| 1 | Abrogation of Hn RNP L enhances anti-PD-1 therapy efficacy via diminishing PD-L1 and promoting CD8^(+) T cell-mediated ferroptosis in castration-resistant prostate cancer显示文摘Owing to incurable castration-resistant prostate cancer(CRPC)ultimately developing after treating with androgen deprivation therapy(ADT),it is vital to devise new therapeutic strategies to treat CRPC.Treatments that target programmed cell death protein 1(PD-1)and programmed death ligand-1(PD-L1)have been approved for human cancers with clinical benefit.However,many patients,especially prostate cancer,fail to respond to anti-PD-1/PD-L1 treatment,so it is an urgent need to seek a support strategy for improving the traditional PD-1/PD-L1 targeting immunotherapy.In the present study,analyzing the data from our prostate cancer tissue microarray,we found that PD-L1 expression was positively correlated with the expression of heterogeneous nuclear ribonucleoprotein L(Hn RNP L).Hence,we further investigated the potential role of Hn RNP L on the PD-L1 expression,the sensitivity of cancer cells to T-cell killing and the synergistic effect with anti-PD-1 therapy in CRPC.Indeed,Hn RNP L knockdown effectively decreased PD-L1 expression and recovered the sensitivity of cancer cells to T-cell killing in vitro and in vivo,on the contrary,Hn RNP L overexpression led to the opposite effect in CRPC cells.In addition,consistent with the previous study,we revealed that ferroptosis played a critical role in T-cell-induced cancer cell death,and Hn RNP L promoted the cancer immune escape partly through targeting YY1/PD-L1 axis and inhibiting ferroptosis in CRPC cells.Furthermore,Hn RNP L knockdown enhanced antitumor immunity by recruiting infiltrating CD8^(+)T cells and synergized with anti-PD-1 therapy in CRPC tumors.This study provided biological evidence that Hn RNP L knockdown might be a novel therapeutic agent in PD-L1/PD-1 blockade strategy that enhanced anti-tumor immune response in CRPC. | Xumin Zhou Libin Zou Hangyu Liao Junqi Luo Taowei Yang Jun Wu Wenbin Chen Kaihui Wu Shengren Cen Daojun Lv Fangpeng Shu Yu Yang Chun Li Bingkun Li Xiangming Mao | 2022 | Acta Pharmaceutica Sinica B2022,12,2: | 4 |
| 2 | Pristine PN junction toward atomic layer devices显示文摘In semiconductor manufacturing,PN junction is formed by introducing dopants to activate neighboring electron and hole conductance.To avoid structural distortion and failure,it generally requires the foreign dopants localize in the designated micro-areas.This,however,is challenging due to an inevitable interdiffusion process.Here we report a brand-new junction architecture,called'layer PN junction',that might break through such limit and help redefine the semiconductor device architecture.Different from all existing semiconductors,we find that a variety of van der Waals materials are doping themselves from n-to p-type conductance with an increasing/decreasing layer-number.It means the capability of constructing homogeneous PN junctions in monolayers'dimension/precision,with record high rectification-ratio(>10^(5))and low cut-off current(<1 pA).More importantly,it spawns intriguing functionalities,like gate-switchable-rectification and noise-signal decoupled avalanching.Findings disclosed here might open up a path to develop novel nanodevice applications,where the geometrical size becomes the only critical factor in tuning charge-carrier distribution and thus functionality. | Hui Xia Man Luo Wenjing Wang Hailu Wang Tianxin Li Zhen Wang Hangyu Xu Yue Chen Yong Zhou Fang Wang Runzhang Xie Peng Wang Weida Hu Wei Lu | 2022 | Light(Science & Applications)2022,11,7: | 4 |
| 3 | Regulating the growth of lithium dendrite by coating an ultra-thin layer of gold on separator for improving the fast-charging ability of graphite anode显示文摘With the ever-growing application of lithium-ion batteries(LIBs), their fast-charging technology has attracted great interests of scientists. However, growth of lithium dendrites during fast charge of the bat teries with high energy density may pose great threats to the operation and cause serious safety issues Herein, we prepared a functional separator with an ultra-thin(20 nm) layer of Au nanoparticles deposited by evaporation coating method which could regulate growth direction and morphology of the lithium dendrites, owing to nearly zero overpotential of lithium meal nucleation on lithiated Au. Once the Li den drites are about to form on the graphite anode during fast charging(or lithiation), they plate predomi nantly on the Au deposited separator rather than on the graphite. Such selective deposition does no compromise the electrochemical performance of batteries under normal cycling. More importantly, i enables the better cycling stability of batteries at fast charge condition. The Li/Graphite cells with Au nanoparticles coated separator could cycle stably with a high areal capacity retention of 90.5% over 95 cycles at the current density of 0.72 m A cm^(-2). | 8 | Oxaliplatin plus irinotecan vs irinotecan as second-line treatment in pancreatic cancer patients:a randomized–controlled open-label Phase II study显示文摘Background:Limited second-line therapeutic options are available for metastasis pancreatic cancer(mPC).We aimed to explore the efficacy and safety of oxaliplatin plus irinotecan(IROX)in mPC patients.Methods:This is an open-label,Phase 2,randomized study of mPC patients(aged 18–75 years)who failed when using gemcitabine plus S-1 as first-line therapy.Block randomization with a block size of four was used to randomly assign patients(1:1)between October 2015 and December 2017 to receive either IROX(oxaliplatin 85mg/m2 and irinotecan 160mg/m2)or irinotecan monotherapy(irinotecan 180mg/m^(2))until disease progression,unacceptable adverse events,or consent withdrawal.The primary end point was overall survival,and the secondary end points were progression-free survival,overall response rate,and adverse event rate.Results:A total of 74 patients were enrolled in this study,including 44 males and 30 females,with an average age of 61 years.The median overall survival was 10.2 and 6.7 months(adjusted hazard ratio[HR],0.7;95%confidence interval[CI],0.4–1.2;P=0.20)and the median progression-free survival was 5.1 and 2.3 months(adjusted HR,0.4;95%CI,0.2–0.6;P<0.01)in the IROX group and irinotecan group,respectively.The overall response rates were 18.4%(7/38)in the IROX group and 5.5%(2/36)in the irinotecan group(P=0.06).Grade 3–4 adverse events occurred in 34%(13/38)of patients in the IROX group and 19%(7/36)of patients in the irinotecan group(P=0.15).Conclusions:IROX had no significant survival benefit over irinotecan monotherapy in our study.However,IROX reduced the risk of disease progression by 60%,with acceptable toxicity. | Hangyu Zhang Zhou Tong Lulu Liu Qihan Fu Xudong Zhu Xiaomeng Dai Xuanwen Bao Weijia Fang Yi Zheng Peng Zhao | 2023 | Gastroenterology Report2023,11,1: | 0 |
| 9 | Intra-seasonal variability of the abyssal currents in COMRA's contract area in the Clarion-Clipperton Zone显示文摘In this paper,the intra-seasonal variability of the abyssal currents in the China Ocean Mineral Resources Association(COMRA)polymetallic nodule contact area,located in the western part of the Clarion and Clipperton Fraction Zone in the tropical East Pacific,is investigated using direct observations from subsurface mooring instruments as well as sea-surface height data and reanalysis products.Mooring observations were conducted from September 13,2017 to August 15,2018 in the COMRA contact area(10°N,154°W).The results were as follows:(1)At depths below 200 m,the kinetic energy of intra-seasonal variability(20-100 d)accounts for more than 40%of the overall low-frequency variability,while the ratio reaches more than 50%below 2000 m.(2)At depths below 200 m,currents show a synchronous oscillation with a characteristic time scale of 30 d,lasting from October to the following January;the energy of the 30-d oscillation increases with depth until the layer of approximately 4616 m,and the maximum velocity is approximately 10 cm/s.(3)The 30-d oscillation of deep currents is correlated with the tropical instability waves in the upper ocean. | Fangfang Kuang Jing Cha Junpeng Zhang Aijun Pan Hangyu Chen Xiwu Zhou Chunsheng Jing Xiaogang Guo | 2022 | Acta Oceanologica Sinica2022,41,11: | 0 |
| 10 | Observation of Magnetic Dipole Forbidden Transitions in LHD and Its Application to Burning Plasma Diagnostics显示文摘Magnetic dipole forbidden (M1) transition was studied in large helical device (LHD)and F-, Si- and Ti-like M1 transitions are successfully observed for highly ionized Ar, Kr, Mo andXe ions. The wavelengths measured in visible range for the heavy elements, which are carefullydetermined with extremely small uncertainties of 0.02~0.05 A as a standard wavelength of usualelectric dipole (E1) plasma emissions, are compared with theoretical predictions. The result showsa good agreement with recent Hatree-Fock calculation including semi-empirical adjustment. TheM1 intensity for the F-like ions is examined by analyzing the intensity ratio of M1 to E1. Densitydependence of the ratio is experimentally verified by comparing with collisional- radiative modelcalculation on level population. The M1/E1 line ratio for the F-like ions is applied to the(He^(2+)) particle diagnostics in ITER, in which a steady-state operation of burning plasmas basedon D-T fusion reaction is expected with α particle heating. Unfortunately, the present estimationsuggests a negative result for the α particle measurement because the ratio is largely enhanced bythe collisional excitation with bulk ions due to high ion temperature of ITER of 10 keV as assumedand the resultant effect of the collisional excitation with α particles becomes less. | 11 | Configurable circular-polarization-dependent optoelectronic silent state for ultrahigh light ellipticity discrimination显示文摘Filterless light-ellipticity-sensitive optoelectronic response generally has low discrimination,thus severely hindering the development of monolithic polarization detectors.Here,we achieve a breakthrough based on a configurable circular-polarization-dependent optoelectronic silent state created by the superposition of two photoresponses with enantiomerically opposite ellipticity dependences.The zero photocurrent and the significantly suppressed noise of the optoelectronic silent state singularly enhance the circular polarization extinction ratio(CPER)and the sensitivity to light ellipticity perturbation.The CPER of our device approaches infinity by the traditional definition.The newly established CPER taking noise into account is 3-4 orders of magnitude higher than those of ordinary integrated circular polarization detectors,and it remains high in an expanded wavelength range.The noise equivalent light ellipticity difference goes below 0.009° Hz-1/2 at modulation frequencies above 1000 Hz by a light power of 281 uW.This scheme brings a leap in developing monolithic ultracompact circular polarization detectors. | Yonghao Bu Xiansong Ren Jing Zhou Zhenhan Zhang Jie Deng Hangyu Xu Runzhang Xie Tianxin Li Weida Hu Xia Guo Wei Lu Xiaoshuang Chen | 2023 | Light(Science & Applications)2023,12,8: | 0 |
| 12 | Genetic differentiation and diversity of SARS-CoV-2 Omicron variant in its early outbreak显示文摘The recently emerged Omicron variant of severe acute respiratory syndrome coronavirus 2(SARS‐CoV‐2)has quickly spread around the world.Although many consensus mutations of the Omicron variant have been recognized,little is known about its genetic variation during its transmission in the population.Here,we comprehensively analyzed the genetic differentiation and diversity of the Omicron variant during its early outbreak.We found that Omicron achieved more structural variations,especially deletions,on the SARS‐CoV‐2 genome than the other four variants of concern(Alpha,Beta,Gamma,and Delta)in the same timescale.In addition,the Omicron variant acquired,except for 50 consensus mutations,seven great new non‐synonymous nucleotide substitutions during its spread.Three of them are on the S protein,including S_A701V,S_L1081V,and S_R346K,which belong to the receptor‐binding domain(RBD).The Omicron BA.1 branch could be divided into five divergent groups spreading across different countries and regions based on these seven novel mutations.Furthermore,we found that the Omicron variant possesses more mutations related to a faster transmission rate than the other SARS‐CoV‐2 variants by assessing the relationship between the genetic diversity and transmission rate.The findings indicated that more attention should be paid to the significant genetic differentiation and diversity of the Omicron variant for better disease prevention and control. | Shenghui Weng Jingzhe Shang Yexiao Cheng Hangyu Zhou Chengyang Ji Rong Yang Aiping Wu | 2022 | Biosafety and Health2022,4,3: | 0 |
