| 3 | Molecular analysis of the chloroplast Cu/Zn-SOD gene(AhCSD2) in peanut显示文摘Superoxide dismutase(SOD, EC 1.15.1.1) plays a key role in response to drought stress, and differences in SOD activity changes among cultivars are important under drought conditions. We obtained the full-length DNA of the chloroplast Cu/Zn-SOD gene(Ah CSD2)from 11 allotetraploid cultivars and 5 diploid wild species in peanut. BLAST search against the peanut genome showed that the Ah CSD2 genes g CSD2-1 and g CSD2-2 are located at the tops of chromosome A03(A genome) and B03(B genome), respectively, and both contain 8exons and 7 introns. Nucleotide sequence analyses indicated that g CSD2-2 sequences were identical among all the tested cultivars, while g CSD2-1 sequences showed allelic variations.The amino acid sequences deduced from g CSD2-1 and g CSD2-2 both contain a chloroplast transit peptide and are distinguished by 6 amino acid(aa) residue differences. The other 2aa residue variations in the mature peptide regions give rise to three-dimensional structure changes of the protein deduced from the genes g CSD2-1 and g CSD2-2. Sequences analyses of cultivars and wild species showed that g CSD2-2 of Arachis hypogaea and g Aip CSD2(Arachis ipaensis) are identical, and despite the abundant polymorphic loci between g CSD2-1 of A.hypogaea and sequences from A genome wild species, the deduced amino acid sequence of Ah CSD2-1(A. hypogaea) is identical to that of Adu CSD2(Arachis duranensis), whereas Aco CSD2(Arachis correntina) and Aca CSD2(Arachis cardenasii) both have 2 aa differences in the transit peptide region compared with Ah CSD2-1(A. hypogaea). Based on the Peanut Genome Project, promoter prediction revealed many stress-related cis-acting elements within the potential promoter regions(pp-A and pp-B). pp-A contains more binding sites for drought-associated transcriptional factors than pp-B. | 4 | Engineering rock mechanics practices in the underground powerhouse at JinpingⅠhydropower station显示文摘Based on the analyses of data obtained from the underground powerhouse at Jinping Ⅰ hydropower station,a comprehensive review of engineering rock mechanics practice in the underground powerhouse is first conducted.The distribution of strata,lithology,and initial geo-stress,the excavation process and corresponding rock mass support measures,the deformation and failure characteristics of the surrounding rock mass,the stress characteristics of anchorage structures in the cavern complex,and numerical simulations of surrounding rock mass stability and anchor support performance are presented.The results indicate that the underground powerhouse of Jinping Ⅰ hydropower station is characterized by high to extremely high geo-stresses during rock excavation.Excessive surrounding rock mass deformation and high stress of anchorage structures,surrounding rock mass unloading damage,and local cracking failure of surrounding rock masses,etc.,are mainly caused by rock mass excavation.Deformations of surrounding rock masses and stresses in anchorage structures here are larger than those found elsewhere:20%of extensometers in the main powerhouse record more than 50 mm with the maximum at around 250 mm observed in the downstream sidewall of the transformer hall.There are about 25%of the anchor bolts having recorded stresses of more than 200 MPa.Jinping Ⅰ hydropower plant is the first to have an underground powerhouse construction conducted in host rocks under extremely high geo-stress conditions,with the ratio of rock mass strength to geo-stress of less than 2.0.The results can provide a reference to underground powerhouse construction in similar geological conditions. | Aiqing Wu Jimin Wang Zhong Zhou Shuling Huang Xiuli Ding Zhihong Dong Yuting Zhang | 2016 | Journal of Rock Mechanics and Geotechnical Engineering2016,8,5: | 4 |

| 7 | Delineating spatiotemporal and hierarchical development of human fetal innate lymphoid cells显示文摘Whereas the critical roles of innate lymphoid cells(ILCs)in adult are increasingly appreciated,their developmental hierarchy in early human fetus remains largely elusive.In this study,we sorted human hematopoietic stem/progenitor cells,lymphoid progenitors,putative ILC progenitor/precursors and mature ILCs in the fetal hematopoietic,lymphoid and non-lymphoid tissues,from 8 to 12 post-conception weeks,for single-cell RNA-sequencing,followed by computational analysis and functional validation at bulk and single-cell levels.We delineated the early phase of ILC lineage commitment from hematopoietic stem/progenitor cells,which mainly occurred in fetal liver and intestine.We further unveiled interleukin-3 receptor as a surface marker for the lymphoid progenitors in fetal liver with T,B,ILC and myeloid potentials,while IL-3RA–lymphoid progenitors were predominantly B-lineage committed.Notably,we determined the heterogeneity and tissue distribution of each ILC subpopulation,revealing the proliferating characteristics shared by the precursors of each ILC subtype.Additionally,a novel unconventional ILC2 subpopulation(CRTH2–CCR9+ILC2)was identified in fetal thymus.Taken together,our study illuminates the precise cellular and molecular features underlying the stepwise formation of human fetal ILC hierarchy with remarkable spatiotemporal heterogeneity. | Chen Liu Yandong Gong Han Zhang Hua Yang Yang Zeng Zhilei Bian Qian Xin Zhijie Bai Man Zhang Jian He Jing Yan Jie Zhou Zongcheng Li Yanli Ni Aiqing Wen Yu Lan Hongbo Hu Bing Liu | 2021 | Cell Research2021,31,10: | 2 |
