22018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 5Paleoproterozoic reworking of ancient crust in the Cathaysia Block,South China:Evidence from zircon trace elements,U-Pb and Lu-Hf isotopes显示文摘A combined study of zircon LA-ICP-MS U-Pb dating, trace elements and Hf isotope was carried out for gneissic granite from the Sanzhishu area in Jingning, SW Zhejiang Province. Nearly all the zircons separated from the granite exhibited oscillatory zoning and high Th/U ratios (>0.1). The REE profile showed a pronounced positive Ce anomaly, negative Eu anomaly and an enrichment of HREE, which are typical characteristics of magmatic zircon. Thirteen concordant or nearly concordant analytical data yielded a weighted mean 207Pb/206Pb age of 1860±13 Ma (MSWD=0.084), representing the formation age of the granite. The magmatic zircons had negative εHf(t) values of -15.6 to -10.0 and two-stage Hf model ages of 3.1 to 3.4 Ga, indicating that the granites were formed by reworking of ancient crust. The major- and trace-element data indicate that the gneissic granites are metaluminous high-K calc-alkaline rocks and exhibit the same geochemical characteristics as aluminous A-type granites, implying the emplacement of the granite in a post-orogenic extensional tectonic setting. 6Rapid HPLC Method for Determination of 12 Isoflavone Components in Soybean Seeds显示文摘It is important to determine the isoflavone components by high-performance liquid chromatography (HPLC) for the molecular assistant selection of isoflavone in soybean. Based on the standard samples of 12 isoflavone components, the isoflavone components were analyzed using the determination of absorbance peaks method by HPLC. The results showed that there were different maximum ultraviolet (UV) absorbance for the aglycones of daidzein, glycitein, and genistein, which were at 250, 257, and 260 nm, respectively. A linear gradient elution of acetonitrile (13-30%) containing 0.1% acetic acid as a mobile phase was applied on a YMC-C18 column at 35℃. The 12 isoflavone components were determined using the UV detector by HPLC. 7Differential expression of serum miR-126,miR-141 and miR-21 as novel biomarkers for early detection of liver metastasis in colorectal cancer显示文摘Objective:MicroRNAs(miRNAs) have potential as diagnostic biomarkers in cancer.Evaluation of the association between miRNA expression patterns and early detection of liver metastasis in colorectal cancer(CRC) has not been reported.Methods:We investigated the expression of metastasis-associated miRs-31,335,206,141,126,200b,200c,21,Let7a,Let7b and Let7c in localized,liver-metastatic and other organ-metastatic CRC(OM-CRC).Expressions of target miRNAs in serum were evaluated in 116 consecutive localized CRC(L-CRC),72 synchronous liver-metastatic CRC(SLM-CRC) and 36 other OM-CRC by quantitative real-time PCR.Results:Seven of 11 tested miRNAs could be detected from serum.Four miRNAs,miR-126,Let-7a,miR-141 and miR-21 were identified as metastasis-associated miRNAs.Compared with L-CRC,significant upregulated expression was observed for miR-141 and miR-21 in SLM-CRC and OM-CRC,down-regulated expression was observed for miR-126 in SLM-CRC and OM-CRC,and up-regulated expression of Let-7a in OM-CRC.The receiver operating characteristic(ROC) curve showed serum miR-126 had a cut-off [log 10 relative quantity(log 10 RQ)=–0.2005] with 77.78% sensitivity and 68.97% specificity with an area under curve(AUC) of 0.7564,miR-141 had a cut-off(log 10 RQ=–0.2285) with 86.11% sensitivity and 76.11% specificity with an AUC of 0.8279,and miR-21 had a cut-off(log 10 RQ=–0.1310) with 73.61% sensitivity and 66.38% specificity with an AUC of 0.7479.Conclusions:We identified liver metastasis-associated miRNAs,suggesting serum miR-126,miR-141 and miR-21 may be novel biomarkers for clinical diagnosis of early stage liver-metastatic CRC.Jie Yin Zhigang Bai Jianning Song Yun Yang Jin Wang Wei Han Jun Zhang Hua Meng Xuemei Ma Yao Yang Tingting Wang Weirong Li Zhongtao Zhang 2014Chinese Journal of Cancer Research2014,26,1:22
8Genome Engineering in Rice Using Cas9 Variants that Recognize NG PAM Sequences显示文摘CRISPR/Cas9 genome editing relies on sgRNA-target DNA base pairing and a short downstream PAM sequence to recognize target DNA. The strict protospacer adjacent motif (PAM) requirement hinders applications of the CRISPR/Cas9 system since it restricts the targetable sites in the genomes. xCas9 and SpCas9-NG are two recently engineered SpCas9 variants that can recognize more relaxed NG PAMs, implying a great potential in addressing the issue of PAM constraint. Here we use stable transgenic lines to evaluate the efficacies of xCas9 and SpCas9-NG in performing gene editing and base editing in rice. We found that xCas9 can efficiently induce mutations at target sites with NG and GAT PAM sequences in rice. However, base editors containing xCas9 failed to edit most of the tested target sites. SpCas9-NG exhibited a robust editing activity at sites with various NG PAMs without showing any preference for the third nucleotide after NG. Moreover, we showed that xCas9 and SpCas9-NG have higher specificity than SpCas9 at the CGG PAM site. We further demonstrated that different forms of cytosine or adenine base editors containing SpCas9-NG worked efficiently in rice with broadened PAM compatibility. 10Transanal natural orifice specimen extraction for laparoscopic anterior resection in rectal cancer显示文摘AIM:To investigate whether transanal natural orifice specimen extraction(NOSE)is a better technique for rectal cancer resection.METHODS:A prospectively designed database of a consecutive series of patients undergoing laparoscopic low anterior resection for rectal cancer with various tumor-node-metastasis classifcations from March 2011to February 2012 at the First Affliated Hospital of Sun Yat-Sen University was analyzed.Patient selection for transanal specimen extraction and intracorporeal anastomosis was made on the basis of tumor size and distance of rectal lesions from the anal verge.Demographic data,operative parameters,and postoperative outcomes were assessed.RESULTS:None of the patients was converted to laparotomy.Respectively,there were 16 cases in the low anastomosis and fve in the ultralow anastomosis groups.Mean age of the patients was 45.4 years,and mean body mass index was 23.1 kg/m2.Mean distance of the lower edge of the lesion from the anal verge was 8.3 cm.Mean operating time was 132 min,and mean intraoperative blood loss was 84 mL.According to the principle of rectal cancer surgery,we performed D2 lymph node dissection in 13 cases and D3 in eight.Mean lymph nodes harvest was 17.8,and the number of positive lymph nodes was 3.4.Median hospital stay was 6.7 d.No serious postoperative complication occurred except for one anastomotic leakage.All patients remained disease free.Mean Wexner score was 3.7 at11 mo after the operation.CONCLUSION:Transanal NOSE for total laparoscopic low/ultralow anterior resection is feasible,safe and oncologically sound.Further studies with long-term outcomes are needed to explore its potential advantages.Fang-Hai Han Li-Xin Hua Zhi Zhao Jian-Hai Wu Wen-Hua Zhan 2013World Journal of Gastroenterology2013,19,43:19

18Field evaluation of effects of transgenic cry1Ab/cry1Ac,cry1C and cry2A rice on Cnaphalocrocis medinalis and its arthropod predators显示文摘The impacts of transgenic Bt rice on target pests and their predators need to be clarified prior to the commercialization of Bt rice.In this study,the percentages of folded leaves of three transgenic Bt rice lines and non-transgenic parental rice line caused by Cnaphalocrocis medinalis were studied over two successive growing seasons.In addition,the population densities,relative abundance and population dynamics of C.medinalis and four species of its natural arthropod predators were investigated at three sites in China.The results showed that rice line significantly affected the percentages of folded leaves and population densities of C.medinalis larvae.Significantly higher percentages of folded leaves were observed on the non-transgenic rice compared with the three transgenic Bt rice on most sampling dates.Significantly higher densities of C.medinalis larvae and higher relative abundance of C.medinalis within phytophages were found on non-transgenic rice compared with three transgenic Bt rice at different sites across the study period.The population dynamics of C.medinalis larvae were significantly affected by rice line,rice line×sampling date,rice line×year,rice line×sampling date×year.However,there was little,if any,significant difference in the relative abundance,population density and population dynamics of the four arthropod predators between the three Bt rice lines and non-transgenic rice.The results of this study indicate that the Bt toxin in transgenic Bt rice can effectively suppress the occurrence of C.medinalis,but has no significant effects on the occurrence of the four predatory arthropod species.XU XueLiang HAN Yu WU Gang CAI WanLun YUAN BenQi WANG Hui LIU FangZhou WANG ManQun HUA HongXia 2011Science China(Life Sciences)2011,54,11:14
