1Multimodality treatment in hepatocellular carcinoma patients with tumor thrombi in portal vein显示文摘AIM To compare the therapeutic effect andsignificances of multimodality treatment forhepatocellular carcinoma (HCC) with tumorthrombi in portal vein (PVTT).METHODS HCC patients (n = 147) with tumortrombi in the main portal vein or the first branchof portal vein were divided into four groups bythe several therapeutic methods. There wereconservative treatment group in 18 out ofpatients (group A); and hepatic artery ligation(HAL) and/or hepatic artery infusion (HAl)group in 18 patients (group B), in whompostoberative chemoembolization was doneperiodically; group of removal of HCC with PVTTin 79 (group C) and group of transcatheterhepatic arterial chemoembolization (TACE) orHAl and/or portal vein infusion (PVI) afteroperation in 32 (group D).RESULTS The median survival period was 12months in our series and the 1-, 3-, and 5-yearsurvival rates were 44.3%, 24.5% and 15.2%,respectively. The median survival times were 2,5, 12 and 16 months in group A, B, C and D,respectively. The 1-, 3- and 5-year survival rateswere 5.6%, 0% and 0% in group A; 22.2%,5.6% and 0% in group B; 53.9%, 26.9% and16.6% in group C; 79.3%, 38.9% and 26.8% ingroup D, respectively. 2A three-dimensional variational ocean data assimilation system:Scheme and preliminary results显示文摘A new 3DVAR-based Ocean Variational Analysis System (OVALS) is developed. OVALS is capable of assimilating in situ sea water temperature and salinity observations and satellite altimetry data. As a component of OVALS, a new variational scheme is proposed to assimilate the sea surface height data. This scheme considers both the vertical correlation of background errors and the nonlinear temperature-salinity relationship which is derived from the generalization of the linear balance constraints to the nonlinear in the 3DVAR. By this scheme, the model temperature and salinity fields are directly adjusted from the altimetry data. Additionally, OVALS can assimilate the temperature and salinity profiles from the ARGO floats which have been implemented in recent years and some temperature and salinity data such as from expendable bathythermograph, moored ocean buoys, etc. A 21-year assimilation experiment is carried out by using OVALS and the Tropical Pacific circulation model. The results show that the assimilation system may effectively improve the estimations of temperature and salinity by assimilating all kinds of observations. 3A study on relationship of nitric oxide,oxidation,peroxidation,lipoperoxidation with chronic cholecystitis显示文摘AIM To study relationship of injury induced bynitric oxide,oxidation,peroxidation,lipoperoxidation with chronic cholecystitis.METHODS The values of plasma nitric oxide(P-NO),plasma vitamin C(P-VC),plasma vitamin E(P-VE),plasma β-carotene(P-β-CAR),plasmalipoperoxides(P-LPO),erythrocyte superoxidedismutase(E-SOD),erythrocyte catalase(E-CAT),erythrocyte glutathione peroxidase(E-GSH-Px)activities and erythrocyte lipoperoxides(E-LPO)level in 77 patients with chronic cholecystitisand 80 healthy control subjects were determined,differences of the above average values betweenthe patient group and the control group anddifferences of the average values betweenpreoperative and postoperative patients wereanalyzed and compared,linear regression andcorrelation of the disease course with the abovedetermination values as well as the stepwiseregression and correlation of the course with thevalues were analyzed.RESULTS Compared with the control group,theaverage values of P-NO,P-LPO,E-LPO weresignificantly increased(P<0.01),and of P-VC, P-VE,P-β-CAR,E-SOD,E-CAT and E-GSH-Pxdecreased(P<0.01)in the patient group.Theanalysis of the linear regression and correlationshowed that with prolonging of the course,thevalues of P-NO,P-LPO and E-LPO in the patientswere gradually ascended and the values of P-VC,P-VE,P-β-CAR,E-SOD,E-CAT and E-GSH-Pxdescended(P<0.01).The analysis of thestepwise regression and correlation indicated thatthe correlation of the course with P-NO,P-VE andP-β-CAR values was the closest.Compared withthe preoperative patients,the average values of P-NO,P-LPO and E-LPO were significantlydecreased(P<0.01)and the average values of P-VC,E-SOD,E-CAT and E-GSH-Px in postoperativepatients increased(P<0.01)in postoperativepatients.But there was no significant difference inthe average values of P-VE,P-β-CAR preoperativeand postoperative patients.CONCLUSION Chronic cholecystitis could inducethe increase of nitric oxide,oxidation,peroxidation and lipoperoxidation.Jun Fu Zhou Dong Cai You Gen Zhu Jin Lu Yang Cheng Hong Peng Yang Hai Yu 2000World Journal of Gastroenterology2000,6,4:36
4Th22 cells are associated with hepatocellular carcinoma development and progression显示文摘Objective: IL-22-producing CD4+ T helper cells(Th22 cells) have been identified as major inducers of tissue inflammation and immune responses. Currently, no previous study explored the role of Th22 cells in the pathogenesis of hepatocellular carcinoma(HCC). The study aimed to determine the biological function of Th22 cells and its effector IL-22 in HCC patients.Methods: Forty-five HCC patients and 19 healthy controls were recruited and their peripheral blood was collected. The fresh HCC tissues, adjacent HCC tissues and ten normal liver tissues were also collected. Flow cytometry analysis was used to determine the frequencies of circulating Th22 cells and Th17 cells. Serum IL-22 levels were tested by enzyme-linked immunosorbent assay(ELISA). Immunohistochemical staining and real-time polymerase chain reaction(PCR) were used to detect IL-22 protein and mRNA in tissues specimens, respectively.Results: Circulating Th22 cells, Th17 cells and serum IL-22 levels were significantly elevated in HCC patients compared with those of healthy controls(P<0.001). Th22 cells were showed to be positively correlated with IL-22 in HCC patients(P<0.05), but not in healthy controls. No significant differences were found in HCC patients with HBeAg positivity or negativity in term of Th22 cells and serum IL-22 levels. The expression of IL-22 protein and mRNA was highest in HCC tissues, followed by adjacent HCC tissues and normal liver tissues. 5DNA-based vaccination induces humoral and cellular immune responses against hepatitis B virus surface antigen in mice without activation of Cmyc显示文摘AIM To develop a safe and effective DNAvaccine for inducing humoral and cellularimmunological responses against hepatitis Bvirus surface antigen(HBsAg).METHODS BALB/c mice were inoculated withNV-HB/s,a recombinant plasmid that had beeninserted S gene of hepatitis B virus genome andcould express HBsAg in eukaryotes.HBsAgexpression was measured by ABC immunohis-tochemical assay,generation of anti-HBs byELISA and cytotoxic T lymphocyte(CTL),byMTT method,existence of vaccine DNA bySouthern blot hybridization and activation ofoncogene C-myc by in situ hybridization,RESULTS With NV-HB/s vaccination byintramuscular injection,anti-HBs was initiallypositive 2 weeks after inoculation while all micetested were HBsAg positive in the muscles.Thetiters and seroconversion rate of anti-HBs weresteadily increasing as time went on and weredose-dependent.All the mice inoculated with100 μg NV-HB/s were anti-HBs positive onemonth after inoculation,the titer was 1:1024 ormore.The humoral immune response was similarinduced by either intramuscular or intradermalinjection.CTL activities were much stronger(45.26%)in NV-HB/s DNA immunized mice as compared with those(only 6%)in plasma-derived HBsAg vaccine immunized mice.Twomonths after inoculation,all muscle sampleswere positive by Southern-blot hybridization forNV.HB/s DNA detection,but decreased to 25%and all were undetectable by in situhybridization after 6 months.No oncogene C-myc activation was found in the muscle ofinoculation site.CONCLUSION NV-HB/s could generatehumoral and cellular immunological responsesagainst HBsAg that had been safely expressed insitu by NV-HB/s vaccination.Lian San Zhao Shan Qin Tao You Zhou Hong Tang Li Liu Bing Jun Lei 2000World Journal of Gastroenterology2000,6,2:24
6Screening for gastric cancer in China:Advances,challenges and visions显示文摘Gastric cancer(GC)is one of the major cancers in China and all over the world.Most GCs are diagnosed at an advanced stage with unfavorable prognosis.Along with some other countries,China has developed the government-funded national screening programs for GC and other major cancers.GC screening has been shown to effectively decrease the incidence of and mortality from GC in countries adopting nationwide screening programs(Japan and Korea)and in studies based on selected Chinese populations.The screening of GC relies mostly on gastroendoscopy,the accuracy,reliability and safety of which have been indicated by previous studies.However,considering its invasive screening approach,requirements on skilled endoscopists and pathologists,and a high cost,developing noninvasive methods to amend endoscopic screening would be highly needed.Numerous studies have examined biomarkers for GC screening and the combination of biomarkers involving pepsinogen,gastrin,and Helicobacter pylori antibodies has been proposed for risk stratification,seeking to narrow down the high-risk populations for further endoscopy.Despite all the achievements of endoscopic screening,evidence on appropriate screening age,intervals for repeated screening,novel biomarkers promoting precision prevention,and health economics need to be accumulated to inform policymakers on endoscopic screening in China.With the guide of Health China 2030 Planning Outline,we have golden opportunities to promote prevention and control of GC.In this review,we summarize the characteristics of screening programs in China and other East Asian countries and introduce the past and current approaches and strategies for GC screening,aiming for featuring the latest advances and key challenges,and illustrating future visions of GC screening.Xiaohan Fan Xiangxiang Qin Yang Zhang Zhexuan Li Tong Zhou Jingying Zhang Weicheng You Wenqing Li Kaifeng Pan 2021Chinese Journal of Cancer Research2021,33,2:21

11Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. 12Detection of Genetically Modified Crops by Combination of Multiplex PCR and Low-density DNA Microarray显示文摘Objective To develop a technique for simultaneous detection of various target genes in Roundup Ready soybean by combining multiplex PCR and low-density DNA microarray. Methods Two sets of the multiplex PCR system were used to amplify the target genes in genetically modified (GM) soybean. Seventeen capture probes (PCR products) and 17 pairs of corresponding primers were designed according to the genetic characteristics of Rroundup Ready soybean (GTS40-3-2), maize (Mon810, Nk603, GA21), canola (T45, MS1/RF1), and rice (SCK) in many identified GM crops. All of the probes were categorized and identified as species-specific probes. One negative probe and one positive control probe were used to assess the efficiency of all reactions, and therefore eliminate any false positive and negative results. After multiplex PCR reaction, amplicons were adulterated with Cy5-dUTP and hybridized with DNA microarray. The array was then scanned to display the specific hybridization signals of target genes. The assay was applied to the analysis of sample of certified transgenic soybean (Roundup Ready GTS40-3-2) and canola (MS1/RF1). Results A combination technique of multiplex PCR and DNA microarray was successfully developed to identify multi-target genes in Roundup Ready soybean and MS1/RF1 canola with a great specificity and reliability. Reliable identification of genetic characteristics of Roundup Ready of GM soybean from genetically modified crops was achieved at 0.5% transgenic events, indicating a high sensitivity. 15Molecular Epidemiological Characteristics of Streptococcus pyogenes Strains Involved in an Outbreak of Scarlet Fever in China,2011显示文摘Objective To investigate molecular characterization of streptococcus pyogenes isolates involved in an outbreak of scarlet fever in China in 2011.Methods Seventy-four Streptococcal pyogenes involved in an outbreak of scarlet fever were isolated from pediatric patients in the areas with high incidence in China from May to August of 2011.Emm genotyping,pulsed-field gel electrophoresis (PFGE),superantigen (SAg) genes and antimicrobial susceptibility profiling were analyzed for these isolates.Results A total of 4 different emm types were identified.Emm12 was the most prevalent type which contained four predominating PFGE patterns corresponding to four different virulence and superantigen profiles.Emm12 (79.7%) and emml (14.9%) accounted for approximately 94%of all the isolates.The speA gene was all negative in emm12 isolates and positive in emml isolates.All strains were resistant to erythromycin,and 89.4%of them were resistant to erythromycin,tracycline,and clindamycin simultaneously.Conclusion Several highly diversified clones with a high macrolide resistance rate comprise a predominant proportion of circulating strains,though no new emm type was found in this outbreak.The data provide a baseline for further surveillance of scarlet fever,which may contribute to the explanation of the outbreak and development of a GAS vaccine in China.YOU Yuan Hai SONG Yan Yan YAN Xiao Mei WANG Hai Bin ZHANG Meng Han TAO Xiao Xia Li Lei Lei ZHANG Yu Xin JIANG Xi Hong ZHANG Bing Hua ZHOU Hao XIAO Di JIN Lian Mei FENG Zi Jian LUO Feng Ji ZHANG Jian Zhong 2013Biomedical and Environmental Sciences2013,26,11:15

18Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection.
