| 6 | Dual gRNAs guided CRISPR/Cas9 system inhibits hepatitis B virus replication显示文摘AIM: To screen and investigate the effective g RNAs against hepatitis B virus(HBV) of genotypes A-D.METHODS: A total of 15 g RNAs against HBV of genotypes A-D were designed. Eleven combinations of two above g RNAs(dual-g RNAs) covering the regulatory region of HBV were chosen. The efficiency of each g RNA and 11 dual-g RNAs on the suppression of HBV(genotypes A-D) replication was examined by the measurement of HBV surface antigen(HBs Ag) or e antigen(HBe Ag) in the culture supernatant. The destruction of HBV-expressing vector was examined in Hu H7 cells co-transfected with dual-g RNAs and HBVexpressing vector using polymerase chain reaction(PCR) and sequencing method, and the destruction of ccc DNAwas examined in Hep AD38 cells using KCl precipitation, plasmid-safe ATP-dependent DNase(PSAD) digestion, rolling circle amplification and quantitative PCR combined method. The cytotoxicity of these g RNAs was assessed by a mitochondrial tetrazolium assay.RESULTS: All of g RNAs could significantly reduce HBs Ag or HBe Ag production in the culture supernatant, which was dependent on the region in which g RNA against. All of dual g RNAs could efficiently suppress HBs Ag and/or HBe Ag production for HBV of genotypes A-D, and the efficacy of dual g RNAs in suppressing HBs Ag and/or HBe Ag production was significantly increased when compared to the single g RNA used alone. Furthermore, by PCR direct sequencing we confirmed that these dual g RNAs could specifically destroy HBV expressing template by removing the fragment between the cleavage sites of the two used g RNAs. Most importantly, g RNA-5 and g RNA-12 combination not only could efficiently suppressing HBs Ag and/or HBe Ag production, but also destroy the ccc DNA reservoirs in Hep AD38 cells.CONCLUSION: These results suggested that CRISPR/Cas9 system could efficiently destroy HBV expressing templates(genotypes A-D) without apparent cytotoxicity. Firstly,we derive the Fourier spectral transfer form of the solution to the definition problem,which describes the one-dimensional seismic wave field. Then based on the special models of the source,path and local site,the physical random function model of ground motions is obtained whose physical parameters are random variables. The superposition method of narrow-band harmonic wave groups is improved to synthesize ground motion samples. Finally,an application of this model to simulate ground motion records in 1995 Kobe earthquake is described. | 9 | Microstructure and mechanical properties of NZ30K alloy by semi-continuous direct chill and sand mould casting processes显示文摘The Mg-3.0Nd-0.2Zn-0.4Zr (NZ30K) alloys were prepared by direct-chill casting (DCC) and sand mould casting (SMC) processes,respectively and their microstructures and mechanical properties were investigated.The results indicate that casting method plays a remarkable influence on the microstructure and mechanical properties of as-cast NZ30K alloy.The grain size increases from 35-40μm in the billets made by the DCC to about 100-120μm in the billets by the SMC.The aggregation of Mg12Nd usually found at the triple joints of grain boundaries in the billets prepared by SMC while is not observable from the billets by DCC.The tensile strengths and elongations of the billets are 195.2 MPa and 15.5% by DCC,and 162.5 MPa and 3.2% by SMC,respectively.The tensile strength of the alloy by DCC is remarkably enhanced by T6 heat treatment,which reached 308.5 MPa.Fracture surfaces of NZ30K alloy have been characterized as intergranular fracture by SMC and quasi-cleavage fracture by DCC,respectively. | Zheng Xingwei Dong Jie Liu Wencai Ding Wenjiang | 2011 | China Foundry2011,8,1: | 25 |
| 10 | Tillage and straw mulching impacts on grain yield and water use efficiency of spring maize in Northern Huang–Huai–Hai Valley显示文摘A two-year field experiment(2012–2013) was conducted to investigate the effects of two tillage methods and five maize straw mulching patterns on the yield, water consumption,and water use efficiency(WUE) of spring maize(Zea mays L.) in the northern Huang–Huai–Hai valley of China. Compared to rotary tillage, subsoil tillage resulted in decreases in water consumption by 6.3–7.8% and increases in maize yield by 644.5–673.9 kg ha-1, soil water content by 2.9–3.0%, and WUE by 12.7–15.2%. Chopped straw mulching led to higher yield,soil water content, and WUE as well as lower water consumption than prostrate whole straw mulching. Mulching with 50% chopped straw had the largest positive effects on maize yield, soil water content, and WUE among the five mulching treatments. Tillage had greater influence on maize yield than straw mulching, whereas straw mulching had greater influence on soil water content, water consumption, and WUE than tillage. | 12 | Contributions of climate and human activities to changes in runoff of the Yellow and Yangtze rivers from 1950 to 2008显示文摘Runoffs in the Yellow River and Yangtze River basins,China,have been changing constantly during the last half century.In this paper,data from eight river gauging stations and 529 meteorological stations,inside and adjacent to the study basins,were analyzed and compared to quantify the hydrological processes involved,and to evaluate the role of human activities in changing river discharges.The Inverse Distance Weighted(IDW)interpolation method was used to obtain climatic data coverage from station observations.According to the runoff coefficient equation,the effect of human activities and climate can be expressed by changes in runoff coefficients and changes in precipitation,respectively.Annual runoff coefficients were calculated for the period 1950-2008,according to the correlation between respective hydrological series and regional precipitation.Annual precipitation showed no obvious trend in the upper reaches of the Yellow River but a marked downward trend in the middle and downstream reaches,with declines of 8.8 and 9.8 mm/10 a,respectively.All annual runoff series for the Yellow River basin showed a significant downward trend.Runoff declined by about 7.8 mm/10 a at Sanmenxia and 10.8 mm/10 a at Lijin.The series results indicated that an abrupt change occurred in the late 1980s to early 1990s.The trend of correlations between annual runoff and precipitation decreased significantly at the Yellow River stations,with rates ranging from 0.013/10 a to 0.019/10 a.For the hydrologic series,all precipitation series showed a downward trend in the Yangtze River basin with declines ranging from about 24.7 mm/10 a at Cuntan to 18.2 mm/10 a at Datong.Annual runoff series for the upper reaches of the Yangtze River decreased significantly,at rates ranging from 9.9 to 7.2 mm/10 a.In the middle and lower reaches,the runoff series showed no significant trend,with rates of change ranging from 2.1 to 2.9 mm/10 a.Human activities had the greatest influence on changes in the hydrological series of runoff,regardless of whether the effect was negative or positive.During 1970-2008,human activities contributed to 83% of the reduction in runoff in the Yellow River basin,and to 71% of the increase in runoff in the Yangtze River basin.Moreover,the impacts of human activities across the entire basin increased over time.In the 2000s,the impact of human activities exceeded that of climate change and was responsible for 84% of the decrease and 73% of the increase in runoff in the Yellow River and Yangtze River basins,respectively.The average annual runoff from 1980 to 2008 fell by about 97%,83%,83%,and 91%,compared with 1951-1969,at the Yellow River stations Lanzhou,Sanmenxia,Huayuankou and Lijin,respectively.Most of the reduction in runoff was caused by human activities.Changes in precipitation also caused reductions in runoff of about 3%,17%,17%,and 9% at these four stations,respectively.Falling precipitation rates were the main explanation for runoff changes at the Yangtze River stations Cuntan,Yichang,Hankou,and Datong,causing reductions in runoff of 89%,74%,43%,and 35%,respectively.Underlying surface changes caused decreases in runoff in the Yellow River basin and increases in runoff in the Yangtze River basin.Runoff decreased in arid areas as a result of increased water usage,but increased in humid and sub-humid areas as a result of land reclamation and mass urbanization leading to decreases in evaporation and infiltration. | WANG Yan DING YongJian YE BaiSheng LIU FengJing WANG Jie WANG Jie | 2013 | Science China Earth Sciences2013,56,8: | 21 |

| 14 | Precise nanomedicine for intelligent therapy of cancer显示文摘Precise nanomedicine has been extensively explored for efficient cancer imaging and targeted cancer therapy, as evidenced by a few breakthroughs in their preclinical and clinical explorations. Here, we demonstrate the recent advances of intelligent cancer nanomedicine, and discuss the comprehensive understanding of their structure-function relationship for smart and efficient cancer nanomedicine including various imaging and therapeutic applications, as well as nanotoxicity. In particular, a few emerging strategies that have advanced cancer nanomedicine are also highlighted as the emerging focus such as tumor imprisonment, supramolecular chemotherapy, and DNA nanorobot. The challenge and outlook of some scientific and engineering issues are also discussed in future development. | 15 | Origin of the Newly Discovered Zhunuo Porphyry Cu-Mo-Au Deposit in the Western Part of the Gangdese Porphyry Copper Belt in the Southern Tibetan Plateau,SW China显示文摘The newly discovered Zhunuo porphyry Cu-Mo-Au deposit is located in the western part of the Gangdese porphyry copper belt in southern Tibet, SW China. The granitoid plutons in the Zhunuo region are composed of quartz diorite porphyry, diorite porphyry, granodiorite porphyry, biotite monzogranite and quartz porphyry. The quartz diorite porphyry yielded zircon U-Pb ages of 51.9±0.7 Ma(Eocene) using LA-ICP-MS, whereas the diorite porphyry, granodiorite porphyry, biotite monzogranite and quartz porphyry yielded ages ranging from 16.2±0.2 to 14.0±0.2 Ma(Miocene). CuMo-Au mineralization is mainly hosted in the Miocene granodiorite porphyry. Samples from all granitoid plutons have geochemical compositions consistent with high-K calc-alkaline series magmatism. The samples display highly fractionated light rare-earth element(REE) distributions and heavy REE distributions with weakly negative Eu anomalies on chondrite-normalized REE patterns. The trace element distributions exhibit positive anomalies for large-ion lithophile elements(Rb, K, U, Th and Pb) and negative anomalies for high-field-strength elements(Nb and Ti) relative to primitive mantlenormalized values. The Eocene quartz diorite porphyry yielded εNd(t) values ranging from-3.6 to-5.2,(^(87)Sr/^(86)Sr)i values in the range 0.7046–0.7063 and initial radiogenic Pb isotopic compositions with ranges of 18.599–18.657 ^(206)Pb/^(204)Pb, 15.642–15.673 ^(207)Pb/^(204)Pb and 38.956–39.199 ^(208)Pb/^(204)Pb. In contrast, the Miocene granitoid plutons yielded ε_(Nd)(t) values ranging from-6.1 to-7.3 and(87Sr/86Sr)i values in the range 0.7071–0.7078 with similar Pb isotopic compositions to the Eocene quart diorite. The Sr-Nd-Pb isotopic compositions of the rocks are consistent with formation from magma containing a component of remelted ancient crust. Zircon grains from the Eocene quartz diorite have ε_(Hf)(t) values ranging from-5.2 to +0.9 and two-stage Hf model ages ranging from 1.07 to 1.46 Ga, while zircon grains from the Miocene granitoid plutons have ε_(Hf)(t) values from-9.9 to +4.2 and two-stage Hf model ages ranging from 1.05–1.73 Ga, indicating that the ancient crustal component likely derives from Paleo- to Mesoproterozoic basement. This source is distinct from that of most porphyry Cu-Mo-Au deposits in the eastern part of the Gangdese porphyry copper belt, which likely originated from juvenile crust. | 16 | Effect of L-NAME on nitric oxide and gastrointestinal motility alterations in cirrhotic rats显示文摘AIM: To invsstigare the effect of L-NAME on nitric oxide andgastriubtestubal motility alterations in cirrhotic ratsMETHODS: Rats with cirrhosis induced by carbontetrachloride were randomly divided into two groups, one( n= 13) receiving 0. 5 mg@ kg-1 per clay of NG-nitro-L-argininemethyl ester (L-NAME), a nitric oxide synthase inhibitor,for 10 days, whereas the other group ( n = 13) and control( n = 10) rats were administrated the same volume of 9 g@ L-1saline.Half gastric emptying time and 2 h residual rate weremeasured by SPECT, using 99m Tc-DTPA-labeled bariumsuifate as test meal. Gastrointestinal transition time wasrecorded simultaneously. Serum concentration of nitrcoxide (NO) was determined by the kinetic cadmiunreduction and colorimetric methods. ImmunohistochemicalSABC method was used to observe the expression anddistribution of three types of nitric oxide synthase (NOS)isoforms in the mt gastrointestinal tract. Western blot wasused to detect expression of gastrointestinal NOS isoforms.RESULTS: Half gastric emptying time and trans-gastrointestinal time were significantly prolonged( 124.0 ± 26.4min; 33.7± 8.9min;72.1 ± 15.3 min; P<0.01), (12.4±0.5h; 9.5±0.3 h; 8.2±0.8 h; P<0.01), 2h residual rate wasraised in cirrhotic rots than in controls and cirrhotic ratstreated with L-NAME(54.9± 7.6 % ,13.7 ± 3.2 %, 34.9± 10.3%, P< 0.01). Serum concentration of NO was significantlyincreased in cirrhotic rots than in the other groups (8.20 ± 2.48)μmol@L-1, (5.94± 1.07) μmol@L-1 ,and control (5.66± 1.60) tμmol@L-1, P< 0.01. NOS staining intensities which weremainly located in the gastrointestinal tissues were markedlylower in cirrhotic rats than in the controls and cirrhotic ratsafter treated with L- NAME.CONCLUSION: Gastrointestinal motility was remarkablyinhibited in cirrhotic rats, which could he alleviated by L-NAME. | 17 | Integrative analysis of aberrant Wnt signaling in hepatitis B virus-related hepatocellular carcinoma显示文摘AIM: To comprehensively understand the underlying molecular events accounting for aberrant Wnt signaling activation in hepatocellular carcinoma(HCC).METHODS: This study was retrospective. The HCC tissue specimens used in this research were obtained from patients who underwent liver surgery. The Catalogue of Somatic Mutations in Cancer(COSMIC) database was searched for the mutation statuses of CTNNB1, TP53, and protein degradation regulator genes of CTNNB1. Dual-luciferase reporter assay was performed with TOP/FOP reporters to detect whether TP53 gain-of-function(GOF) mutations could enhance the transcriptional activity of Wnt signaling. Methylation sensitive restriction enzyme-quantitative PCR was used to explore the methylation status of Cp G islands located in the promoters of APC, SFRP1, and SFRP5 in HCCs with different risk factors. Finally, nestedreverse transcription PCR was performed to examine the integration of HBx in front of LINE1 element and the existence of HBx-LINE1 chimeric transcript in Hepatitis B virus-related HCC. All results in this article were analyzed with the software SPSS version 19.0 for Windows, and different groups were compared by χ2 test as appropriate.RESULTS: Based on the data from COSMIC database, compared with other solid tumors, mutation frequency of CTNNB1 was significantly higher in HCC(P < 0.01). The rate of CTNNB1 mutation was significantly less frequent in Hepatitis B virus-related HCC than in other etiologies(P < 0.01). Dual-luciferase reporter system and TOP/FOP reporter assays confirmed that TP53 GOF mutants were able to enhance the transcriptional ability of Wnt signaling. An exclusive relationship between the status of TP53 and CTNNB1 mutations was observed. However, according to the COSMIC database, TP53 GOF mutation is rare in HCC, which indicates that TP53 GOF mutation is not a reason for the aberrant activation of Wnt signaling in HCC. APC and AXIN1 were mutated in HCC. By using methylation sensitive restriction enzyme-quantitative PCR, hypermethylation of APC was detected in HCC with different risk factors, whereas SFRP1 and SFRP5 were not hypermethylated in any of the HCC etiologies, which indicates thatthe mutation of APC and AXIN1, together with the methylation of APC could take part in the overactivation of Wnt signaling.
