2OSBPL2-disrupted pigs recapitulate dual features of human hearing loss and hypercholesterolaemia显示文摘Oxysterol binding protein like 2(OSBPL2), an important regulator in cellular lipid metabolism and transport, was identified as a novel deafness-causal gene in our previous work. To resemble the phenotypic features of OSBPL2 mutation in animal models and elucidate the potential genotypephenotype associations, the OSBPL2-disrupted Bama miniature(BM) pig model was constructed using CRISPR/Cas9-mediated gene editing, somatic cell nuclear transfer(SCNT) and embryo transplantation approaches, and then subjected to phenotypic characterization of auditory function and serum lipid profiles. The OSBPL2-disrupted pigs displayed progressive hearing loss(HL) with degeneration/apoptosis of cochlea hair cells(HCs) and morphological abnormalities in HC stereocilia, as well as hypercholesterolaemia. High-fat diet(HFD) feeding aggravated the development of HL and led to more severe hypercholesterolaemia. 5Comprehensive study of crystalline AlN/sapphire templates after high-temperature annealing with various sputtering conditions显示文摘High-quality AlN/sapphire templates were fabricated by the combination of sputtering and high-temperature(HT)annealing.The influence of sputtering parameters including nitrogen flux,radio frequency power,and substrate temperature on the crystalline quality and surface morphology of annealed AlN films were investigated.With lower substrate temperature,lower power,and lower N2 flux,the full width at half maximum of the X-ray rocking curve for AlN(0002)and(102)were improved to 97.2 and 259.2 arcsec after high-temperature annealing.This happens because the increased vacancy concentration of sputtered AlN films can facilitate the annihilation of dislocations by increasing the recovery rate during HT annealing.Step and step-bunching morphologies were clearly observed with optimized sputtering conditions.Wen Gu Zhibin Liu Yanan Guo Xiaodong Wang Xiaolong Jia Xingfang Liu Yiping Zeng Junxi Wang Jinmin Li Jianchang Yan 2020Journal of Semiconductors2020,41,12:1

7Design of a wideband common-mode filter using approximate closed-loop pattern of compact C-shaped defected ground structure显示文摘A compact common-mode filter is proposed to suppress common-mode noise for application of high-speed differential signal traces. The filter adopts one big C-shaped defected ground structure(DGS) cell in the left of ground plane and two small C-shaped DGS cells with opposite direction in the right of ground plane. Because these DGS cells have different dimensions, the filter has three adjacent equivalent resonant points, which can suppress wideband common-mode noise effectively. The left C-shaped DGS and its adjacent C-shaped DGS cell form an approximate closed structure, which can efficiently reduce the influence of the mutual capacitance. The filter provides a common-mode suppression from 3.6 GHz to 14.4 GHz over 15 dB while it has a small size of 10 mm × 10 mm. The fractional bandwidth of the filter is 120%, and the differential signals still keep good signal integrity. 10Identification of A-to-I RNA editing profiles and their clinical relevance in lung adenocarcinoma显示文摘Adenosine-to-inosine(A-to-I)RNA editing is a widespread posttranscriptional modification that has been shown to play an important role in tumorigenesis.Here,we evaluated a total of 19,316 RNA editing sites in the tissues of 80 lung adenocarcinoma(LUAD)patients from our Nanjing Lung Cancer Cohort(NJLCC)and 486 LUAD patients from the TCGA database.The global RNA editing level was significantly increased in tumor tissues and was highly heterogeneous across patients.The high RNA editing level in tumors was attributed to both RNA(ADAR1 expression)and DNA alterations(mutation load).Consensus clustering on RNA editing sites revealed a new molecular subtype(EC3)that was associated with the poorest prognosis of LUAD patients.Importantly,the new classification was independent of classic molecular subtypes based on gene expression or DNA methylation.We further proposed a simplified model including eight RNA editing sites to accurately distinguish the EC3 subtype in our patients.The model was further validated in the TCGA dataset and had an area under the curve(AUC)of the receiver operating characteristic curve of 0.93(95%CI:0.91-0.95).In addition,we found that LUAD cell lines with the EC3 subtype were sensitive to four chemotherapy drugs.These findings highlighted the importance of RNA editing events in the tumorigenesis of LUAD and provided insight into the application of RNA editing in the molecular subtyping and clinical treatment of cancer.Cheng Wang Mingtao Huang Congcong Chen Yuancheng Li Na Qin Zijian Ma Jingyi Fan Linnan Gong Hui Zeng Liu Yang Xianfeng Xu Jun Zhou Juncheng Dai Guangfu Jin Zhibin Hu Hongxia Ma Fengwei Tan Hongbing Shen 2022Science China(Life Sciences)2022,65,1:1

13SARS-CoV-2 Surveillance Through China Influenza Surveillance Information System—China,December 1,2022 to February 12,2023显示文摘Introduction:The World Health Organization(WHO)proposed using influenza surveillance systems to carry out coronavirus disease 2019(COVID-19)surveillance due to the similarity between the two diseases in some respiratory symptoms.To assess the prevalence of COVID-19,we analyzed the influenzalike illness(ILI)and positive rate of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)detections in ILI patients reported to the influenza Surveillance Information System(CNISIS)since late 2022.Methods:Data related to ILI were reported by national surveillance sentinel hospitals.Positive testing for SARS-CoV-2 and influenza viruses was conducted using real-time reverse transcription polymerase chain reaction(rRT-PCR)detection by the national influenza surveillance network laboratories.Surveillance data were reported to CNISIS.Results:Beginning on December 12,2022(Week 50),the ILI percentage increased dramatically,peaking in Week 51 at 12.1%.Subsequently,the ILI percentage began to decline rapidly from Week 52,2022,and by Week 6,2023(February 6-12),the ILI and ILI percentage had returned to the levels observed at the beginning of December 2022.From December 1,2022 to February 12,2023,115,844 specimens were tested for both SARS-CoV-2 and influenza virus.Of these,30,381(26.2%)were positive for SARSCoV-2 and 1,763(1.5%)were positive for influenza virus.The positive rate of SARS-CoV-2 tests peaked at 74.1%around December 23 and 25.Conclusions:Sentinel-based surveillance,previously established for influenza,is an effective way to track the circulation trend of SARS-CoV-2 during community-level epidemics.There was no coprevalence of SARS-CoV-2 and influenza virus during the outbreak of SARS-CoV-2,even during the winter influenza season.However,it is important to remain vigilant for the potential rise of influenza activities following the COVID-19 epidemic.Xiaoxu Zeng Yiran Xie Xiaokun Yang Zhibin Peng Jing Tang Lei Yang Shuxia Lin Jun Han Yanping Zhang Dayan Wang 2023China CDC weekly2023,5,7:1
14Renin--angiotensin system inhibitor is associated with the reduced risk of all-cause mortality in COVID-19 among patients with/without hypertension显示文摘Consecutively hospitalized patients with confirmed coronavirus disease 2019(COVID-19)in Wuhan,China were retrospectively enrolled from January 2020 to March 2020 to investigate the association between the use of renin–angiotensin system inhibitor(RAS-I)and the outcome of this disease.Associations between the use of RAS-I(angiotensin-converting enzyme inhibitor(ACEI)or angiotensin receptor blocker(ARB)),ACEI,and ARB and in-hospital mortality were analyzed using multivariate Cox proportional hazards regression models in overall and subgroup of hypertension status.A total of 2771 patients with COVID-19 were included,with moderate and severe cases accounting for 45.0%and 36.5%,respectively.A total of 195(7.0%)patients died.RAS-I(hazard ratio(HR)=0.499,95%confidence interval(CI)0.325–0.767)and ARB(HR=0.410,95%CI 0.240–0.700)use was associated with a reduced risk of all-cause mortality among patients with COVID-19.For patients with hypertension,RAS-I and ARB applications were also associated with a reduced risk of mortality with HR of 0.352(95%CI 0.162–0.764)and 0.279(95%CI 0.115–0.677),respectively.RAS-I exhibited protective effects on the survival outcome of COVID-19.ARB use was associated with a reduced risk of all-cause mortality among patients with COVID-19.Huai-yu Wang Suyuan Peng Zhanghui Ye Pengfei Li Qing Li Xuanyu Shi Rui Zeng Ying Yao Fan He Junhua Li Liu Liu Shuwang Ge Xianjun Ke Zhibin Zhou Gang Xu Ming-hui Zhao Haibo Wang Luxia Zhang Erdan Dong 2022Frontiers of Medicine2022,16,1:0
15Optimal decomposition level selection approach in wavelet threshold denoising algorithm for ECG signal显示文摘The effect of electrocardiogram(ECG)signal wavelet denoising depends on the optimal configuration of its control parameters and the selection of the optimal decomposition level.Nevertheless,the existing optimal decomposition level selection scheme has some problems,such as lack of reliable theoretical guidance and insufficient accuracy,which need to be solved urgently.To solve this problem,this paper proposes an optimal decomposition level selection method based on multi-index fusion,which is used to select the optimal decomposition level for wavelet threshold denoising of ECG signal.In the stage of index selection,in order to overcome the limitation of a single evaluation index,the optimal multi-evaluation index is selected through the joint analysis of the geometric and physical significance of traditional evaluation indexes.In the stage of index fusion,based on the method of weighting the selected multiple indexes by the information entropy weight method and the coefficient of variation method,an optimal decomposition level selection method based on the evaluation index Z is proposed to improve the accuracy of the optimal decomposition level selection.Finally,extensive experiments are carried out on the real ECG signal from the Massachusetts Institute of Technology-Beth Israel Hospital(MIT-BIH)arrhythmia database and simulated signal to test the performance of the proposed method.The experimental results show that the accuracy of this method is superior to other related methods,and it can achieve better denoising effect of ECG signal.Yao Yindi Yi Jun Zeng Zhibin Li Xiong Wang Chen Li Yuhang 2023The Journal of China Universities of Posts and Telecommunications2023,30,4:0
16Wideband bandstop defected ground structure filter with enhanced common-mode suppression using split ring显示文摘An effective method to broaden the rejection bandwidth of dump-shaped defected ground structure(DGS) filter design is proposed in the paper. One traditional dumbbell-shaped DGS is split by using one split ring, and reconstructed into two different compact DGSs. The distance between both of the DGSs is so close that there exists mutual inductance, which can be utilized to broaden the rejection bandwidth effectively. The effect of different parameters of the split ring on the rejection bandwidth and depth of the filter is analyzed. Simulated results demonstrate that, with the same area of 5.5mm by 10 mm, rejection bandwidth of the proposed filter is from 4.4 GHz to 10.5 GHz for the common-mode noise(CM) while that of the traditional dumbbell-shaped DGS is from 5.3 GHz to 7.3 GHz. Compared to other schematics adopting multiple DGS to broaden bandwidth, it can broaden the bandwidth by dividing the existed DGS into multiple parts and reconstructing a new filter. 18Propofol inhibits cells migration and invasion via HOTAIR/miR-93/HIF-1α-mediated lactate secretion in colon cancer显示文摘Propofol,a common intravenous anesthetic used in clinical,has been shown to regulate cells proliferation,inflammation,angiogenesis and metastasis as well as exerting anti-cancer effect in several different types of cancer.However,the functions and mechanisms of propofol in lactate secretion and invasion of cancer cells remain unknown.The primary aim of this study was to investigate the role of propofol in cells migration and invasion of colon cancer(CRC).Scratches assay,Transwell assay were used to detect colon cells migration and invasion.Realtime Quantitative Polymerase Chain Reaction(RT-qPCR),Western blotting were utilized to detect the expression of related molecules.Dual-luciferase assay was performed to identify transcriptional activity of target genes,RNA pulldown assay was used to confirm the interactions of RNAs.Results indicated that propofol inhibited cell migration and invasion through suppressing lactic acid production in CRC.Moreover,propofol reduced lactic acid production by down-regulating the expression of hypoxia inducible factor-1α(HIF-1α)and monocarboxylate transporter 4(MCT4).In addition,propofol inhibited the expression of HIF-1αand cells migration/invasion via suppressing the interaction of HOX transcript antisense RNA(HOTAIR)with miR-93.The present study indicated that propofol inhibited colon cancer cell migration and invasion by reducing lactic acid production and the expression of HIF-1αand MCT4 via HOTAIR/miR-93 axis.These data may provide potential metabolic targets for CRC treatment.RUONAN GU WENJING GUO WENYANG WANG GUIHUAN LI XIAOJU LAI ZHIBIN HUANG WANLU ZHAO ZHICONGWU HUA CHEN WENYANG LUO FANGYIN ZENG FAN DENG 2021BIOCELL2021,45,6:0

20Effects of the Fermentation of Lophatherum gracile Brongn on Its Active Ingredients of Total Flavonoids and Total Polysaccharide显示文摘[Objectives] In order to explore the effect of fermentation on Lophatherum gracile Brongn. [Methods] The single factor experiment was applied to screen the fermentation conditions of L. gracile Brongn,and the effect of fermentation on the contents of total flavonoids and total polysaccharides was explored. [Results] The experiment defined the best fermentation conditions as: keeping natural state of p H,temperature 35℃,inoculation amount 6%,rotation speed 210 r/min,fermentation time 36 h. The results showed that the content increase of total flavonoids in the fermentation group was 5. 59% higher than the that in the non-fermentation group.
