2Combinations of Hd2 and Hd4 genes determine rice adaptability to Heilongjiang Province, northern limit of China显示文摘Heading date is a key trait in rice domestication and adaption, and a number of quantitative trait loci(QTLs)have been identified. The rice(Oryza sativa L.) cultivars in the Heilongjiang Province, the northernmost region of China,have to flower extremely early to fulfill their life cycle.However, the critical genes or different gene combinations controlling early flowering in this region have not been determined. QTL and candidate gene analysis revealed that Hd2/Ghd7.1/Os PRR37 plays a major role in controlling rice distribution in Heilongjiang. Further association analysis with a collection of rice cultivars demonstrated that another three major QTL genes(Hd4/Ghd7, Hd5/DTH8/Ghd8, and Hd1)also participate in regulating heading date under natural long day(LD) conditions. Hd2/Ghd7.1/Os PRR37 and Hd4/Ghd7 are two major QTLs and function additively. With the northward rice cultivation, the Hd2/Ghd7.1/Os PRR37 and Hd4/Ghd7 haplotypes became non-functional alleles. Hd1 might be non-functional in most Heilongjiang rice varieties,implying that recessive hd1 were selected during local rice breeding. Non-functional Hd5/DTH8/Ghd8 is very rare, but constitutes a potential target for breeding extremely early flowering cultivars. 3Population Genomic Analysis and De Novo Assembly Reveal the Origin of Weedy Rice as an Evolutionary Game显示文摘Crop weediness,especially that of weedy rice(Oryza sativa f.spontanea),remains mysterious.Weedy rice possesses robust ecological adaptability;however,how this strain originated and gradually formed proprietary genetic features remains unclear?Here,we demonstrate that weedy rice at Asian high latitudes(WRAH)is phylogenetically well defined and possesses unselected genomic characteristics in many divergence regions between weedy and cultivated rice.We also identified novel quantitative trait loci underlying weedy-specific traits,and revealed that a genome block on the end of chromosome 1 is associated with rice weediness.To identify the genomic modifications underlying weedy rice evolution,we generated the first de novo assembly of a high-quality weedy rice genome(WR04-6),and conducted a comparative genomics study between WR04-6 with other rice reference genomes.Multiple lines of evidence,including the results of demographic scenario comparisons,suggest that differentiation between weedy rice and cultivated rice was initiated by genetic improvement of cultivated rice and that the essence of weediness arose through semi-domestication.A plant height model further implied that the origin of WRAH can be modeled as an evolutionary game and indicated that strategy-based selection driven by fitness shaped its genomic diversity.Jian Sun Dianrong Ma Liang Tang Minghui Zhao Guangchen Zhangh Wenjia Wang Jiayu Song Xiang Li Zimeng Liu Wenxing Zhang Quan Xu Yuncheng Zhou Jianzhong Wu Toshio Yamamoto Fei Dai Yan Lei Song Li Gang Zhou Hongkun Zheng Zhengjin Xu Wenfu Chen 2019Molecular Plant2019,12,5:9
4Novel circular RNA expression profile of uveal melanoma revealed by microarray显示文摘Objective: The present study aimed to investigate circular RNA(circRNA) expression in uveal melanoma(UM).Methods: First,we used microarray to compare the expression profiles of circRNA in five UM samples and five normal uvea tissues.Next,bioinformatics analyses,including gene ontology(GO) analysis and pathway analysis,were applied to study these differentially expressed circRNAs to predict pathogenic pathways that may be involved.Quantitative real-time polymerase chain reaction(qRT-PCR) in 20 UM samples and 20 normal uvea samples was used to confirm the circRNA expression profiles obtained from the microarray data.Finally,we analyzed the interaction between validated circRNAs and their potential cancer-associated miRNA targets.Results: In total,50,579 circRNAs [fold change(FC) ≥2.0; P<0.05],including 20,654 up-regulated and 29,925 down-regulated circRNAs,were identified as differentially expressed between UM tissues and normal uvea tissues.We used qRT-PCR to verify seven dysregulated circRNAs indicated by the microarray data,including hsa_circ_0119873,hsa_circ_0128533,hsa_circ_0047924,hsa_circ_0103232,hsa-circRNA10628-6,hsa_circ_0032148 and hsa_circ_0133460,which may be promising candidates to study future molecular mechanisms.Conclusions: This study explored,for the first time,the abnormal expression of circRNAs in UM and described the expression profile of circRNAs,providing a new potential target for the mechanism of UM and future treatment of UM.Xuan Yang Yang Li Yueming Liu Xiaolin Xu Yingzhi Wang Yanni Yan Wenjia Zhou Jingyan Yang Wenbin Wei 2018Chinese Journal of Cancer Research2018,30,6:6
5Supersaturation controlled growth of MAFAPbI3 perovskite film for high efficiency solar cells显示文摘Controlling the nucleation and growth of organic-inorganic hybrids perovskite is of key importance to improve the morphology and crystallinity of perovskite films. However, the growth mechanism of perovskite films based on classical crystallization theory is not fully understood. Here, we develop a supersaturation controlled strategy(SCS) to balance the nucleation and crystal growth speeds. By this strategy, we are able to find an ideal supersaturation region to realize a balance of nucleation and crystal growth, which yields highly crystallized perovskite films with micrometer-scale grains. Besides, we provide a thoughtful analysis of nucleation and growth based on the fabrication of the perovskite films. As a result, the highest photovoltaic power conversion efficiencies(PCE) of 19.70% and 20.31% are obtained for the planar and the meso-superstructured devices, respectively. 9Structural and biochemical studies of RIG-I antiviral signaling显示文摘Retinoic acid-inducible gene I(RIG-I)is an important pattern recognition receptor that detects viral RNA and triggers the production of type-I interferons through the downstream adaptor MAVS(also called IPS-1,CARDIF,or VISA).A series of structural studies have elaborated some of the mechanisms of dsRNA recognition and activation of RIG-I.Recent studies have proposed that K63-linked ubiquitination of,or unanchored K63-linked polyubiquitin binding to RIG-I positively regulates MAVS-mediated antiviral signaling.Conversely phos-phorylation of RIG-I appears to play an inhibitory role in controlling RIG-I antiviral signal transduction.Here we performed a combined structural and biochemical study to further define the regulatory features of RIG-I signaling.ATP and dsRNA binding triggered dimeriza-tion of RIG-I with conformational rearrangements of the tandem CARD domains.Full length RIG-I appeared to form a complex with dsRNA in a 2:2 molar ratio.Com-pared with the previously reported crystal structures of RIG-I in inactive state,our electron microscopic struc-ture of full length RIG-I in complex with blunt-ended dsRNA,for the first time,revealed an exposed active conformation of the CARD domains.Moreover,we found that purified recombinant RIG-I proteins could bind to the CARD domain of MAVS independently of dsRNA,while S8E and T170E phosphorylation-mimick-ing mutants of RIG-I were defective in binding E3 ligase TRIM25,unanchored K63-linked polyubiquitin,and MAVS regardless of dsRNA.These findings suggested that phosphorylation of RIG inhibited downstream signaling by impairing RIG-I binding with polyubiquitin and its interaction with MAVS.Miao Feng Zhanyu Ding Liang Xu Liangliang Kong Wenjia Wang Shi Jiao Zhubing Shi Mark IGreene Yao Cong Zhaocai Zhou 2013Protein & Cell2013,4,2:2
10Compound Danshen Dripping Pill inhibits hypercholesterolemia/atherosclerosis-induced heart failure in ApoE and LDLR dual deficient mice via multiple mechanisms显示文摘Heart failure is the leading cause of death worldwide.Compound Danshen Dripping Pill(CDDP)or CDDP combined with simvastatin has been widely used to treat patients with myocardial infarction and other cardiovascular diseases in China.However,the effect of CDDP on hypercholesterolemia/atherosclerosis-induced heart failure is unknown.We constructed a new model of heart failure induced by hypercholesterolemia/atherosclerosis in apolipoprotein E(ApoE)and LDL receptor(LDLR)dual deficient(ApoE^(–/–)LDLR^(–/–))mice and investigated the effect of CDDP or CDDP plus a low dose of simvastatin on the heart failure.CDDP or CDDP plus a low dose of simvastatin inhibited heart injury by multiple actions including anti-myocardial dysfunction and anti-fibrosis.Mechanistically,both Wnt and lysine-specific demethylase 4A(KDM4A)pathways were significantly activated in mice with heart injury.Conversely,CDDP or CDDP plus a low dose of simvastatin inhibited Wnt pathway by markedly up-regulating expression of Wnt inhibitors.While the anti-inflammation and anti-oxidative stress by CDDP were achieved by inhibiting KDM4A expression and activity.In addition,CDDP attenuated simvastatin-induced myolysis in skeletal muscle.Taken together,our study suggests that CDDP or CDDP plus a low dose of simvastatin can be an effective therapy to reduce hypercholesterolemia/atherosclerosis-induced heart failure.Yanfang Yang Ke Feng Liying Yuan Yuxin Liu Mengying Zhang Kaimin Guo Zequn Yin Wenjia Wang Shuiping Zhou He Sun Kaijing Yan Xijun Yan Xuerui Wang Yajun Duan Yunhui Hu Jihong Han 2023Acta Pharmaceutica Sinica B2023,13,3:2

20Manipulating the ionic nanophase of Nafion by in-situ precise hybridization with polymer quantum dot towards highly enhanced fuel cell performances显示文摘Hierarchical-structure materials hold great promise for numerous applied domains such as fuel cell,sensor,and optic.However,the developments are significantly impeded by the lack of efficient strategy permitting precise and efficient decoration of specific confined space.Here,an in-situ precise hybridization strategy is proposed to efficiently manipulate the nanostructure of membrane nanochannels.Typically,Nafion ionic nanochannels are impregnated with precursors via heat swelling,followed by microwave-assisted condensation to form polymer quantum dot network.The formation of polymer quantum dot network significantly improves the stability and functionality of ionic nanophase(i.e.,ionic nanochannel).This helps hybrid membrane achieving enhanced proton conduction and methanol barrier properties,resulting in over ten times increase in proton/methanol selectivity.These then impart prominent device performances for both hydrogen and methanol fuel cells with the elevation of~100%.Importantly,such function manipulation of ionic nanochannels is achieved with fully maintaining function of backbone nanophase.Besides,the regulation of physical topology and chemical environment of ionic nanochannel also brings optimization of gas and ion separation properties.This facile and versatile strategy may open up a new avenue for decorating confined space of many hierarchical-structure materials.Wenjia Wu Zhuofan Zhou Yan Wang Yatao Zhang Yong Wang Jingtao Wang Yecheng Zou 2022Nano Research2022,15,5:1
