| 1 | Pericytes augment glioblastoma cell resistance to temozolomide through CCL5-CCR5 paracrine signaling显示文摘Glioblastoma(GBM)is a prevalent and highly lethal form of glioma,with rapid tumor progression and frequent recurrence.Excessive outgrowth of pericytes in GBM governs the ecology of the perivascular niche,but their function in mediating chemoresistance has not been fully explored.Herein,we uncovered that pericytes potentiate DNA damage repair(DDR)in GBM cells residing in the perivascular niche,which induces temozolomide(TMZ)chemoresistance.We found that increased pericyte proportion correlates with accelerated tumor recurrence and worse prognosis.Genetic depletion of pericytes in GBM xenografts enhances TMZ-induced cytotoxicity and prolongs survival of tumor-bearing mice.Mechanistically,C-C motif chemokine ligand 5(CCL5)secreted by pericytes activates C-C motif chemokine receptor 5(CCR5)on GBM cells to enable DNA-dependent protein kinase catalytic subunit(DNA-PKcs)-mediated DDR upon TMZ treatment.Disrupting CCL5-CCR5 paracrine signaling through the brain-penetrable CCR5 antagonist maraviroc(MVC)potently inhibits pericyte-promoted DDR and effectively improves the chemotherapeutic efficacy of TMZ.GBM patient-derived xenografts with high CCL5 expression benefit from combined treatment with TMZ and MVC.Our study reveals the role of pericytes as an extrinsic stimulator potentiating DDR signaling in GBM cells and suggests that targeting CCL5-CCR5 signaling could be an effective therapeutic strategy to improve chemotherapeutic efficacy against GBM. | Xiao-Ning Zhang Kai-Di Yang Cong Chen Zhi-Cheng He Qiang-Hu Wang Hua Feng Sheng-Qing Lv Yan Wang Min Mao Qing Liu Yao-Yao Tan Wen-Ying Wang Tian-Ran Li Lin-Rong Che Zhong-Yi Qin Ling-Xiang Wu Min Luo Chun-Hua Luo Yu-Qi Liu Wen Yin Chao Wang Hai-Tao Guo Qing-Rui Li Bin Wang Wei Chen Shuang Wang Yu Shi Xiu-Wu Bian Yi-Fang Ping | 2021 | Cell Research2021,31,10: | 5 |
| 2 | Microwave ablation of primary breast cancer inhibits metastatic progression in model mice via activation of natural killer cells显示文摘Surgery is essential for controlling the symptoms and complications of stage IV breast cancer.However,locoregional treatment of primary tumors often results in distant progression,including lung metastasis,the most common type of visceral metastasis.As a minimally invasive thermal therapy,microwave ablation(MWA)has been attempted in the treatment of breast cancer,but the innate immune response after MWA has not yet been reported.Using two murine models of stage IV breast cancer,we found that MWA of primary breast cancer inhibited the progression of lung metastasis and improved survival.NK cells were activated after MWA of the primary tumor and exhibited enhanced cytotoxic functions,and the cytotoxic pathways of NK cells were activated.Depletion experiments showed that NK cells but not CD4+or CD8+T cells played a pivotal role in prolonging survival.Then,we found that compared with surgery or control treatment,MWA of the primary tumor induced completely different NK-cell-related cytokine profiles.Macrophages were activated after MWA of the primary tumor and produced IL-15 that activated NK cells to inhibit the progression of metastasis.In addition,MWA of human breast cancer stimulated an autologous NK-cell response.These results demonstrate that MWA of the primary tumor in metastatic breast cancer inhibits metastatic progression via the macrophage/IL-15/NK-cell axis.MWA of the primary tumor may be a promising treatment strategy for de novo stage IV breast cancer,although further substantiation is essential for clinical testing. | Muxin Yu Hong Pan Nan Che Li Li Cong Wang Yue Wang Ge Ma Mengjia Qian Jiawei Liu Mingjie Zheng Hui Xie Lijun Ling Yi Zhao Xiaoxiang Guan Qiang Ding Wenbin Zhou Shui Wang | 2021 | Cellular & Molecular Immunology2021,18,9: | 5 |
| 3 | Microsatellite DNA Variation of the Gametophyte Clones Isolated from Introduced Laminaria japonica (Phaeophyta) and L. longissima of China and Varieties Derived from them显示文摘The variation of 90 Laminaria gametophyte clones representing the introduced Laminaria japonica (Group 1) and Laminaria longissima (Group 2), the varieties of L. japonica (Group 3) and the varieties derived from interspecific hybrids (Group 4) was determined with 18 microsatellite markers. The allelic diversity and Nei's gene diversity of Group 1 were significantly higher than those of Group 2 (2.9 vs. 1.8 and 0.414 vs. 0.161, respectively), demonstrating that the variation of the introduced L. japonica is richer than that of L. longissima. Both allelic diversity and Nei's gene diversity of Group 3 were lower than those of Group 1, indicating that only a portion of variation of L. japonica was incorporated into the varieties of L. japonica. Significant genetic differentiation was detected between four groups and between female (Population 1) and male (Population 2) gametophyte clones in each group. The variation among groups accounted for 39.95%, while that among populations accounted for 21.65% of the total. | 5 | A transient method for measuring the gas volume fraction in a mixed gas-liquid flow using acoustic resonance spectroscopy显示文摘In this paper, the feasibility of measuring the gas volume fraction in a mixed gas-liquid flow by using an acoustic resonant spectroscopy (ARS) method in a transient way is studied theoretically and experimentally. Firstly, the effects of sizes and locations of a single air bubble in a cylindrical cavity with two open ends on resonant frequencies are investigated numerically. Then, a transient measurement system for ARS is established, and the trends of the resonant frequencies (RFs) and resonant amplitudes (RAs) in the cylindrical cavity with gas flux inside are investigated experimentally. The measurement results by the proposed transient method are compared with those by steady-state ones and numerical ones. The numerical results show that the RFs of the cavity are highly sensitive to the volume of the single air bubble. A tiny bubble volume perturbation may cause a prominent RF shift even though the volume of the air bubble is smaller than 0.1% of that of the cavity. When the small air bubble moves, the RF shift will change and reach its maximum value as it is located at the middle of the cavity. As the gas volume fraction of the two-phase flow is low, both the RFs and RAs from the measurement results decrease dramatically with the increasing gas volume, and this decreasing trend gradually becomes even as the gas volume fraction increases further. These experimental results agree with the theoretical ones qualitatively. In addition, the transient method for ARS is more suitable for measuring the gas volume fraction with randomness and instantaneity than the steady-state one, because the latter could not reflect the random and instant characteristics of the mixed fluid due to the time consumption for frequency sweeping. | 6 | Experimental studies on the effects of porosity on acoustic resonance spectroscopy for synthetic porous rock samples in a cylindrical resonant cavity显示文摘Based on our previous work on acoustic resonance spectroscopy (ARS) measurement, the ARSs in low frequency modes were measured for 10 synthetic oil-saturated rock samples with the porosities rang-ing from 22% to 30%, and the effects of porosity on ARS of the synthetic porous rock samples in a cylindrical resonant cavity were studied. The experimental results show that as the location of a syn-thetic rock sample changes from the top to the middle of the resonant cavity, the resonance frequency (RF) will gradually increase and the corresponding resonance amplitude (RA) will decrease. In the middle of the cavity, the RF reaches its maximum, while the RA reaches its minimum. When the syn-thetic rock porosity increases, the RF will decrease and the RA will decrease. Furthermore, the RF of a synthetic rock sample was numerically calculated with an average volume model in the resonant cavity. Our numerical results show that as the location of the sample changes from the top to the middle of the cavity, the RF gradually increases and the RF reaches its maximum value in the middle of the cavity. However, the RF decreases as the porosity of the sample increases in the middle of the cavity. | 7 | A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. | 9 | Joint Deep Matching Model of OCT Retinal Layer Segmentation显示文摘Optical Coherence Tomography(OCT)is very important in medicine and provide useful diagnostic information.Measuring retinal layer thicknesses plays a vital role in pathophysiologic factors of many ocular conditions.Among the existing retinal layer segmentation approaches,learning or deep learning-based methods belong to the state-of-art.However,most of these techniques rely on manual-marked layers and the performances are limited due to the image quality.In order to overcome this limitation,we build a framework based on gray value curve matching,which uses depth learning to match the curve for semi-automatic segmentation of retinal layers from OCT.The depth convolution network learns the column correspondence in the OCT image unsupervised.The whole OCT image participates in the depth convolution neural network operation,compares the gray value of each column,and matches the gray value sequence of the transformation column and the next column.Using this algorithm,when a boundary point is manually specified,we can accurately segment the boundary between retinal layers.Our experimental results obtained from a 54-subjects database of both normal healthy eyes and affected eyes demonstrate the superior performances of our approach. | Mei Yang Yuanjie Zheng Weikuan Jia Yunlong He Tongtong Che Jinyu Cong | 2020 | Computers, Materials & Continua2020,,6: | 0 |

report 98 taxa distributed in two phyla,seven classes,26 orders and 50 families which are described and illustrated.Taxa in this study were collected from Australia,Brazil,Burkina Faso,Chile,China,Cyprus,Egypt,France,French Guiana,India,Indonesia,Italy,Laos,Mexico,Russia,Sri Lanka,Thailand,and Vietnam.There are 59 new taxa,39 new hosts and new geographical distributions with | 12 | Single virus tracking of Ebola virus entry through lipid rafts in living host cells显示文摘Ebola virus(EBOV)is one of the most pathogenic viruses in humans which can cause a lethal hemorrhagic fever.Understanding the cellular entry mechanisms of EBOV can promote the development of new therapeutic strategies to control virus replication and spread.It has been known that EBOV virions bind to factors expressed at the host cell surface.Subsequently,the virions are internalized by a macropinocytosis-like process,followed by being trafficked through early and late endosomes.Recent researches indicate that the entry of EBOV into cells requires integrated and functional lipid rafts.Whilst lipid rafts have been hypothesized to play a role in virus entry,there is a current lack of supporting data.One major technical hurdle is the lack of effective approaches for observing viral entry.To provide evidence on the involvement of lipid rafts in the entry process of EBOV,we generated the fluorescently labeled Ebola virus like particles(VLPs),and utilized single-particle tracking(SPT)to visualize the entry of fluorescent Ebola VLPs in live cells and the interaction of Ebola VLPs with lipid rafts.In this study,we demonstrate the compartmentalization of Ebola VLPs in lipid rafts during entry process,and inform the essential function of lipid rafts for the entry of Ebola virus.As such,our study provides evidence to show that the raft integrity is critical for Ebola virus pathogenesis and that lipid rafts can serve as potential targets for the development of novel therapeutic strategies. | Jin Cong Che Bin Guo Zhengyuan Li Chuan Liu Yang Wu Wei Wang Shiwen Li Dexin Cui Zongqiang Liang Mifang | 2020 | Biosafety and Health2020,2,1: | 0 |
