1The Assessment of Global Surface Temperature Change from 1850s:The C-LSAT2.0 Ensemble and the CMST-Interim Datasets显示文摘Based on C-LSAT2.0,using high-and low-frequency components reconstruction methods,combined with observation constraint masking,a reconstructed C-LSAT2.0 with 756 ensemble members from the 1850s to 2018 has been developed.These ensemble versions have been merged with the ERSSTv5 ensemble dataset,and an upgraded version of the CMSTInterim dataset with 5°×5°resolution has been developed.The CMST-Interim dataset has significantly improved the coverage rate of global surface temperature data.After reconstruction,the data coverage before 1950 increased from 78%−81%of the original CMST to 81%−89%.The total coverage after 1955 reached about 93%,including more than 98%in the Northern Hemisphere and 81%−89%in the Southern Hemisphere.Through the reconstruction ensemble experiments with different parameters,a good basis is provided for more systematic uncertainty assessment of C-LSAT2.0 and CMSTInterim.In comparison with the original CMST,the global mean surface temperatures are estimated to be cooler in the second half of 19th century and warmer during the 21st century,which shows that the global warming trend is further amplified.The global warming trends are updated from 0.085±0.004℃(10 yr)^(–1)and 0.128±0.006℃(10 yr)^(–1)to 0.089±0.004℃(10 yr)^(–1)and 0.137±0.007℃(10 yr)^(–1),respectively,since the start and the second half of 20th century.Wenbin SUN Qingxiang LI Boyin HUANG Jiayi CHENG Zhaoyang SONG Haiyan LI Wenjie DONG Panmao ZHAI Phil JONES 2021Advances in Atmospheric Sciences2021,38,5:8
2Regulation of the inflammatory cycle by a controllable release hydrogel for eliminating postoperative inflammation after discectomy显示文摘Surgery is the final choice for most patients with intervertebral disc degeneration(IDD).Operation-caused trauma will cause inflammation in the intervertebral disc.Serious inflammation will cause tissue defects and induce tissue degeneration,IDD recurrence and the occurrence of other diseases.Therefore,we proposed a scheme to treat recurrence after discectomy by inhibiting inflammation with an aspirin(ASP)-loaded hydrogel to restore the mechanical stability of the spine and relieve local inflammation.ASP-liposomes(ASP-Lips)were incorporated into a photocrosslinkable gelatin-methacryloyl(GelMA)via mixing.This material can effectively alleviate inflammation by inhibiting the release of high mobility group box 1(HMGB1)from the nucleus to the cytoplasm.We further assessed the expression of inflammatory cytokines,such as interleukin 6(IL-6)and tumor necrosis factor-α(TNF-α),and degeneration-related factors,such as type II collagen(COL-2),Aggrecan,matrix metallopeptidases-3(MMP-3),MMP-13,a disintegrin and metalloproteinase with thrombospondin motifs-4(ADAMTS-4)and ADAMTS-5 in rat nucleus pulpous cells.The level of IDD was analyzed through H&E,safranin-O staining and immunohistochemistry in rabbit samples.In vitro,we found that ASP-Lip@GelMA treatment significantly decreased inflammatory cytokines,MMP-3 and-13,and ADAMTS-4 and-5 and up-regulated COL-2 and Aggrecan via the inhibited release of HMGB-1 from the nucleus.In vivo,ASP-Lip@GelMA can effectively inhibit inflammation of local tissue after disc surgery and fill local tissue defects.This composite hydrogel system is a promising way to treat the recurrence of IDD after surgery without persistent complications.Yu Liu Jiacheng Du Peng Peng Ruoyu Cheng Jiayi Lin Congxin Xu Huilin Yang Wenguo Cui Haiqing Mao Yuling Li Dechun Geng 2021Bioactive Materials2021,6,1:6

5Typing of biliary tumor thrombus influences the prognoses of patients with hepatocellular carcinoma显示文摘Objective:To establish a new classification of biliary tumor thrombus(BTT).Methods:Overall survival of patients with BTT was first used to determine whether it correlated with current hepatocellular carcinoma staging systems.Univariate and multivariate analyses were used to determine factors affecting the overall survival(OS)to form the basis of our new classification for BTT.Results:All 6 international staging systems showed overlapping survival curves.Univariate followed by multivariate analyses showed that total bilirubin and intrahepatic/extrahepatic BTT were significant risk factors of OS.Based on these data,a new BTT classification was defined as:TypeⅠ:intrahepatic BTT;and TypeⅡ:extrahepatic BTT involving a common bile duct or common hepatic duct.TypeⅠwas further subdivided into type Ia:BTT involving a second-order intrahepatic duct or above,and type Ib:BTT involving a first-order intrahepatic duct.TypeⅡwas further subdivided into typeⅡa and typeⅡb using a cut-off total bilirubin(TB)>300μmol/L.The numbers(percentages)of patients with typesⅠandⅡBTT were 69(34.2%)and 133(65.8%),respectively.The median OS of typeⅠpatients was significantly higher than that of typeⅡpatients(37.5 months vs.23.2 months;P=0.002).Using subgroup analyses,OS outcomes were significantly different between the subgroups of typeⅡb and type IIa,although there was no significant difference between the type Ia and type Ib subgroups(P=0.07).Conclusions:A new BTT classification was established to predict prognoses of HCC patients with BTT who underwent liver resection.Juxian Sun Jiayi Wu Chang Liu Jie Shi Yonggang Wei Jianyin Zhou Zhibo Zhang Wan Yee Lau Maolin Yan Shuqun Cheng 2021Cancer Biology & Medicine2021,18,3:3
6Moisture-resistant,stretchable NO_(x)gas sensors based on laser-induced graphene for environmental monitoring and breath analysis显示文摘The accurate,continuous analysis of healthcare-relevant gases such as nitrogen oxides(NOx)in a humid environment remains elusive for low-cost,stretchable gas sensing devices.This study presents the design and demonstration of a moisture-resistant,stretchable NO_(x)gas sensor based on laser-induced graphene(LIG).Sandwiched between a soft elastomeric substrate and a moisture-resistant semipermeable encapsulant,the LIG sensing and electrode layer is first optimized by tuning laser processing parameters such as power,image density,and defocus distance.The gas sensor,using a needlelike LIG prepared with optimal laser processing parameters,exhibits a large response of 4.18%o ppm^(-1)to NO and 6.66%o ppm^(-1)to NO_(2),an ultralow detection limit of 8.3 ppb to NO and 4.0 ppb to NO_(2),fast response/recovery,and excellent selectivity.The design of a stretchable serpentine structure in the LIG electrode and strain isolation from the stiff island allows the gas sensor to be stretched by 30%.Combined with a moisture-resistant property against a relative humidity of 90%,the reported gas sensor has further been demonstrated to monitor the personal local environment during dfferent times of the day and analyze human breath samples to classify patients with respiratory diseases from healthy volunteers.Moisture-resistant,stretchable NO_(x)gas sensors can expand the capability of wearable devices to detect biomarkers from humans and exposed environments for early disease diagnostics.Li Yang Guanghao Zheng Yaoqian Cao Chuizhou Meng Yuhang Li Huadong Ji Xue Chen Guangyu Niu Jiayi Yan Ye Xue Huanyu Cheng 2022Microsystems & Nanoengineering2022,8,4:2
7A distinct D1-MSN subpopulation down-regulates dopamine to promote negative emotional state显示文摘Dopamine(DA)level in the nucleus accumbens(NAc)is critical for reward and aversion encoding.DA released from the ventral mesencephalon(VM)DAergic neurons increases the excitability of VM-projecting D1-dopamine receptor-expressing medium spiny neurons(D1-MSNs)in the NAc to enhance DA release and augment rewards.However,how such a DA positive feedback loop is regulated to maintain DA homeostasis and reward-aversion balance remains elusive.Here we report that the ventral pallidum(VP)projection of NAc D1-MSNs(D1^(NAc-VP))is inhibited by rewarding stimuli and activated by aversive stimuli.In contrast to the VM projection of D1-MSN(D1^(NAc-VM)),activation of D1^(NAc-VP) projection induces aversion,but not reward.D1^(NAc-VP) MSNs are distinct from the D1^(NAc-VM) MSNs,which exhibit conventional functions of D1-MSNs.Activation of D1^(NAc-VP) projection stimulates VM GABAergic transmission,inhibits VM DAergic neurons,and reduces DA release into the NAc.Thus,D1^(NAc-VP) and D1^(NAc-VM) MSNs cooperatively control NAc dopamine balance and reward-aversion states.Zhiyuan Liu Qiumin Le Yanbo Lv Xi Chen Jian Cui Yiming Zhou Deqin Cheng Chaonan Ma Xiujuan Su Lei Xiao Ruyi Yang Jiayi Zhang Lan Ma Xing Liu 2022Cell Research2022,32,2:2
8Integrated dynamic wet spinning of core-sheath hydrogel fibers for optical-to-brain/tissue communications显示文摘Hydrogel optical light-guides have received substantial interest for applications such as deep-tissue biosensors,optogenetic stimulation and photomedicine due to their biocompatibility,(micro)structure control and tissue-like Young’s modulus.However,despite recent developments,large-scale fabrication with a continuous synthetic methodology,which could produce core-sheath hydrogel fibers with the desired optical and mechanical properties suitable for deep-tissue applications,has yet to be achieved.In this study,we report a versatile concept of integrated light-triggered dynamic wet spinning capable of continuously producing core-sheath hydrogel optical fibers with tunable fiber diameters,and mechanical and optical propagation properties.Furthermore,this concept also exhibited versatility for various kinds of core-sheath functional fibers.The wet spinning synthetic procedure and fabrication process were optimized with the rational design of the core/sheath material interface compatibility[core=poly(ethylene glycol diacrylate-co-acrylamide);sheath=Ca-alginate],optical transparency,refractive index and spinning solution viscosity.The resulting hydrogel optical fibers exhibited desirable low optical attenuation(0.18±0.01 d B cm^(-1) with 650 nm laser light),excellent biocompatibility and tissue-like Young’s modulus(<2.60 MPa).The optical waveguide hydrogel fibers were successfully employed for deep-tissue cancer therapy and brain optogenetic stimulation,confirming that they could serve as an efficient versatile tool for diverse deep-tissue therapy and brain optogenetic applications.Guoyin Chen Gang Wang Xinrong Tan Kai Hou Qingshuo Meng Peng Zhao Shun Wang Jiayi Zhang Zhan Zhou Tao Chen Yanhua Cheng D Benjamin S.Hsiao Elsa Reichmanis Meifang Zhu 2021National Science Review2021,8,9:2
9Proteomic analysis of overweight/obesity and related abnormal glucose and lipid metabolism caused by phlegm-dampness retention显示文摘Objective:To investigate the proteomic characteristics of overweight/obesity and related abnormal glucose and lipid metabolism caused by phlegm-dampness retention to identify related biomarkers.Methods:Seventy-one subjects were enrolled in the study.We assessed blood glucose,blood lipids,body mass index(BMI),and phlegm-dampness pattern,which was confirmed by a traditional Chinese medicine clinician.Of the participants,we included healthy participants with normal weight(NW,n=23),overweight/obese participants with normal metabolism(ONM,n=19),overweight/obese participants with pre-diabetes(OPD,n=12),and overweight/obese participants with marginally-elevated blood lipids(OML,n=17).Among them,the ONM,OPD,and OML groups were diagnosed with phlegmdampness pattern.The data-independent acquisition(DIA)method was first used to analyze the plasma protein expression of each group,and the relevant differential proteins of each group were screened.The co-expressed proteins were evaluated by Venn analysis.The pathway analyses of the differential proteins were analyzed using Ingenuity Pathway Analysis(IPA)software.Parallel reaction monitoring(PRM)was used to verify the differential and common proteins in each group.Results:After comparing ONM,OPD,and OML groups with NW group,we identified the differentially expressed proteins(DEPs).Next,we determined the DEPs among OPD,OML,and ONM groups.Using Venn analysis of the DEPs in each group,24 co-expressed proteins were screened.Two co-expressed proteins were verified by PRM.IPA analysis showed that pathways including LXR/RXR activation,acute phase response signaling,and FXR/RXR activation were common to all three groups of phlegmdamp overweight/obesity participants.However,the activation or inhibition of these pathways was different among the three groups.Conclusion:Participants with overweight/obesity have similar proteomic characteristics,though each type shows specific proteomic characteristics.Two co-expressed proteins,VTN and ORM1,are potential biomarkers for glucose and lipid metabolism diseases with overweight/obesity caused by phlegmdampness retention.Jiayi Ma Shuxian Sun Cheng Ni Lingru Li Jing Xia Houqin Li Huirong Song Xujun Heng Dandan Hu Yuanyuan Li 2021Journal of Traditional Chinese Medical Sciences2021,8,3:1

14Pan-cancer analysis of DNA epigenetic modifications by hydrophilic interaction liquid chromatography-tandem mass spectrometry显示文摘Accumulating evidence in recent years indicates that DNA methylation(5-methyl-2-deoxycytidine,5-mdC) and hydroxymethylation(5-hydroxymethyl-2-deoxycytidine, 5-hmd C) have been implicated in various biological processes, and the aberrations of these DNA cytosine modifications is tightly associated with cancer. N6-methyl-2-deoxyadenosine(m~6dA), as a newly discovered epigenetic modification in genome of mammals, has been demonstrated to play vital regulatory roles in tumorigenesis. However, the content information of m~6dA in human tumor tissues is still limited and pan-cancer analysis of these DNA epigenetic modifications is lacked. Herein, we developed a sensitive and robust stable isotopediluted hydrophilic interaction liquid chromatography-tandem mass spectrometry(HILIC-MS/MS) method for accurate quantification of m~6dA, 5-mdC and 5-hmdC in genomic DNA from 82 pairs of human tumor tissues and matched tumor-adjacent normal tissues. The types of tumors included esophagus cancer, lung cancer, breast cancer, liver cancer, pancreatic cancer, gastric cancer, stromal tumor and colorectal cancer.Compared to the normal tissues, we revealed the level of m6dA was increased in tumor tissues of esophagus cancer, lung cancer and liver cancer, whereas the level of m~6dA was diminished in tumor tissues of pancreatic cancer and gastric cancer;while the contents of 5-mdC and 5-hmdC exhibited significant decrease in tumor tissues of most types of cancer. It is worth noting that we revealed, for the first time,the content of genomic m~6dA in pancreatic cancer, stromal tumor and colorectal cancer. The significant changes of these DNA epigenetic modifications indicate they may serve as indicators of cancers. 15Organic photovoltaic device enhances the neural differentiation of rat bone marrow-derived mesenchymal stem cells显示文摘Electrical stimulation is known to be involved in stem cell differentiation,particularly neural differentiation.Various electrical stimulation systems and devices have been developed for neural tissue engineering.The organic photovoltaic materials PM6 and Y6 have showed high-power conversion efficiency.In this study,we used PM6 and Y6 to develop an organic photovoltaic device(OPD)to supply electrical stimulation.The photoelectric stimulation by the OPD showed no impact on cell viability.We tested the neural differentiation potential of rat bone marrow-derived mesenchymal stem cells(rBMSCs)under light induced electrical stimulation.The changes in cell morphology suggested that photoelectric stimulation significantly increased the neurite length and the number of extremities of differentiated neural cells.In addition,genes of neuron markers and neurotrophic factors were upregulated when rBMSCs were under photoelectric stimulation.Furthermore,the calcium influx of differentiated cells responding to acetylcholine and the phosphorylation of extracellular-signal-regulated kinase(ERK)1 and 2,protein kinase B(AKT)and mammalian target of rapamycin(mTOR)were significantly elevated after photoelectric stimulation.These findings demonstrated that PM6:Y6 based OPD could provide photoelectric stimulation to enhance rBMSCs neural differentiation,which might be an alternative approach to electrically manipulate stem cells differentiation into neural cells in vitro.Hong Cheng Yan Huang Jiayi Qian Fanzhe Meng Yubo Fan 2022Medicine in Novel Technology and Devices2022,,4:0
