| 1 | Antihepatoma effect of alpha-fetoprotein antisense phosphorothioate oligodeoxyribonucleotides in vitro and in mice显示文摘AIM To evaluate antihepatoma effect ofantisense phosphorothioate oligodeo-xyribonucleotides (S-ODNs) targeted to alpha-fetoprotein (AFP) genes in vitro and in nudemice.METHODS AFP gene expression was examinedby immunocytochemical method or enzyme-linked immunosorbent assay. Effect of S-ODNson SMMC-7721 human hepatoma cell growth invitro was determined using microculturetetrazolium assay. In vivo antitumor activitiesof S-ODNs were monitored by measuring tumorweight differences in treated and control micebearing SMMC-7721 xenografts. Induction of cellapoptosis was evaluated by fluorescence-activated cell sorter (FACS) analysis.RESULTS Antisense S-ODN treatment led toreduced AFP gene expression. Specificantisense S-ODNs, but not control S-ODNs,inhibited the growth of heaptoma cells in vitro.In vivo. only antisense S-ODNs exhibitedobvious antitumor activities. FACS analysisrevealed that the growth inhibition by antisenseS. | 2 | Growth-inhibiting effects of taxol on human liver cancer in vitro and in nude mice显示文摘AIM To investigate the effects of taxol onSMMC-7721 human hepatoma and itsmechanisms.METHODS In vitro cell growth was assessedby trypan blue exclusion method.Experimentalhepatoma model was established by seedingSMMC-7721 cells subcutaneously into Balb/c(nu/nu)nude mice.In vivo tumor growth wasdetermined by measurement of tumor diameterwith Vernier calipers.The syntheses of DNA,RNA and protein were analyzed by incorporationof ~3H-thymidine,~3H-uridine and ~3H-leucinerespectively.Using light and electronmicroscopes to observe the morphologicalchanges of cells including mitosis andapoptosis.RESULTS Taxol was effective against SMMC-7721 human hepatoma cell growth in the rangesof 2.5 nmol/L-10 nmol/L with mitotic arrestand apoptosis in vitro.DNA,RNA and proteinsyntheses in cells were also obviouslysuppressed by in vitro treatment of taxol for72 h.Taxol at 2.5 nmol/L reduced ~3H-thymidineuptake to about 34% of the control value(P<0.05).Increasing the dose of taxol to20 nmol/L resulted in a greater decrease in ~3H-thymidine incorporation to 60% of the controlvalue(P<0.01).At a concentration of 20 nmol/L,the ~3H-uridine and ~3H-leucine uptakeswere reduced to 52%(P<0.05)and 63%(P<0.01),respectively.In vivo,taxolsignificantly inhibited SMMC-7721 tumor growthat 10 mg/kg,i.p.,once daily for 10 d.A morethan 90% decrease in tumor volume wasobserved by day 11(P<0.01)similarly withmitotic arrest and cell apoptosis.CONCLUSION Taxol has a marked anticanceractivity in SMMC-7721 human hepatoma both invitro and in nude mice.Its mechanisms might beassociated with mitotic arrest,subsequently,apoptosis of the hepatoma cells.No obvioustoxicity was observed with in vivoadministration of taxoi. | Jin Hui Yuan Ru Ping Zhang Ru Gang Zhang Li Xia Guo Xing Wang Wang Dan Luo Yong Xie Hong Xie | 2000 | World Journal of Gastroenterology2000,6,2: | 18 |
| 3 | Grape Seed Proanthocyanidin Extract Alleviates Arsenic-induced Oxidative Reproductive Toxicity in Male Mice显示文摘Objective To determine the ability of grape seed proanthocyanidin extract(GSPE) in alleviating arsenic-induced reproductive toxicity. Methods Sixty male Kunming mice received the following treatments by gavage: normal saline solution(control); arsenic trioxide(ATO; 4 mg/kg); GSPE(400 mg/kg); ATO+GSPE(100 mg/kg); ATO+GSPE(200 mg/kg) and ATO+GSPE(400 mg/kg). Thereafter, the mice were sacrificed and weighed, and the testis was examined for pathological changes. Nuclear factor(erythroid-derived 2)-like 2(Nrf2), heme oxygenase 1(HO1), glutathione S-transferase(GST), NAD(P)H dehydrogenase, and quinone 1(NQO1) expression in the testis was detected by real-time PCR. Superoxide dismutase(SOD), glutathione(GSH), total antioxidative capability(T-AOC), malondialdehyde(MDA), 8-hydroxydeoxyguanosine(8-OHd G), and reproductive indexes were analyzed. Results ATO-treated mice showed a significantly decreased sperm count and testis somatic index and activity levels of SOD, GSH, and T-AOC than control group. Compared to the ATO-treated group, ATO +GSPE group showed recovery of the measured parameters. Mice treated with ATO+high-dose GSPE showed the highest level of m RNA expression of Nrf2, HO, NQO1, and GST. It was expressed mainly in the inclusion body. The gene product was solubilized by guanidine hydrochloride, refolded by conventional dilution method, and purified using SP-sepharose cation chromatography.RESULTS: The expression vector B3 (scdsFv)-SEA-PETwas constructed, the expression product existed mainly in the inclusion body, the refolding product retained the binding ability of the single-chain antibody and had cytotoxic effect on HT-29 colon carcinoma cells. | 15 | The leaching of additive-derived flame retardants(FRs) from plastics in avian digestive fluids:The significant risk of highly lipophilic FRs显示文摘The exposure to plastic debris and associated pollutants for wildlife is of urgent concern,but little attention has been paid on the transfer of plastic additives from plastic debris to organisms.In the present study,the leaching of incorporated flame retardants (FRs),including polybrominated diphenyl ethers (PBDEs),alternative brominated FRs (AFRs),and phosphate flame retardants (PFRs),from different sizes of recycled acrylonitrile-butadiene-styrene (ABS) polymer were investigated in avian digestive fluids.The impact of co-ingested sediment on the leaching of additive-derived FRs in digestive fluids was also explored.In the recycled ABS,BDE 209 (715 μg/g) and 1,2-bis(2,4,6-tribromophenoxy) ethane (BTBPE,1766 μg/g) had the highest concentrations among all target FRs.The leaching proportions of FRs were higher in finer sizes of ABS.The leaching proportions of FRs from recycled ABS increased with elevated logKow of FRs.In the tests with coexisted ABS and sediment,hexato deca-BDEs,BTBPE,and decabromodiphenyl ethane (DBDPE) migrated from ABS to sediment,which resulted in the less bioaccessible fractions of these FRs in gut fluids.More lipophilic chemicals tended to be adsorbed by sediment from ABS.The results suggest the migration of additive-derived FRs from plastics to other indigestible materials in digestive fluids.The findings in this study provide insights into the transfer of additive-derived FRs from plastics to birds,and indicate the significant contribution of FR-incorporated plastics to bioaccumulation of highly lipophilic FRs. | Huiying Guo Xiaobo Zheng Shuling Ru Xiaojun Luo Bixian Mai | 2019 | Journal of Environmental Sciences2019,31,11: | 5 |

| 18 | Numerical simulation of spatial distributions of mining-induced stress and fracture fields for three coal mining layouts显示文摘In this study, the spatial distributions of stress and fracture fields for three typical underground coal mining layouts, i.e. non-pillar mining(NM), top-coal caving mining(TCM) and protective coal-seam mining(PCM), are modeled using discrete element software UDEC. The numerical results show that different mining layouts can lead to different mining-induced stress fields, resulting in diverse fracture fields. For the PCM, the mining influenced area in front of the mining faces is the largest, and the stress concentration factor in front of the mining faces is the lowest. The spatial shapes of the mining-induced fracture fields under NM, TCM and PCM differ, and they are characterized by trapezoidal, triangular and tower shapes, respectively. The fractal dimensions of mining-induced fractures of the three mining layouts decrease in the order of PCM, TCM and NM. It is also shown that the PCM can result in a better gas control effect in coal mines with high outburst potential. Methods A total of 2 873 men and 5 559 women were included in this cross-sectional study. Metabolic syndrome(MetS) was diagnosed according to the definition of Chinese Diabetes Society in 2004. The relation between WC and MetS was analyzed by multivariate logistic regression analysis. The optimal WC cut-off values were identified using the area under the ROC curve and the different diagnostic criteria for central obesity were compared. Results The WC was the risk factor for MetS independent of BMI, blood glucose, blood lipid, and blood pressure. The optimal WC cut-off value was 83.8 cm and 91.1 cm for identifying MetS in women and men, respectively. Compared with 80 cm and 85 cm for women and men, 85 cm and 90 cm had a higher Youden index for identifying all metabolic risk factors and MetS in women and men.
