1The Reference Genome Sequence of Scutellaria baicalensis Provides Insights into the Evolution of Wogonin Biosynthesis显示文摘Scutellaria baicalensis Georgi is important in Chinese traditional medicine where preparations of dried roots,'Huang Qin,' are used for liver and lung complaints and as complementary cancer treatments. We report a high-quality reference genome sequence for S. baicalensis where 93% of the 408.14-Mb genome has been assembled into nine pseudochromosomes with a super-N50 of 33.2 Mb. Comparison of this sequence with those of closely related species in the order Lamiales, sesamum indicum and Salvia splendens,revealed that a specialized metabolic pathway for the synthesis of 4'-deoxyflavone bioactives evolved in the genus Scu-tellaria. We found that the gene encoding a specific cinnamate coenzyme A ligase likely obtained its newfunc- tion following recent mutations, and that four genes encoding enzymes in the 4'-deoxyflavone pathway are present as tandem repeats in the genome of S. baicalensis. Further analyses revealed that gene duplications, segmental duplication, gene amplification, and point mutations coupled to gene neo- and subfunctionaliza-tions were involved in the evolution of 4'-deoxyflavone synthesis in the genus Scutellaria. Our study not only provides significant insight into the evolution of specific flavone biosynthetic pathways in the mint family, Lamiaceae, but also will facilitate the development of tools for enhancing bioactive productivity by metabolic engineering in microbes or by molecular breeding in plants. 2Tandem catalysis in electrochemical CO_(2) reduction reaction显示文摘Electrochemical CO_(2) reduction reaction(CO_(2)RR)is an attractive pathway for closing the anthropogenic carbon cycle and storing intermittent renewable energy by converting CO_(2) to valuable chemicals and fuels.The production of highly reduced carbon compounds beyond CO and formate,such as hydrocarbon and oxygenate products with higher energy density,is particularly desirable for practical applications.However,the productivity towards highly reduced chemicals is typically limited by high overpotential and poor selectivity due to the multiple electron-proton transfer steps.Tandem catalysis,which is extensively utilized by nature for producing biological macromolecules with multiple enzymes via coupled reaction steps,represents a promising strategy for enhancing the CO_(2)RR performance.Improving the efficiency of CO_(2)RR via tandem catalysis has recently emerged as an exciting research frontier and achieved significant advances.Here we describe the general principles and also considerations for designing tandem catalysis for CO_(2)RR.Recent advances in constructing tandem catalysts,mainly including bimetallic alloy nanostructures,bimetallic heterostructures,bimetallic core-shell nanostructures,bimetallic mixture catalysts,metal-metal organic framework(MOF)and metal-metallic complexes,metal-nonmetal hybrid nanomaterials and copper-free hybrid nanomaterials for boosting the CO_(2)RR performance are systematically summarized.The study of tandem catalysis for CO_(2)RR is still at the early stage,and future research challenges and opportunities are also discussed.Yating Zhu Xiaoya Cui Huiling Liu Zhenguo Guo Yanfeng Dang Zhanxi Fan Zhicheng Zhang Wenping Hu 2021Nano Research2021,14,12:5
3GSH-triggered sequential catalysis for tumor imaging and eradication based on star-like Au/Pt enzyme carrier system显示文摘Distinctively different metabolism between tumor cells and normal cells endows tumor tissues unique microenvironment.In this regard,we have successfully prepared a sequential catalytic platform based on Au/Pt star for tumor theragnostic.The multifunctional probes consisted of a gold/platinum star-shaped core(Au/Pt star)conjugated with a GSH-sensitive disulfide bond(S–S),a targeting ligand(rHSA-FA),a near-infrared fluorophore(IR780)and glucose oxidase(GOx).When systemically administered in a xenografted murine model,the probes specifically targeted the tumor sites.As the disulfide linker was cleaved by intracellular GSH,the IR780 molecules could be released for photo-thermal therapy&photodynamic therapy(PTT&PDT)and imaging.Subsequently,the Pt nanolayer of the Au/Pt star and the GOx formed a sequential catalytic system:GOx effectively catalyzed intracellular glucose by consuming oxygen to generate H2O2 and enhance the local acidity,and the Pt layer exhibited peroxidase-like property to catalyze H2O2 producing toxic·OH for tumor oxidative damage.Here we demonstrated that our probes simultaneously possessed a GSH-sensitive release,real-time imaging ability,and synergetic cancer starving-like therapy/enzyme oxidative therapy/PTT/PDT features,which provides a potential strategy for effective tumor theragnostic.Amin Zhang Qian Zhang Gabriel Alfranca Shaojun Pan Zhicheng Huang Jin Cheng Qiang Ma Jie Song Yunxiang Pan Jian Ni Lijun Ma Daxiang Cui 2020Nano Research2020,13,1:4
4A four-gene signature-derived risk score for glioblastoma:prospects for prognostic and response predictive analyses显示文摘Objective: Glioblastoma(GBM) is the most common primary malignant brain tumor regulated by numerous genes, with poor survival outcomes and unsatisfactory response to therapy.Therefore, a robust, multi-gene signature-derived model is required to predict the prognosis and treatment response in GBM.Methods: Gene expression data of GBM from TCGA and GEO datasets were used to identify differentially expressed genes(DEGs)through DESeq2 or LIMMA methods.The DEGs were then overlapped and used for survival analysis by univariate and multivariate COX regression.Based on the gene signature of multiple survival-associated DEGs, a risk score model was established,and its prognostic and predictive role was estimated through Kaplan–Meier analysis and log-rank test.Gene set enrichment analysis(GSEA) was conducted to explore high-risk score-associated pathways.Western blot was used for protein detection.Results: Four survival-associated DEGs of GBM were identified: OSMR, HOXC10, SCARA3, and SLC39A10.The four-gene signature-derived risk score was higher in GBM than in normal brain tissues.GBM patients with a high-risk score had poor survival outcomes.The high-risk group treated with temozolomide chemotherapy or radiotherapy survived for a shorter duration than the low-risk group.GSEA showed that the high-risk score was enriched with pathways such as vasculature development and cell adhesion.Western blot confirmed that the proteins of these four genes were differentially expressed in GBM cells.Conclusions: The four-gene signature-derived risk score functions well in predicting the prognosis and treatment response in GBM and will be useful for guiding therapeutic strategies for GBM patients.Mianfu Cao Juan Cai Ye Yuan Yu Shi Hong Wu Qing Liu Yueliang Yao Lu Chen Weiqi Dang XiangZhang Jingfang Xiao Kaidi Yang Zhicheng He Xiaohong Yao Yonghong Cui Xia Zhang Xiuwu Bian 2019Cancer Biology & Medicine2019,16,3:2
5Landscape and Fruit Developmental Regulation of Alternative Splicing in Tomato by Genome-Wide Analysis显示文摘In eukaryotes, alternative splicing(AS) is one of the posttranscriptional regulatory mechanisms that play important roles by generating transcriptome diversity. To obtain a global view of AS and its dynamics during tomato fruit development, we analyzed the AS events using a large amount of transcriptome datasets. Same with other plant species, about half of the expressed multiexonic genes were alternatively spliced in tomato. Besides that, our further analyzation of RNA-seq datasets of ovule and pericarp at early fruit developmental stages reveals that the dynamic alteration of AS events occurred in specific tissues and AS was regulated spatially and temporally during early fruit development in tomato. By investigating the sequence variations at splice sites causing differential AS events between tomato cultivar ‘Moneymaker' and wild species Solanum pimpinellifolium PI365967, we uncover that AS may play the regulatory roles during domestication of tomato. 6Fine-tuning inverse metal-support interaction boosts electrochemical transformation of methanol into formaldehyde based on density functional theory显示文摘Different from traditional metal-support heterogenous catalysts,inverse heterogeneous catalysts,in which the surface of metal is decorated by metal oxide,have recently attracted increasing interests owing to the unique interracial effect and electronic structure.However,a deep insight into the effect of metaloxide interaction on the catalytic performance still remains a great challenge.In our work,an inverse hematite/palladium(Fe_(2) O_(3)/Pd) hybrid nanostructure,i.e.,the active Fe_(2) O_(3) ultrathin oxide layers partially covering on the surface of Pd nanoparticles(NPs),exhibited superior electrocatalytic performance towards methanol oxidation reaction(MOR) as compared to the bare Pd NPs based on density functional theory calculation.The charge could transfer from Pd to Fe_(2) O_(3) driven by the built-in potential at the interface of Pd and Fe_(2) O_(3),which favors the downshift of d band center of Pd.With the assistance of interfacial hydroxyl OH*,the cleavage of O—H and C—H in CH_(3) OH could take place much easily with lower barrier ene rgy on Fe_(2) O_(3)/Pd than that on pure Pd via two electrons transferring reaction pathways.Our results highlight that the syne rgy of Pd and Fe_(2) O_(3) at the interface could facilitate the electrochemical transformation of methanol into formaldehyde assisted with interfacial hydroxyl OH*.Wenjuan Yang Junjun Li Xiaoya Cui Chenhuai Yang Yiting Liu Xianwei Zeng Zhicheng Zhang Qitao Zhang 2021Chinese Chemical Letters2021,32,8:2
7Heterojunction interface editing in Co/NiCoP nanospheres by oxygen atoms decoration for synergistic accelerating hydrogen and oxygen evolution electrocatalysis显示文摘Controllable designing of well-defined heterojunction nanostructures provides an insightful strategy for accelerating the kinetics of the hydrogen and oxygen evolution reactions(HER/OER),but such task is still challenging.Herein,we proposed a protocol of heterojunction interface editing(HIE)strategy by oxygen atoms decoration for synergistic boosting electrocatalytic HER and OER performances.A novel Co/NiCoP nanospheres(NSs)heterojunction was synthesized by crystal seed template transformation method with Ni_(5)P_(4) microspheres as seeds.The effective oxygen atoms interface editing increased the oxidation state of Co atoms and prolonged the Co-P bond length of Co/NiCoP NSs heterojunction,thus the electron localization on P sites was enhanced,leading to the dramatically elevated HER and OER performances simultaneously.The as-constructed O-Co/NiCoP NSs show excellent electrocatalytic activity with 361 and 430 mV vs.reversible hydrogen electrode(RHE)to arrive high current density of 300 mA·cm^(-2)for HER and OER in 1 M KOH as well as good stability.The proposed HIE concept could provide a new perspective on the catalyst design for energy conversion systems.Yan Lin Xiaojiao Cui Yilin Zhao Zhicheng Liu Guoxin Zhang Yuan Pan 2023Nano Research2023,16,7:1
