| 1 | Changes in sea ice kinematics in the Arctic outflow region and their associations with Arctic Northeast Passage accessibility显示文摘Amplification of climate warming in the Arctic is causing a dramatic retreat of sea ice, which means the Arctic sea routes are becoming increasingly accessible. This study used a satellite-derived sea ice motion product to quantify the kinematic features of sea ice in the Arctic outflow region which specially referred to the Fram Strait and to the north of the Northeast Passage(NEP). An observed trend of increased southward sea ice displacement from the central Arctic to the Fram Strait indicated enhancement of the Transpolar Drift Stream(TDS). In the regions to the north of the NEP, the long-term trend of northward sea ice speed in the Kara sector was +0.04 cm/s per year in spring. A significant statistical relationship was found between the NEP open period and the northward speed of the sea ice to the north of the NEP. The offshore advection of sea ice could account for the opening of sea routes by 33% and 15% in the Kara and Laptev sectors, respectively. The difference in sea level pressure across the TDS,i.e., the Central Arctic Index(CAI), presented more significant correlation than for the Arctic atmospheric Dipole Anomaly index with the open period of the NEP, and the CAI could explain the southward displacement of sea ice toward the Fram Strait by more than 45%. | 3 | Provenance of Lower Carboniferous Bauxite Deposits in Northern Guizhou,China:Constraints from Geochemistry and Detrital Zircon U-Pb Ages显示文摘The Lower Carboniferous Jiujialu Formation bauxite deposits of northern Guizhou Province,China,are a Kazakhstan subtype of karst bauxite deposits.The provenance of the Jiujialu Formation has long been debated,with uncertainty about the formation of the bauxite deposits.Here we report new geochemical data that indicate the affinity between the Lower-Middle Ordovician clastic rocks and argillaceous dolostones and the overlying Carboniferous bauxite deposits,all of which are characterized by high Al_(2)0_(3),K2_(O),and∑REE contents,flat post-Archean Australian shale(PAAS)-normalized REE patterns,and uniform immobile element ratios(TiO2/Al_(2)0_(3),Nb/TiO2,and Zr/TiO2).Their similar detrital zircon age distributions further indicate the link between the bauxite deposits and the clastic rocks and argillaceous dolostones.Zircon age spectra of clastic rocks of the Lower Silurian Hanchiatien Formation in northern Guizhou match those of the bauxite deposits,with a maximum age peak at-980 Ma and other secondary age peaks,suggesting these clastic rocks may represent the provenance of the bauxite deposits.The youngest detrital zircons(-445 Ma)occur only in the bauxite deposits and are probably sourced from K-bentonite beds of the Ordovician-Silurian tran-sition.Our analyses indicate that the source materials of the bauxite deposits in the Jiujialu Formation are of mixed provenance:Lower-Middle Ordovician aluminosilicate rocks and argillaceous dolostones of the underlying strata,and Lower Silurian clastic rocks and K-bentonite from adjacent areas.A comparison of Early Carboniferous bauxitic provenances in northern and central Guizhou indicates that paleotopography was the major factor controlling the provenance of these bauxite deposits. | Guolin Xiong Wenchao Yu Yuansheng Du Shenfu Weng Dawei Pang Xusheng Deng Jintao Zhou | 2021 | Journal of Earth Science2021,32,1: | 3 |
| 4 | Characterization of the unprecedented polynya events north of Greenland in 2017/2018 using remote sensing and reanalysis data显示文摘Based on an ice concentration threshold of 90%,it has been identified that two polynya events occurred in the region north of Greenland during the 2017/2018 ice season.The winter event lasted from February 20 to March 3,2018 and the summer event persisted from August 2 to September 5,2018.The minimum ice concentration derived from Advanced Microwave Scanning Radiometer 2(AMSR2)observations was 72%and 65%during the winter and summer events,respectively.The occurrence of both events can be related to strengthened southerly winds associated with an increased east-west zonal surface level air pressure gradient across the north Greenland due to perturbation of mid-troposphere polar vortex.The relatively warm air temperature during the 2017/2018 freezing season in comparison with previous years,together with the occurrence of the winter polynya,formed favourable pre-conditions for ice field fracturing in summer,which promoted the formation of the summer polynya.Diminished southerly winds and increased cover of new ice over the open water were the dominant factors for the disappearance of the winter polynya,whereas increased ice inflow from the north was the primary factor behind the closure of the summer polynya.Sentinel-1 Synthetic Aperture Radar(SAR)images were found better suited than AMSR2 observations for quantification of a new ice product during the polynya event because the SAR images have high potential for mapping of different sea ice regimes with finely spatial resolution.The unprecedented polynya events north of Greenland in 2017/2018 are important from the perspective of Arctic sea ice loss because they occurred in a region that could potentially be the last“Arctic sea ice refuge”in future summers. | Ruibo Lei Dawei Gui Zhouli Yuan Xiaoping Pang Ding Tao Mengxi Zhai | 2020 | Acta Oceanologica Sinica2020,39,9: | 3 |

| 6 | Molecular Cloning and Bioinformatics Analysis of TypeⅢSecretion System Effector Protein Va1686 Gene of Vibrio alginolyticus显示文摘In this study,Va1686 gene was cloned from Vibrio alginolyticus. The total length of the gene is 1 164 bp,and it could encode 387 amino acids. The physicochemical properties,protein structure,genetic evolutionary relationship and antigenic characteristics of the effect protein Va1686 of V. alginolyticus HY9901 type Ⅲ secretion system were studied and analyzed by bioinformatics methods and tools. The results showed that Va1686 is a stable hydrophilic and acidic protein without a transmembrane region and a signal peptide,and secondary structure to α-helix. The evolutionary analysis showed that V. alginolyticus HY9901 and V.harveyi were clustered together,which indicated that the genetic relationship between the two species was the closest. Va1686 contains a Fic superfamily conserved domain associated with cell division. Bioinformatics analysis showed that the B-cell preponderant epitopes of Va1686 might be localized in the regions of 48-49,82-85,125-126,150-153,185-186,236-237 and so on. The 3D structure model of Va1686 subunit was simulated by SWISS-MODEL software and it was found that the vop S of V. parahaemolyticus was similar and the similarity was 89. 46%. | 12 | Construction of Live Attenuated Vaccine against Vib- rio alginolyticus and the Evaluation on Its Immuno- protection Effect显示文摘Vibrio alginolyticus,a gram-negative bacterium has been described to be one of the most common and economically important aquatic pathogens of fish and shellfish. Vaccine immunization is an effective approach preventing V. alginolyticus infection. Attenuated vaccine stimulates systemic immune response in the host,but few attenuated vaccine against V. alginolyticus is available. The type III secretion system( T3SS),an important pathogenic factor of V. alginolyticus,is used by bacterial pathogens to inject effector proteins into the cytoplasm of their host cells. The T3 SS forms a structure called needle complex with a multi-ring base that spans the bacteria and a needle-like extension that protrudes several nanometers from the bacterial surface. vscO locates at the ' needle' site of T3 SS,playing the role of escorting the molecular chaperone and effector proteins into host cells and further inducing the death of host cells. In this paper,an in-frame deletion mutant of vscO was constructed using overlap PCR and homologous recombination technology combining with chloramphenicol( Cm) and sucrose screening. The LD50 changes of ZJ03ΔvscO mutant strains compared with the strain ZJ03 were examined in grouper( Epinephelus coioides). The ZJ03ΔvscO mutant showed about 150 times decrease in virulence in E. coioides compared with wide type ZJ03. After vaccination with ZJ03ΔvscO in E. coioides through injection and immersion,the specific antibody titers were markedly higher than that in the saline control group( P < 0. 05). The titers of injection and immersion group on the forth week reached the maximums at 1∶ 2 048 and 1∶ 128,respectively. The relative percentage survival( RPS) of injection group was 84%,while that in immersion group was 68%.These results indicate that the ZJ03ΔvscO of V. alginolyticus has a high immunogenicity,and can be used as live attenuated vaccine. In addition,RPS may be affected by vaccination and infection methods. | 14 | Analysis of Biological Activity and Immunogenicity of Extracellular Products from Streptococcus iniae显示文摘[Objectives] This study aimed to analyze the biological activity and immunogenicity of extracellular products from Streptococcus iniae. [Methods] S.iniae was incubated with brain heart infusion agar medium( BHIA + 4% calf serum) for 60 h. The bacterial liquid was rinsed with PBS,centrifuged,and filtered through microporous filtering film to collect extracellular products( ECPs). [Results]Extracellular proteinase( ECPase) of S. iniae exhibited amylase,protease,lecitinase,gelatinase,lipase activities and hemolytic activity but had no urease activity. EDTA,DTT and PMSF could reduce ECPase activity to 72. 4%,77. 6%and 72. 4%,respectively. Cu^(2+),Ca^(2+),K+and Mg^(2+)exhibited an inhibitory effect on ECPase activity,whereas Fe^(3+),Co^(2+)and Mn^(2+)could activate ECPase activity. ECPs had good heat stability and exhibited relatively high activities under alkaline conditions. The optimal temperature for ECPs was 55 ℃. Two-dimensional electrophoresis was performed to analyze the main protein of ECPs. The results indicated that there are 12 main bands of ECPs,and the molecular weights mainly ranged between 28-68 kD a. About 120 protein spots were detected,and the molecular weights mainly ranged between 26-95 kD a. The mouse anti-S. iniae was used for Western-blot analysis of ECPs,and the results showed that there were four proteins,with molecular weights of 26,37,95,and 97 kD a,respectively. The pathogenicity assay indicated that ECPs of S. iniae were highly pathogenic to tilapia. The mortality rate of tilapia was enhanced as the concentration of ECPs increased. | 16 | Molecular Cloning and Bioinformatics Analysis of kdpD Gene from Vibrio alginolyticus显示文摘In this study,histidine kinase gene kdpD in the two-component regulatory system of Vibrio alginolyticus strain HY9901 was cloned for bioinformatics analysis. Sequence analysis revealed that kdpD gene( GenB ank accession number: KJ544668) was 1 374 bp in length,encoding a putative protein of 457 amino acids. The predicted molecular weight( MW) of KdpE was 51. 60 kD with a theoretical isoelectric point( pI) of 6. 02. Using SignalP 4. 0,TMHMM Server 2. 0 and SoftB erry-Psite software,it was predicted that KdpE protein was equally located in Golgi apparatus,plasma membrane and endoplasmic reticulum( 33. 3%),which did not contain a signal peptide but contained three transmembrane domains. KdpE protein had five casein kinase II phosphorylation sites,five protein kinase C phosphorylation sites and one cA MP- and cG MP-dependent protein kinase phosphorylation site. A phylogenetic tree was constructed by MEGA 5. 0 software,which revealed that KdpD from V. alginolyticus had close genetic relationship with corresponding proteins from V. campbellii and V. parahaemolyticus. | 18 | Dynamical investigation of tunable magnetism in Au@Ni-carbide nanocrystals by a combined soft and hard X-ray absorption spectroscopy显示文摘Nickel based magnetic nanocrystals have been widely applied in magnetic and catalytic facilities.Tunable magnetic properties of nickel can be easily obtained via non-magnetic doping or phase transformation.However,phase transformation from face centered cubic(fcc)to hexagonal close packed(hcp)induced magnetism adjustment of Ni are always confused with nickel carbide(Ni_(3)C),due to the similar atomic structures of hcp-Ni and Ni3C.Here,we present series of Au@Ni-carbide magnetic materials achieved from the controlled carbonation of Au@Ni core-shell structures,whose magnetism is tunable by adjusting the amount of carbon in the Ni layer.Ex-situ hard X-ray absorption spectroscopy(XAS)at the metal K edge and soft XAS at both metal L edge and carbon K edge provide solid evidence for the carbonation process from fcc-Ni to Ni_(x)C,rather than phase transformation to hcp-Ni.Further investigation reveals that the magnetism of the hybrids is mainly contributed from the residual fcc-Ni.The result represents an accurate and effective way to distinguish hexagonal Ni_(3)C from hcp-Ni,and provides the pathway to control magnetism of Ni-based materials for applications. | Weifeng Huang Jianxin Kang Tingwen Chen Dawei Pang Lihua Wang Hang Wei Changchun Yang Dongfeng Zhang Lin Guo | 2022 | Nano Research2022,15,5: | 0 |
