| 1 | Expressions of type I collagen, α2 integrin and β1 integrin in sclera of guinea pig with defocus myopia and inhibitory effects of bFGF on the formation of myopia显示文摘AIM:To investigate the expressions of type I collagen, α2 integrin and β1 integrin in the posterior sclera of guinea pigs with defocus myopia and whether basic fibroblast growth factor (bFGF) injection inhibits the formation and development of myopia by upregulating the expression of type I collagen, α2 integrin and β1 integrin. METHODS:After 14 days of treatment, the refractive state and axial length were measured and the levels of type I collagen, α2 integrin and β1 integrin were assayed in the posterior sclerae of groups of guinea pigs that wore a monocular-7D polymethylmethacrylate (PMMA) lens or had -7D lens wear followed by the peribulbar injection of Phosphate Buffer Solution (PBS) or bFGF. The untreated fellow eye served as a control. Guinea pigs with no treatment served as normal group. ·RESULTS:The results showed that 14 days of monocular defocus increased axial eye length and refraction, while bFGF delivery inhibited them markedly. Further, it was also found that the monocular-7D lens could decrease the levels of type I collagen, α2 integrin and β1 integrin expressions, while, unlike PBS, bFGF increased them significantly in comparison to contralateral control eyes and normal eyes. CONCLUSION:bFGF can prevent the formation anddevelopment of defocus myopia by upregulating the expressions of type I collagen, α2 integrin and β1 integrin. | 2 | Antioxidant and Anti-inflammatory Capacity of Ferulic Acid Released from Wheat Bran by Solid-state Fermentation of Aspergillus niger显示文摘Objective A strain of Aspergillus niger(A. niger), capable of releasing bound phenolic acids from wheat bran, was isolated. This strain was identified by gene sequence identification. The antioxidant and anti-inflammatory capacity of ferulic acid released from wheat bran by this A. niger strain(FA-WB) were evaluated. Methods Molecular identification techniques based on PCR analysis of specific genomic sequences were conducted; antioxidant ability was examined using oxygen radical absorbance capacity(ORAC), cellular antioxidant activity(CAA) assays, and erythrocyte hemolysis assays. RAW264.7 cells were used as a model to detect anti-inflammatory activity. Results The filamentous fungal isolate was identified to be A. niger. ORAC and CAA assay showed that FA-WB had better antioxidant activity than that of the ferulic acid standard. The erythrocyte hemolysis assay results suggested that FA-WB could attenuate AAPH-induced oxidative stress through inhibition of reactive oxy gen species(ROS) generation. FA-WB could significantly restore the AAPH-induced increase in intracellular antioxidant enzyme activities to normal levels as well as inhibit the intracellular malondialdehyde formation. TNF-?, IL-6, and NO levels indicated that FA-WB can inhibit the inflammation induced by lipopolysaccharide(LPS). | 3 | Seroepidemiological Investigation of Lyme Disease and Human Granulocytic Anaplasmosis among People Living in Forest Areas of Eight Provinces in China显示文摘Objective Lyme disease and Human granulocytic anaplasmosis are tick‐borne diseases caused by Borrelia burgdorferi and Anaplasma phagocytophilum respectively. We have investigated infection and co-infection of the two diseases in the population of forest areas of eight provinces in China by measuring seroprevalence of antibodies against B. burgdorferi and A. phagocytophilum. Methods Forest areas in 8 provinces were chosen for investigation using whole sampling and questionnaire survey methods. 3 669 serum samples from people in the forest areas were tested for the presence of antibodies by indirect immunofluorescent assay (IFA). Results Seroprevalence against B. burgdorferi was 3% to 15% and against A. phagocytophilum was 2% to 18% in the study sites in the 8 provinces in China. We also found co-infection of B. burgdorferi and A. phagocytophilum in 7 of the 8 provinces (the exception being the Miyun area in Beijing). The seroprevalence for both B. burgdorferi and A. phagocytophilum was significantly higher among people exposed to ticks than among people who were not exposed to ticks. | 4 | Hangzhou criteria as downstaging criteria in hepatocellular ca显示文摘Background:The downstaging of hepatocellular carcinoma(HCC)has been confirmed to benefit liver transplantation(LT)patients whose tumors are beyond the transplantation criteria.Milan criteria(MC),a tumor size and number-based assessment,is currently used as the endpoint in these patients.However,many studies believe that tumor biological behavior should be added to the evaluation criteria for downstaging efficacy.Hence,this study aimed to explore the feasibility of Hangzhou criteria(HC),which introduced tumor grading and alpha-fetoprotein in addition to tumor size and number,as an endpoint of downstaging.Methods:We performed a multicenter and retrospective study of 206 patients accepted locoregional therapy(LRT)as downstaging/bridge treatment prior to LT in three centers of China.Results:Recipients were divided into four groups:failed downstaging to the HC(group A,n=46),successful downstaging to the HC(group B,n=30),remained within the HC all the time(group C,n=113),and tumor progressed(group D,n=17).The 3-year HCC recurrence probabilities of groups B and C were not significantly different(10.3%vs.11.6%,P=0.87).The HCC recurrent rate was significantly higher in group A(52.3%)compared with that in group B/C(P<0.05).Seven patients(7/76,9.2%)whose tumor exceeded the the HC were successfully downstaged to the MC,and 39.5%(30/76)to the the HC.In group B,23 patients remained beyond the MC and their survivals were as well as those of patients within the MC.Conclusions:Compared to the MC,HC downstaging criteria can give more HCC patients access to LT and furthermore,the outcome of these patients is the same as those matching MC downstaging criteria.Hangzhou downstaging criteria therefore is applicable in clinical practice. | Qi-Fan Zhan Sun-Bin Ling Yi-Nan Deng Qiao-Nan Shan Qian-Wei Ye Sheng-Jun Xu Guang-Jiang Jiang Di Lu Xu-Yong Wei Li Zhuang Wu Zhang Tian Shen Bei-Ni Cen Hai-Yang Xie Ji-Min Liu Jian Wu Shu-Sen Zheng Yang Yang Xiao Xu | 2020 | Hepatobiliary & Pancreatic Diseases International2020,19,4: | 11 |

| 6 | Establishment of an efficient transformation protocol and its application in marine-derived Bacillus strain显示文摘Marine-derived Bacillus strains have been proved to be a very promising source for natural product leads.However,transformation of environmental strains is much more difficult than that of domesticated strains.Here,we report the development of an efficient and robust electroporation-based transformation system for marine-derived Bacillus marinus B-9987,which is a macrolactin antibiotics producer and a very promising biological control agent against fungal plant diseases.The transformation efficiency was greatly enhanced 103-fold by using unmethylated plasmid to bypass modification-restriction barrier,and using glycine betaine to protect cells from electrical damages during electroporation.Addition of HEPES and 2 mmol L?1MgCl2 further improved the efficiency by additional 2-fold,with a maximum value of 7.1×104 cfu/μg pHT3101.To demonstrate the feasibility and efficiency of the protocol,a green fluorescent protein reporter system was constructed;furthermore,phosphopantetheinyl transferase gene sfp,which is essential to the biosynthesis of polyketides and nonribosomal peptides,was overexpressed in B-9987,leading to increased production of macrolactin A by about 1.6-fold.In addition,this protocol is also applicable to marine-derived Bacillus licheniforms EI-34-6,indicating it could be a reference for other undomesticated Bacillus strains.To our knowledge,this is the first report regarding the transformation of marine-derived Bacillus strain. | LIU Yang ZHENG Hua ZHAN GuiHua QIN Wen TIAN Li LI WenLi | 2014 | Science China(Life Sciences)2014,57,6: | 9 |

| 9 | Preparation of Polyimide/BaTiO_3/Ag Nanocomposite Films via in situ Technique and Study of Their Dielectric Behavior显示文摘Triphase polyimide nanocomposite films were fabricated using barium titanate(BaTiO3) with high dielectric constant and silver(Ag) with high conductivity as fillers. In situ method was utilized to obtain the homogeneous dispersion of nanoparticles. The in situ polymerization of polyimide precursor-poly(amic acid) was performed in the presence of BaTiO3 particles. Silver compound 1,1,1-trifluoro-2,4-pentadionato silver(I) was added into the BaTiO3 containing poly(amic acid) solution to achieve silver nanoparticles via in situ self metallization technique. The thermally induced reduction converted silver(I) to metallic silver with concomitant imidization of poly(amic acid) to polyimide. Both BaTiO3 and silver nanoparticles were uniformly dispersed in the polyimide substrate. The dependence of dielectric behavior on the BaTiO3 and Ag contents was studied. | 10 | Response of Endogenous Salicylic Acid and Jasmonates to Mechanical Wounding in Pea Leaves显示文摘The roles of on endogenous jasmonates(JAs) and salicylic acid(SA) in wounding response were investigated. Pea(Pisum sativum L.) seedlings were treated with three different methods including mechanical wounding,JAs application,and SA application. The contents of endogenous JAs and SA,as well as the activities of the related enzymes were detected by enzyme-linked immunosorbent assay(ELISA),high performance liquid chromatography(HPLC),and spectrophotometer,respectively. The results showed that endogenous JA rapidly accumulated within 30 min after wounding. The increase in the activities of both lipoxygenase(LOX) and allene oxide synthase(AOS) lagged behind JAs burst. A second slight increase in JAs level was observed at 24 h after wounding treatment,and at the same time point,higher activities of LOX and AOS were also detected. Endogenous free SA content decreased accompanied with JAs burst. Effects of exogenous JA application were similar to those of wounding treatment on endogenous SA level and phenylalanine ammonia lyase(PAL) activity,whereas exogenous SA application led to the significant inhibition of LOX and AOS activities and the decrease of endogenous JAs level at the early stage of treatment. | 12 | Effects of preventive acupuncture or moxibustion at the Guanyuan acupoint on expression of interleukin-2 and its receptors in hypothalamus, serum, and uterus of menopausal rats显示文摘BACKGROUND: Previous studies have indicated preventive acupuncture and moxibustion at the Guanyuan point have positive effects on the immune system of menopausal rats. OBJECTIVE: To observe the effects of preventive acupuncture or moxibustion at the Guanyuan point on interleukin-2 (IL-2) and its receptor (IL-2R mRNA) levels in hypothalamus, serum, and uterus of menopausal rats. DESIGN: Randomized control animal experiment. SETTING: School of Acupuncture and Moxibustion, Beijing University of Chinese Medicine. MATERIALS: This study was performed at the Laboratory of Beijing University of Chinese Medicine from November 2004 to July 2005. The vaginal smear method was used to determine the estrus cycle of 9.5-month-old Sprague Dawley (SD) female rats. Three continuous estrus cycles were measured, and 160 rats with estrus cycle disorder, weighing (300 ± 20) g, were included in this study. The rats were randomly divided into four groups: preventive acupuncture group (n =48), preventive moxibustion group (n =48), model groups (n =48), and 10-month-old group (n =16). Sixteen 3.5-month-old healthy SD female rats, weighing (240 ± 20) g, were also assigned to the young group. All rats were clean grade and were purchased from the Laboratory Animal Center, Beijing University of Chinese Medicine. The experiment was in accordance with animal ethics standards. METHODS: Acupuncture was applied to the preventive acupuncture group at 10 months of age. The needle was inserted upward and perpendicularly two fen to a point that was two fen below the Guanyuan point. The rats freely moved around during needle retention (20 minutes). Acupuncture was administered twice a week and continued for eight weeks. A moxa cone (small) moxibustion was applied to the preventive moxibustion group at 10 months of age. The rats were caught gently and kept at the same position as the preventive acupuncture group. The moxa cone was ignited and held to the Guanyuan point directly. Only one moxa cone was used each time. After moxibustion, the local skin turned slightly red. Moxibustion was used twice a week and continued for eight weeks. Nothing except gently catching and lying was administered to the model group and the young group. MAIN OUTCOME MEASURES: Radioimmunoassay, in situ hybridization, and similar methods were employed to detect IL-2 in serum and the uterus, as well as IL-2R mRNA expression in the hypothalamus and uterus arcuate nuclei, of the preventive acupuncture group, preventive moxibustion group, model groups (at 12, 14, and 16 months), 10 month old group (at 10 months), and the young group (at 4 months). RESULTS: All 176 rats were included in the final analysis. ① IL-2R mRNA: IL-2R mRNA expression decreased significantly in the uterus of 12-, 14-, and 16-month-old model groups and in the hypothalamus of 14- and 16-month-old model groups (P < 0.05-0.01). Compared to the same month-aged model groups, the expression of IL-2R mRNA increased significantly in the hypothalamus and uterus of 16-month-old preventive acupuncture or moxibustion groups (P < 0.05-0.01).② IL-2: IL-2 in serum of 12- and 14-month-old model groups and in the uterus of 12-, 14-, and 16-month-old model groups decreased significantly compared to the young group (P < 0.05-0.01). IL-2 serum levels increased significantly in the 16-month-old group with preventive moxibustion; in addition, IL-2 expression levels increased significantly in the uterus of the 12-month-old group with preventive moxibustion, as well as the 12- and 16-month-old groups with preventive acupuncture (P < 0.05-0.01). CONCLUSION: As a rat ages and becomes menopausal, IL-2R mRNA expression in the hypothalamus anduterus, as well as IL-2 levels in the serum and uterus, decrease. However, preventive acupuncture and preventive moxibustion can repress the decrease to some extent. | 13 | High expression of FABP4 in colorectal cancer and its clinical significance显示文摘Objective:To investigate the relationship between the fatty acid-binding protein 4(FABP4)and colorectal cancer(CRC).Methods:Using an enzyme-linked immunosorbent assay(ELISA),we measured the expression of FABP4 in plasma of50 patients who underwent surgery for CRC from October 2017 to May 2018 and 50 healthy controls.The content of the visceral fat area(VFA)as seen with abdominal computed tomography(CT)scanning was measured by ImageJ software.The expression levels of FABP4,E-cadherin,and Snail proteins in CRC and adjacent tissues were determined by immunohistochemistry.Results:The mean concentration of plasma FABP4 of CRC patients was higher than that of the control group(22.46 vs.9.82 ng/m L;P<0.05).The concentration of plasma FABP4 was related to the tumor,node,metastatis(TNM)stage and lymph node metastasis and was independent of age,body mass index(BMI),tumor size and location,and the degree of differentiation of CRC.The concentration of plasma FABP4 was positively correlated with high VFA and lipoprotein-a(LPA)(P<0.05);but it was not correlated with total cholesterol(TG),total triglyceride(TC),low-density lipoprotein(LDL),high-density lipoprotein(HDL),or apolipoprotein AI(Apo-AI).The expression of FABP4 protein in CRC tissues was positively correlated with the degree of CRC differentiation,tumor stage,and lymph node metastasis.The level of FABP4 protein was negatively correlated with E-cadherin protein(r=-0.3292,P=0.0196)and positively correlated with Snail protein(r=0.5856,P<0.0001).Conclusions:High LPA and VFA were risk factors for increased plasma FABP4 in CRC patients.FABP4 protein was highly expressed in CRC tissues and associated with TNM stage,differentiation,and lymph node metastasis of CRC.The level of FABP4 in CRC tissue was correlated with E-cadherin and Snail expression,suggesting that FABP4 may promote CRC progression related to epithelial-mesenchymal transition(EMT). | Yan ZHANG Wenjia ZHANG Min XIA Zhujun XIE Fangmei AN Qiang ZHAN Wenying TIAN Tianyue ZHU | 2021 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,2: | 5 |
| 14 | Fiber damage of machine-harvested cotton before ginning and after lint cleaning显示文摘Machine harvesting increases the foreign matter content of seed cotton. Excessive cleaning causes fiber damage and economic loss. Most trading companies in the Xinjiang Uygur Autonomous Region, China have indicated reluctance to use machine-harvested cotton. The first objective was to determine how the fiber quality was affected by the ginning and lint cleaning and how the fiber damage during levels of lint cleaning changed. The second objective was to determine the optimum number of lint cleaners for machine-harvested cotton based on fiber damage. Cotton samples were collected from 13 fields and processed in seven ginneries between 2013 and 2015. The results indicated that ginning and lint cleaning didn't have significant effect on fiber strength and significantly affected both fiber length and short fiber index. Fiber length was reduced by more than 1.00 mm from six of 13 fields after lint cleaning, then the damage rate on short fiber index from 11 of 13 fields was more than 20%. The third lint cleaning caused great fiber damage, reducing fiber length by 0.35 mm and increasing short fiber index by 0.65%. | 15 | Nanocrystal Model for Liquid Metals and Amorphous Metals显示文摘A nanocrystal model for liquid metals and amorphous metals has been developed. With the nanocrystal model, the broadening peak profiles (BPPs) of Cu, Al, Al65Cu20Fe15 alloy, Cu70Ni30 alloy and Fe50Si50 alloy were gained by broadening the X-ray diffraction (XRD) peaks of a crystal lattice. By comparing the BPPs with the XRD intensity curves measured on the liquid metals, it is found that the BPPs are closely in agreement with the XRD intensity curves, respectively, except the Fe50Si50 alloy. | 17 | Substantial transition to clean household energy mix in rural China显示文摘The household energy mix has significant impacts on human health and climate,as it contributes greatly to many health-and climate-relevant air pollutants.Compared to the well-established urban energy statistical system,the rural household energy statistical system is incomplete and is often associated with high biases.Via a nationwide investigation,this study revealed high contributions to energy supply from coal and biomass fuels in the rural household energy sector,while electricity comprised∼20%.Stacking(the use of multiple sources of energy)is significant,and the average number of energy types was 2.8 per household.Compared to 2012,the consumption of biomass and coals in 2017 decreased by 45%and 12%,respectively,while the gas consumption amount increased by 204%.Increased gas and decreased coal consumptions were mainly in cooking,while decreased biomass was in both cooking(41%)and heating(59%).The time-sharing fraction of electricity and gases(E&G)for daily cooking grew,reaching 69%in 2017,but for space heating,traditional solid fuels were still dominant,with the national average shared fraction of E&G being only 20%.The non-uniform spatial distribution and the non-linear increase in the fraction of E&G indicated challenges to achieving universal access to modern cooking energy by 2030,particularly in less-developed rural and mountainous areas.In some non-typical heating zones,the increased share of E&G for heating was significant and largely driven by income growth,but in typical heating zones,the time-sharing fraction was<5%and was not significantly increased,except in areas with policy intervention.The intervention policy not only led to dramatic increases in the clean energy fraction for heating but also accelerated the clean cooking transition.Higher income,higher education,younger age,less energy/stove stacking and smaller family size positively impacted the clean energy transition. | Guofeng Shen Rui Xiong Yanlin Tian Zhihan Luo Bahabaike Jiangtulu Wenjun Meng Wei Du Jing Meng Yuanchen Chen Bing Xue Bin Wang Yonghong Duan Jia Duo Fenggui Fan Lei Huang Tianzhen Ju Fenggui Liu Shunxin Li Xianli Liu Yungui Li Mu Wang Ying Nan Bo Pan Yanfang Pan Lizhi Wang Eddy Zeng Chao Zhan Yilin Chen Huizhong Shen Hefa Cheng Shu Tao | 2022 | National Science Review2022,9,7: | 4 |
| 18 | Electropolishing of titanium alloy under hydrodynamic mode显示文摘Titanium(Ti) alloys are widely used in aerospace industry due to the low density and high corrosion resistance. However, machining and polishing remain great challenges because of the hardness and chemical stability. With a home-made electrochemical machining workstation, cyclic voltammetry is performed at a wide potential range of [0 V, 20 V] to record the details of passivation and depassivation processes under a hydrodynamic mode. The results show that the thickness of viscous layer formed on the alloy surface plays a crucial effect on the electropolishing quality. | 20 | CaMKIIα-driven,phosphatase-checked postsynaptic plasticity via phase separation显示文摘Ca^(2+)/calmodulin-dependent kinase Ila(CaMKIla)is essential for synaptic plasticity and learning by decoding synaptic Ca^(2+)oscillations.Despite decades of extensive research,new mechanisms underlying CaMKIIa's function in synapses are still being discovered.Here,we discover that Shank3 is a specific binding partner for autoinhibited CaMKIla.We demonstrate that Shank3 and GluN2B,via combined actions of Ca^(2+)and phosphatases,reciprocally bind to CaMKIla.Under basal condition,CaMKIla is recruited to the Shank3 subcompartment of postsynaptic density(PSD)via phase separation.Rise of Ca^(2+)concentration induces GluN2Bmediated recruitment of active CaMKIla and formation of the CaMKIIa/GluN2B/PSD-95 condensates,which are autonomously dispersed upon Ca^(2+)removal.Protein phosphatases control the Ca^(2+)-dependent shuttling of CaMKIla between the two PSD subcompartme nts and PSD condensate formation.Activation of CaMKIla further enlarges the PSD assembly and in duces structural LTP.Thus,Ca^(2+)-induced and phosphatase-checked shuttling of CaMKIla between distinct PSD nano-domains can regulate phase separation-mediated PSD assembly and synaptic plasticity. | Qixu Caic Menglong Zeng Xiandeng Wu Haowei Wu Yumeng Zhan Ruijun Tian Mingjie Zhang | 2021 | Cell Research2021,31,1: | 3 |
