1AXL is a candidate receptor for SARS-CoV-2 that promotes infection of pulmonary and bronchial epithelial cells显示文摘The current coronavirus disease 2019(COVID-19)pandemic presents a global public health challenge.The viral pathogen responsible,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),binds to the host receptor ACE2 through its spike(S)glycoprotein,which mediates membrane fusion and viral entry.Although the role of ACE2 as a receptor for SARS-CoV-2 is clear,studies have shown that ACE2 expression is extremely low in various human tissues,especially in the respiratory tract.Thus,other host receptors and/or co-receptors that promote the entry of SARS-CoV-2 into cells of the respiratory system may exist.In this study,we found that the tyrosine-protein kinase receptor UFO(AXL)specifically interacts with the N-terminal domain of SARS-CoV-2 S.Using both a SARS-CoV-2 virus pseudotype and authentic SARS-CoV-2,we found that overexpression of AXL in HEK293T cells promotes SARS-CoV-2 entry as efficiently as overexpression of ACE2,while knocking out AXL significantly reduces SARS-CoV-2 infection in HI 299 pulmonary cells and in human primary lung epithelial cells.Soluble human recombinant AXL blocks SARS-CoV-2 infection in cells expressing high levels of AXL.The AXL expression level is well correlated with SARS-CoV-2 S level in bronchoalveolar lavage fluid cells from COVID-19 patients.Taken together,our findings suggest that AXL is a novel candidate receptor for SARS-CoV-2 which may play an important role in promoting viral infection of the human respiratory system and indicate that it is a potential target for future clinical intervention strategies.Shuai Wang Zongyang Qiu Yingnan Hou Xiya Deng Wei Xu Tingting Zheng Peihan Wu Shaofang Xie Weixiang Bian Chong Zhang Zewei Sun Kunpeng Liu Chao Shan Aifu Lin Shibo Jiang Youhua Xie Qiang Zhou Lu Lu Jing Huang Xu Li 2021Cell Research2021,31,2:39
2Chemical characteristics of size-resolved aerosols in winter in Beijing显示文摘Size-resolved aerosols were continuously collected by a Nano Sampler for 13 days at an urban site in Beijing during winter 2012 to measure the chemical composition of ambient aerosol particles. Data collected by the Nano Sampler and an ACSM(Aerodyne Aerosol Chemical Speciation Monitor) were compared. Between the data sets,similar trends and strong correlations were observed,demonstrating the validity of the Nano Sampler. PM10 and PM2.5concentrations during the measurement were 150.5 ± 96.0 μg/m3(mean ± standard variation)and 106.9 ± 71.6 μg/m3,respectively. The PM2.5/PM10 ratio was 0.70 ± 0.10,indicating that PM2.5dominated PM10. The aerosol size distributions showed that three size bins of 0.5–1,1–2.5 and 2.5–10 μm contributed 21.8%,23.3% and 26.0% to the total mass concentration(TMC),respectively. OM(organic matter) and SIA(secondary ionic aerosol,mainly SO42-,NO3-and NH4+) were major components of PM2.5. Secondary compounds(SIA and secondary organic carbon) accounted for half of TMC(about 49.8%) in PM2.5,and suggested that secondary aerosols significantly contributed to the serious particulate matter pollution observed in winter. Coal burning,biomass combustion,vehicle emissions and SIA were found to be the main sources of PM2.5. 3Monovalent cation perm-selective membranes(MCPMs)'New developments and perspectives显示文摘As one of the most typical and promising membrane processes, electrodialysis(ED) technique plays a more and more significant role in industrial separation. Especially, the separation of monovalent cations and multivalent cations is currently a hot topic, which is not only desirable for many industries but also challenging for academic explorations. The main aim of the present contribution is to view the advances of a wide variety of monovalent cation perm-selective membranes(MCPMs) and their preparation technologies including(1) covalent crosslinking,(2) surface modification,(3) polymer blending,(4) electrospinning,(5) nanofiltration alike membrane,and(6) organic–inorganic hybrid. The relevant advantages and disadvantages with respect to some specific cases have been discussed and compared in detail. 4Recent progress and perspective of electrochemical CO_(2) reduction towards C_(2)-C_(5) products over non-precious metal heterogeneous electrocatalysts显示文摘Electroreduction of carbon dioxide(CO_(2)ER)into value-added chemical compounds has presented as a promising route for renewable carbon cycle,which alleviates global warming concern.Compared with traditional C1 products,high-value multicarbon products converted from atmospheric CO_(2) via CO_(2)ER have attracted dramatic interest due to their significant economic efficiency,however desired catalytic selectivity of multicarbon products is difficult to achieve because of the high thermodynamic barriers and complex reaction pathways.To replace currently used precious-metal based catalysts,developing highly efficient and precious-metal-free CO_(2)ER catalysts based on earth abundant elements is the top priority to meet the requirements of industrialization.Although certain progress has been made,there are still few systematic reports on the non-precious metal heterogeneous(NPMH)CO_(2)ER electrocatalysts for efficient conversion of CO_(2) to multicarbon products.Herein,we summarize the latest research advances in recent developments of NPMH electrocatalysts,including nanostructured Cu,Cu-based bimetallic catalysts,Cu-based complexes,and carbon-based Cu-free catalysts for electroreduction of CO_(2) into high-value multicarbon products.The corresponding CO_(2)ER performances are discussed in the order of the types of multicarbon products,specifically for ethanol(C2H5OH),ethylene(C2H4),ethane(C2H6),acetic acid(CH3COOH),propanol(C3H7OH),and other O^(2+)products with a special attention paid to understand the structure-activity relationship.Moreover,key strategies and characterization techniques for catalytic mechanism insights,and unsolved issues and future trends for enhancing the CO_(2)ER performance of NPMH electrocatalysts are highlighted,which provides a constructive guidance on the development of CO_(2)ER electrocatalysts with high activity and selectivity for multicarbon products.Jiayi Chen Tingting Wang Zhongjian Li Bin Yang Qinghua Zhang Lecheng Lei Pingyun Feng Yang Hou 2021Nano Research2021,14,9:7

7Regulation of superoxide flashes by transient and steady mitochondrial calcium elevations显示文摘The mitochondria play essential roles in both intracellular calcium and reactive oxygen species signaling.As a newly discovered universal and fundamental mitochondrial phenomenon,superoxide flashes reflect transient bursts of superoxide production in the matrix of single mitochondria.Whether and how the superoxide flash activity is regulated by mitochondrial calcium remain largely unknown.Here we demonstrate that elevating mitochondrial calcium either by the calcium ionophore ionomycin or by increasing the bathing calcium in permeabilized HeLa cells increases superoxide flash incidence,and inhibition of the mitochondrial calcium uniporter activity abolishes the flash response.Quantitatively,the superoxide flash incidence is correlated to the steady-state mitochondrial calcium elevation with 1.7-fold increase per 1.0?F/F0 of Rhod-2 signal.In contrast,large mitochondrial calcium transients(e.g.,peak△F/F0~2.8,duration^2 min)in the absence of steady-state elevations failed to alter the flash activity.These results indicate that physiological levels of sustained,but not transient,mitochondrial calcium elevation acts as a potent regulator of superoxide flashes,but its mechanism of action likely involves a multi-step,slow-onset process.JIAN ChongShu HOU TingTing YIN RongKang CHENG HePing WANG XianHua 2014Science China(Life Sciences)2014,57,5:5
8Automatic analytical approach for the determination of 12 illicit drugs and nicotine metabolites in wastewater using on-line SPE-UHPLC-MS/MS显示文摘In this study,we developed a novel on-line solid phase extraction(SPE)-ultra-high-performance liquid chromatography tandem mass spectrometry(UHPLC-MS/MS)-based analytical method for simultaneously quantifying 12 illicit drugs and metabolites(methamphetamine,amphetamine,morphine,codeine,6-monoacetylmorphine,benzoylecgonine,3,4-methylenedioxymethamphetamine,3,4-methylenedioxyamphetamine,cocaine,ketamine,norketamine,and methcathinone)and cotinine(COT)in wastewater samples.The analysis was performed by loading 2 m L of the sample onto an Oasis hydrophilic-lipophilic balance cartridge and using a cleanup step(5%methanol)to eliminate interference with a total run time of 13 min.The isotope-labeled internal standard method was used to quantify the target substances and correct for unavoidable losses and matrix effects during the on-line SPE process.Typical analytical characteristics used for method validation were sensitivity,linearity,precision,repeatability,recovery,and matrix effects.The limit of detection(LOD)and limit of quantification(LOQ)of each target were set at 0.20 ng/L and 0.50 ng/L,respectively.The linearity was between 0.5 ng/L and250 ng/L,except for that of COT.The intra-and inter-day precisions were<10.45%and 25.64%,respectively,and the relative recovery ranged from 83.74%to 162.26%.The method was used to analyze various wastewater samples from 33 cities in China,and the results were compared with the experimental results of identical samples analyzed using off-line SPE.The difference rate was between 19.91%and-20.44%,and the error range could be considered acceptable.These findings showed that on-line SPE is a suitable alternative to off-line SPE for the analysis of illicit drugs in samples.Jingyuan Wang Likai Qia Chenzhi Hou Tingting Zhang Mengyi Chen Haitao Meng Mengxiang Su Hui Xu Zhendong Hua Youmei Wang Bin Di 2021Journal of Pharmaceutical Analysis2021,11,6:5
9NDUFAB1 confers cardio-protection by enhancing mitochondrial bioenergetics through coordination of respiratory complex and supercomplex assembly显示文摘The impairment of mitochondrial bioenergetics,often coupled with exaggerated reactive oxygen species (ROS) production, is a fundamental disease mechanism in organs with a high demand for energy, including the heart. Building a more robust and safer cellular powerhouse holds the promise for protecting these organs in stressful conditions. Here, we demonstrate that NADH:ubiquinone oxidoreductase subunit AB1 (NDUFAB1), also known as mitochondrial acyl carrier protein, acts as a powerful cardio-protector by conferring greater capacity and efficiency of mitochondrial energy metabolism. In particular, NDUFAB1 not only serves as a complex I subunit, but also coordinates the assembly of respiratory complexes I, II, and III, and supercomplexes, through regulating iron-sulfur biosynthesis and complex I subunit stability. Cardiac-specific deletion of Ndufab1 in mice caused defective bioenergetics and elevated ROS levels, leading to progressive dilated cardiomyopathy and eventual heart failure and sudden death. Overexpression of Ndufab1 effectively enhanced mitochondrial bioenergetics while limiting ROS production and protected the heart against ischemia-reperfusion injury. 10Factors affecting directional migration of bone marrow mesenchymal stem cells to the injured spinal cord显示文摘Microtubule-associated protein 1B plays an important role in axon guidance and neuronal migration. In the present study, we sought to discover the mechanisms underlying microtubule-associated protein 1B mediation of axon guidance and neuronal migration. We exposed bone marrow mesenchymal stem cells to okadaic acid or N-acetyl-D-erythro-sphingosine(an inhibitor and stimulator, respectively, of protein phosphatase 2A) for 24 hours. The expression of the phosphorylated form of type I microtubule-associated protein 1B in the cells was greater after exposure to okadaic acid and lower after N-acetyl-D-erythro-sphingosine. We then injected the bone marrow mesenchymal stem cells through the ear vein into rabbit models of spinal cord contusion. The migration of bone marrow mesenchymal stem cells towards the injured spinal cord was poorer in cells exposed to okadaic acid- and N-acetyl-D-erythro-sphingosine than in non-treated bone marrow mesenchymal stem cells. Finally, we blocked phosphatidylinositol 3-kinase(PI3K) and extracellular signal-regulated kinase 1/2(ERK1/2) pathways in rabbit bone marrow mesenchymal stem cells using the inhibitors LY294002 and U0126, respectively. LY294002 resulted in an elevated expression of phosphorylated type I microtubule-associated protein 1B, whereas U0126 caused a reduction in expression. 11Spatial differences in soybean bioactive components across China and their influence by weather factors显示文摘Bioactive components are partially responsible for the nutritional and health benefits of soybeans. Four major bioactive components: isoflavones, oligosaccharides, phospholipids,and saponins, were quantified in 763 soybean samples collected from widely distributed regions across China from 2010 to 2013. A majority of the tested bioactive components showed generally declining trends from the north(high latitude) to the south(low latitude).A positive relationship between total oligosaccharides(TO) and altitude was observed. Total isoflavones(TI), phospholipids(TP) and TO were negatively correlated with cumulative temperature above or equal to 15 °C(AT15) and mean daily temperature(MDT), but positively correlated with diurnal temperature range(DTR) and hours of sunshine(HS).Total saponins(TS) were negatively correlated with MDT but positively correlated with rainfall(RF), whereas TO were negatively correlated with RF. Path-coefficient analysis showed that, besides genotype differences, temperature and HS during the reproductive period influenced TI and TP contents, while temperature and RF influenced TS and TO. The effects of weather factors on soybean bioactive components in diverse regions of China were characterized. 12Ultra‐low cost Ti powder for selective laser melting additive manufacturing and superior mechanical properties associated显示文摘One of the bottleneck issues for commercial scale-up of Ti additive manufacturing lies in high cost of raw material, i.e. the spherical Ti powder that is often made by gas atomization. In this study, we address this significant issue by way of powder modification & ball milling processing, which shows that it is possible to produce printable Ti powders based on ultra- low cost, originally unprintable hydrogenation-dehydrogenation (HDH) Ti powder. 14The September 16, 2015 Mw 8.3 Illapel, Chile Earthquake: characteristics of tsunami wave from near-field to far-field显示文摘On September 16, 2015, an earthquake with magnitude of M_w 8.3 occurred 46 km offshore from Illapel, Chile,generating a 4.4-m local tsunami measured at Coquimbo. In this study, the characteristics of tsunami are presented by a combination of analysis of observations and numerical simulation based on sources of USGS and NOAA. The records of 16 DART buoys in deep water, ten tidal gauges along coasts of near-field, and ten coastal gauges in the far-field are studied by applying Fourier analyses. The numerical simulation based on nonlinear shallow water equations and nested grids is carried out to provide overall tsunami propagation scenarios, and the results match well with the observations in deep water and but not well in coasts closed to the epicenter. Due to the short distance to the epicenter and the shelf resonance of southern Peru and Chile, the maximum amplitude ranged from 0.1 m to 2 m, except for Coquimbo. In deep water, the maximum amplitude of buoys decayed from9.8 cm to 0.8 cm, suggesting a centimeter-scale Pacific-wide tsunami, while the governing period was 13–17 min and 32 min. Whereas in the far-field coastal region, the tsunami wave amplified to be around 0.2 m to 0.8 m,mostly as a result of run-up effect and resonance from coast reflection. 16Development of a widely targeted volatilomics method for profiling volatilomes in plants显示文摘Volatile organic compounds play essential roles in plant environment interactions as well as determining the fragrance of plants.Although gas chromatography-mass spectrometry-based untargeted metabolo-mics is commonly used to assess plant volatiles,it suffers from high spectral convolution,low detection sensitivity,a limited number of annotated metabolites,and relatively poor reproducibility.Here,we report a widely targeted volatilomics(WTV)method that involves using a“targeted spectra extraction”algorithm to address spectral convolution,constructing a high-coverage MS2 spectral tag library to expand volatile annotation,adapting a multiple reaction monitoring mode to improve sensitivity,and using regression models to adjust for signal drift.The newly developed method was used to profile the volatilome of rice grains.Compared with the untargeted method,the newly developed WTV method shows higher sensitivity(for example,the signal-to-noise ratio of guaicol increased from 4.1 to 18.8),high annotation coverage(the number of annotated volatiles increased from 43 to 132),and better reproducibility(the number of volatiles in quality control samples with relative standard deviation value below 30.0%increased from 14 to 92 after normalization).Using the WTV method,we studied the metabolic responses of tomato to environmental stimuli and profiled the volatilomes of different rice accessions.The results identified benzothiazole as a potential airborne signal priming tomato plants for enhanced defense and 2-nonanone and 2-heptanone as novel aromatic compounds contributing to rice fragrance.These case studies suggest that the widely targeted volatilomics method is more efficient than those currently used and may considerably promote plant volatilomics studies.Honglun Yuan Guangping Cao Xiaodong Hou Menglan Huang Pengmeng Du Tingting Tan Youjin Zhang Haihong Zhou Xianqing Liu Ling Liu Yiding Jiangfang Yufei Li Zhenhuan Liu Chuanying Fang Liqing Zhao Alisdair R.Fernie Jie Luo 2022Molecular Plant2022,15,1:2
17Ginger-indirect moxibustion plus acupuncture versus acupuncture alone for chronic fatigue syndrome:a randomized controlled trial显示文摘OBJECTIVE:To assess the efficacy and safety of gingerindirect moxibustion for chronic fatigue syndrome(CFS).METHODS:In this central randomized,controlled trial,290 CFS participants were recruited and randomly allocated to group A(ginger-indirect moxibustion plus acupuncture)or group B(acupuncture alone).The study consisted of a treatment period of 8 weeks with a total of 24 treatments(3 sessions per week,every other day),and a follow-up period of 12 weeks.The outcome was measured by Fatigue Severity Scale(FSS),Psychological Health Report(SPHERE),the Self-rating depression scale(SDS)and the Hamilton anxiety scale(HAMA)at baseline,2,4,6,8,12 and 20 weeks.RESULTS:With the treatment undergoing,the changes of FSS,SPHERE,SDS and HAMA scores in both groups increased gradually,and the effect maintained at the 12th week.Between groups,significantly higher score changes were seen in group A in FSS after 4 weeks treatment(11.94 vs 9.12,95%CI:0.94,4.7)and in SPHERE after 2 weeks treatment(3.7 vs 2.27,95%CI:0.56,2.31).But for SDS and HAMA,the improvement did not differ significantly between groups.No severe adverse events were reported.CONCLUSION:Ginger-indirect moxibustion is a safe and effective intervention to relieve fatigue and accompanying physical symptoms of CFS.MA Tingting WU Jie YANG Lijie FENG Fen YANG Huilin ZHANG Jinhua ZHONG Yanjin NING Qing HUANG Lirong LIN Youbing YAN Jue CHEN Guiquan HOU Tianshu WANG Li REN Yuanfang TAN Jing 2022Journal of Traditional Chinese Medicine2022,42,2:2
18Oxygen vacancies induced narrow band gap of BiOCl for efficient visible-light catalytic performance from double radicals显示文摘In this work,a high-efficiency photocatalytic BiOCl material with a visible light absorption range was successfully prepared by one-pot molecular self-assembly and particle recrystallization method at room temperature.In the process of crystal growth,tartaric acid,as a structure control agent,gradually transforms the stacked two-dimensional nano-sheet-like BiOCl into a hierarchical structure composed of petallike nano-sheets through hydrogen bonding.Besides,the acid etching of organic carboxylic acid on the crystal surface increases the number of micropores and mesopores,thereby the reaction interface.The thiourea(TU)molecules adsorbed on the BiOCl surface with a strong electronic effect introduce oxygen vacancies(OVs)under the condition of low oxygen content.The synergistic effect of hierarchical structure and OVs reduces the recombination of photogenerated carriers,but absorbs more O_(2)and OH−to generate a large number of superoxide radicals(·O_(2)−)and hydroxyl radicals(·OH)effectively.The photocatalytic performance of the synthesized BiOCl material has been significantly improved,and it can effectively degrade 94.15%of rhodamine B(RhB)within 20 min.Furthermore,90.95%of tetracycline(TC),93.76%of ciprofloxacin(CIP),and 85.53%of methyl orange(MO)can be removed in 80 min.Therefore,our work provides an effective method for preparing BiOCl with visible light catalytic activity,which,of course,can be used to treat and repair actual environmental problems under mild conditions.Changyan Chen Ting Jiang Jianhua Hou Tingting Zhang Geshan Zhang Yongcai Zhang Xiaozhi Wang 2022Journal of Materials Science & Technology2022,,19:2
19Analysis of the relationship between bile duct and duodenal microbiota reveals that potential dysbacteriosis is the main cause of primary common bile duct stones显示文摘Bacteria play an important role in the formation of primary Common Bile Duct(CBD)stones.However,the composition and function of the microbiota of bile duct in patients with primary CBD stones remained to be explored.We utilized the 16S rRNA gene high-throughput sequencing technology to analyze the microbial diversity and community composition of biliary and duodenal microbiota in 15 patients with primary CBD stones and 4 patients without biliary tract diseases.Alpha diversity analysis showed that the microbiota richness was similar in bile and intestinal fluid;Beta diversity analysis showed that there were differences in the composition between biliary microbiota and the duodenal microbiota,but the abundance of the main groups showed similarities.The composition of the biliary microbiota from gallstone patients was more complex,as was the duodenal microbiota.Proteobacteria and Firmicutes were the dominant bacteria at phylum level,accounting for at least 75%of the total reads in each subgroup.Pseudomonas and EscherichiaShigella were the major genus among subgroups,but Escherichia-Shigella had increased abundance in duodenal microbiota with primary choledocholithiasis,which may play an important role in stone formation.It is noteworthy that Clostridiumsensu_stricto,Lachnospiraceae_UCG-008,Butyrivibrio and Roseburia which could produce short chain fatty acids(SCFAs),were significantly decreased in biliary microbiota with primary CBD stones(p<0.05).Our study provided new insights into the compositional of normal biliary microbiota.The micro-ecology of biliary and duodenal in patients with stones is complex and closely related,and there is a potential for dysbacteriosis.The decrease in abundance of certain major acid-producing bacteria affects the health of the biliary tract and thus leads to the formation of stones.Zhitang Lyu Tingting Yu Lichao Zhang Xiaona Xu Yijun Zhang Jihong Li Zhirong Li Wei Zhang Senlin Hou 2021Synthetic and Systems Biotechnology2021,6,4:2
