| 1 | GSDMB promotes non-canonical pyroptosis by enhancing caspase-4 activity显示文摘Gasdermin B (GSDMB) has been reported to be associated with immune diseases in humans, but the detailed molecular mechanisms remain unsolved. The N-terminus of GSDMB by itself, unlike other gasdermin family proteins, does not induce cell death. Here, we show that GSDMB is highly expressed in the leukocytes of septic shock patients, which is associated with increased release of the gasdermin D (GSDMD) N-terminus. GSDMB expression and the accumulation of the N-terminal fragment of GSDMD are induced by the activation of the non-canonical pyroptosis pathway in a human monocyte cell line. The downregulation of GSDMB alleviates the cleavage of GSDMD and cell death. Consistently, the overexpression of GSDMB promotes GSDMD cleavage, accompanied by increased LDH release. We further found that GSDMB promotes caspase-4 activity, which is required for the cleavage of GSDMD in non-canonical pyroptosis, by directly binding to the CARD domain of caspase-4. | 2 | Chemical characterization and source identification of PM_(2.5) in Luoyang after the clean air actions显示文摘Luoyang is a typical heavy industrial city in China,with a coal-dominated energy structure and serious air pollution.Following the implementation of the clean air actions,the physic-ochemical characteristics and sources of PM_(2.5) have changed.A comprehensive study of PM_(2.5) was conducted from October 16,2019 to January 23,2020 to evaluate the effectiveness of previous control measures and further to provide theory basis for more effective policies in the future.Results showed that the aerosol pollution in Luoyang in autumn and win-ter is still serious with the average concentration of 91.1 μg/m^(3),although a large reduction(46.9%)since 2014.With the contribution of nitrate increased from 12.5%to 25.1%and sul-fate decreased from 16.7%to 11.2%,aerosol pollution has changed from sulfate-dominate to nitrate-dominate.High NO_(3)^(-)/SO_(4)^(2-)ratio and the increasing of NO_(3)^(-)/SO_(4)^(2-)ratio with the aggravation of pollution indicating vehicle exhaust playing an increasingly important role in PM_(2.5) pollution in Luoyang,especially in the haze processes.Secondary inorganic ions contributed significantly to the enhancement of PM_(2.5) during the pollution period.The high value of Cl^(-)/Na^(+)and EC concentration indicate coal combustion in Luoyang is still serious.The top three contributor sources were secondary inorganic aerosols(33.3%),coal combus-tion(13.6%),and industrial emissions(13.4%).Close-range transport from the western and northeastern directions were more important factors in air pollution in Luoyang during the sampling period.It is necessary to strengthen the control of coal combustion and reduce vehicle emissions in future policies. | Min Xu Zirui Liu Bo Hu Guangxuan Yan Jianan Zou Shuman Zhao Jingxiang Zhou Xianhui Liu Xueping Zheng Xiaoyan Zhang Jing Cao Mengshuang Guan Yirong Lv Yanyun Zhang | 2022 | Journal of Environmental Sciences2022,34,5: | 1 |
| 3 | Multiplexed imaging of tumor immune microenvironmental markers in locally advanced or metastatic non-small-cell lung cancer characterizes the features of response to PD-1 blockade plus chemotherapy显示文摘Background:Although programmed cell death 1(PD-1)blockade plus chemotherapy can significantly prolong the progression-free survival(PFS)and overall survival(OS)in first-line settings in patients with driver-negative advanced non-small-cell lung cancer(NSCLC),the predictive biomarkers remain undetermined.Here,we investigated the predictive value of tumor immune microenvironmental marker expression to characterize the response features to PD-1 blockade plus chemotherapy.Methods:Tumor tissue samples at baseline were prospectively collected from 144 locally advanced or metastatic NSCLC patients without driver gene alterations who received camrelizumab plus chemotherapy or chemotherapy alone.Tumor immune microenvironmental markers,including PD-1 ligand(PDL1),CD8,CD68,CD4 and forkhead box P3,were assessed using multiplex immunofluorescence(mIF)assays.Kaplan-Meier curveswere used to determine treatment outcome differences according to their expression status.Mutational profiles were compared between tumors with distinct expression levels of these markers and their combinations.Results:Responders had significantly higher CD8/PD-L1(P=0.015)or CD68/PD-L1 co-expression levels(P=0.021)than non-responders in the camrelizumab plus chemotherapy group,while no difference was observed in the chemotherapy group.Patients with high CD8/PD-L1 or CD68/PD-L1 co-expression level was associated with significantly longer PFS(P=0.002,P=0.024;respectively)and OS(P=0.006,P=0.026;respectively)than those with low co-expression in camrelizumab plus chemotherapy group.When comparing survival in the camrelizumab plus chemotherapy with chemotherapy by CD8/PD-L1 co-expression stratification,significantly better PFS(P=0.003)and OS(P=0.032)were observed in high co-expression subgroups.The predictive value of CD8/PD-L1 and CD68/PD-L1 co-expression remained statistically significant for PFS and OS when adjusting clinicopathological features.Although the prevalence of TP53 or KRAS mutations was similar between patients with and without CD8/PD-L1 or CD68/PD-L1 co-expression,the positive groups had a significantly higher proportion of TP53/KRAS co-mutations than the negative groups(both 13.0%vs.0.0%,P=0.023).Notably,enriched PI3K(P=0.012)and cell cycle pathway(P=0.021)were found in the CD8/PD-L1 co-expression group.Conclusion:Tumor immune microenvironmental marker expression,especially CD8/PD-L1 or CD68/PD-L1 co-expression,was associated with the efficacy of PD-1 blockade plus chemotherapy as first-line treatment in patients with advanced NSCLC. | Fengying Wu Tao Jiang Gongyan Chen Yunchao Huang Jianying Zhou Lizhu Lin Jifeng Feng Zhehai Wang Yongqian Shu Jianhua Shi Yi Hu Qiming Wang Ying Cheng Jianhua Chen Xiaoyan Lin Yongsheng Wang Jianan Huang Jiuwei Cui Lejie Cao Yunpeng Liu Yiping Zhang Yueyin Pan Jun Zhao LiPing Wang Jianhua Chang Qun Chen Xiubao Ren Wei Zhang Yun Fan Zhiyong He Jian Fang Kangsheng Gu Xiaorong Dong Tao Zhang Wei Shi Jianjun Zou Xuejuan Bai Shengxiang Ren Caicun Zhou | 2022 | Cancer Communications2022,42,12: | 1 |

| 6 | Isolation, Identification and Pathogenicity Evaluation of Pathogens of Soybean Bacterial Diseases显示文摘In order to research soybean bacterial diseases in Heilongjiang Province,a bacterial strain was isolated from diseased leaves of Suinong 14 collected from Harbin Yanjiagang farm. The pathogen had been carried out antibiotic resistance identification,molecular identification,re-inoculation identification and pathogenicity identification respectively. By Gram staining,pathogen inoculation test,16 S r DNA molecular identification,the pathogen was determined as Pseudomonas syringae and named Psgneau 2. The isolated strain presented resistance to 25,50 and 100 μg/m L ampicillin and carbenicillin,but susceptible to spectinomycin,rifampicin,kanamycin,gentamicin,tetracycline and chloramphenicol. | 7 | A soybean NAC homolog contributes to resistance to Phytophthora sojae mediated by dirigent proteins显示文摘Phytophthora sojae infection severely impairs soybean production. We previously identified a dirigent protein, Gm DRR1(Glycine max Disease Resistant Response 1), that increases soybean resistance to P.sojae. However, the molecular basis of Gm DRR1 function remained largely uncharacterized. In the present study, analysis of Gm DRR1-RNAi, Gm DRR1-overexpressing, and CRISPR/Cas9-derived Gmdrr1 mutant lines revealed that Gm DRR1 expression significantly restricted P. sojae growth. Combining coimmunoprecipitation with liquid chromatography–tandem mass spectrometry revealed a Gm DRR1-interacting protein, Gm DRR2, which is homologous to Gm DRR1. An E-coniferyl alcohol coupling assay indicated that Gm DRR1 promotes the synthesis of(+)-pinoresinol, which helps to protect plants from P. sojae. The Gm NAC1(Glyma.05 G025500) transcription factor bound to the Gm DRR1 promoter both in vitro and in vivo to upregulate Gm DRR1 expression. Soybean resistance to P. sojae was increased by overexpression of Gm NAC1. Our findings suggest a novel signaling pathway involving a NAC transcription factor that mediates soybean resistance to P. sojae.
