulted in remarkable improvement of hyperlipidemia.Once daily oral administration of 326E for 24 weeks prevented the occurrence of atherosclerosis in ApoE^(-/-)mice to a greater extent than that of BA treatment.Taken together, our data suggest that inhibition of ACLY by 326E represents a promising strategy for the treatment of hypercholesterolemia. | Zhifu Xie Mei Zhang Qian Song Long Cheng Xinwen Zhang Gaolei Song Xinyu Sun Min Gu Chendong Zhou Yangming Zhang Kexin Zhu Jianpeng Yin Xiaoyan Chen Jingya Li Fajun Nan | 2023 | Acta Pharmaceutica Sinica B2023,13,2: | 1
