2SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality.Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li 2019Horticulture Research2019,6,1:39
3Diagenetic fluids evolution and genetic mechanism of tight sandstone gas reservoirs in Upper Triassic Xujiahe Formation in Sichuan Basin, China显示文摘The reservoirs of the Upper Triassic Xujiahe Formation in Sichuan Basin have the characteristics of low compositional maturity, low contents of cements and medium textural maturity. The general physical properties of the reservoirs are poor, with low porosity and low permeability, and there are only a few reservoirs with medium porosity and low permeability in local areas. Based on the diagenetic mineral association, a diagenetic sequence of cements is established: early calcites (or micrite siderites) →first quartz overgrowth→chlorite coatings→dissolution of feldspars and debris→chlorite linings→ second quartz overgrowth (quartz widen or filled in remain intergranular pores and solution pores)→dissolution→third quartz overgrowth (quartz filled in intergranular and intragranular solution pores)→intergrowth (ferro) calcites→dolomites→ferro (calcites) dolomites→later dissolution→veins of quartz and calcites formation. Mechanical compaction is the main factor in making the reservoirs tight in the basin, followed by the second and third quartz overgrowth. In a long-term closed system, only feld-spars and some lithic fragments are dissolved by diagenetic fluids, while intergranular cements such as quartz and calcit are not dissolved and thus have little influence on the porosity of the Xujiahe Formation. This is the third factor that may have kept the sandstones of Xujiahe Formation tight finally. 8Polyamine Accumulation in Transgenic Tomato Enhances the Tolerance to High Temperature Stress显示文摘Polyamines play an important role in plant response to abiotic stress.S-adenosyl-l-methionine decarboxylase(SAMDC) is one of the key regulatory enzymes in the biosynthesis of polyamines.In order to better understand the effect of regulation of polyamine biosynthesis on the tolerance of high-temperature stress in tomato,SAMDC cDNA isolated from Saccharomyces cerevisiae was introduced into tomato genome by means of Agrobacterium tumefaciens through leaf disc transformation.Transgene and expression was confirmed by Southern and Northern blot analyses,respectively.Transgenic plants expressing yeast SAMDC produced 1.7-to 2.4-fold higher levels of spermidine and spermine than wild-type plants under high temperature stress,and enhanced antioxidant enzyme activity and the protection of membrane lipid peroxidation was also observed.This subsequently improved the efficiency of CO2 assimilation and protected the plants from high temperature stress,which indicated that the transgenic tomato presented an enhanced tolerance to high temperature stress(38℃) compared with wild-type plants.Our results demonstrated clearly that increasing polyamine biosynthesis in plants may be a means of creating high temperature-tolerant germplasm.Lin Cheng Yijing Zou Shuli Ding Jiajing Zhang Xiaolin Yu Jiashu Cao Gang Lu 2009Journal of Integrative Plant Biology2009,51,5:16
9Changes of the proportion and mortality of pulmonary thromboembolism in hospitalized patients from 1974 to 2005显示文摘Background Pulmonary thromboembolism (PTE) has become a common disease that severely endangers people’s health. This study analysed the changes in proportion and mortality of PTE in hospitalized patients to provide data for prevention and management of the disease. Methods The data of 763 hospitalized patients with PTE from 1974 to 2005 in Fuwai Hospital were analysed. Results During the 1970s, 0.27% of patients in a cardiovascular hospital had PTE (< 5 cases per year); while so far this century the proportion is 0.94% (48 to 113 per year). The mortality of hospitalized PTE patients fell from 20.00% in the 1970s to 4.10% this century. Prior to 1990, the mortality of hospitalized PTE patients was 12.50%, and in the years after 1990 only 3.40%. The difference was statistically significant (P<0.005). People with this disease were mostly between the ages of 30 and 69 years. Men were most susceptible between the ages of 30 and 69 years, while women between the ages of 40 and 69 years. Men contracted PTE 10 years earlier than women. The mortality of male PTE patients was 4.70%, not significantly different from female patients, 5.06% (0.500.1). More people contracted the disease in winter than in other seasons (P<0.05). 10Effect and mechanism of Irbesartan on occurrence of ventricular arrhythmias in rats with myocardial ischemia through connexin43(cx43)显示文摘Objective: To explore the effect and mechanism of angiotensin Ⅱ receptor blockers-Irbesartan on occurrence of ventricular arrhythmias in rats with myocardial ischemia. Methods: Rats with embryonic cardiomyocytes-H9c2 were randomly divided into control group, ischemia group, Irbesartan group and Irbesartan+ischemia group. The cell viability of rats in each group was tested using MTT. Real-time PCR was employed to detect the expression of connexin43(Cx43) mR NA and western blot to detect the expression of Cx43 and phosphorylated Cx43. SD rats were randomly divided into the sham-operation group(SO), myocardial infarction group(MI), Irbesartan group and MI+ Irbesartan group, with 10 rats in each group. HE staining was employed to observe the change in the pathomorphology of left ventricular tissue and TUNEL method to analyze the cell apoptosis in the tissue. The immunofluorescence was adopted to observe the expression and distribution of Cx43 in the left ventricular myocardium and study the change in the expression of Cx43 in the cardiac muscular tissue at mR NA and protein level. Results: The intervention of Irbesartan in the condition of ischemia indicated the significant decrease in the number of necrotic cells. The expression of Cx43 was significantly decreased under the culture of ischemia(P<0.05), but in the presence of Irbesartan, the expression of Cx43 was increased compared with the ischemia group(P<0.01). The results of WB assay showed the similar trend of change at mRNA level. There was the significant difference in the score of ventricular arerythmia between MI group and SO group(P<0.01). The incidence of ventricular tachycardia or ventricular fibrillation was significantly increased compared with the one in SO group(P<0.05). There was the significant difference in the overall score between MI+Irbesartan group and MI group(P<0.05). The expression of Cx43 in the cardiac muscular tissue in MI group was significantly decreased(P<0.01 vs SO group). But the expression of Cx43 was increased after the treatment with Irbesartan. Conclusions: Irbesartan can inhibit the injury of H9c2 cardiomyocytes and the decreased expression of Cx43 that are induced by the ischemic myocardial infarction. 11Meta‐analysis and candidate gene mining of low‐phosphorus tolerance in maize显示文摘Plants with tolerance to low‐phosphorus(P) can grow better under low‐P conditions, and understanding of genetic mechanisms of low‐P tolerance can not only facilitate identifying relevant genes but also help to develop low‐P tolerant cultivars. QTL meta‐analysis was conducted after a comprehensive review of the reports on QTL mapping for low‐P tolerance‐related traits in maize. Meta‐analysis produced 23 consensus QTL(cQTL), 17 of which located in similar chromosome regions to those previously reported to influence root traits. Meanwhile, candidate gene mining yielded 215 genes, 22 of which located in the cQTL regions.These 22 genes are homologous to 14 functionally characterized genes that were found to participate in plant low‐P tolerance, including genes encoding miR399s, Pi transporters and purple acid phosphatases. 13Development of sintered Ni-Cu wicks for loop heat pipes显示文摘The present study fabricated a series of capillary wicks for loop heat pipes(LHPs),using two different methods,the cold-pressing sintering and direct loose sintering,and experimentally investigated the effect of different methods,compositions and sintering parameters on their properties in terms of porosity,permeability and pore radius.Porosity and pore radius were measured by the Archimedes method and Scanning Electron Microscope(SEM),respectively.Permeability of the wicks was compared by calculation using empirical equation.Results show that capillary wicks were successfully fabricated by using two different methods;the optimal capillary wick was found to be sintered at 650℃ for 30min,using direct loose sintering technique,with 90% nickel and 10% copper.The wicks could reach the porosity of 70.07% and the permeability of 10-13m2 order,with mean pore radius of 0.54μm.XIN GongMing CUI KeHang ZOU Yong CHENG Lin 2009Science China(Technological Sciences)2009,52,6:11
14Exogenous application of a low concentration of melatonin enhances salt tolerance in rapeseed(Brassica napus L.) seedlings显示文摘Melatonin is a naturally occurring compound in plants. Here, we tested the effect of exogenous melatonin on rapeseed(Brassica napus L.) grown under salt stress. Application of 30 μmol L^(–1) melatonin alleviated salt-induced growth inhibition, and the shoot fresh weight, the shoot dry weight, the root fresh weight, and the root dry weight of seedlings treated with exogenous melatonin increased by 128.2, 142.9, 122.2, and 124.2%, respectively, compared to those under salt stress. In addition, several physiological parameters were evaluated. The activities of antioxidant enzymes including peroxidase(POD), catalase(CAT) and ascorbate peroxidase(APX) were enhanced by 16.5, 19.3, and 14.2% compared to their activities in plants without exogenous melatonin application under salt stress, while the H_2O_2 content was decreased by 11.2% by exogenous melatonin. Furthermore, melatonin treatment promoted solute accumulation by increasing the contents of proline(26.8%), soluble sugars(15.1%) and proteins(58.8%). The results also suggested that higher concentrations(>50 μmol L^(–1)) of melatonin could attenuate or even prevent the beneficial effects on seedling development. In conclusion, application of a low concentration of exogenous melatonin to rapeseed plants under salt stress can improve the H_2O_2-scavenging capacity by enhancing the activities of antioxidant enzymes such as POD, CAT and APX, and can also alleviate osmotic stress by promoting the accumulation of osmoregulatory substances such as soluble proteins, proline, and water soluble glucan. 15Mass production of 2D materials by intermediate-assisted grinding exfoliation显示文摘The scalable and high-efficiency production of 2D materials is a prerequisite to their commercial use.Currently,only graphene and graphene oxide can be produced on a ton scale,and the inability to produce other 2D materials on such a large scale hinders their technological applications.Here we report a grinding exfoliation method that uses micro-particles as force intermediates to resolve applied compressive forces into a multitude of small shear forces,inducing the highly efficient exfoliation of layer materials.The method;referred to as intermediate-assisted grinding exfoliation(iMAGE);can be used for the large-scale production of many 2D materials.As an example;we have exfoliated bulk h-BN into 2D h-BN with large flake sizes;high quality and structural integrity,with a high exfoliation yield of 67%,a high production rate of 0.3 g h^-1 and a low energy consumption of 3.01 x 106 J g^-1.The production rate and energy consumption are one to two orders of magnitude better than previous results.Besides h-BN,this iMAGE technology has been used to exfoliate various layer materials such as graphite,black phosphorus,transition metal dichalcogenides;and metal oxides;provings universality.Molybdenite concentrate,a natural low-cost and abundant mineral;was used as a demo for the large-scale exfoliation production of 2D M0S2 flakes.Our work indicates the huge potential of the iMAGE method to produce large amounts of various 2D materials;which paves the way for their commercial application.Chi Zhang Junyang Tan Yikun Pan Xingke Cai Xiaolong Zou Hui-Ming Cheng Bilu Liu 2020National Science Review2020,7,2:10

18Effects of yeast and yeast cell wall polysaccharides supplementation on beef cattle growth performance, rumen microbial populations and lipopolysaccharides production显示文摘This experiment was conducted to investigate the effects of live yeast and yeast cell wall polysaccharides on growth performance,rumen function and plasma lipopolysaccharides(LPS) content and immunity parameters of beef cattle.Forty Qinchuan cattle were randomly assigned to one of four treatments with 10 replicates in each treatment.The dietary treatments were: control diet(CTR),CTR supplemented with 1 g live yeast(2×10^10 live cell g^-1 per cattle per day(YST1),CTR supplemented with 2 g live yeast per cattle per day(YST2) and CTR supplemented with 20 g of yeast cell wall polysaccharides(30.0%≤β-glucan≤35.0%,and 28.0%≤mannanoligosaccharide≤32.0%) per cattle per day(YCW).The average daily gain was higher(P=0.023) and feed conversion ratio was lower(P=0.042) for the YST2 than the CTR.The digestibility of neutral detergent fiber(P=0.039) and acid detergent fiber(P=0.016) were higher in yeast supplemented groups.The acetic acid:propionic acid of the YST2 was lower compared with the CTR(P=0.033).Plasma LPS(P=0.032),acute phase protein haptoglobin(P=0.033),plasma amyloid A(P=0.015) and histamine(P=0.038) were lower in the YST2 compared with the CTR.The copies of fibrolytic microbial populations such as Fibrobacter succinogenes S85,Ruminococcus albus 7 and Ruminococcus flavefaciens FD-1 of the YST2 were higher(P<0.001),while the copies of typical lactate producing bacteria Streptococcus bovis JB1 was lower(P<0.001) compared with the CTR.Little differences were observed between the CTR,YST1 and YCW in growth performance,ruminal fermentation characteristics,microbial populations,immunity indices and total tract nutrient digestibility.It is concluded that the YST2 could promote fibrolytic microbial populations,decrease starch-utilizing bacteria,reduce LPS production in the rumen and LPS absorption into plasma and decrease inflammatory parameters,which can lead to an improvement in growth performance in beef cattle.PENG Quan-hui CHENG Long KANG Kun Tian Gang Mohammad Al-Mamun XUE Bai WANG Li-zhi ZOU Hua-wei Mathew Gitau Gicheha WANG Zhi-sheng 2020Journal of Integrative Agriculture2020,19,3:9

20Cogitation on developing a dynamic model of soil wind erosion显示文摘Studies on soil wind erosion began with single factors affecting soil wind erosion; with increasing quantities of data being accumulated,the wind erosion equation(WEQ),the revised wind erosion equation(RWEQ),the wind erosion prediction system(WEPS),and other soil wind erosion models have been successively established,and great advances have been achieved.Here we briefly review the soil wind erosion research course and analyze the advantages and disadvantages of the current soil wind erosion models.From the perspective of the dynamics of wind erosion,we classified the factors affecting soil wind erosion into three categories,namely,wind erosivity factors(WEF),soil antierodibility factors(SAF),and roughness interference factors(RIF).We proposed the concept of a standard plot of soil wind erosion to solve the problem of uncertainty of the soil wind erosion modulus on a spatial scale,and provided methods to set similarity conditions in wind tunnel simulation experiments and to convert the spatial scale of the wind erosion modulus from the standard plot to a large scale field.We also proposed a conceptual model on the basis of the dynamics of soil wind erosion with the theoretical basis that wind produces a shear force on the soil surface.This shear force is partitioned by barely erodible soil surfaces and roughness elements on the ground,and the amount of soil loss by wind should be calculated by comparing the shear force of the wind on barely erodible soil surfaces with the anti-erosion force of the surface soil.One advantage of this conceptual model is that the calculated soil wind erosion modulus is not subject to changes of spatial scale.Finally,we recommended continual improvement of the existing models while also establishing new models.ZOU XueYong ZHANG ChunLai CHENG Hong KANG LiQiang WU YongQiu 2015Science China Earth Sciences2015,58,3:9
