1Efficient generation of transgenic chickens using the spermatogonial stem cells in vivo and ex vivo transfection显示文摘The highly efficient novel methods to produce transgenic chickens were established by directly in-jecting the recombinant plasmid containing green fluorescent protein (GFP) gene into the cock's testis termed as testis-medianted gene transfer (TMGT), and transplanting transfected spermatogonial stem cells (TTSSCs). For the TMGT approach,four dosages of pEGFP-N1 DNA/cationic polymer complex were injected intratesticularly. The results showed: (1) 48 h after the injection,the percentages of testis cells expressing GFP were 4.0%, 8.7%, 10.2% and 13.6% in the 50, 100, 150 and 200 μg/mL group, re-spectively. The difference from the four dosage groups was significant (P<0.05). On day 25 after the injection, a dosage-dependent and time-dependent increase in the number of transgenic sperm was observed. The percentages of gene expression reached the summit and became stable from day 70 to 160, being 12.7%, 12.8%, 15.9% and 19.1%, respectively. The difference from the four dosage groups was also significant (P<0.05). (2) 70 d after the injection, strong green fluorescent could be observed in the seminiferous tubules by whole-mount in-situ hybridization. (3) 70 d after the injection, the semen was collected and used to artificially inseminate wild-type females. The blastoderms of F1 and F2 transgenic chicken expressed GFP were 56.2% (254/452) and 53.2% (275/517), respectively. The detec-tion of polymerase chain reaction (PCR) of F1 and F2 transgenic chicken blood genomic DNA showed that 56.5% (3/23) of F1 and 52.9% (9/17) of F2 were positive. Southern blot showed GFP DNA was in-serted in their genomic DNAs. (4) Frozen whole mount tissue sections of F1 and F2 transgenic chicken liver, heart, kidney and muscle showed that the rates of green fluorescent positive were between 50.0% and 66.7%. (5) With the TTSSCs method, SSCs ex vivo transfected with GFP were transplanted into recipient roosters whose endogenic SSCs had been resoluted. The donor SSCs settled and GFP ex-pression became readily detectable in the frozen whole mount tissue sections of recepient testes. Moreover, sperms carrying GFP could be produced normally. The results of artificially inseminating wild-type females with these sperms showed 12.5% (8/64) of offspring embryo expressed GFP and 11.1% (2/18) hatched chicks were tested transgenic. 2Bio-inspired polygonal skeleton structure for vibration isolation:Design,modelling,and experiment显示文摘Inspired by the safe landing of a cat falling from a high altitude,a bio-inspired polygonal skeleton(BIPS) structure is proposed,and its nonlinear characteristics are systematically studied to explore its potential application in the suppression of vibration. The polygon is formed by the skeleton structure of the cat’s entire body and the ground. The BIPS system consists of two symmetrical bionic legs with three robs(as skeleton) and four horizontal springs(as muscle). Two bionic legs are connected through the bearing platform(as spine),which could adjust the distance between the two bionic legs. A theoretical model is developed to characterize its stiffness nonlinearity through geometrical and mechanical analysis. Parameter analysis reveals that the BIPS structure has diverse stiffness,including nonlinear positive stiffness and negative stiffness. By imitating adjustment of leg posture and telescopic function of the spine(control the distance between legs),these flexible stiffness properties can be adjusted by structure parameters. In addition,the load capacity and working range can also be designed by the length of the bars,the initial angle,the mounting position,and the spring stiffness. The experimental setup is established,and the vibration isolation performance under various excitation is tested. The experimental results verify the accuracy of the dynamic model and also show that the proposed BIPS structure can suppress the vibration effectively under a variety of excitations. 3Design,modeling and experimental investigation of a magnetically modulated rotational energy harvester for low frequency and irregular vibration显示文摘Vibration energy harvesting is a promising approach for sustainable energy generation from ambience to meet the development of self-powered systems.Here,we propose a novel compact non-resonant magnetically modulated rotational energy harvester(MMR-EH)for low frequency and irregular vibration.Through the rational arrangement of multiple magnetic fields in space,a ring route with low potential energy is established.A movable magnet can be non-contact modulated by the magnetic force to move along the ring route under irregular vibration,which is instrumental in electromechanical energy conversion.A dynamic model of the MMR-EH is developed based on the energy method and verified experimentally.The effects of key parameters on the magnetically modulated route are analysed.The simulation and experimental results demonstrate that the MMR-EH can effectively harvest the energy from ultra-low frequency(3 Hz)and irregular vibration.At a reciprocating vibration frequency of 10 Hz and an amplitude of 20 mm,the harvester can produce an average power of 0.29 mW.ZHAO LinChuan ZOU HongXiang GAO QiuHua YAN Ge WU ZhiYuan LIU FengRui WEI KeXiang YANG Bin ZHANG WenMing 2020Science China(Technological Sciences)2020,63,10:5

6Identification of a novel seed size associated locus SW9-1 in soybean显示文摘Seed size is one of the vital traits determining seed appearance, quality, and yield. Untangling the genetic mechanisms regulating soybean 100-seed weight (100-SW), seed length and seed width across environments may provide a theoretical basis for improving seed yield. However, there are few reports related to QTL mapping of 100-SW across multiple ecological regions. In this study, 21 loci associated with seed size traits were identified using a genome-wide association of 5361 single nucleotide polymorphisms (SNPs) across three ecoregions in China, which could explain 8.12%–14.25% of the phenotypic variance respectively. A new locus, named as SW9-1 on chromosome 9 that explained 10.05%–10.93% of the seed weight variance was found significantly related to seed size traits, and was not previously reported. The selection effect analysis showed that SW9-1 locus has a relatively high phenotypic effect (13.67) on 100-SW, with a greater contribution by the accessions with bigger seeds (3.69) than the accessions with small seeds (1.66). Increases in seed weight were accompanied by increases in the frequency of SW9-1T allele, with >90% of the bred varieties with a 100-SW >30 g carrying SW9-1T. Analysis of SW9-1 allelic variation in additional soybean accessions showed that SW9-1T allele accounting for 13.83% of the wild accessions, while in 46.55% and 51.57% of the landraces and bred accessions, respectively, this results indicating that the SW9-1 locus has been subjected to artificial selection during the early stages of soybean breeding, especially the utilization of SW9-1T in edamame for big seed. These results suggest that SW9-1 is a novel and reliable locus associated with seed size traits, and might have an important implication for increasing soybean seed weight in molecular design breeding. 7Magnetic levitation using diamagnetism:Mechanism,applications and prospects显示文摘As a typical contact-free manipulation technique that removes friction and contamination risk,levitation has gradually become a preferred candidate for various applications.Magnetic levitation using diamagnetism,beyond Earnshaw’s theorem,is a kind of passive stable levitation that can be achieved at normal temperatures with no energy input.Appealingly,most seemingly nonmagnetic materials can be levitated in a magnetic field and can stabilize free levitation of magnetic materials.This review focuses on the fundamental principles of magnetic levitation using diamagnetism,with emphasis on its burgeoning applications.The theoretical basis associated with the magnetic levitation using diamagnetism is discussed by elucidating the characteristics of diamagnetic materials,and the key levitation mechanisms are clarified.Afterwards,state-of-the-art applications in various aspects,including sensing and measurement,actuating and micromanipulation,energy harvesting and magnetic gravity compensation,are summarized and compared.Finally,the review concludes with a brief outlook on future perspectives.GAO QiuHua YAN Han ZOU HongXiang LI WenBo PENG ZhiKe MENG Guang ZHANG WenMing 2021Science China(Technological Sciences)2021,64,1:3
8Analysis of single frequency modulation for passive hydrogen maser显示文摘Based on the theory of the passive hydrogen maser,along with the technology of frequency modulation and modulation transfer spectroscopy, the theoretical expression of the single frequency modulation for the passive hydrogen maser and the function of the cavity and H line error signals separation are derived, which are basically coincident with the experiment. The absorption and dispersion spectrum curves with different resonance widths show that the cavity and hydrogen transition serve as discriminators, and the two error signals can be separated.Through the calculations of the two error signals in the passive hydrogen maser, it analyzes the traditional method of the two error signals separation, and then describes a new improved method for the passive hydrogen servo loops consisting in the use of a single modulation frequency and frequency discrimination. 9Feasibility Study of Aluminum Hydroxide Sol as Adjuvant to Prepare Stable Vaccine Using Newcastle Disease Virus as a Model显示文摘Aluminum hydroxide sol (consisting of aluminum hydroxide nano-particles) prepared by a hydrothermal method was used as nano-aluminum adjuvant adsorbed the inactived Newcastle disease virus (NDV). The zeta potential of aluminum hydroxide colloidal particles,the inactivated NDV and the nano-aluminum adjuvant NDV are +48.16 mV,?13.8 mV and +39.97 mV,respec-tively. The nano-aluminum adjuvant vaccine was transparent and stable without precipitate,whereas the conventional aluminum ad-juvant vaccine was turbid and a white precipitate was visible soon. The hemagglutination inhibition (HI) titers of the nano-aluminum adjuvant group were generally higher than those of the conventional aluminum group except the titer on day 29. 14Micron-to nano-pore characteristics in the shale of Longmaxi Formation,southeast Sichuan Basin显示文摘For shale of Lower Silurian Longmaxi Formation in Chongqing,southeast Sichuan Basin,characteristics of micro-nano pores in marine shale reservoirs were well studies by means of Field-Emission Scanning Electron Microscope and Low-temperature Low-pressure Adsorption Experiment of CO_(2)and N_(2).Results showed that six types of pore were developed in the shale of Longmaxi Formation,i.e.,organic pores,intergranular pores,intragranular pores,intercrystalline pores,dissolution pores and microfractures,among which the organic pores and intragranular pores in interlayers of clay minerals were most developed,and a plenty of dissolution pores were also well developed because of high thermal evolution degree.BET specific surface area of the shale in Longmaxi Formation ranged from 3.5 to 18.1 m^(2)/g,BJH total pore volume was from 0.00234 to 0.01338 cm^(3)/g,DA specific surface area of micropores vaired from 1.3 to 7.3 m^(2)/g,and DA pore volume ranged from 0.00052 to 0.00273 cm^(3)/g.The specific surface area of micropores in the shale accounted for 23.1%-80.2%of total specific surface area with an average of 50.3%,and the pore volume of micropores accounted for 12.1%-48.5%of total pore volume with an average of 32.3%.Micropore was the main storage space in shale reservoir for methane adsorption,that because capacities of specific surface area provided by micropores were considerably greater than those provided by mesopores and macropores.Pore size distribution of the shale was complex,and multiple different peaks occurred in the pore size curves,showing two or three peaks in the range from 0 to 100 nm and four peaks occasionally.TOC had a good linear relationship with pore structure parameters of micropores,mesopores t macropores and total pores in the shale,indicating that TOC was the most important control factor for micron-to nano-pore structure in the shale.After normalization of pore structure parameters to TOC,the pore structure parameters of total pores and mesopores t macropores,had positive linear relationships with content of clay minerals but negative linear relationships with content of brittle minerals,indicating that clay minerals and brittle minerals mainly controlled development of mesopores and macropores in the shale.Wenming Ji Yan Song Zhenxue Jiang Mianmo Meng Qingxin Liu Fenglin Gao 2017Petroleum Research2017,2,2:1

19Consistent Alterations of Human Fecal Microbes After Transplantation into Germ-free Mice显示文摘Fecal microbiota transplantation(FMT)of human fecal samples into germ-free(GF)mice is useful for establishing causal relationships between the gut microbiota and human phenotypes.However,due to the intrinsic differences between human and mouse intestines and the different diets of the two organisms,it may not be possible to replicate human phenotypes in mice through FMT;similarly,treatments that are effective in mouse models may not be effective in humans.In this study,we aimed to identify human gut microbes that undergo significant and consistent changes(i.e.,in relative abundances)after transplantation into GF mice in multiple experimental settings.We collected 16S rDNA-seq data from four published studies and analyzed the gut microbiota profiles from 1713 human–mouse pairs.Strikingly,on average,we found that only 47%of the human gut microbes could be re-established in mice at the species level,among which more than 1/3 underwent significant changes(referred to as“variable taxa”).Most of the human gut microbes that underwent significant changes were consistent across multiple human–mouse pairs and experimental settings.Consequently,about 1/3 of human samples changed their enterotypes,i.e.,significant changes in their leading species after FMT.Mice fed with a controlled diet showed a lower enterotype change rate(23.5%)than those fed with a noncontrolled diet(49.0%),suggesting a possible solution for rescue.Most of the variable taxa have been reported to be implicated in human diseases,with some recognized as the causative species.Our results highlight the challenges of using a mouse model to replicate human gut microbiota-associated phenotypes,provide useful information for researchers using mice in gut microbiota studies,and call for additional validations after FMT.An online database named FMT-DB is publicly available at http://gffzz31fc3efd07a444b1houwqcx5965cf6vn0.ffgz.tsg.suse.edu.cn/#/.Yanze Li Wenming Cao Na L Gao Xing-Ming Zhao Wei-Hua Chen 2022Genomics, Proteomics & Bioinformatics2022,20,2:0
