1A Resource for Inactivation of MicroRNAs Using Short Tandem Target Mimic Technology in Model and Crop Plants显示文摘microRNAs (miRNAs)are endogenous small non-coding RNAs that bind to mRNAs and target them for cleavage and/or translational repression,leading to gene silencing.We previously developed short tandem target mimic (STTM)technology to deactivate endogenous miRNAs in Arabidopsis.Here,we created hundreds of STTMs that target both conserved and species-specific miRNAs in Arabidopsis,tomato,rice,and maize,providing a resource for the functional interrogation of miRNAs.We not only revealed the functions of several miRNAs in plant development,but also demonstrated that tissue-specific inactivation of a few miRNAs in rice leads to an increase in grain size without adversely affecting overall plant growth and development.RNA-seq and small RNAseq analyses of STTM156/157 and STTM165/166 transgenic plants revealed the roles of these miRNAs in plant hormone biosynthesis and activation,secondary metabolism,and ion-channel activity-associated electrophysiology,demonstrating that STTM technology is an effective approach for studying miRNA functions.To facilitate the study and application of STTM transgenic plants and to provide a useful platform for storing and sharing of information about miRNA-regulated gene networks,we have established an online Genome Browser (http://gffzzc750bc0108fa4044sov9b5oncuvw66x0n.ffgz.tsg.suse.edu.cn/designindex2.php) to display the transcriptomic and miRNAomic changes in STTMinduced miRNA knockdown plants.Ting Peng Mengmeng Qiao Haiping Liu Sachin Teotia Zhanhui Zhang Yafan Zhao Bobo Wang Dongjie Zhao Lina Shi Cui Zhang Brandon Le Kestrel Rogers Chathura Gunasekara Haitang Duan Yiyou Gu Lei Tian Jinfu Nie Jian Qi Fanrong Meng Lan Huang Qinghui Chen Zhenlin Wang Jinshan Tang Xiaoqing Tang Ting Lan Xuemei Chen Hairong Wei Quanzhi Zhao Guiliang Tang 2018Molecular Plant2018,11,11:7
2Development of a Rapid Multi-residue Assay for Detecting β-lactams Using Penicillin Binding Protein 2x显示文摘Objective To develop a rapid multi-residue assay for detecting 16 demanded by the European Union (EU). Methods A recombinant penicillin-binding protein(PBP)2x* from Streptococcus pneumoniae R6 was expressed in vitro and six β-lactams were conjugated to HRP by four methods.A rapid multi-residue assay for β-lactams was established with PBP2x* and HRP-conjugate. Results PBP2x* was expressed and purified successfully and the ideal HRP-conjugate was identified. The multi-residue assay was developed.After optimization,penicillin G,ampicillin,amoxicillin, cloxacillin,dicloxacillin,oxacillin,nafcillin,cephalexin,ceftiofur,cefaIonium,cefquinome,cefazolin, cefoperazone,cephacetrile,and cephapirin can be detected at levels below MRL in milk with simple pretreatment. Conclusion This assay developed can detect all 16 β-lactams demanded by the European Union(EU). 3Comparative QTL analysis of maize seed artificial aging between an immortalized F_2 population and its corresponding RILs显示文摘Seed aging decreases the quality and vigor of crop seeds,thereby causing substantial agricultural and economic losses in crops.To identify genetic differences in seed aging between homozygotes and heterozygotes in maize,the seeds of a set of recombinant inbred lines(RILs) and an immortalized F_2(IF_2) population were subjected to artificial aging treatments for 0,2,3,and 4 days under 45℃ and 85%relative humidity and seed vigor was then evaluated in a field experiment.Seed vigor of all entries tested decreased sharply with longer aging treatment and seed vigor decreased more slowly in heterozygotes than in homozygotes.Forty-nine QTL were detected for four measured seed vigor traits in the RIL(28QTL) and IF_2(21 QTL) populations.Only one QTL,qGP5,was detected in both populations,indicating that the genes involved in anti-aging mechanisms differed between inbred lines and hybrids.Several QTL were identified to be responsible for multiple seed vigor traits simultaneously in the RIL and IF_2 populations under artificial aging conditions.These QTL may include major genes for seed vigor or seed aging.QTL qVI4 b and qGE3 a detected in the RIL population coincided with genes ZmLOX1 and ZmPLD1 in the same respective chromosomal regions.These QTL would be useful for screening for anti-aging genes in maize breeding.Bin Wang Zhanhui Zhang Zhiyuan Fu Zonghua Liu Yanmin Hu Jihua Tang 2016The Crop Journal2016,4,1:5
4MiR319-targeted OsTCP21 and OsGAmyb regulate tillering and grain yield in rice显示文摘Multiple genes and microRNAs(miRNAs)improve grain yield by promoting tillering.MiR319s are known to regulate several aspects of plant development;however,whether miR319s are essential for tillering regulation remains unclear.Here,we report that miR319 is highly expressed in the basal part of rice plant at different development stages.The miR319 knockdown line Short Tandem Target Mimic 319(STTM319)showed higher tiller bud length in seedlings under low nitrogen(N)condition and higher tiller bud number under high N condition compared with the miR319a-overexpression line.Through targets prediction,we identified OsTCP21 and OsGAmyb as downstream targets of miR319.Moreover,OsTCP21 and OsGAmyb overexpression lines and STTM319 had increased tiller bud length and biomass,whereas both were decreased in OsTCP21 and OsGAmyb knockout lines and OE319a.These data suggest that miR319 regulates rice tiller bud development and tillering through targeting OsTCP21 and OsGAmyb.Notably,the tiller number and grain yield increased in STTM319 and overexpression lines of OsTCP21 and OsGAmyb but decreased in OE319a and knockout lines of OsTCP21 and OsGAmyb.Taken together,our findings indicate that miR319s negatively affect tiller number and grain yield by targeting OsTCP21 and OsGAmyb,revealing a novel function for miR319 in rice.Rongna Wang Xiuyan Yang Shuang Guo Zhaohui Wang Zhanhui Zhang Zhongming Fang 2021Journal of Integrative Plant Biology2021,63,7:4
5Biodegradable intestinal stents: A review显示文摘Biodegradable stents are an attractive alternative to self-expanding metal stents in the treatment of intestinal strictures. Biodegradable stent can be made of biodegradable polymers and biodegradable metals(magnesium alloys). An overview on current biodegradable intestinal stents is presented. The future trends and perspectives in the development of biodegradable intestinal stents are proposed. For the biodegradable polymer intestinal stents, the clinical trials have shown promising results, although improved design of stents and reduced migration rate are expected. For the biodegradable magnesium intestinal stents, results of preliminary studies indicate magnesium alloys to have good biocompatibility. 6Safety and efficacy of anti-EGFR monoclonal antibody (SCT200) as second-line therapy in advanced esophageal squamous cell carcinoma显示文摘Objective:The mainstay treatment of esophageal squamous cell carcinoma(ESCC)involves chemotherapy and immunotherapy.However,alternative therapies are required for patients who are refractory or intolerant to existing therapies.Methods:In this single-arm,multicenter,open-label phase Ib study,30 patients received an intravenous infusion of SCT200,an antiepidermal growth factor receptor(EGFR)monoclonal antibody,6.0 mg/kg once a week for 6 weeks,followed by 8.0 mg/kg once every 2 weeks until disease progression or intolerable toxicity.The primary endpoint was the objective response rate(ORR).The secondary endpoints were progression-free survival(PFS),overall survival(OS),and safety.Results:Thirty patients were enrolled between July 2018 and May 2019.The ORR was 16.7%(95%CI:5.6%–34.7%).The median PFS and OS were 3.1 months(95%CI:1.5–4.3)and 6.8 months(95%CI:4.7–10.1),respectively.A numerical difference without any statistical significance in ORR was observed in patients with different EGFR expressions(≥50%:25.0%vs.<50%:0%,P=0.140)or TP53 mutation abundance(<10%:23.8%vs.≥10%:0%,P=0.286).Improved median PFS(3.4 vs.1.4 months,P=0.006)and OS(8.0 vs.4.2 months,P=0.027)were associated with TP53 mutation abundance of<10%.The most common treatment-related adverse events of grade 3 or 4(occurring in≥2 patients)were hypomagnesemia[7(23.3%)]and rash[2(6.7%)].No treatmentrelated death occurred.Conclusions:SCT200 monotherapy as the second-or further-line treatment for advanced ESCC showed favorable efficacy,with an acceptable safety profile.TP53 mutation abundance might serve as a potential predictive biomarker.Ming Bai Meng Wang Ting Deng Yuxian Bai Kai Zang Zhanhui Miao Wenlin Gai Liangzhi Xie Yi Ba 2022Cancer Biology & Medicine2022,19,3:2
7Synthesis of a Novel Co-B/CTAB Catalyst via Solid-state-reaction at Room Temperature for Hydrolysis of Ammonia-borane显示文摘Cobalt-boride(Co-B)is emerging as one of the promising materials in the base-hydrolytic dehydrogena-tion of ammonia-borane(AB).In order to avoid the low specific area and poor catalytic capacity of Co-B catalyst caused by aggregation arising from the strong reducing property and rapid reaction condensation of sodium borohy-dride(NaBHa),novel cobalt boride/cetyltrimethylammonium bromide(Co-B/CTAB)catalyst was obtained via solid-state grinding at room temperature,and the catalyst was further characterized by XRD,SEM,XPS and BET.The hydrogen generation rate(HGR)was then determined by the hydrolysis reaction of AB.The SEM images indicate that a lot of irregular folds and curled edges are formed on the sample with a maximum surface area of 145.57 m2/g,thus possibly resulting in the high hydrogen production(HGR was 10.68 L.minlg),which may be attributed to CTAB that provide favorable large specific surface area and abundant porous structure.Additionally,catalyst will not be affected by solvants during solid-state reaction.As a diluent,the surfactant CTAB hindered the reaction rate of sodium borohydride reduction to cobalt boride and obtained the novel catalyst with a large specific surface area.HU Haibin LONG Bo JIANG Yifan SUN Shichang Ibrahim LAWAN ZHOU Weiming- ZHANG Mingxin WANG Liwei ZHANG Fan YUAN Zhanhui 2020Chemical Research in Chinese Universities2020,36,6:2

13Biodegradable magnesium alloy with eddy thermal effect for effective and accurate magnetic hyperthermia ablation of tumors显示文摘Magnetic hyperthermia therapy(MHT)is able to ablate tumors using an alternating magnetic field(AMF)to heat up magnetocaloric agents(e.g.magnetic nanoparticles)administered into the tumors.For clinical applications,there is still a demand to find new magnetocaloric agents with strong AMF-induced heating performance and excellent biocompatibility.As a kind of biocompatible and biodegradable material,magnesium(Mg)and its alloys have been extensively used in the clinic as an implant metal.Herein,we discovered that the eddy thermal effect of the magnesium alloy(MgA)could be employed for MHT to effectively ablate tumors.Under low-field-intensity AMFs,Mg A rods could be rapidly heated,resulting in a temperature increase in nearby tissues.Such AMF-induced eddy thermal heating of MgA could not only be used to kill tumor cells in vitro,but also be employed for effective and accurate ablation of tumors in vivo.In addition to killing tumors in mice,we further demonstrated that VX2 tumors of much larger sizes growing in rabbits after implantation of Mg A rods could also be eliminated after exposure to an AMF,illustrating the ability of MgA-based MHT to kill large-sized tumors.Moreover,the implanted MgA rods showed excellent biocompatibility and~20%of their mass was degraded within three months.Our work thus discovered for the first time that non-magnetic biodegradable Mg A,an extensively used implant metal in clinic,could be used for effective magnetic thermal ablation of tumors under a low-field-intensity AMF.Such a strategy could be readily translated into clinical use.Nailin Yang Fei Gong Liang Cheng Huali Lei Wei Li Zongbin Sun Caifang Ni Zhanhui Wang Zhuang Liu 2021National Science Review2021,8,1:1
