| 1 | Establishment and application of distant hybridization technology in fish显示文摘Hybridization is widely used. However, for a long time, systematic theories and technologies related to hybridization in fish have been lacking. In this study, through long-term systematic research, we investigated and obtained the main rules regarding inheritance and reproduction related to fish distant hybridization. Furthermore, we established one-step and multistep breeding technologies that were suitable for interspecific hybridization and intraspecific hybridization. Simultaneously, we used these two breeding technologies to produce a batch of diploid fish lineages and tetraploid fish lineages and improved fishes. In addition, we widely discuss the methods, technologies and results of hybridization breeding, referring to the domestic and foreign literature on fish hybridization. | 2 | The RNA-binding protein Musashi2 governs osteoblast-adipocyte lineage commitment by suppressing PPARγsignaling显示文摘Osteoporosis caused by aging is characterized by reduced bone mass and accumulated adipocytes in the bone marrow cavity. How the balance between osteoblastogenesis and adipogenesis from bone marrow mesenchymal stem cells(BMSCs) is lost upon aging is still unclear. Here, we found that the RNA-binding protein Musashi2(Msi2) regulates BMSC lineage commitment. Msi2 is commonly enriched in stem cells and tumor cells. We found that its expression was downregulated during adipogenic differentiation and upregulated during osteogenic differentiation of BMSCs. Msi2 knockout mice exhibited decreased bone mass with substantial accumulation of marrow adipocytes, similar to aging-induced osteoporosis. Depletion of Msi2 in BMSCs led to increased adipocyte commitment. Transcriptional profiling analysis revealed that Msi2 deficiency led to increased PPARγ signaling.RNA-interacting protein immunoprecipitation assays demonstrated that Msi2 could inhibit the translation of the key adipogenic factor Cebpα, thereby inhibiting PPAR signaling. Furthermore, the expression of Msi2 decreased significantly during the aging process of mice, indicating that decreased Msi2 function during aging contributes to abnormal accumulation of adipocytes in bone marrow and osteoporosis. | 3 | Poly-PR in C9ORF72-Related Amyotrophic Lateral Sclerosis/Frontotemporal Dementia Causes Neurotoxicity by Clathrin-Dependent Endocytosis显示文摘GGGGCC repeat expansions in the C9 ORF72 gene are the most common cause of amyotrophic lateral sclerosis and frontotemporal dementia(c9 ALS/FTD). It has been reported that hexanucleotide repeat expansions in C9 ORF72 produce five dipeptide repeat(DPR) proteins by an unconventional repeat-associated non-ATG(RAN)translation. Within the five DPR proteins, poly-PR and poly-GR that contain arginine are more toxic than the other DPRs(poly-GA, poly-GP, and poly-PA). Here, we demonstrated that poly-PR peptides transferred into cells by endocytosis in a clathrin-dependent manner, leading to endoplasmic reticulum stress and cell death. In SH-SY5 Y cells and primary cortical neurons, poly-PR activated JUN amino-terminal kinase(JNK) and increased the levels of p53 and Bax. The uptake of poly-PR peptides by cells was significantly inhibited by knockdown of clathrin or by chlorpromazine, an inhibitor that blocks clathrin-mediated endocytosis. Inhibition of clathrin-dependent endocytosis by chlorpromazine significantly blocked the transfer of poly-PR peptides into cells, and attenuated poly-PRinduced JNK activation and cell death. | 4 | An application of the LTP_DSEF model to heavy precipitation forecasts of landfalling tropical cyclones over China in 2018显示文摘Recently, a track-similarity-based Dynamical-Statistical Ensemble Forecast(LTP_DSEF) model has been developed in an attempt to predict heavy rainfall from Landfalling Tropical cyclones(LTCs). In this study, the LTP_DSEF model is applied to predicting heavy precipitation associated with 10 LTCs occurring over China in 2018. The best forecast scheme of the model with optimized parameters is obtained after testing 3452 different schemes for the 10 LTCs. Then, its performance is compared to that of three operational dynamical models. Results show that the LTP_DSEF model has advantages over the three dynamical models in predicting heavy precipitation accumulated after landfall, especially for rainfall amounts greater than 250 mm. The model also provides superior or slightly inferior heavy rainfall forecast performance for individual LTCs compared to the three dynamical models. In particular, the LTP_DSEF model can predict heavy rainfall with valuable threat scores associated with certain LTCs, which is not possible with the three dynamical models. Moreover, the model can reasonably capture the distribution of heavier accumulated rainfall, albeit with widespread coverage compared to observations. | 6 | Prognosis of gestational choriocarcinoma diagnosed incidentally during laparoscopy for a presumed cornual pregnancy:a report of five cases显示文摘The clinical presentations of gestational choriocarcinoma vary markedly, and a misdiagnosis could be made in atypical patients if simply relying on clinical features. Laparoscopic resection of uterine mass lesion is rarely used in gestational choriocarcinoma diagnosis because of the fear of heavy blood loss and distant metastasis. Five patients who were preoperatively diagnosed as having cornual pregnancy underwent laparoscopic resection of mass lesion and then proved to have gestational choriocarcinoma based on pathological examinations. Chemotherapy was started within two days after surgery, and the rate of complete remission was 100%. The mean follow-up time was 29.8±19.1 months, and no patient showed signs of relapse. | 11 | A New Discovery of Ag-Pb-Zn Mineralization via Modern Portable Analytical Technology and Stream Sediment Data Processing Methods in Dajiacuo Area, Western Tibet(China)显示文摘Tibet, which is rich in mineral resources, is a treasure trove for geological explorers. However, prospecting work has been slow, especially in the western part, due to the precipitous terrain, changeable climate and low access. Hence, modern advanced field analytical technology and effective data processing methods play significant roles in rapid and efficient exploration in Tibet. In this paper, spectrum-area fractal modeling and portable X-ray fluorescence analysis(pXRFA) were used to identify and verify geochemical anomalies associated with Ag-Pb-Zn mineralization based on a stream-sediment dataset of 39 elements in the Dajiacuo-Xurucuo region of western Tibet. First, staged factor analysis(SFA) was used to obtain the Ag-Pb-Zn-Cd geochemical assemblage. Second, the first-factor pattern obtained using SFA was dissociated by a spectrum-area(S-A) fractal model and a digital elevation model(DEM)-based geochemical model(DGM) was constructed. Finally, the sections of Ag, Cd, Pb, and Zn were obtained using pXRFA. The results show that Ag-Pb-Zn-Cd enrichment zones were mostly located around the contact belt of volcanic rocks and intrusions, or along SE-NW trending faults. Considering the variable terrain and catchment basin, the extension of long axes of Ag-Pb-Zn-Cd anomalies into higher elevation areas that are favorable for Ag-Pb-Zn mineralization should be investigated. Anomaly maps created with the aid of a DGM show promising potential for mineralization in the Dajiacuo-Xurucuo region, and abundant Ag-Pb-Zn mineralization was identified with the assistance of pXRFA in the source areas for the geochemical anomalies in the Dajiacuo. We conclude that SFA and the S-A fractal model constitute a valid tool to identify or verify geochemical anomalies in areas of low-density stream-sediment sampling.
