| 1 | The impacts of conservation agriculture on crop yield in China depend on specific practices, crops and cropping regions显示文摘For smooth and wide application of conservation agriculture(CA), remaining uncertainties about its impacts on crop yield need to be reduced. Based on previous field experiments in China, a meta-analysis was performed to quantify the actual impacts of CA practices(NT: no/reduced-tillage only, CTSR: conventional tillage with straw retention, NTSR: NT with straw retention) on crop yields as compared to conventional tillage without straw retention(CT).Although CA practices increased crop yield by 4.6% on average, there were large variations in their impacts. For each CA practice, CTSR and NTSR significantly increased crop yield by 4.9%and 6.3%, respectively, compared to CT. However, no significant effect was found for NT. Among ecological areas, significant positive effects of CA practices were found in areas with an annual precipitation below 600 mm. Similar effects were found in areas with annual mean air temperature above 5 °C. For cropping regions, CA increased crop yield by 6.4% and 5.5%compared to CT in Northwest and South China, respectively, whereas no significant effects were found in the North China and Northeast China regions. Among crops, the positive effects of CA practices were significantly higher in maize(7.5%) and rice(4.1%) than in wheat(2.9%). NT likely decreased wheat yield. Our results indicate that there are great differences in the impacts of CA practices on crop yield, owing to regional variation in climate and crop types. CA will most likely increase maize yield but reduce wheat yield. | 2 | Organic amendments increase corn yield by enhancing soil resilience to climate change显示文摘A 22-year field experiment was conducted in Gongzhuling, Jilin province, China to investigate corn yield response to fertilization practice. Compared to an unfertilized control(CK), all fertilization treatments, including inorganic nitrogen fertilizer only(N), balanced inorganic fertilizers(NPK), NPK plus corn straw(SNPK), and NPK plus farmyard manure(MNPK), resulted in significant increases in corn yield. However, only organic matter amendments sustained increasing yield trends, with annual rates of 0.137 and 0.194 t ha-1for the SPNK and MNPK treatments, respectively(P < 0.05). During the 22 years, the daily mean, maximum and minimum temperatures increased by 0.50, 0.53, and 0.46 °C per decade, whereas precipitation displayed no significant change but showed large seasonal variation. According to a regression analysis, increased air temperature exerted positive effects on corn yields under the SNPK and the MNPK treatments. Under both treatments,soil organic carbon contents and soil nutrient availabilities increased significantly compared to their initial levels in 1990, whereas soil bulk density and total porosity changed slightly under the two treatments, which showed higher soil water storage than other treatments. In contrast, significant increases in soil bulk density and decreases in soil total porosity and soil nutrient availability were observed under the CK, N and NPK treatments. The contributions of soil fertility to corn yield were 28.4%, 37.9%, 38.4%, 39.0%,and 42.9% under CK, N, NPK, SNPK, and MNPK treatments, respectively, whereas climate changes accounted for 27.0%, 14.6%, 12.4%, 11.8%, and 10.8%. | 4 | Intraoperative ultrasound assistance in resection of intracranial meningiomas显示文摘Objective: Intracranial meningiomas, especially those located at anterior and middle skull base, are difficult to be completely resected due to their complicated anatomy structures and adjacent vessels. It's essential to locate the tumor and its vessels precisely during operation to reduce the risk of neurological deficits. The purpose of this study was to evaluate intraoperative ultrasonography in displaying intracranial meningioma and its surrounding arteries, and evaluate its potential to improve surgical precision and minimize surgical trauma. Methods: Between December 2011 and January 2013, 20 patients with anterior and middle skull base meningioma underwent surgery with the assistance of intraoperative ultrasonography in the Neurosurgery Department of Shanghai Huashan Hospital. There were 7 male and 13 female patients, aged from 31 to 66 years old. Their sonographic features were analyzed and the advantages of intraoperative ultrasonography were discussed. Results: The border of the meningioma and its adjacent vessels could be exhibited on intraoperative ultrasonography. The sonographic visualization allowed the neurosurgeon to choose an appropriate approach before the operation. In addition, intraoperative ultrasonography could inform neurosurgeons about the location of the tumor, its relation to the surrounding arteries during the operation, thus these essential arteries could be protected carefully. Conclusions: Intraoperative ultrasonography is a useful intraoperative technique. | 10 | Mettl3 deficiency leads to the upregulation of Cav1.2 and increases arrhythmia susceptibility in mice显示文摘Methyltransferase-like 3(Mettl3)is a component of methyltransferase complex that mediates m6A modification of RNAs,and participates in multiple biological processes.However,the role of Mettl3 in cardiac electrophysiology remains unknown.This study aims to explore the ventricular arrhythmia susceptibility of Mettl3+/–mice and the underlying mechanisms.Mice were anesthetized with 2%avertin(0.1 mL/10 g body weight)for echocardiography and programmed electrical pacing.Whole-cell patch clamp technique was used to examine the electrophysiological property of cardiomyocytes.The expression of Cav1.2 was determined by qRT-PCR and western blot analysis.The m6A medication of mRNA was examined by MeRIP-Seq and MeRIP-qPCR.No differences are found in the morphology and function of the hearts between Mettl3+/–mice and wild-type(WT)controls.The QT and QTc intervals of Mettl3+/–mice are significantly longer.High-frequency electrical stimulation showed that heterozygous knockout of Mettl3 increases ventricular arrhythmia susceptibility.The whole-cell patch-clamp recordings showed that the APD is prolonged in Mettl3+/–ventricular myocytes and more EADs were observed.The density of ICa-L is substantially increased in ventricular myocytes of Mettl3+/–mice.The pore-forming subunit of L-type calcium channel Cav1.2 is upregulated in Mettl3+/–mice,while the mRNA of its coding gene CACNA1C does not change.MeRIP-Seq and MeRIP-qPCR showed that the m6A methylation of CACNA1C mRNA is decreased in cultured Mettl3-knockdown cardiomyocytes and Mettl3+/–hearts.Collectively,deficiency of Mettl3 increases ventricular arrhythmia susceptibility due to the upregulation of Cav1.2 by reducing m6A modification on CACNA1C mRNA in mice.This study highlights the role of m6A modification in the regulation of cardiac electrophysiology. | Ling Shi Xuexin Jin Zheng Li Rui Gong Yang Guo Jiudong Ma Yang Zhang Benzhi Cai Baofeng Yang Dongmei Gong Zhenwei Pan | 2022 | Acta Biochimica et Biophysica Sinica2022,54,2: | 1 |

| 13 | Effects of cyclooxygenase 2 inhibitor on growth-associated protein 43 and nerve growth factor expression in dorsal root ganglion during neuropathic pain development显示文摘BACKGROUND:Inflammatory responses in injured nerves have been recognized as important factors for initially sensitizing nociceptive neurons.Cyclooxygenase(COX) is the rate-limiting enzyme in prostaglandin synthesis,and COX-2 inhibitor is involved in mechanisms of analgesia and anti-inflammation. OBJECTIVE:To investigate the effects of COX-2 inhibitor on thermal and mechanical hyperalgesia, as well as expression of growth associated protein 43(GAP-43) and nerve growth factor(NGF) in dorsal root ganglion,in a rat model of neuropathic pain due to chronic constriction injury. DESIGN,TIME AND SETTING:A randomized,controlled,comparison study that was performed at the Surgical Department and Pathological Laboratory,Second Affiliated Hospital of Shantou University Medical College from September 2006 to September 2007. MATERIALS:COX-2 inhibitor,lornoxicam,was purchased from Nycomed Pharmaceutical(Austria); rabbit anti-GAP-43,and rabbit anti-NGF polyclonal antibodies were purchased from Boster,Wuhan, China. METHODS:A total of 50 adult,Wistar rats were randomly assigned to four groups:normal control (n = 5),model(n = 15),normal saline control(n = 15),and lornoxicam treatment(n = 15).With exception of the control group,the sciatic nerve of all rats was loosely ligated to establish a model of chronic constriction injury.The model rats were divided into three subgroups according to varying post-operative survival periods:3,7 and 14 days(n = 5),respectively.Rats in the lornoxicam treatment group were intraperitoneally injected with 1.3 mg/kg lornoxicam every 12 hours throughout the entire experimental procedure.Rats in the normal saline control group were intraperitoneally injected with 1.3 mL/kg saline. MAIN OUTCOME MEASURES:Immunohistochemistry revealed expression of GAP-43 and NGF in the L5 dorsal root ganglions.Mechanical withdrawal threshold and thermal withdrawal latency were used to observe neurological behavioral changes in rats. RESULTS:The relative gray values of GAP-43- and NGF-positive neurons in the model group were remarkably increased compared with the normal control rats(P<0.01),while the relative gray values in the lornoxicam treatment group were significantly less than the model and normal saline control groups(P<0.01).Mechanical withdrawal threshold and thermal withdrawal latency gradually decreased with increasing injury time in the model,normal saline control,and lornoxicam treatment groups,and were significantly less than the normal control group(P<0.05).In addition,mechanical withdrawal threshold and thermal withdrawal latency were significantly greater in the lornoxicam treatment group compared with the model and normal saline control groups(P<0.05). Compounds 1 − 5 were successfully resolved by chiral high-performance liquid chromatography (HPLC), yielding five pairs of enantiomers: (+)- and (−)-asponchimides A−E ( 1a / 1b − 5a / 5b ). Their structural identities were discerned by extensive spectroscopic analyses, including high-resolution mass spectrometry (HRMS), ultraviolet-visible (UV-Vis) spectroscopy, infrared (IR) spectroscopy, and nuclear magnetic resonance (NMR), and their absolute configurations were determined by electronic circular dichroism (ECD) calculations. Compounds 1 − 5 are pioneering instances of NADA trimers featuring a Δ7 double bond.
